Characterization of a fad3 cDNA Encoding Microsomal Fatty Acid Desaturase from Arabidopsis thaliana (Arabidopsis thaliana로부터 지방산 불포화효소 유전자의 분석)

  • For the molecular genetic study of cold tolerance mechanism in plants, a cDNA encoding fatty acid desaturase (fad3), converting linoleic acid (18:2, $\omega$-6) to linolenic acid (18:3, $\omega$-3), was isolated from $\lambda$ZAPII Arabidopsis thaliana cDNA expression library by plaque hybridization using fad3 cDNA probe derived from Brassica napus. A 1.8 kb-EcoRI fragment from a lambda clone showing a strong positive hybridization signal was subcloned into pGEM7 and analyzed for its nucleotide sequence. From deduced amino acid sequences, the fad3 gene was revealed to have an open reading frame(ORF) consisting of 386 amino acids with a molecular mass of 44,075 Da. The fad3 gene was compared to chloroplast $\omega$-3 fatty acid desaturase (fad7) and endoplasmic reticulum Δ12 fatty acid desaturase (fad2) to show 70% and 58% amino acid sequence homology, respectively, Especially, amino acids of internal (82 to 151) and carboxy terminal (276 to 333) regions were highly conserved, implying their requisite role for enzymatic functioning of fatty acid desaturases. IPTG-induced fad3 cDNA expression in E. coli cells was suggested to be toxic to bacterial growth.

  • Until now, pure drinking water grnerally menas the water without taste, odor, general bacteria, coliform, and other exotic substance. Such a definition has been changing recently due to the finding of numerous other inorganic and organic substances unknown to us. 10 years ago, major causes of death were infectious agents and parasites contained in water, but recently, it has become apparent that some substances contained in drinking water cause cancer and heart diseases. We must drink about 2L of water everyday in order to maintain healthy condition. Waters used for drinking include tap water, well water, spring water, filtered water, etc., but the quality of drinking water has more polluted due to the industrial development and population increase. For example, industrial waste waters from industrial plants pollute the water supply sources ; toxic substances contained in the waste waters pollute the ground water sources by penetrating the geological strata, and municipal, livestock, public building waste waters also pollute the water supply sources. Sometimes, the polluted surface waters were announced to be polluted by various kinds of orgainc substance, and it is reported that the pollution of ground water by orga nic substances has few in number but high in its concectration comparing with those of surface water. As the water quality pollution level increases, so the amount of disinfectant also increase. For example, chlorine solution, one of widely used disinfectants, creates trihalomethane(THM), a carcinogen, and halogen compounds. According to Oliver, through chlorine disinfection process, humine substance and chlorine create bolatile organic halide and nonvolatile organic halide by chemical reaction. There are tens or hundreds filtering devices, but filtering principles and maintenance metjhods are different, so their efficiency tests are needed. According to Smith, the effeciency tests aginst over 30 Ameican filtering devices show that 10 devices can remove 85% of volatile organics and further studies on filtered waters are underway. In consideration of important impacts of polluted drinking water on national health, authors studied the state of water quality pollution against tap water used as drinking water, filtration device passed water, ground water, and conserved drinking water ; tested the efficiency of filtration devices for tap water ; tried to sep up the detection method by using ion chromatography based on negative ion and positive ion by using single column, and attemped the simple filtration method for general households.

  • Glyphosate is the active ingredient in widely used herbicides. It acts through inhibition of the shikimate metabolic pathway in plants. This pathway does not exist in mammals, however, so glyphosate is presumably less toxic to humans. Nevertheless, fatal cases of glyphosate poisoning in humans have still occurred. Cases of glyphosate poisoning reported in the previous literature were almost always caused by intentional ingestion. Therefore, intramuscular injection of glyphosate with suicidal intent has not been reported. We report a case of 43-year-old man with poisoning due to intramuscular injection of glyphosate herbicide. He was admitted to the emergency department with a chilling sensation, local hotness, swelling, and tenderness at the site of glyphosate injection. He was treated with intravenous antibiotics and analgesics for 10 days and was discharged without any other complication.

  • Modern chemical plants including petroleum refinery and gas industries have evolved into more complex and specialized. In these industrial complexes, it is important to maintain acceptable safety level protecting from various potential disasters caused by fire, explosion and the leakage of toxic materials. Recently possibility and consequence of accidents are increasing in the industrial process. So there is a trade-off between the plant operation efficiency and safety level. In this study SIF(Safety instrument Functions) was incorporated into SIL(Safety Integrity Levels). As a result, the safety level was upgraded by designing resonable allocation of safety instruments.

Evaluation of a Rapid Immunochromatography Technique for Determination of Cd in Soils and Rice Grains (면역크로마토그래피를 이용한 토양 및 쌀의 Cd 간이진단법 평가)

  • Cadmium is known to be very toxic to human health and can be relative easily translocated from soils in plants. Therefore, a rapid method for screening Cd in soils and crops has become more and more important. For this reason, we examined a rapid immunochromatograpy (ICG) test kit which uses antigen-antibody reaction based on immunoassay and chromatography. Soils and rice grains collected from mine waste-contaminated sites were determined for their Cd contents using this kit. For comparison purposes, 0.1 M HCl and ICP-OES were employed as a conventional extraction and determination method. Cadmium contents in rice grains determined using ICG technique were $0.46{\sim}2.39mg\;kg^{-1}$ and Cd contents determined using 0.1 M HCl and ICP-OES were $0.52{\sim}1.97mg\;kg^{-1}$. The correlation between these two Cd contents were statistically significant ($r^2$=0.930). The results of Cd contents in soils also showed a statistically significant relationship between these two methods ($r^2$=0.975). On the basis of these results, ICG technique can be applied to rapidly quantify Cd in crops and soils. However, further research is necessary to apply ICG technique for the field screening.

Risk Assessment on Nitrate and Nitrite in Vegetables Available in Korean Diet

  • Nitrate is a naturally occurring compound in plant and can be converted to nitrite in the nitrogen cycle and vice versa. Therefore, it is easy to find nitrate in plants including vegetables. Especially, it is known that high levels of nitrate found in leafy vegetables. Nitrate itself is relatively non-toxic but its metabolites and reaction products such as nitrite, nitric oxide and N-nitroso compounds, may produce adverse health effects such as methaemoglobinaemia and carcinogenesis. To execute the risk assessment of dietary nitrate and nitrite for the intake of vegetables, it is investigated that the levels of nitrate and nitrite in 23 vegetables (798 samples). The range of concentration were 0-6,719mg/kg for nitrate and 0-1,635mg/kg for nitrite, respectively. The Estimated daily intakes (EDI) were 0.85-1.38 mg/kg body weight/day for nitrate and 0.02-0.03 mg/kg body weight/day for nitrite by ages. We conclude that there are no health concerns for eating various vegetables since the EDI were below the Tolerable Daily Intake (3.7 mg/kg body weight/day for nitrate, 0.06 mg/kg body weight/day for nitrite) level established by WHO.

Chemical Properties of Soil Solution under Larix leptolepis, Pinus koraiensis, and Quercus mongolica Stands (일본잎갈나무, 잣나무, 신갈나무 임분 토양의 층위별 토양수의 화학적 특성)

  • Soil solutions were collected by zero-tension lysimeters at Kwangju, Kyunggi Province to estimate differences in ion concentration among species and horizons. Zero-tension lysimeters were installed in O, A, and B horizons in Larix leptolepis, Pinus koraiensis, and Quercus mongolica stands. Soil solution samples were collected from September 1996 through August 1998 and analyzed for $K^+$, $NA^+$, $NH_4^+$, $Mg^{2+}$, $Ca^{2+}$, $Al^{3+}$, $Cl^-$, $SO_4^{2-}$, $NO_3^-$, and $PO_4^{3-}$. The experimental site had high nitrogen loading from the atmosphere, and $NO_3^-$ was positively correlated with $K^+$ and $Mg^{2+}$. However, $NO_3^-$ and $NH_4^+$ showed a positive correlation only in the O horizon of Q. mongolica stand. Mg^(2+) deficit in the soil was predicted owing to the positive relationship of $Mg^{2+}$ with $NO_3^-$. Concentrations of $K^+$ and $Ca^{2+}$in soil solution were estimated higher in L. leptolepis than in other stands because of high leaching from the plant. Concentration of $Al^{3+}$ in soil solution was negatively correlated with soil solution pH. Mean soil solution pH of A and B horizons in P. koraiensis was lower than 4.7, however the $Al^{3+}$ concentration was lower than the toxic level to plants.

  • The wetlands have functions those were retention of diverse biota, purification of water quality, control of climate and flood, eco-tourism and supply of agricultural water, and that was the ecosystem of high biodiversity as the zone of transition between inland and water. Therefore, this study showed the conservation and management plan by analyzing in the abiotic and biotic environment of forest swamp, Gyeongsangbuk-do. The results were as follows. Examining the management plan of the abiotic environment, there will need to establish the hydrological plan in continuous expediting the supply of water to maintain a humid soil of swamp, and to try to find the organic farming, use of low toxic agrichemicals and so on to prevent occurring a non-point source pollutants. To prevent changing the flowing of ground water and inflow of earth and sand in modifying the land character, there will be needed to restrict the construction of farmland and slope around swamp. To manage the biotic environment, there needed to offer the habitat by removing the regular interval and individual of a dead tree, and to improve the growth environment of vegetation. Because the naturalized plants disturb the natural vegetation, they will be removed. The afforested trees like Robinia pseudoacacia and Amorpha truticosa will be renewed to prevent the heterogeneity of landscape ecology, and the active conservation plan on wetland species will be established. As this study was carried out to study on the partial swamp, Gyeongsanbuk-do, the ecological environments distributing a swamp in Korea show a some problem. In the future, the study will accomplish to study the accurately and objectively ecological environment and management of a swamp by analyzing the extensive sites.

In Rappaccini's Daughter, he explains how the power of men influences a woman's life and drives her to death. Her father, Rappaccini, cultivates plants in his garden that are toxic and conducts a scientific experiment that gives his daughter Beatrice a fatal level of toxicity. He insists that this experiment was performed to protect Beatrice, but ultimately, it causes her death. Giovanni, who falls in love with Beatrice, provided an antidote in the attempt to detoxify her, but it resulted in her death. Finally, Baglioni used Giovanni to steer Beatrice to drink the antidote to defend his social status. The three men's selfishness and jealousy led to the demise of Beatrice, who eventually died from the selfish power of men and not due to her toxicity.

  • Although gut microbial functions have been analyzed through cultivation of isolated microbes, molecular analysis without cultivation is becoming a popular approach in recent years. Gene cloning studies have partially revealed the mechanisms involved in fiber digestion of individual microbe. The molecular approach finally made it possible to analyze full genomes of the representative rumen cellulolytic bacteria Fibrobacter and Ruminococcus. The coming database may contain useful information such as regulation of gene expression relating to fiber digestion. Meanwhile, unculturable bacteria are still poorly characterized, even though they are main constituents of gut microbial ecosystem. The molecular analysis is essential to initiating the studies on these unculturable bacteria. The studies dealing with rumen and large intestine are revealing considerable complexity of the microbial ecosystems with many undescribed bacteria. These bacteria are being highlighted as possibly functional members contributing to feed digestion. Manipulation of gut bacteria and gut ecology for improving animal production is still at challenging stage. Bacteria newly introduced in the rumen, whether they are genetically modified or not, suffer from poor survival. In one of these attempts, Butyrivibrio fibrisolvens expressing a foreign dehalogenase was successfully established in sheep rumen to prevent fluoroacetate poisoning. This expands choice of forages in tropics, since many tropic plants are known to contain the toxic fluoroacetate. This example may promise the possible application of molecular breeding of gut bacteria to the host animals with significance in their health and nutrition. When inoculation strategies for such foreign bacteria are considered, it is obvious that we should have more detailed information of the gut microbial ecology.
