토양을 비롯하여 어류, 패류, 조류, 포유류의 소화기관으로부터 Clostridium botulinum 분리를 시도하였다. 총 158개 분리원을 screening한 결과 10개 시료로부터 Clostridium botulinum 분포 가능성을 확인하였으며 6개 시료로부터 Clostridium botulinum을 순수 분리하여 3.8% 분리율을 나타내었다. 분리 균주의 형태적 특징, 배양상의 특성 및 생화학적 특성 등을 표준 균주의 특성과 비교하고 항혈청에 의한 중화시험을 실시하여 분리균주를 동정하였다. Egg york agar에서의 opalescence 생성, 탄수화물 이용성, Egg york GAM 배지상에서의 pearly layer 생성 등으로부터 Clostridium botulinum으로 동정할 수 있었으며 trypsin에 의한 toxicity 활성화, type E 항혈청에 의한 opalescence 생성억제 및 mouse 방어효과가 인정되어 type E 로 동정하였다. 국내에서 시판되고 있는 수 종의 식품을 대상으로 Clostridium botulinum 의 toxin 생성능을 비교하였던 바 식품의 종류, 사용균주에 따라 toxin 생성량에 현저한 차이가 있었다. 분리균주 type E 의 경우 어패류통조림, ham 식품에서 많은 양의 toxin 이 생성되었으며 sausage, 과일통조림 식품에서는 비교적 적었다. 그러나 type A 의 경우에는 어패류, ham , sausage 식품에서 상당량의 toxin 이 생성되었으며 과일통조림에서도 비교적 맣은 양의 toxin이 생성되었다 .
The effect of AF-toxin I produced by the strawberry pathotype of Alternaria alternata on the protein synthesis of susceptible strawberry protoplasts was examined by using the radiolabeled amino acids. The incorporation of the radiolabeled amino acids into newly synthesized proteins in the strawberry protoplasts was stimulated by the toxin treatment at relatively low concentrations (2.2$\times$10-11 to 2.2$\times$10-9 M), but not at higher concentrations (2.2$\times$10-8 to 2.2$\times$10-6 M). An one-dimensional SDS-polyacrylamide gel electrophoresis revealed no detectable differences in the proteins synthesized in both the toxintreated and untreated protoplasts. The susceptible strawberry protoplasts were treated with AF-toxin I and stained with Fluostain I to detect the extracellular polysaccharides. The toxin treatment induced the accumulation of extracellular polysccharides in a dose-dependent manner. These results indicate a transient activation of cellular metabolism in the susceptible cells by the toxin exposure.
The role of botulinum toxin as a therapeutic agent is expanding rapidly in otolaryngology. Botulinum toxin is a protease that blocks the release of acetylcholine from nerve terminals. Its effects are transient and nondestructive, and largely limited to the area in which it is administered These effects are also graded according to the dose, allowing for individualized treatment of patients and disorders. Botulinum toxin has been used primarily to treat disorders of excessive or inappropriate muscle contraction. In the field of otolaryngology, these include spasmodic dysphonia, oromandibular dystonia, and blepharospasm, vocal tics and stuttering, cricopharyngeal achalasia, various tremors and tics, hemifacial spasm, temporomandibular joint disorders and a number of cosmetic applications. Botulinum toxin treatment has recently begun to show some benefit in the control of pain from migraine and tension headache. It may also prove useful in the control of autonomic dysfunction, as in Frey syndrome, sialorrhea, and rhinorrhea. In over 20 yews of use in humans, botulinum toxin has accumulated a considerable safety record, and in many cases represents relief for thousands of patients unaided by other therapy.

The present study was performed to assess the activity of the botulinum toxin A on breast cancer cells. The T47D cell line was exposed to diverse concentrations of the botulinum toxin A and cell viability and apoptosis were estimated using MTT and propidium iodine/annexin V methods, respectively. Botulinum toxin A exerted greater cytotoxic activity in T47D cells in comparison with MCF10A normal cells; this appeared to be via apoptotic processes caspase-3 and -7. In conclusion, botulinum toxin A induces caspase-3 and -7 dependent apoptotic processes in the T47D breast cancer cell line.

Recombinant antibody-toxin is a bifunctional protein that binds and kills a target cell expressing a specific antigen on the surface of the cell, and its structure is chimeric, in which a toxin is fused to an antigen-binding domain such as scFv or Fab. Divalent antibody-toxin molecules showed higher cytotoxicities against cancer cell lines than monovalent molecules. However, the yields of the divalent molecules were very low. In this study, we introduced the CH2, CH3, or CH2-CH3 (=Fc) domain of antibody in the middle of the Fab-toxin between the hinge region of human IgG1 and the toxin domain to increase the yield. The covalently bonded dimer could be formed by three disulfide bridges from cysteine residues in the hinge region. The molecule with the CH3 domain showed about 3-fold higher dimerization yield than previously constructed Fab-toxin molecules, while maintaining the cytotoxic activity comparable to that of scFv-toxin. However, the introduction of CH2 or Fc domain to the same position showed little effect on the dimerization yield. We also observed that the introduction of the CH3 region made it possible to form noncovalently associated dimer molecules.
Purpose: The purpose of this report is to present a case of persistent parotid fistula treated successfully with preoperative botulinum toxin type A injection into the parotid parenchyma, followed by fistulectomy. Methods: A 72-year-old female patient presented to the hospital with a 5-month history of clear, watery discharge from a tiny opening on the left cheek, which increased during food intake. A chemistry test of the fluid revealed an high amylase level. An ultrasonography of left parotid gland showed a $1.13{\times}0.6cm$ sized fistula. After demarcating the left parotid gland with assistance of ultrasonography, a total 40 units of botulinum toxin type A (Botox, Allergan, Irvine, CA) was injected into 4 subdivisions of the left parotid gland. The clear serous discharge ceased completely on the 5th day after botulinum toxin injection. On the 7th day, a fistulectomy was performed under the local anesthesia. Results: The parotid fistula healed completely without complications. During the 6-month follow up period, there was no discharge from the cheek. Conclusion: On the basis of our experience with type A botulinum toxin as a local anticholinergic agent in treating parotid fistula, preoperative botulinum toxin A injection seems to be very useful to prevent recurrence after fistulectomy.
Toxin-antitoxin (TA) system은 박테리아와 고세균에서 진화적으로 보존되어 흔히 발견되는 유전적 모듈이다. 기본적으로 이 시스템은 세포 내 toxin과 그들의 억제자로 작용하는 antitoxin으로 구성되어있으며, 현재 총 다섯가지 유형으로 구분된다. 공통적으로 toxin은 스트레스 조건에서 활성화됨으로써 세포 내 다양한 과정을 억제하는 활성을 가지는데 이는 결과적으로 세포 사멸 혹은 가역적인 생장 저해를 일으킨다. Toxin의 이러한 효과들은 유전자 발현의 조절, 성장 조절, programmed cell arrest, programmed cell death, persister cell의 형성, 박테리오파지 방어기작, 가동성 유전인자의 안정화, 플라스미드 유지 기작 등 다양한 생리학적 역할을 나타낸다. 그러므로 TA system은 일반적인 스트레스 반응모듈로서 여겨진다. 하지만 이를 역이용한다면 TA system으로부터 toxin을 활성화 시키는 인자를 개발하여 새로운 항균 물질로 이용할 수 있다. 그뿐만 아니라 TA system은 toxin의 세포 사멸 효과를 이용하여 원하는 타겟 유전자가 존재하는 세포만 선택적으로 살아남도록 하는 효율적인 클로닝 전략에 이용될 수 있다. 또한, toxin의 서열 특이적 리보핵산 가수분해효소 활성을 이용하여 타겟 단백질 이외의 단백질 합성을 막아 효과적인 단일 단백질 대량 생산을 위해서도 이용할 수 있다. 더 나아가 일부 TA system의 toxin은 진핵 세포에서도 세포 독성을 나타내기 때문에 암세포, 바이러스 감염 세포에서 toxin의 발현을 유도하여 세포사멸을 일으킴으로써 인간의 질병 치료로 이어질 수 있다.
This study is to investigate the biological, biochemical and cytotoxic effects of puffer fish (Arothron stellatus) toxin extracts under in-vitro condition. Extracted toxins from various organs of puffer fish were purified by using active charcoal column, and Bio-gel-P2 column chromatography. The lethality of toxin was tested in crabs, which consists of neurotoxic compounds. The degree of the brine shrimp lethality assay was found directly proportional to the concentration of the toxin extracts, which was well supported by hemolytic assay. The experimental results suggested that the gonad was found higher toxins than the liver and muscles. The mortality rate of brine shrimp nauplii was increased with the raise of concentrations of toxin level. Among the different doses and time dependent cytotoxic effect of human cervical carcinoma (HeLa) cells were showed $4.0{\mu}g/mL$ of toxin, which was effectively inhibited cancer cell proliferation. HPLC and TLC analysis was revealed that the A. stellatus toxin contains tetrodotoxin (TTX), related compounds 4-epi TTX and anhydro-TTX. The present results suggested that the A. stellatus contain TTX as a major and anh-TTX as a minor toxin. It could be the potential candidate in the field of anticancer drug discovery against human cervical cancer cells. The present data is confirming that the puffer fish toxin as an interesting source of novel bioactive natural compounds with potent applications in pharmacology.
BT toxin is produced by a soil bacterium Bacillus thuringiensis and has long been used as a biological insecticide without any competition. Recently, Photorhabdus, a symbiotic bacterium from entomopathogenic nematodes, family Heterorhabditae, has been researched and discussed as alternatives to B. thuringiensis. Photorhabdus, which lives in the gut of entomopathogenic nematodes, is a highly virulent pathogen of a wide range of insect larvae. When an insect is infected by the nematodes, the bacteria are released into the cadaver, and produce a number of insecticidal toxins. The biological role of the different Photorhabdus toxins in the infection process is still unclear. Photorhabdus toxin complex (Tc) is highly secreted gut-active toxin and has been characterized as a potent three-component (A, B and C) insecticidal protein complex. These components are necessary for full oral activity against insect larvae. The Photorhabdus PirAB binary toxins exhibit a potent injectable activity for Galleria mellonella larvae, and have oral toxicity against mosquitoes and caterpillar pest Plutella xylostella. Other toxin, 'makes caterpillars floppy' (Mcf) showed injectable activity on caterpillars. Recombinant Mcf triggers apoptosis in both insect hemocytes and the midgut epithelium and carries a BH3 domain. In this review, the relationship between the Photorhabdus and the nematode is discussed and recent important insecticidal toxins from Photorhabdus are described.
Botulinum toxin-A, a neurotoxin derived from Clostridia Botulinum, has been injected into the laryngeal muscle(s) for the treatment of the spasmodic dysphonia at the Voice Clinic, Yonsei Institute of Logopedics and phoniatrics since December 1995. We analyzed 355 patients with spasmodic dysphonia, using Botox register review. In the 355 patients, female is 86.8%. male is 13.2%. 305 patients (85.9%) had adductor type of spasmodic dysphonia and 35 patients (9.9%) were vocal tremor type and 15 patients were abduction and mixed type. Botulinum toxin type-A (Botox) injection using EMG was most frequently conducted as 587 cases, comparing with flexible nasopharyngoscopy gudied injection (68cases) and tele- laryn-goscopy guided injection (31cases). In the respect of frequency of Botox injection, 137 patients(38.6%) were injected one time but 1 patient was injected 17times. The mean dose of Botox is 6.2U. Clinically, initial dose of Botulinum toxin-A was high dose (7-8U) but current dose is small dose (3U). And the mean duration of Botox injection is 6.4 month. In conclusion, to optimize effect of the treatment for spasmodic dysphonia, Botulinum toxin-A injection is combined with voice therapy.
