Morphological and Cultural Characteristics of Trichoderma spp. Associated with Green Mold of Oyster Mushroom in Korea

  • A total of 179 isolates of Trichoderma spp. were collected from oyster mushroom substrates in Korea. On the basis of morphological and cultural characteristics, Trichoderma isolates were divided into seven groups, namely T. atroviride, T. citrinoviride, T. harzianum, T. longibrachiatum, T. virens, and two unidentified species, referred to as Trichoderma sp. 1 and 2. The predominant species was Trichoderma sp. 2 (n=86) followed by Trichoderma sp. 1 (n=52). Trichoderma sp. 1 and 2 were morphologically distinct not only from the other species of Trichoderma reported but also from each other in the characteristics such as mycelial growth rate, colony appearance, shape of conidia and conidiophores and branching pattern of phialides, although branching pattern of phialides of Trichoderma sp. 1 was similar to that of T. harzianum. In virulence test, the degree for compost colonization of Trichoderma sp. 2 was significantly greater than that of the other Trichoderma species. Trichoderma sp. 2 was found to be the main cause of green mold disease in oyster mushroom production. More work including molecular characterization is needed to confirm the species of Trichoderma sp. 1 and 2.

  • Trichoderma spp.는 white biotechnology에서 이용되는 대표적인 미생물로서 이들이 강력하게 분비 생산하는 효소들은 산업적으로 매우 중요하다. 본 연구에서는 amylase, pectinase, cellobiohydrolase 및 xylanase 분비활성이 높은 것으로 밝혀진 Trichoderma sp. KACC 40541균주에 대한 Agrobacterium이용 형질전환을 수행하였으며 균주개량을 위한 효율적인 유전자도입 방법을 제시하였다. 특히 형질전환을 위하여서는 균사체에 대한 적정 농도의 NaOH처리가 매우 효과적임을 보여주었다.

  • Cellulose를 분해하는 균주를 자연토양으로부터 분리하여 형태적 관찰, 생리적 특성 등을 검토하였고 F.P Cellulase, CMCase, ${\beta}-glucosidase$의 활성을 측정하였다. 11속, 21종이 동정되었으며 속분류검색에 있어 기본적인 차이점을 보이는 5종을 선정하였다. 동정 결과는 Acremonium sp., Trichoderma viride., 미동정된 Trichoderma SP., Verticillium intertexum., verticillium malthousei 이었다. 효소활동은 Trichoderma viride로 동정된 균주가 비교적 높아보였다.

  • 마늘 흑색썩음균핵병균 (Sclerotium cepivrum, Sclerotium sp.)에 대하여 생물적 방제제 Trichoderma harzianum 23 WP와 Bacillus subtilis 122 WP를 제제화하였다. 2003 년에 포장시험결과에서 태안지역은 남도마늘의 흑색썩음균핵 병균 (Sclerotium cepivrum)에 Trichoderma harzianum 23 WP 와 Bacillus subtilis 122 WP를 습분의 처리한 결과는 무처리 10.9% 비하여 4.1% 와 6.2%로 병 발생이 감소하였다. 서산지역은 서산종 마늘의 흑색썩음균핵병균 (Sclerotium sp.)에 Trichoderma harzianum 23 WP 와 Bacillus subtilis 122 WP를 습분의 처리한 결과는 무처리 17.8% 비하여 1.2% 와 2.6%로 병 발생이 감소하였다. 마늘수량은 두 지역에서 무처리에 비하여 증수되었다. 그러므로 Trichoderma harzianum 23 WP 와 Bacillus subtilis 122 WP는 마늘 흑색썩음병에 생물농약으로서 방제가능성을 나타내었다.

  • We describe two new Trichoderma species associated with oyster mushroom in Korea. Trichoderma green mould has been one of the most serious diseases of oyster mushroom in Korea. Of these the predominant species are two unrecorded species. We designed as Trichoderma sp. Korean type 1 (Th K1) and Trichoderma sp. Korean type 2 (Th K2), respectively. Th K1 and Th K2 can be distinguished from previously reported Trichoderma species as well as each other in morphological characteristics including growth rate at 35$^{\circ}C$, colony morphology, conidia shape and branch pattern of phialides. Sequence of the ITS region of rDNA, the protein coding translation elongation factor gene(EF-1${\alpha}$), and RNA polymeraseII (RPB2) not only clearly separated Trichoderma sp. Korean types from their closely related T. harzianum biotype but also distinguished them from each other. Analyses of the EF-1${\alpha}$ and RPB2 sequences were found to be more useful for establishing systematic relationships among Trichoderma isolates than those of the ITS sequence.   • In order to obtain a good microorganism capable of degrading microcrystaline cellulose (avicel), the screening test was carried out from soil and brown-rot wood. 8 strains which had good avicel-hydrolyzing activity were isolated. Among them, HK 47 which exhibited the highest avicel hydrolyzing activity was identified as Trichoderma sp. HK 47. Maximum avicel-hydrolyzing enzyme production from Trichoderma sp. HK 47 was obtained with the optimum medium contained carboxymethylcellulose 1.5% as carbon source, NaN030.75% as nitrogen source, KH2P040.5%, MgSO4.7H2O 0.1%, Tween 800.005% (V/V) during stationary cultivation at pH 6.0, 3$0^{\circ}C$ In this case, the production of avicel-hydrolyzing enzyme was 0.028 U/ml.

Trichoderma sp.의 중복기생에 대하여 (Hyperparawitism of Trichoderma sp.)

  • Attempts were made to develope the method of biological control by application of hyperparasitism on plant disease. The hyperparasitic fungi used in this work was Trichoderma sp. which was isolated from the ginseng growing soil, and the host fungi were Fusarium oxysporum Schlecht and Glomerella cingulata(St) Spau. et Schr. The hyperparasitic fungi identified as Trichoderma viride. It was observed that the hyperparasitic fungi either contact and penetrate into the hyphae of the host or inhibit the growth of host finally destroy of the host cells.

Trichoderma속의 염색체(染色體)에 관한 연구(硏究) (Chromosomal Studies on the Genus Trichoderma)

  • ATCC로 부터 구입한 11종(種)과 우리나라의 토양에서 분리 동정한 6종(種)의 Trichoderma를 실험(實驗) 재료(材料)로 하였다. 2% malt extract agar 배지(培地)에서 배양(培養)하면서 균사의 영양핵에서의 핵분열을 관찰하였다. 염색(染色)은 Giemsa staining solution으로 염색(染色)하였고 그들의 염색체(染色體) 수(數)를 세었다. 염색체의 형태(形態)는 다른 균류와 마찬가지로 점(點) 모양(dot-like)의 것이 대부분(大部分)이었다. 염색체(染色體)의 크기는 매우 작아서 $0.2{\sim}0.3\;{\mu}$정도이다. 염색체(染色體)의 수는 $5{\sim}10$개로서 n=5인 종(種)이 2종(種)(Trichoderma pseudokoningil, T. saturnisporum)이고, n=6인 종(種)은 8종(種)(T. koningil, T. viride, T. hamatum, T. longibranchiatum, T. sp. No.40, T. sp. No.58 T. sp. No.237, T. sp. No.241)으로 가장 많았다. n=7인 종(種)은 2종(種)(T. reesei, T. harzianum, T. polysporium, T. virigatum, T. sp. No.196)으로 관찰되었다. 이상의 17종(種)을 비교 검토하여 보면 Trichoderma속의 기본(基本) 염색체(染色體) 수(數)는 $4{\sim}10$개 사이에 있을 것으로 생각되며 그 중에서 가장 많은 수(數)를 나타탠 n=6을 기본(基本) 염색체수(染色體數)로 추론하였다.

Specificity of ${\beta}$-Mannanase from Trichoderma sp. for Amorphophallus konjac Glucomannan

  • Five oligosaccharides were isolated from the hydrolysate of konjac (Amorphophallus konjac) glucomannan by a purified ${\beta}$-mannanase from Trichoderma sp. These oligosaccharides were identified as M-M, G-M, M-G-M, M-G-M-M, and M-G-G-M; where G- and M- represent ${\beta}$-1,4-D-glucopyranosidic and ${\beta}$-1,4-D-mannopyranosidic linkages, respectively. The mode of action of the mannanase on the glucomannan is discussed on the basis of the structure of the above oligosaccharides.

Trichoderma sp. MR-93 균주가 생산하는 Isocyanide 계열의 Melanin 생성 저해물질 (Production of the Isocyanide Inhibitor of Melanin Biosynthesis by Trichoderma sp. MR-93)

  • During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR-93 which was capable of producing high level of an inhibitor was selected from plant leaf. Based on taxonomic studies, the fungus could be classified as a strain of Trichoderma sp.. The active compound (MR-93D) was purified from the culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatography and HPLC. The inhibitor was identified as 4-hydroxy-8-isocyano-l-oxaspiro[4-4]cyclonon-8-en-2- one by spectroscopic methods of UV, $^{1}$H-NMR, ESIMS and IR. MR-93D showed a strong tyrosinase inhibitory activity with 0.03 $\mu$g/m of IC$_{50}$ value. It also inhibited melanin biosynthesis with 35 mm inhibition zone at 30 $\mu$g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work.
