Unusual bloom of toxic cyanobacteria in water bodies have drawn attention of environmentalists world over. Major bloom of Anabaena, Microcystis in water storage reservoir, rivers and lake leading to adverse health effects have been reported from Australia, England and many part of the world. These cyanobacterial cells can release intercellular matter like toxin in water and these intercellular matter can increase the concentration of organic matter. Cellysis can occur when algal cells meet the disinfectants like chlorine in water treatment plant and the resultant rising up of DOC(Dissolved Organic Carbon) or TOC(Total Organic Carbon) can increase the formation of disinfection by products. Disinfectants that kill microorganisms react with the organic or inorganic matter in raw water. In general disinfectants oxidize the matter in raw water and the resultant products can be harmful to human. There are always conflict about which is more important, disinfection or minimizing disinfection by products. The best treatment process for raw water is the process of the lowest disinfection by products and also the the lowest microorganism. In this study the cultured cells, Microcytis Aeruginosa(MA), Anabaena Flos-aquae(AF), Anabaena Cylindrica(AC), and the cells obtained in Daechung Dam(DC) whose dominant species was Anabaena Cylindrica were subjected to chlorination. Chlorination oxidizes inorganic and organic compounds and destruct live cells in raw water. Chloroform was analyzed for the cultured cells which were treated with $20mg/\ell$ dose of chlorine. In general chloroform is easily formed when dissolved organic matter react with chlorine. The cultured cells contributes the concentration of dissolved organic carbon and also that of total organic carbon which might be potent precusors of chloroform formed. The correlations of the concentration of chloroform, DOC and TOC were investigate in this study.

The present study aimed to analyze the metaproteome of the microbial community comprising harmful algal bloom (HAB) in the Daechung reservoir, Korea. HAB samples located at GPS coordinates of 36°29'N latitude and 127°28'E longitude were harvested in October 2013. Microscopic observation of the HAB samples revealed red signals that were presumably caused by the autofluorescence of chlorophyll and phycocyanin in viable cyanobacteria. Metaproteomic analysis was performed by a gelbased shotgun proteomic method. Protein identification was conducted through a two-step analysis including a forward search strategy (FSS) (random search with the National Center for Biotechnology Information (NCBI), Cyanobase, and Phytozome), and a subsequent reverse search strategy (RSS) (additional Cyanobase search with a decoy database). The total number of proteins identified by the two-step analysis (FSS and RSS) was 1.8-fold higher than that by one-step analysis (FSS only). A total of 194 proteins were assigned to 12 cyanobacterial species (99 mol%) and one green algae species (1 mol%). Among the species identified, the toxic microcystin-producing Microcystis aeruginosa NIES-843 (62.3%) species was the most dominant. The largest functional category was proteins belonging to the energy category (39%), followed by metabolism (15%), and translation (12%). This study will be a good reference for monitoring ecological variations at the meta-protein level of aquatic microalgae for understanding HAB.
The effect of chlorination on disinfection byproducts (DBPs) production from bloom-forming freshwater algae including 7 strains of cyanobacteria and 6 strains of diatoms was investigated. The release and degradation of hepatotoxin (microcystins) by the chlorination on Microcystis under differential condition of the chlorination time and dose were also investigated. The disinfection byproducts formation potentials (DBPFP) of cyanobacterial species and diatoms were ranged from 0.017 to $0.070{\mu}mol\;DBPs/mg$ C and from 0.129 to $0.708{\mu}mol\;DBPs/mg$ C respectively. Among three major groups of DBPs, haloacetonitrils (HANs) was major product in most test strains except Aphanizomenon sp. and Oscillatoria sp. Haloacetic acids (HAAs) was less than 5 % of total DBPs. Chloroform and dichloroacetonitril (DCAN) were dominant compounds in trihalomethanes (THMs) and HANs respectively. After 4 hours chlorination of toxic Microcystis aeruginosa under the dose range of 0.5 to $10mg\;Cl_2/L$, the concentration of intracellular microcystins decreased, but dissolved dissolved microcystins concentration increased with the treatment of more than $3mg\;Cl_2/L$. However the total amount of microcystins was almost constant even at $10mg\;Cl_2/L$ of chlorination. To conclude, our results indicate that the chlorination causes algal cell lysis and release of intracellular microcystins in the intact form to surrounding waters.
A MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) induced chromium resistant strain ($Cr^{r}18$) of unicellular cyanobacterium Anacystis nidulans has been isolated and characterized. The resistant strain could grow (although restricted to $50\%$ of control) in chromium concentration (180${\mu}M$) lethal to the wild-type. Sublethal ($160{\mu}M$) concentration of $Cr^{6+}$ significantly reduced (13-$37.5$) all the photosynthetic pigments of A. nidulans with maximum reduction in phycoerythrin followed by ChI $\alpha$. Pigments of A. nidulans were drastically decreased in lethal concentration of Cr^{6+} with maximum reduction in phycoerythrin ($75\%$) and allophycocyanin ($67.5\%$). Resistant strain $Cr^{r}18$ resisted toxic effects of sublethal and lethal concentrations of $Cr^{6+}$ on photosynthetic pigments as revealed by less decrease in pigments as compared to A. nidulans. Effect of $Cr^{6+}$ stress was also studied on nitrogen assimilation and phosphate uptake. Sublethal concentration of $Cr^{6+}$ drastically reduced ($71.5\%$) nitrate uptake by A. nidulans while a decrease of $29\%$ was observed in strain $Cr^{r}18$. Short (2 day) exposure of A. nidulans and its resistant strain $Cr^{r}18\;to\;Cr^{6+}$ did not affect nitrate reductase and glutamine synthetase (transferase), whereas longer (10 day) exposure to $Cr^{6+}$ lowered activities of both enzymes in A. nidulans but not significantly in the strain $Cr^{r}18$. Ammonium uptake by both strains was not affected by $Cr^{6+}$. Thus, $Cr^{6+}$ affected photosynthetic pigments, nitrogen assimilation, and phosphate uptake of A. nidulans, while strain $Cr^{r}18$ was able to resist toxic effects of the metal. Advantages of using strain $Cr^{r}18$ for bioremediation purposes have been evaluated by studying $Cr^{6+}$ removal from the solution. Resistant strain $Cr^{r}18$ was able to remove $33\%$ more $Cr^{6+}$ than A. nidulans and thus it can prove to be a good candidate for bioremediation of $Cr^{6+}$ from polluted waters.
본 연구에서는 CyanoHABs를 제어하기 위해 주요 우점종인 Microcystis aeruginosa에 대한 항균 펩타이드의 살조 활성을 조사하고, 구조적 특이성을 바탕으로 새로운 M. aeruginosa 제어 펩타이드를 제작하였다. 본 실험에서 CA-MA 유래 펩타이드 CMA1, CMA2와 Helicobacter pylori 유래 펩타이드 HPA3P, HPA3NT3는 처리 48시간 후 각각 67.3, 73.1, 76.7, 69.8%의 최대 살조 효율을 보였다. 또한, 신규 펩타이드 K160242~5는 처리 48시간 후 각각 64.0, 64.1, 66.4, 70.1%의 최대 살조 효율을 보였다. CA-MA 유래 펩타이드는 처리 24시간 이후 세포의 재성장이 관찰되었으므로 세포 표면과의 정전기 인력을 통해 세포를 응집하고 간접적으로 제어하는 것으로 조사되었다. 반면에, 양친매성 펩타이드는 세포의 재성장이 관찰되지 않았으므로 세포 응집과 더불어 세포 내 침투를 통해 직 간접적으로 세포를 제어하는 것으로 추정되었다. 또한, 펩타이드의 살조 기작 및 효율에는 구성 아미노산의 종류, 수, 구조 및 펩타이드의 분자량, 처리 농도 등이 복합적으로 영향을 미치는 것으로 나타났다. 그러나 펩타이드를 이용한 CyanoHABs의 제어 가능성을 제고하기 위해서는 정확한 기작과 관련 인자들의 확인 및 증명이 요구된다.
잡식성 및 플랑크톤 섭식 어류(Carassius gibelio langsdorfi, Hypophthalmichthys molitrix)의 간접노출(CCMF, HCMF) 농도차(0, 10, 50%)에 따른, 남조 Microcystis aeruginosa의 생체량과 세포내부와 외부의 마이크로시스틴(microcystin, MC) 함량을 1일 간격으로 관찰하였다. 실험기간동안 M. aeruginosa균주의 세포내에 함유된 MC의 양은 대조군 보다 모든 처리군에서 증가한 것으로 나타났다(CCMF1, P=0.015; CCMF2, P<0.001; HCMF1, P< 0.001; HCMF2, P<0.001). 처리군간의 비교에서는 CCMF1의 세포내 MC함량 보다 CCMF2의 세포내 MC 함량이 통계적으로 유의한 수준으로 증가하였다(P=0.023). 또한 HCMF2의 세포내 MC함량이 HCMF1의 MC함유량보다 증가한 것으로 나타났다(P<0.001). M. aeruginosa균주의 세포외 MC함량은 대조군과 CCMF1과 CCMF2에서 차이를 보이지 않았고, HCMF1과 HCMF2의 세포외 MC함량이 대조군보다 높은 것으로 나타났다(HCMF1, P=0.003; HCMF2, P<0.001). 본 연구결과, 독성 Microcystis의 경우 어류의 분비화학물질(kairomone) 농도에 따라 세포 내, 외부의 독성이 증가될 가능성이 있으며, 부영양호에서 생물적 조절을 통한 조류저감을 실시할 경우 Microcystis의 독소 변화를 고려해야 할 것으로 보인다.
우리나라 4대강 사업에 의해 건설된 낙동강의 달성보와 함안보, 금강의 세종보와 백제보, 영산강의 송촌보와 죽산보를 대상으로 3차원 수리-수질모델을 이용하여 가동보 운영에 따른 수질 및 녹조 현상 예측 결과를 비교 분석하였다. 가동보를 전면 개방하였을 때, 금강과 영산강에서는 상류에 위치한 세종보와 승촌보에서 연중 및 하절기 Chl-a 농도와 남조류 농도가 감소되는 것으로 예측되었으나, 하류에 위치하는 백제보와 죽산보에서는 하절기에 모두 증가되는 것으로 나타났다. 금강과 영산강에서 수문이 닫힌 경우, 상류에서는 체류시간이 증가하여 조류가 성장에 필요한 용존성 무기인을 충분히 섭취하면서 조류 농도가 증가하지만, 하류에서는 용존성 무기인이 고갈되어 상대적으로 조류의 성장이 제한되는 현상이 발생하는 것으로 분석된다. 반면 낙동강의 달성보 및 함안보에서는 연평균과 하절기 평균 농도가 모두 증가하는 것으로 나타났다. 낙동강에서는 금강과 영산강과 같이 용존성 무기인의 농도가 심각하게 증가하지 않은 반면 수문 개방으로 수심이 감소함에 따라 조류가 이용하는 평균 광량이 증가하면서, 조류의 성장이 증가한 것으로 분석된다. 결과적으로 수문을 전면 개방 또는 폐쇄하였을 경우, 녹조현상 발생 특성은 위치에 따라 수리학적 체류시간, 용존성 무기인 농도 및 수위변화에 따른 광량의 변화 등에 의해 서로 다르게 나타나는 것으로 분석된다.
구슬말(Nostoc commune Vaucher ex Bornet & Flahault)은 이형세포를 갖는 특징으로 다른 목들과 구분되는 남조류의 일종으로 극지방에서 열대지역까지 광범위한 지역에 분포하며 단세포가 연결되어 형성된 수많은 trichome들이 점액질에 둘러 쌓인 형태로 커다란 군체를 형성한다. 주로 토양, 암반, 잔디 위 등에 서식한다고 알려져 있으나 흔히 관찰되지 않기 때문에 현재 연구가 거의 없는 실정이다. 이에 본 연구에서는 토양 남조류인 N. commune HCW0811을 분리 및 동정하였으며 항염증 활성을 조사 하고자 하였다. 그 결과 N. commune HCW0811는 LPS로 유도된 RAW 264.7세포에서 80%이상의 세포 생존율을 나타내었으며 NO, PGE2 및 TNF-α, IL-6, IL-1β의 생성을 효과적으로 억제하였다. 또한 western blot assay를 통해 iNOS, COX-2 및 MAP kinase (p38, ERK1/2, JNK)와 NF-κB 세포내 신호전달 경로에서의 단백질 발현을 조사한 결과 이들의 발현이 유의하게 억제됨을 확인하였다. 본 연구에서는 이러한 결과를 근거하여 HCW0811가 다양한 염증 인자를 표적으로 하는 피부 면역 질환을 포함한 염증성 질환의 예방과 치료를 위한 항염증 기능성 화장품 및 식품소재로의 개발가능성을 제시한다.
