The Inhibitory Effect of Baicalin on the Short-Term Food Intake in C57BL/6J Mice

  • Appetite is inhibited by the anorexigenic neuropeptides POMC (proopiomelanocortin) and CART (cocaine-amphetamine-regulated transcript) in the hypothalamus. The present study was performed to examine the inhibitory effects of baicalin against food intake and the upregulation of POMC/CART. Short-term food intake (48 h) was significantly inhibited by treatment with baicalin (10 mg/kg, p<0.05) in C57BL/6 mice. Immunohistochemical analysis showed that baicalin upregulated POMC and CART levels in the arcuate nucleus of the hypothalamus. These effects were also examined using an in vitro system. pPOMC-Luc or pCART-Luc plasmids were transformed into mouse N29-2 neuronal and human SH-SY5Y cells, and the activities of baicalin were examined in these cells. Baicalin increased POMC and CART promoter-driven luciferase activity in a dose-dependent manner without cytotoxic effects. These results suggest that baicalin downregulates short-term food intake while upregulating POMC and CART expression.

  • 초고속 통신망의 확대 적용으로 인터넷의 빠른 성장과 함께 음성과 비디오 그리고 데이터를 통합하고자 하는 노력이 시도 되고 있다. VoIP(Voice over IP)는 IP를 이용하여 음성과 데이터를 패킷 형태로 통합하여 실시간으로 전송하는 기술이다[1]. 패킷 네트워크에서 VoIP 시그널링 기술을 이용하면 망 자원의 효율적 이용 및 PSTN에 가까운 음질 그리고 인터넷과 연계한 다양한 음성서비스 지원(H.323, SIP, MGCP 등 다양한 신호처리 지원)이 가능하다. 본 논문에서는 VoIP망에서 IP기반 녹취 시스템을 설계 및 구현한다. 녹취 시스템은 고객과 상담원의 통화 내용을 자동으로 녹음하여 보관함으로써 고객의 요구사항을 명확히 파악할 수 있으며 녹취 데이터의 통계 자료 제공으로 효율적인 관리가 지원되고 선택 녹취, 스케줄링 녹취, 상담원의 평가 자료를 제공하여 고객 관리의 질적인 향상을 지원한다. 본 논문의 녹취 시스템은 고객과의 통화 내용을 녹취하여 서버의 녹취 DB에 저장하여 관리하는 녹취 시스템으로 모든 네트워크 환경에서 사용할 수 있으며 CTI와 연동하여 효율적이고 체계적인 녹취 시스템 구국이 가능하다.

Modulation of the Somatotropic Axis in Periparturient Dairy Cows

  • This review focuses on modulation of growth hormone (GH) and its downstream actions on periparturient dairy cows undergoing physiological and metabolic adaptations. During the periparturient period, cows experience a negative energy balance implicating that the feed intake does not meet the total energy demand for the onset of lactation. To regulate this metabolic condition, key hormones of somatotropic axis such as GH, IGF-I and insulin must coordinate adaptations required for the preservation of metabolic homeostasis. The hepatic GHR1A transcript and GHR protein are reduced at parturition, but recovers on postpartum. However, plasma IGF-I concentration remains low even though hepatic abundance of the GHR and IGF-I mRNA return to pre-calving value. This might be caused by alternation in IGFBPs and ALS genes, which consequently affect the plasma IGF-I stability. Plasma insulin level declines in a parallel manner with the decrease in plasma IGF-I after parturition. Increased GH stimulates the lipolytic effects and hepatic glucose synthesis to meet the energy requirement for mammary lactose synthesis, suggesting that GH antagonizes insulin-dependent glucose uptake and attenuates insulin action to decrease gluconeogenesis.

Code-Switching of English Learners in the TEE Program

  • Research on strategic Code-Switching (CS) of second language learners in teaching English in English (TEE) program provides an elaborate framework for analyzing how learners manage to express themselves in spite of their limited knowledge of the target language. This research presupposes that L2 learners' CS presents innovative solutions for communicative strategy, and that CS used as communicative strategy can promote L2 learners' language acquisition. The major questions of current research involve examining the significant patterns of different functions of CS in L2 learners' interaction and investigating L2 learners' CS styles according to the different functions of CS. The implication of CS utility is regarded as a teaching technique in the TEE program. Recorded transcript is analyzed to trace the same pattern and the categorization of CS as well as to recognize the functions of CS and their ratio. Hence, this leads to the conclusion that learners' negotiation between code selection and communication intention occurs in patterns. The learners' CS tends to be predictable, reproductive, and systematic, as one of the language acquisition phases. Therefore, the attention to the CS in the TEE program should be redirected in communication substantiality toward the principles of pragmatics. As an additional advantage of the CS analysis, this research elaborates on a conceptual acceptance of CS as a set of learners' strategies in the TEE program.

RNase P-dependent Cleavage of Polycistronic mRNAs within Their Downstream Coding Regions in Escherichia coli

  • M1 RNA, the catalytic subunit of Escherichia coli RNase P, is an essential ribozyme that processes the 5' leader sequence of tRNA precursors (ptRNAs). Using KS2003, an E. coli strain generating only low levels of M1 RNA, which showed growth defects, we examined whether M1 RNA is involved in polycistronic mRNA processing or degradation. Microarray analysis of total RNA from KS2003 revealed six polycistronic operon mRNAs (acpP-fabF, cysDNC, flgAMN, lepAB, phoPQ, and puuCBE) showing large differences in expression between the adjacent genes in the same mRNA transcript compared with the KS2001 wild type strain. Model substrates spanning an adjacent pair of genes for each polycistronic mRNA were tested for RNase P cleavage in vitro. Five model RNAs (cysNC, flgMN, lepAB, phoPQ, and puuBE) were cleaved by RNase P holoenzyme but not by M1 RNA alone. However, the cleavages occurred at non-ptRNA-like cleavage sites, with much less efficiency than the cleavage of ptRNA. Since cleavage products generated by RNase P from a polycistronic mRNA can have different in vivo stabilities, our results suggest that RNase P cleavage may lead to differential expression of each cistron.

Analysis on Reports of Qualitative Research Published in Domestic Korean Medicine Journals (국내 한의학 학회지에 게재된 질적 연구 보고 분석)

  • Objectives This study was aimed to analyze the reports of qualitative research published in domestic Korean medicine journals.Methods Eleven published qualitative research studies were selected for analysis. Selected reports were analyzed by Consolidated Criteria for Reporting Qualitative Research (COREQ), a reporting guideline for qualitative research.Results In the 1st domain, descriptions of personal characteristics, interviewer's bias or assumption were insufficient. In the 2nd domain, descriptions such as presence of non-participants, research questions for the interview guidelines, data saturation and transcript return to participants were insufficient. In the 3rd domain, descriptions of items related to number of data coder, participants evaluating the finding, and clarity of minor themes were insufficient.Conclusions Qualitative studies in Korean Medicine have increased. COREQ should be considered by investigators who report qualitative research, in order to improve the quality of research.

Misexpression of AtTX12 encoding a Toll/interleukin-1 receptor domain induces growth defects and expression of defense-related genes partially independently of EDS1 in Arabidopsis

  • In this study, a tissue-specific GAL4/UAS activation tagging system was used for the characterization of genes which could induce lethality when ubiquitously expressed. A dominant mutant exhibiting stunted growth was isolated and named defective root development 1-D (drd1-D). The T-DNA tag was located within the promoter region of AtTX12, which is predicted to encode a truncated nucleotide-binding leucine-rich repeat (NLR) protein, containing a Toll/interleukin-1 receptor (TIR) domain. The transcript levels of AtTX12 and defense-related genes were elevated in drd1-D, and the misexpression of AtTX12 recapitulated the drd1-D phenotypes. In the presence of ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1), a key transducer of signals triggered by TIR-type NLRs, a low-level of AtTX12 misexpression induced strong defective phenotypes including seedling lethality whereas, in the absence of EDS1, a high-level of AtTX12 misexpression induced weak growth defects like dwarfism, suggesting that AtTX12 might function mainly in an EDS1-dependent and partially in an EDS1-independent manner.

  • The global regulator, GacS (global antibiotic and cyanide sensor kinase), was required for the increased resistance to hydrogen peroxide occurring as cultures of the rhizobacterium, P. chlororaphis O6, matured. Specific stationary-phase peroxidase and catalase isozymes were absent in the GacS mutant, whereas a manganese-superoxide dismutase isozyme was expressed earlier and to a great extent than wild type. In the wild type cell, transcript accumulation of rpoS was higher in late logarithmic-phase cells than cells from mid logarithmic- or stationary-phase. Transcripts from rpoS in the GacS mutant were reduced in each of these growth phases compared to the wild type expression. The down stream sequence from rpoS lacked sequences encoding a small RNA, rsmZ, found in other pseudomonads and implicated in control of genes activated by the GacS system. These findings suggest that GacS-mediated regulation of RpoS plays role in control of oxidative stress in P. chlororaphis O6 by as yet an unknown mechanism.

Effects of Chitosan on the Differentiation of MDPC-23 Cells

  • The effects of chitosan upon the experimentally induced differentiation of MDPC-23 cells, derived from mouse dental papilla cells, were investigated by RT-PCR, observations of cell morphology and Alizaline red-S staining. Chitosan was found to significantly increase and accelerate the expression of ALP mRNA but decrease the ColI transcript levels, as compared with the control, in a time-dependent manner during the differentiation of MDPC-23 cells. Chitosan also significantly downregulated ON mRNA expression and accelerated mineralization in differentiating MDPC-23 cells. These results suggest that chitosan facilitates odontoblast differentiation and mineralization and may have potential clinical applications as a dentin regeneration material.

Expression of ${\alpha}$-Galactosidase Gene from Leuconostoc mesenteroides SY1 in Lactobacillus brevis 2.14

  • ${\alpha}$-Galactosidase gene (aga) from Leuconostoc mesenteroides SY1 was expressed in a heterologous host, Lactobacillus brevis 2.14 using an Escherichia coli-Leuconostoc shuttle vector, pSJE. pSJEaga (pSJE carrying aga) was introduced into Lactobacillus brevis 2.14 by electroporation and transformation efficiency was $1.1{\times}10^3$ per ${\mu}g$ DNA. L. brevis transformants (TFs) showed higher ${\alpha}$-galactosidase (${\alpha}$-Gal) activities than cells containing pSJE. Transcription levels of aga in L. brevis 2.14 grown on different carbon sources (1%, w/v) were examined by slot blot analysis. Aga transcript levels and ${\alpha}$-Gal activities were higher in cells grown on melibiose, raffinose, and galactose than cells on glucose, sucrose, and fructose. Western blot result showed that L. brevis 2.14 harboring pSJEaga produced much more ${\alpha}$-Gal when grown on melibiose than on glucose.
