변형 키토산의 항균효과 (The Anti-Microbial Activity of Modified Chitosan.)

  • 게나 새우 등의 갑각류의 껍질에서 주로 얻어지는 생체적합성이 뛰어난 천연고분자인 chitosan에 음이온 수용성의 함인계 단량체인 mono(2-methacryloyl oxyethyl) acid phosphate를 그라프트 중합시켜 새로운 유도체인 chitosan-g-MAP을 합성하였다. 그라프트중합에서 개시제로는 ceric ammonium nitrate을 사용하여 개시제의 농도, 단량체의 농도, 반응온도, 반응시간에 따른 그라프트율, 단량체의 그라프트효율 및 단량체의 총전환율의 변화를 조사하여 최적조건을 규명하였다. 그라프트율은 개시제농도 3.5$\times$$10^{-3}$ M, 단량체농도 0.19 M, 반응온도 4$0^{\circ}C$에서 가장 높은 값을 나타내었고, 반응시간 4시간이 최적조건 이었다. Chitosan의 유리 아민의 함량과 그라프트된 단량체의 종류에 따른 항균성을 Candida albicans, Trichophyton rubrum 및 Trichophyton violaceum에 대하여 조사한 결과, pH 5.75에서 가장 우수한 항균능이 발현되고, 항균제의 농도에 따른 영향에서는 변형chitosan은 C. albicans와 T. violaceum에 대하여 100%의 우수한 항균력을 나타낸 반면, 탈아세틸화도가 다른 DA70과 DA-90인 chitosan은 T. rubrum균에 대해서만 변형 chitosan 보다 우수한 항균성을 나타내었다.

삼나무 정유의 표재성 진균에 대한 항진균 활성 (Antifungal activity of essential oil from Cryptomeria japonica against dermatophytic fungi)

  • 본 연구에서는 표재성 진균에 의한 피부질환 개선용 소재로의 개발을 위해 삼나무 정유의 표재성 알레르기성 진균, Trichophyton schoenleinii, Trichophyton tonsurans, Trichophyton mentagrophytes (interdigitale), Trichophyton rubrum, Epidermophyton floccosum, Aspergillus fumigatus에 대한 항진균 활성을 평가하여 삼나무 정유의 표재성 진균에 의한 피부질환 개선 소재로의 개발 가능성을 평가하고자 하였다. 항진균 활성 평가는 삼나무 정유의 MIC (Minimum Inhibitory Concentration) 측정과 TLC bioassay, 한천희석법을 통해 측정하였다. 또한 활성 성분의 주성분을 확인하기 위해 GC/MS 분석을 실시하였다. 삼나무 정유 crude oil의 MIC는 모든 균에서 500 ppm 이하의 농도 값을 나타냈으며, TLC bioassay를 통해 spot 1에서 항진균 효과를 확인하였다. 한천희석법을 통한 항진균 활성은 A. fumigatus를 제외한 모든 진균에 대해 삼나무 정유 crude oil의 경우, 500 ppm 이상에서 100%의 활성을 나타냈고, 분획 D의 경우, 100 ppm에서도 100%의 활성을 나타냈다. TLC bioassay를 통해 항진균 활성을 나타낸 spot 1과 한천희석법을 통해 활성을 나타낸 분획 D를 GC/MS로 분석한 결과 주성분은 elemol, ${\gamma}$-eudesmol, ${\beta}$-eudesmol이었다. 따라서 elemol, eudesmol 등은 표재성 진균에 의한 피부질환의 개선 소재로의 이용 가능성이 충분하다고 사료된다.

Antifungal Activity of Non-thermal Dielectric Barrier Discharge Plasma Against Clinical Isolates of Dermatophytes

  • Dermatophytes can invade in keratinized tissues and cause dermatophytosis [1] that rank among the most widespread and common infectious diseases world-wide. Although several systemically and topically administered drugs with activities against these fungi are available, still complete eradication of some of these infections, is difficult and relapses and remissions are often observed [2,3]. In addition, some people are allergic to many of the available drugs which add complications even more. Therefore, the search for novel, selective and more effective therapy is always required and it may help the clinicians to choose the correct treatment for their patients. Non-thermal plasmas primarily generate reactive species and recently have emerged as an efficient tool for medical applications including sterilization. In this study, we evaluated the ability of non-thermal dielectric barrier discharge (DBD) plasma for the inactivation of clinical isolates of Trichophyton genera, Trichophyton mentagrophytes (T. mentagrophytes) and Trichophyton rubrum (T. rubrum), which cause infections of nails and skin and, are two of the most frequently isolated dermatophytes [4]. Our results showed that DBD plasma has considerable time dependent inactivation potential on both T. mentagrophytes and T. rubrum in-vitro. Furthermore, the mechanisms for plasma based T. mentagrophytes and T. rubrum inactivation and planning for in-vivo future studies will be discussed.

Trichophyton verrucosum var album 의 실험적(實驗的) 감염(感染) 기니픽에 대한 병리학적(病理學的) 관찰(觀察) (Pathological observations of guinea pigs experimentally infected with Trichophyton verrucosum var album)

  • Guinea pigs were cutaneously inoculated with Trichophyton verrucosum var album, that is a common causative fungus of dermatophytosis in cattle. The developmental process of lesions, clinical and histopathological changes and reisolations of the fungi were studied to evaluate the pathogenicity of Trichophyton verrucosum var album in guinea pigs. Results obtained through the experiments were summarized as follows : 1. The incidence of infection of the clipping group was 13(86%) of 15 animals, and that of the plucking group was 14(93%) of 15 animals. In both of the clipping and plucking groups, visible cutaneoas lesions were developed between 4 and 7 days post inoculation(p.i.). The spreading and the climax stages persisted for 4 to 11 and 6 to 12 days, respectively. 2. In macroscopic observations, formation of various degree of erythemas and scales over the inoculated skin sites were observed in the spreading stage. In the climax stage, exudative changes and dark red crusts were formed as typical circular lesions. In the healing stage, the lesions revealed shedding of crust, alopecia and hair regrowth. 3. In histopathological observations, infiltration of inflammatory cells, hyperplasia, microabscesses and keratinous-hyaloid materials of epidermis were observed in the spreading stage. Hyphal invasion was primarily observed at the level of epidermis and pilosebaceous ducts. In the climax stage, the infected epidermis was thick with severe hyperplasia, hyperkeratosis and acanthosis. The microabscesses with fungal hyphae, folliculitis and hyperplasia of external root sheath were observed in the dermis. The fungal hyphae were observed only in the tissues of hair follicles, that were internal root sheath, cuticle, the keratinized portions of cortex and medulla 4. In reisolation of the inoculated fungus, all trials for ten animals showed positive cultures until 25 days p.i.. Afterward, the reisolation rates were gradually decreased, showing all negative after 40 days p.i..

Synergism in Antifungal Activity against Candida and Trichophyton Species in Combination with the Essential Oil of Coriandrum sativum L. and Antibiotics

  • To determine whether the essential oil from Coriandum sativum and its main component, linalool, exhibit antifungal activity, we employed a broth dilution assay and disk diffusion test using common pathogenic Candida and Trichophyton species. Both coriander oil and linalool significantly inhibited growth of the tested fungi, with minimal inhibitory concentrations (MICs) in the range of $0.03{\sim}2mg/ml$. Furthermore, in a checkerboard titer test, both the oil fraction and linalool exhibited synergism when combined with ketoconazole, with resultant FICIs ranging from 0.06 to 0.53. Notably, hyphal formation in C. albicans cells was obviously inhibited by C. sativum essential oil in this experiment.

Trichophyton tonsurans의 진균학적 성상 (Mycologic Characteristics of Trichophyton tonsurans)

  • Trichophyton(T.) tonsurans has been identified for the first time in Korea in 1995 and has been increasing in number of reported in wrestlers, in judo players, and in korean - style wrestlers. Author tried mycological studies with T. tonsurans is isolated in Daegu Korea. The results are as follows: All 16 strains of T. tonsurans tested for nutritional requirement, showed strongly stimulated growth by thiamine, mild to moderate growth on casamino acid agar, poor growth on ammonium nitrates base agar. All 16 strains of T. tonsurans tested showed positive results for hair perforation and urease test. Gross findings of T. tonsurans showed that fine granular surface with light yellow hue or white color and mahogany brown color on the reverse side. Microscopic findings showed that septated hyphae, chlamydoconidia and microcondia. Microconidia were characteristic in their arrangerment and shape : round or oval shaped microconidia laterally to hyphae, some were match - head like terminal swelling.

In vitro Antifungal Activity of Limonene against Trichophyton rubrum

  • In this study, the antifungal activities of limonene against Trichophyton rubrum were evaluated via broth microdilution and vapor contact assays. In both assays, limonene was shown to exert a potent antifungal effect against T. rubrum. The volatile vapor of limonene at concentrations above $1{\mu}l$/800 ml air space strongly inhibited the growth of T. rubrum. The MIC value was 0.5% v/v in the broth microdilution assay. The antifungal activity of limonene against T. rubrum was characterized as a fungicidal effect.

구릿대 뿌리 정유의 항진균 효과 (Antifungal Activities of Essential Oil from the Roots of Angelica dahurica Bentham et Hooker f.)

  • The essential oil was extracted from the roots of Angelica dahurica Bentham et Hooker f. by steam distillation and its composition was analyzed by GC-MS. The antifungal activities were evaluated by micro-dilution method against five Aspergillus and three Trichophyton species. The most abundant component was ${\alpha}$-pinene (17.21%) among 40 compounds identified in this oil. The essential oil fraction of A. dahurica and ${\alpha}$-pinene exhibited marked inhibiting activities against the tested Aspergillus and Trichophyton species with MICs (minimum inhibitory concentrations) between 0.12 mg/ml and 8 mg/ml.
