Phenanthrene-induced changes in blood organic components, transaminase and ALP activities of Paralichthys olivaceus

  • Phenanthrene-induced changes in blood organic components. transaminase and ALP activities of Paralichthys oIivaceus were investigaled. The aim of this study was to obtain a holistic view of the toxic responses, and compensations of fish exposed to waterborne phenanthrene. Fish were exposed to 0, 0.5, 1, 2$\mu$M of phenanthrene for 4 weeks. The most noticeable changes were concentration-dependent increase in levels of blood GOT and GPT activities. At concentrations 1.0 and 2.0 $\mu$M of phenanthrene GOT, GPT and ALP enzyme activities in plasma were increased. Plasma albumin concentration was stable in phenanthrene-exposed fish, however, plasma total protein level was reduced significantly at the 4-week sampling points (2.0$\mu$M). Plasma glucose levels were found to be increased significantly over the cootrol throughout duration of the experiment with phenanthrene. Overall the results suggest that phenanthrene has the potential to alter some physiological functions in olive flounder.

Hypoglycemic Effect of Collybia confluens Exobiopolymer Produced by Submerged Mycelial Culture on Diabetic Rats

  • The hypoglycemic effect of Collybia confluens exobiopolymer was investigated in streptozotocin (STZ)-induced diabetic rats. In a dose-dependent study, the exobiopolymer, at 150 mg/kg body weight (BW) dose substantially lowered the plasma glucose level by 29.3%, as compared to the control group. It also lowered the plasma total cholesterol and triglyceride levels by 23.3 and 30.7%, respectively, and reduced the liver total cholesterol and triglyceride levels by 23.0 and 33.5%, respectively. The activity of alanine transaminase (ALT) and aspartate transaminase (AST) was reduced 34.6% and 23.6% respectively, by the exobiopolymer administration, compared to the control group. The exobiopolymer was found to contain 83.2% carbohydrate and 16.8% protein. The sugar and amino acid of the exobiopolymer were also analyzed in detail.

Effects of the Hovenia dulcis THUMBER var. Koreana Nakai Extracts on Hepatoprotective Activities and Improvement of Hepatofunction

  • The Hovenia extracts showed a significant hepato-protective activity against alcohol and CC4-induced hepatotoxicity. Injection of CC4 and alcohol induced liver injury and increased serum GOT (glutamic oxaloacetic transaminase) and GPT (glutamic pyuvic transaminase) levels in injured rats. The ADH (alcohol dehydrogenase)-like and ALDH (aldehyde dehydrogenase)-like activities in the Hovenia extracts were studied. Their activities in the extracts of Hovenia wood were higher than the extracts of other parts of Hovenia. And then Hovenia wood extracts have high effects on the alcohol and acetealdehyde degradation. The Hovenia extracts were decreased concentration of fusel oil. The ADH-like and ALDH-like activities in the Hovenia wood extract were 1.14 $unit/{\mu}l$, 0.33$unit/{\mu}l$ respectively.

Hepatoprotective and antioxidant activity of the methanolic extract of Mucuna pruriens Bak. roots

  • The methanolic extract of natural roots of Mucuna pruriens was screened for hepatoprotective and antioxidant effects in carbon tetrachloride ($CCl_4$) induced hepatotoxicity in Albino rats. The degree of protection was measured by estimating biochemical parameters like serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, total protein and level of serum bilirubin (both total and direct). The extract also reduced $CCl_4$ induced lipid peroxidation both in-vitro and in-vivo. Hepatoprotective activity of methanolic extract at dose 300 mg/kg body weight, i.p., was comparable with standard drug Silymarin (25 mg/kg, i.p.). Furthermore, histopathological experiments were also carried out to support the study.

  • This study il focused on the hepatopreventive effect in cirrhotic rats induced by N, N -dimethylnitrosamine treatment when organir Lulfur -containing compoundE were orally injected. Biochemical parameters (aspatate transaminase (AST), alanine transaminase (ALT) alkaline phosphatase (ALP), 1-protein and t-bilirubin) were measured in serum injured liver tissue. The increased AST and ALT values were significantly reduced by organic sulfur-containing compounds at the oral dotes of 50 mg/kg. The result of morphological changes have illustrated the accumulation of liver damages, Each as inflammatory cell accumulation and cirrhosis, caused by N, N-dimethylnitrosamine. Also, it was found that liver damages were prevented by the treatment of organic sulfur- containing compounds.

Occurrence of Microcystin-Containing Toxic Water Blooms in Central India

  • Three out of fourteen Microcystis-dominant cyanobacterial blooms in Central India were found to be toxic to mice ($LD_{50}$ ranging from 35-450 mg bloom dry mass/kg body weight). The liver architecture of the treated mice showed characteristic symptoms of hepatotoxicity relative to the untreated controls, with increased enzyme activities of serum lactate dehydrogenase (LDH), serum glutamate oxaloacetate transaminase (SGOT), alkaline phosphatase (ALP), and serum glutamate pyruvate transaminase (SGPT). RP-HPLC revealed the presence of microcystin-LR, microcystin-RR, and desmethyl microcystin-RR in the given region to maximum amounts of 390, 1,030, and $860{\mu}g/g$ bloom dry weight, respectively, corresponding to a maximum of 2.8 mg/l microcystin-LR in the lake water. Further confirmation of the microcystin variants was conducted using a MALDI-TOF MS analysis.

Antioxidant Activities of Decursinol Angelate and Decursin from Angelica gigas Roots

  • The anti-oxidant activities of decursinol angelate (1) and decursin (2) isolated from Angelica gigas were investigated. These two coumarins exhibited decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4-intoxication$ in rats. They also showed increase in anti-oxidant enzyme such as hepatic cytosolic superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-px) in $CCl_4-intoxicated$ rats. These results suggest that decursinol angelate (1) and decursin (2) from A. gigas possess not only the anti-oxidant, but also the hepatoprotective activities in rats.

  • The effect of Dolichos lablab leave extract on enzyme activities in mice blood was investigated in this study to clarify the new useful application of the Plant leaves. There were not significant differences in the enzyme activities in mice blood among treatment fed with the leave extract and non fed control. The feeding treatment with the extract showed a tendency to activity compared to the nun fed control. The activities of glutamate oxaloacetate transaminase, glutamate pyruvate transaminase and lactate dehydrogenase in mice blood at the treatment fed with lead and the leave extract were significantly low compared to their activities at the non fed treatment with the extract, respectively. The choline esterase activity was high at the leave extract feeding. The cadmium dietary treatment showed the same result as the lead treatment. In conclusion, the physiological function of the Jebikong leave was significantly in creased when the mice was stressed by the hear metal intake. Therefore, the plant leave extract would consider the reduction of heart metal effect.

우황과 사향의 간세포 보호효과 (Antihepatotokic Activity of Bezoar Bovis and Moschus)

  • The antihepatotoxic activity of Bezoar Bovis and Moschus was investigated by in vitro assay method using galactosamine and carbon tetrachloride-induced cytotoxicity in primary-cultured rat hepatocytes. The antihepatotoxic activity was evaluated by measuring the level of glutamate pyruvate transaminase and sorbitol dehydrogenase which were released from the necrotic hepatocytes to the culture medium. In galactosamine-intoxicated hepatocytes, the chloroform fraction of Bezoar Bovis reduced the level of glutamate pyruvate transaminase and sorbitol dehydrogenase resulting in 65% and 59% protection, respectively. The n-Hexane fraction of Moschus resulted in 45% and 40% protection, respectively in this system. In the case of carbon tetrachloride-intoxicated rat hepatocytes, Bezoar Bovis did not have significant effect and only the aqueous fraction of Moschus showed 42% and 40% protection, respectively.

Effect of Docosahexaenoic Acid Rich Tuna Orbital Oil on Acute Liver Injury Induced by Carbon Tetrachloride

  • Docosahexaenoic acid (DHA) rich oil was obtained from blue fin tuna (Thunnus thynnus orientalis) orbital tissue with centrifugation of 12,000 rpm under vaccum $(10^{-1}\;Torr)\;at\;4^{\circ}C$. The effect of DHA rich oil (DHA content; $27.8\%$) on $CCl_4-induced$ acute injury was investigated biochemically and histopathologically. Dosage of DHA rich oil on 24h before $ CCl_4-administration$ prevented significantly the increase of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GTP) values. No necrosis of hepatocytes was observed in rat livers treated with DHA oil on 24h prior to $CCl_4-administration$. These results suggested that DHA oil controls the accumulation of fat in the liver and prevented the liver injury.
