Molecular characterization and biological changes caused by Agrobacterium-mediated infiltration of PgTRX1

  • In order to test the functionality of Panax ginseng thioredoxin 1 (PgTRX1) isolated from fermented wild ginseng roots, a transient effect on physiological activity were performed over a short time frame using the Agrobacterium infiltration technique. The PgTRX1 gene isolated from fermented wild ginseng was confirmed to have a size of 579 bp, and the expression of PgTRX1 was the highest in the sample after 6 h of fermentation. As a result of constructing this gene and confirming the infiltration reaction mediated by Agrobacterium in tobacco leaves, it was found that the expression of the NbHSR203j gene was also induced as PgTRX1 expression increased. As a result of measuring the biological activity of the infiltration samples, the total phenol content increased by 35.45±1.84 to 49.01±1.84 ㎍ GAE/mL compared to the control, and the total flavonoid amount of 9.52±0.41 to 9.82±0.25 ㎍ QE/mL was slightly high. From these results, Agrobacterium-mediated PgTRX1 appears to be related to the hypersensitive response induction mechanism of plants and the production of secondary metabolites such as phenolic substances.

Hydrophilic Extracts of the Bark from Six Pinus Species

  • Pine barks are important biomass resources because they are utilised in the production of pine wood and rosins. However, no chemical study has been conducted on the hydrophilic status of pine barks in Indonesia. This aim of this study is to explore the hydrophilic extracts of the barks from six Pinus species (P. elliotii, P. caribeae, P. oocarpa, P. merkusii P. montezumae, and P. insularis). The hydrophilics of pine barks were analysed using gas chromatography-mass spectrometry. The presence of polyphenol contents in the ethanol extracts obtained from the barks of six Pinus species was determined using the tannin-formaldehyde method, Folin-Cioucalteu assay, and vanillin-HCl assay. The ethanol and hot water soluble extractives derived from inner barks were higher in quantity when compared to those derived from the outer bark samples. The polyphenol measurement showed that the highest value of total phenol content was derived from the outer bark of P. montezumae whereas those of the total phenol and tannin- formaldehyde contents were derived from the inner and outer barks of P. oocarpa. GC-MS analysis revealed that nitrogenous compounds are dominant constituents in the inner and outer barks of the six species, followed by sugars and monophenolics, respectively.

The Absorption and Metabolism of Fenobucarb and Carbofuran by Susceptible and Carbamate Insecticide-selected Strains of the Brown Planthopper (Nilaparvata lugens Stal) (Fenobucarb 및 Carbofuran의 저항성 벼멸구 체벽 투과량과 체내대사에 관한 연구)

  • Cuticular penetration and detoxication as mechanisms of resistance to the carbamate insecticides in fenobucarb-selected($R_{f}$) and carbofuran-selected($R_{c}$) strains of the brown planthopper (N. lugens Stal) were investigated. Rates of penetration were not significantly different in the susceptible and resistant strains. However, total amount of excretion of the $R_{f}$ and $R_{c}$ strains were much larger than that of the susceptible strain. Fenovucarb and carbofuran were in vivo metabolizd much faster in the $R_{f}$ strain than in the susceptible strain. OSBP(o-sec-butyl phenol) and 3-ketocarbofuran phenol were invitro the major metabolites of fenobucarb and carbofuran in the brown planthopper, respectively. Total amount of the two major metabolites were produced abotu 2 times larger in the $R_{f}$ and $R_{c}$ strains compared to the susceptible strain. OSBP and 3-ketocarbofuran phenol were not so toxic to the brown planthopper ($LD_{50}$ >100 $\mu\textrm{g}$/g hopper). Based on our data, detoxication plays a large role in resistance to fenobucarb and carbofuran in the resistant strain of BPH, although several resistance factors maybe involved.

Antioxidant Activities of Various Extracts from Korean Yam (Dioscorea batatas DECNE.)

  • The objective of this study was to investigate the antioxidant activity of Korean yam (Dioscorea batatas $D_{ECNE.}$) by different extraction solvents including 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v). Raw yam was analyzed for its color property, total phenol content and antioxidant activity. Yam possessed high $L^*$ value and $H^{\circ}$ value, which were $81.64{\pm}2.59$ and $83.36{\pm}0.15$, respectively. Raw yam was found to have great antioxidant activity evaluated through ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power. Total phenol contents of various extracts from raw yam increased in the following order: 70% methanol extract ($37.62{\pm}0.88mg\;CAE/g$), 70% ethanol extract ($43.38{\pm}0.66mg\;CAE/g$) and CM extract ($67.17{\pm}0.12mg\;CAE/g$), respectively. The same trend was also could be found in the ABTS radical scavenging activity, DPPH radical scavenging activity and ferric reducing antioxidant power assays. These results implied that Korean yam might play an important role in antioxidation and serve as the bio-health functional food to take a good part in prevention of human diseases and aging.

Changes in Antioxident Nutrients and Vitamin $B_1$ Contents of Gamma Irradiated Foods Consumed most Frequently in Korea (한국인 다소비 섭취 식품의 방사선 조사후 항산화 영양소와 비타민 $B_1$의 변화)

  • Food irradiation has been steadily increased in many countries concomitantly with increasing international trades and concerns about naturally occurring harmful contaminants of food. Although, irradiation provides an excellent safeguard for the consumer by destroying harmful bacteria, it would be necessary to investigate nutritional adequacy of irradiated foods with a special emphasis on such foods as red pepper powders, cold-water fishes (mackerel), sesame seeds that are widely consumed in Korea. These food items are of special concern due to the fact that most of them are imported and that several nutrients contained in them are sensitively reacting to irradiation. We observed changes in the antioxidant nutrients contents, such as vitamin A, $\beta$-carotene, vitamin C and total phenol, and also vitamin B$_1$ in the foods that were irradiated at different dose levels. The nutrients contents in red pepper were respectively 98.6% and 92.3% for total phenol, and 97.8% and 94.5% for $\beta$-carotene, right after irradiation at the dose of 10 and 20 kGy. The vitamin C contents decreased with irradiation doses up to 76.4% (10 kGy) and 68.5% (20 kGy) and B$_1$ contents decreased to 54.9% and 80.7% of non irradiated red pepper. When the mackerel was irradiated at the applied dose of 3 and 7 kGy, total phenol concentration of the irradiated fish was respectively 91.3% and 94.2% of non irradiated fish. The amounts of vitamin A were 82.6% (3 kGy) and 78.0% (7 kGy) of unirradiated sample and vitamin B$_1$ contents reduced to 30.5% and 51.5%. $\alpha$-Tocopherol content was 33.9% of the non-irradiated one when it was irradiated at 7 kGy. Vitamin A and ${\gamma}$-tocopherol contents in sesame seeds linearly decreased with the applied doses (5 and 10 kGy) Vitamin A contents decreased up to 87.8% (5 kGy) and 82.9% (10 kGy) and that of ${\gamma}$-tocopherol were 72.4% and 66.5% of the unirradiated sample.

Antioxidant Activities and Lipid Peroxidation Inhibition Ability of Gardenia jasminoides Ellis Fructus Seed Extracts (치자(Gardenia jasminoides Ellis fructus) 종자 추출물의 항산화 활성 및 지질과산화 저해능)

  • The purpose of this study was to investigate the bioactivity of extracts from the seeds of Gardenia jasminoides Ellis fructus (GJE) found in Namhae, Korea. Extraction was performed using three solvents, 70% methanol, Distilled Water (DW), and Ethyl Acetate (EA). We determined the total phenol and phytic acid contents of the extracts to evaluate their nitrogen oxide scavenging activity, antioxidant activity, reducing power, and lipid peroxidation inhibition ability. The phytic acid content of GJE was found to be 1.157 mg PAE (Phytic Acid Equivalent) /g DW. The yields of the three extraction processes were as follows: DW, 36.61%; 70% methanol, 30.10%; and EA, 20.40%. The physiological activities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, and 0.6 mg/mL) (p<0.05), but were lower than those of ascorbic acid, BHA, and trolox. Total phenol content was the highest in the 70% methanol extract, followed by DW and EA extracts. Further, nitrogen oxide scavenging activity and antioxidant activity were the highest for the 70% methanol extract followed by DW and EA extracts. Based on these results, the bioactivities of the 70% methanol and DW extracts of GJ seeds were excellent. These extracts can be used as natural antioxidants.

Effects of Thermal Treatment on Antioxidant Activity in Yam (Dioscorea batatas DECNE.)

  • The aim of this study was to investigate the antioxidant activity of thermal treatment yam (Dioscorea batatas DECNE.) in Korea. Thermal treatment yam was extracted by different solvents including 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v). Then color property, total phenol content and antioxidant activity were analyzed. Yam possessed high $L^*$ value and $H\limits^{\circ}$ value, which were $54.92{\pm}2.18$ and $73.20{\pm}0.77$, respectively. Thermal treatment yam exhibited great antioxidant activity evaluated by ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power. Total phenol contents of various extracts from thermal treatment yam increased in the following order: 70% methanol extract ($63.53{\pm}0.33mg\;CAE/g$), 70% ethanol extract ($69.47{\pm}1.00mg\;CAE/g$) and CM extract ($97.49{\pm}0.66mg\;CAE/g$), respectively. The same trend was also could be found in antioxidant activity assays except for reducing power assay. These results implied that these extracts from thermal treatment yam might be useful to take a good part in prevention of human diseases and aging.

Characteristics of Tyrosinase Inhibitory Extract from Ecklonia stolonifera

  • Tyrosinase inhibitory activities of 14 kinds of seaweed, Ecklonia stolonifera, Ecklonia cava, Undaria pinnatiflda, Laminaria japonica, Sargassum fulvellum, Sargassum miyabei, Sargassum thunbergii, Porphyra yezoensis, Gracilaria verrucosa, Carpopeltis affinis, Pachymeniopsis elliptica, Gelidium amansii, Codium fragile and Ulva pertusa were determined using commercially available mushroom tyrosinase in an in vitro assay system. The $1\%$ (w/v) methanol extract from E. stolonifera showed the highest tyrosinase inhibitory activity of $79.0\%$, electron donating activity of $79.0\%$ and total phenol content of 3.75 mg/100g. Ethyl acetate-methanol-water (7 : 2 : 0.2, v/v) fraction $(0.5\%,\;w/v)$ isolated from the methanol extract showed tyrosinase inhibitory activity of $75.9 \%$, electron donating activity of $88.1 \%$ and total phenol content of 4.38 mg/100g. Tyrosinase inhibitory activity was closely associated with total phenol content (R = 0.99) and electron donating activity (R=0.99). Maximum absorption wavelength of the fraction was 218nm and that of phenolic compounds showed about a range from 210 to 220nm. The inhibition mode of the fraction was noncompetitive inhibition.

Quantity Analysis of Nutrients in Soybean Sprouts Cultured with Germanium (게르마늄을 첨가하여 재배한 콩나물에서 영양성분의 함량 분석)

  • This study was carried out to evaluate the nutritional value of soybean sprouts cultured in water with germanium. In the germanium treated soybean sprouts (GSP), the contents of moisture and ash were higher than those of control group (germanium non-treated soybean sprouts), with 88.7~90.2% and 16~24%, respectively. The contents of dietary fiber, vitamin C and total phenol of GSP were also higher than those of control group especially with more than 20~40% in total phenol being contained. Minerals of soybean sprouts were considerably increased during S-day cultivation. Of minerals, the contents of calcium and iron in GSP were higher than in the control. In particular there was no germanium in control group, but on the other hand, GSP had germanium of 159 ppm. Therefore, it may be suggested that, soybean sprout with germanium is better than soybean sprout without germanium in terms of nutrition.

Contents of Isoflavones and Antioxidative Related Compounds in Soybean Leaf, Soybean Leaf Jangachi, and Soybean Leaf Kimchi (콩잎 밑 콩잎 요리의 이소플라본 함량 및 항산화 관련 성분들의 비교)

  • Soybean is an important plant as it is the source of protein and oil as well as various phytochemicals that are related with biological activity. Over the past decades, scientists have conducted considerable research on the physiological properties of soybeans, especially isoflavones, which are the characteristic components in soybeans. However, there is no research on the properties or the bio-functionality of soybean leaf. Jangachi and kimchi are two of the traditional special dishes of Gyungsang Province in Korea which we made from soybean leaves. Depending on the recipe, green or yellow soybean leaves are used for the preparation of these two side dishes. We compared the antioxidative activity and measured the contents of isoflavones, total phenol, chlorophylls, carotenoids, and vitamin C in the ingredients (green and yellow soybean leaf) and the final side dishes (jangachi and kimchi). We Int report that isoflavones were contained in soybean leaf and that jangachi had the highest isoflavone contents among the samples. Yellow soybean leaf contained higher isoflavones than green soybean lear and kimchi. From the TEAC assay results, the sequence or antioxidative activities was yellow soybean leaf > soybean leaf jangachi > green soybean leaf > soybean leaf kimchi. The sequence was the same with total phenol contents, indicating that antioxidative activity is highly related with total phenol level. Chlorophylls, carotenoids and vitamin C existed abundantly in green soybean leaf. In conclusion, soybean leaf could be a good material for health due to the presence of isoflavones and the other useful antioxidants mentioned above.
