갈색 느티만가닥버섯 메탄올 추출물의 항산화 및 tyrosinase 저해 효과 (Antioxidant and tyrosinase inhibitory activity of Hypsizygus marmoreus (brown cultivar) methanol extracts)

  • 본 연구에서는 기능성 식품 및 화장품 소재로써 갈색 느티만가닥버섯(Hypsizygus marmoreus)의 이용가능성을 조사하기 위해서 갈색 느티만가닥버섯 메탄올 추출물의 항산화 활성 및 tyrosinase 저해 효과를 갓(pileus)과 대(stipes)로 분리하여 부위별로 조사하였다. 메탄올 추출물 중 갓과 대의 총 폴리페놀 함량은 각각 $8.7{\pm}3.27$ ug/mg과 $5.6{\pm}2.85$ ug/mg이었고 플라보노이드 함량은 각각 $2.8{\pm}3.81$ ug/mg과 $1.4{\pm}1.95$ ug/mg이었으며 총 폴리페놀과 플라보노이드 함량 모두 대보다 갓에서 높게 나타났다. Tyrosinase 저해 활성은 추출물의 농도에 따라 증가하는 경향을 나타내었으나 양성 대조구로 사용한 2% 알부틴(arbutin)과 비교했을 때 1200 mg/ml의 고농도에서도 갓은 66.9%, 대는 57.97%의 낮은 저해 활성을 나타내었다. 항산화 활성은 DPPH에 의한 라디칼소거 활성을 측정하여 확인하였으며 갓과 대의 DPPH 라디칼 소거능은 20 mg/ml의 농도에서도 각각 52.55%와 30.35%로 낮게 나타났다. 추출물이 B16BL6 mouse melanoma cell에 미치는 영향을 알아보기 위해 WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate)를 이용하여 세포생존율을 조사하였으며 갓과 대 모두 200-2,000 ug/ml의 농도로 처리하였을 때 100% 이상의 생존율을 나타내었고 추출물의 처리 농도가 증가함에 따라 세포생존율도 증가하는 경향을 나타내었으며 양성대조군인 0.04% adenosine을 처리한 경우보다 높게 나타났다.

  • Plant tissue homogenization using a mortar or mechanical equipment has been the preferred method for obtaining high yields of total RNA; this method, however, is both time-consuming and expensive. Additionally, homogenization may generate excessive endogenous RNases, polyphenolics, and other substances that reduce the quality and quantity of RNA. In this study, we describe the microwave irradiation-assisted RNA extraction (MIRE) technique which, without tissue disruption and homogenization, allows for the cost-effective and rapid generation of intact RNA from apple cane shavings and the reliable detection of apple virus by RT-PCR.

Antioxidant and Antiproliferative Activities of Methanolic Extracts from Thirty Korean Medicinal Plants

  • To study the health promoting effects of medicinal plants, 30 medicinal plants commonly available in Korea have been evaluated for their antioxidant compounds and antioxidant and antiproliferative activities. Total polyphenolics and flavonoids in the methanolic extracts were measured by spectrophotometric methods and 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities and chelating effects have been determined for antioxidant activities. Moreover, the effects of medicinal plants on cell proliferation of intestinal (Caco-2) and pituitary (GH3) tumor cells were investigated using thiazolyl blue terazolium bromide (MIT) assay. The methanolic extracts of Pueraria thunbergiana and Artemisiae asiatria contained the highest total polyphenolic and flavonoid contents, respectively. P. thunbergiana exhibited the highest antioxidant activities. A. asiatria showed the strongest antiproliferative activity against Caco-2 and Ponciruc trifoliata Rafin and Lophathrum gracile Bronghiart exhibited the highest activities against GH3. Although there was positive correlation between ABTS radical scavenging activity and polyphenolic contents ($R^2=8189$), no relationship was found between antiproliferative and antioxidant activities.

Stachys riederi var. japonica Extract Reduces Cytochrome C Release from Mitochondria in UVA-irradiated Human Dermal Fibroblasts

  • This study was performed to investigate the cytoprotective effects of Stachys riederi var. japonica ethanol extract (SREE) to control oxidative stress induced by UVA-irradiation by examining antioxidant capacity and gene expression of cytochrome c using human dermal fibroblasts. The total polyphenolics and flavonoids in the SREE were 41.2 and 25.4 mg/g, respectively. At concentrations of 500 and $1000{\mu}g/mL$, the electron-donating ability of SREE was 48.6% and 82.0%, respectively, and the 2,2'-azino-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity was 62.3% and 78.8%, respectively. These findings showed that SREE has a fairly good antioxidant capacity. As determined by an MTT assay, the maximum permissible level for treating SREE to human dermal fibroblasts was shown to be over $200{\mu}g/mL$. SREE ($200{\mu}g/mL$) significantly decreased cytochrome c mRNA and protein expression by 31.1% (p<0.001) and 38.8% (p<0.01), respectively. These findings suggest that SREE may protect human skin cells against mitochondrial-dependent apoptosis. Therefore, SREE seems to be a natural antioxidant to protect cells against oxidative stress induced by UVA-irradiation.

Enhancement of polyphenols, flavonoids and antioxidant activities in water extract of mulberry (Morus alba L.) root bark by steam treatment

  • Different parts of the mulberry plant are described to be potential sources of polyphenolics exhibiting strong antioxidant activity. In this study, we prepared various aqueous extracts of mulberry root bark by subjecting to steam at different temperatures and time intervals (45℃, 15 h; 70℃, 15 h; 95℃, 6 h and 95℃, 15 h) followed by extracting at 80℃ for 1 h. The total polyphenolic content ranged from 66.82-101.20 mg gallic acid equivalent (GE)/g of extract whereas the flavonoids were in the range of 13.03-25.23 mg catechin equivalent (CE)/g of extract. The extracts also exhibited strong antioxidant activities (0.99-1.66 mg trolox equivalent (TE)/g of extract in DDPH assay and 10.65-16.26 mg TE/g of extract in ABTS assay). This study clearly showed an improvement in the antioxidant activity of the water extract of mulberry root bark by the steam treatment, which can be used as a tea or health-promoting materials.

인삼 적변유기에 대한 폴리페놀과 Polyphenol Oxidase의 잠재적 역할 (Potential Role of Polyphenolics and Polyphenol Oxidase on the Induction of Browning in Ginseng Roots)

  • 본 연구는 적변 유발 세균에 의해 발생하는 적변삼과 polyphenol 및 polyphenol oxidase 활성과의 관계를 조사하였으며, 이는 다음과 같다. 1. 적변 유발 균주를 상처낸 건전 인삼 외피에 접종하여 무처리구와 비교하였을 때, 접종일수의 증가에 따라 Hue angle값이 접종 후 1일 101.2, 10일 95.9, 20일 90.1, 30일 60.9로 적색으로 변화되었다. 2. Lysobacter gummosus, Pseudomonas veronii 및 Agrobacterium tumefaciens 균주를 인삼의 상처부위에 접종하였을 때, 총페놀의 함량은 일수가 경과할수록 증가하였다. 3. Lysobacter gummosus, Pseudomonas veronii 및 Agrobacterium tumefaciens 균주를 상처낸 인삼에 접종하였을 때 페놀함량의 증가와 함께 효소의 활성이 증가하였으나 단백질당 PPO의 활성은 감소하였다.

오옥신 처리가 에키네시아의 부정근 및 생리활성물질 생산에 미치는 영향 (Auxin Affects on Production of Adventitious Roots and Secondary Metabolites in Echinacea angustifolia)

  • The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L ${\alpha}$-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.

새싹 밀의 추출용매에 따른 생리활성 평가 (Evaluation of Biological Activities of Wheat Sprouts with Different Extraction Solvents)

  • The purpose of this study was to examine biological activities, including total contents of polyphenol, antioxidant activities, inhibitory activities of tyrosinase, and protective effect against oxidative stress in the HepG2 cells of ethanol extracts from wheat sprout. The antioxidant activity of extracts was determined by ABTS and DPPH radical scavenging activities. Ethanol extracts were tested using different ethanol concentrations (0%, 30%, 50%, 80% and 95%, respectively). The highest amount of total polyphenol was extracted by 50% and 80% ethanol which was 26.3 and 26.8 mg gallic acid equivalents/g sample, respectively. High levels of ABTS and DPPH radical scavenging activity were found in 50% ethanol (26.7 and 15.0 mg TEAC/g sample, respectively) and 80% (24.3 and 16.1 mg TEAC/g sample, respectively) ethanol extracts. Also, 50% and 80% ethanol extracts indicated higher inhibitory activities of tyrosinase compared with other extracts. In the cell-based assay, pre-treatment of the HepG2 cells with wheat sprout extracts prevented the cell damage induced by TBHP (tert-butyl hydroperoxide). The results of this study indicate that wheat sprout has significantly higher diverse biological activities and apparently has significant health benefits.

발아현미 첨가에 따른 국수의 제조 특성과 GABA 함량 및 항산화 활성 변화 (Quality Characteristics and Changes in GABA Content and Antioxidant Activity of Noodle Prepared with Germinated Brown Rice)

  • 발아 전과 후의 현미와 미강을 첨가한 밀가루 복합분으로 만들어진 국수의 품질 특성과 GABA 함량 및 항산화 활성의 변화를 조사하였다. 발아 전후의 현미와 미강을 각각 5, 10, 15% 첨가하여 국수를 제조하였다. 이렇게 제조된 국수는 조리 및 제면특성(cooking loss, swelling index, water holding capacity), 조리에 따른 GABA 함량과 항산화 성분(polyphenol) 및 활성(ABTS와 DPPH의 radical scavenging activities) 변화를 통해 기능성 국수로의 이용가능성을 알아보고자 하였다. 실험결과, 발아 후 rice bran의 첨가에 따른 cooking loss가 증가하는 것으로 관찰되었으며 반면에 swelling index에서는 차이를 나타내지 않았다. 발아 후 현미와 미강을 첨가한 조리 전후 국수에서 발아 전 현미와 미강을 첨가한 국수보다 높은 GABA 함량을 확인하였으며 발아에 따른 polyphenol 함량과 항산화 활성에서는 큰 변화가 없음을 확인할 수 있었다. 따라서 발아현미가 첨가된 국수는 밀가루 국수의 영양학적 가치와 항산화 기능을 향상시킬 수 있는 소재로써 이용 가능할 것으로 생각된다.

추출온도 및 함수율에 따른 오미자 종자유의 이화학적 특성 및 항산화활성 (Physicochemical Characteristics and Antioxidant Activities of Omija (Schizandra chinensis Baillon) Seed Oil Extracted at Different Temperatures and Moisture Contents)

  • Omija seed oil was extracted from Omija seeds, a by-product of Omija, using an eco-friendly pressed extraction method. Physicochemical properties and antioxidant activities of 12 extracts using different combinations of seed moisture content (5, 7.5, and 10%) and extraction temperature (25, 49, 75, and 100℃) were then investigated. The highest extraction yield was 31.33% at moisture of 5% and temperature of 75℃. The lowest acid value was 4.18±0.25 at moisture of 5% and temperature of 49℃or moisture of 7.5% and temperature of 25℃. Peroxide value, iodine value, and saponification value were the lowest at 0.64±0.56 meq/kg, 159.38± 6.03, and 57.60±9.40, respectively, at moisture of 5% and temperature of 25℃. The content of total polyphenolics was the highest at 4,413±125 mg TAE/100 g when the moisture content was 10% and the extraction temperature was 25℃. DPPH radical scavenging activities of oil extracts at 20~100 mg/mL were 28.68±7.30~87.65±2.20%. The maximum antioxidant activity and IC50 were 87.65±2.20% and <20 mg/mL, respectively, for extract obtained at moisture of 10% and temperature at 100℃. As a result, the extraction yield, acid value, peroxide value, iodine value, saponification value, and so on were excellent at moisture content of 5% and extraction temperature of 25℃ or 49℃. However, the content of total polyphenolic compounds and antioxidant activity were the highest at moisture of 10% and extraction temperature of 100℃. In conclusion, extracting Omija seed oil from Omija seeds, a by-product of Omija, is effective with a pressed extraction method.
