  • A simple protocol for the detection of opines, cucumopines and mikimopines using a general horizontal or vertical get electrophoresis system for protein or DNA separation in the laboratory are demonstrated. This electrophoresis method can also be applied to other opines as long as correct detection reagent and buffer system are used.

  • A fair exchange protocol enable two parties to exchange secrets with fairness, so that neither can gain any information advantage by quitting prematurely or otherwise misbehaving. Therefore a fair exchange is the most important for electronic transactions between untrusted parties. To design new fair exchange, after describing basic concepts, definitions and existing protocols and designing a non-interactive OT protocol using ELGamal's public key system, I will design new protocol to support fair exchange. In my designed new protocol, untrusted parties exchange secrets obliviously and verify that their received secrets are true by using transformed Zero Knowledge Interactive Proof extended to duplex. At this time, concerned two parties can't decrypt the other's ciphertext. .After all of the steps, two parties can do it. It is the most important to provide perfect fairness and anonymity to untrusted parties in this protocol.

  • E-mail system has been widely used for information transmission. As multimedia technologies are developed, many Internet services support multimedia. E-mail system only transmits 7bit ASCII data using simple mail transfer protocol(SMTP) specified in RFC 821. E-mail system can't support the authoring and the transmission of multimedia data. In this study multimedia mail authoring system has been designed and implemented in order to provide message integration. Multimedia messages can be authored and transformed into text types with this system. Transformed messages are transmitted from sender's mail transfer agent (MTA) to receiver's MTA by means of SMTP. Messages can be authored in hypertext markup language (HTML) including text, image, audio and video. This system provides integrated messages which consist or various media. BASE64 Encoding method is used in order to transform multimedia messages into text types. Received mails are managed by multimedia user agent (MUA) which is proposed in this study. Convenience for user is improved in this system.

A Comparison of the Phenotypic and Genetic Stability of Recombinant Trichoderma spp. Generated by Protoplast- and Agrobacterium-Mediated Transformation

  • Four different Trichoderma strains, T. harzianum CECT 2413, T. asperellum T53, T. atroviride T11 and T. longibrachiatum T52, which represent three of the four sections contained in this genus, were transformed by two different techniques: a protocol based on the isolation of protoplasts and a protocol based on Agrobacterium-mediated transformation. Both methods were set up using hygromycin B or phleomycin resistance as the selection markers. Using these techniques, we obtained phenotypically stable transformants of these four different strains. The highest transformation efficiencies were obtained with the T. longibrachiatum T52 strain: 65-70 $transformants/{\mu}g$ DNA when transformed with the plasmid pAN7-1 (hygromycin B resistance) and 280 $transformants/l0^7$ spores when the Agrobacterium-mediated transformation was performed with the plasmid pUR5750 (hygromycin B resistance). Overall, the genetic analysis of the transform ants showed that some of the strains integrated and maintained the transforming DNA in their genome throughout the entire transformation and selection process. In other cases, the integrated DNA was lost.

High-efficiency development of herbicide-resistant transgenic lilies via an Agrobacterium-mediated transformation system (고효율의 아그로박테리움 형질전환법을 이용한 제초제저항성 나리 식물체 개발)

  • Transgenic lilies have been obtained using Agrobacterium tumefaciens (AGL1) with the plant scale explants, followed by DL-phosphinothricin (PPT) selection. In this study, scales of lily plants cv. "red flame" were transformed with the pCAMBIA3301 vector containing the gus gene as a reporter and the blpR gene as a selectable marker, as well as a gene of interest showing herbicide tolerance, both driven by the CaMV 35S promoter. Using a 20-minute infection time and a 5-day cultivation period, factors that optimized and demonstrated a high transformation efficiency were achieved. With these conditions, approximately 22-27% efficiency was observed for Agrobacterium-mediated transformation in lilies. After transformation with Agrobacterium, scales of lilies were transferred to MS medium without selective agents for 2 weeks. They were then placed on selection MS medium containing 5 mg/L PPT for a month of further selection and then cultured for another 4-8 weeks with a 4-week subculture regime on the same selection medium. PPT-resistant scales with shoots were successfully rooted and regenerated into plantlets after transferring into hormone-free MS medium. Also, most survived putatively transformed plantlets indicated the presence of the blpR gene by PCR analysis and showed a blue color indicating expression of the gus gene. In conclusion, when 100 scales of lily cv. "red flame" are transformed with Agrobacterium, approximately 22-27 transgenic plantlets can be produced following an optimized protocol. Therefore, this protocol can contribute to the lily breeding program in the future.

  • Bifidobacterium longum KACC 91563 secretes family 5 extracellular solute-binding protein via extracellular vesicle. In our previous work, it was demonstrated that the protein effectively alleviated food allergy symptoms via mast cell specific apoptosis, and it has revealed a therapeutic potential of this protein in allergy treatment. In the present study, we cloned the gene encoding extracellular solute-binding protein of the strain into the histidine-tagged pET-28a(+) vector and transformed the resulting plasmid into the Escherichia coli strain BL21 (DE3). The histidine-tagged extracellular solute-binding protein expressed in the transformed cells was purified using Ni-NTA affinity column. To enhance the efficiency of the protein purification, three parameters were optimized; the host bacterial strain, the culturing and induction temperature, and the purification protocol. After the process, two liters of transformed culture produced 7.15 mg of the recombinant proteins. This is the first study describing the production of extracellular solute-binding protein of probiotic bacteria. Establishment of large-scale production strategy for the protein will further contribute to the development of functional foods and potential alternative treatments for allergies.

  • In this paper, we propose an asynchronous IR-UWB ranging system based on the two-way ranging protocol. The periodic pulse sequence is used to measure a distance between two devices. At the receiver, a received signal is first transformed into a frequency-domain signal using an analog correlator bank and digital signal processing is followed in the frequency-domain. This make it possible for the system to use an ADC with a conversion speed of pulse rate. The proposed algorithm at the receiver side includes a peak detection procedure using mutipath channel compensation and matched filtering, and retransmits a pulse sequence synchronized with the detected peak. The validity of the proposed algorithm is verified from simulation results where the CM1 channel is assumed.

  • In this paper, we propose a web-based content syndication system in which users can easily choose Internet protocol television (IPTV) contents. This system generates personalized content guide to provide a list of IPTV contents with respect to users' interests and statistics information of their online social community. For this, IPTV contents and relevant metadata are collected from various sources and transformed. Then, the service and content metadata are processed by user metadata including audience measurement and community metadata. The metadata flows are separated from content flows of transport network. The implementation of IPTV content syndication system demonstrates how to arrange IPTV contents efficiently from content providers to the end user's screen. We also show that the user metadata including online community information are important for the system's performance and the user's satisfaction.

  • In this paper, we have studied modelling and simulation of computer communication protocols theoretically. After describing a definition and functions of communication protocols, we have classified models for protocol design. And, in those protocol models, by endowing Timed Petri Net (TPN) models with a time function .tau., we have proposed a structural definition of TPN models. Furthermore, in order to complement Petri Net Based models with some problems, we have introduced the Discrete EVent system Specification (DEVS) concept in system simulation field. As an important result of our study, we have presented a theorem, which says that a TPN model becomes a DEVS model, and proved it. According to the theorem, we can perform efficient simulation by using the DEVS model transformed from a TPN model when we intend the TPN model to be simulated, otherwise we design another simulation model for it.

For a specified purpose through P2P, the Peers who make temporary a group delivery a request efficient and safe. And the resources can be jointed common, cooperation and communication. When P2P is developed more, we can expect more formation of online community and development. But to be a safe of personal ID and password in internet, it should be possible to make a key-exchange. In the paper, it suggest P2P security system suitable to personal security that Kerberos be transformed. The user who make community in P2P, have Kerberos Server, and using Physical Address of Ethernet card in personal computer, authenticate users.
