동결건조 비빔밥을 이용한 방사선 멸균 우주식품 개발 (Development of Freeze-dried Bibimbap, Korean Cooked Rice with Red Pepper Paste, as a Space Food Sterilized by Irradiation)

  • This study evaluated the microbial population (total aerobic count, fungi) and organoleptic quality of freeze-dried bibimbap sterilized by gamma irradiation to develop the space food. An irradiation dose above 30 kGy was needed to sterilize the freeze-dried bibimbap, while organoleptic quality of the sample was significantly decreased by irradiation (p<0.05). However, it was observed that scores on overall acceptance of rehydrated bibimbap after irradiation were the highest, when vitamin C and paprika extract were added at 0.1%, respectively. Therefore, it was considered that the freeze-dried bibimbap could be developed as a space food, which meet microbial requirements and organoleptic quality through addition of vitamin C 0.1% and paprika extract 0.1% before gamma irradiation at 30 kGy.

  • 비무장지대 일원의 진균 다양성을 연구하기 위한 일환으로 경기도 연천군 마전리에 있는 산과 황진리에 있는 임진강변에서 11가지 식물체 시료와 토양시료 2개를 채집하여 진균을 분리 동정하였다. 그 결과 총 18속 23종의 진균을 확인하였다. 이 중 Eupenicilliumsaturniforme, Mortierellasclerotiella, M. sossauensis, M. verticillate, Trichoderma hispanicum등 5종의 진균이 국내에 보고가 되어있지 않는 미기록 종이었다. 본 연구에서 이들의 형태적, 분자적 특성을 기술하였다.

A Comparison of the Ability of Fungal Internal Transcribed Spacers and D1/D2 Domain Regions to Accurately Identify Candida glabrata Clinical Isolates Using Sequence Analysis

  • Candida glabrata is the second most prevalent causative agent for candidiasis following C. albicans. The opportunistic yeast, C. glabrata, is able to cause the critical bloodstream infections in hospitalized patients. Conventional identification methods for yeasts are often time consuming and labor intensive. Therefore, recent studies on sequence-based identification have been conducted. Recently, sequencing the D1/D2 domain of the large subunit ribosomal RNA gene and the internal transcribed spacers (ITS) 1 and ITS2 regions of the ribosomal DNA has proven useful for DNA-based identification of most species of fungi. In the present study, therefore, fungal ITS and D1/D2 domain regions were targeted and analyzed by DNA sequencing for the accurate identification of C. glabrata clinical isolates. A total of 102 C. glabrata clinical isolates from various clinical samples including bloodstream, catheterized urine, bile and other body fluids were used in the study. The results of the DNA sequence analysis showed that the mean standard deviation of species identity percent score between ITS and D1/D2 domain regions was $97.8%{\pm}2.9$ and $99.7%{\pm}0.46$, respectively. These results revealed that the D1/D2 domain region might be a better target for identifying C. glabrata clinical isolates based on DNA sequences than the ITS1 and ITS2 regions. However, in order to evaluate the usefulness of D1/D2 domain region for species identification of all Candida species, other Candida species such as C. albicans, C. tropicalis, C. dubliniensis, and C. krusei should be verified in further studies additionally.

경남지역 전통 된장의 숙성기간에 따른 품질 특성 및 미생물 군집 비교 (Quality Characteristics and Comparison of Microbial Community in Traditional Doenjang by Aging Period in Gyeongnam Province)

  • This study investigated the microbial community and quality characteristics of traditional soybean pastes (doenjang) based on the aging period in Gyeongnam province. The samples were collected from seven regions in Gyeongnam Province (Goseong, Hadong, Haman, Hamyang, Sacheon, Sancheong and Uiryeong). Contents of moisture, salinity, pH, and acidity of doenjang after 210 days aging were 56.40~65.21%, 9.05~16.08%, 4.88~6.86 and 0.64~2.14%, respectively. Lightness significantly decreased over the aging period, while the redness tended to increase over the aging period. Yellowness was from 21.39~26.81 to 21.10~28.36. Reducing sugar content was from 0.31~1.45% to 0.11~3.13%. The amino-type content increased from 141.87~495.13 mg/100 g to 328.53~823.67 mg/100 g. In contrast, the ammonia-type content declined from 136.74~364.70 mg/ 100 g to 128.62~331.00 mg/100 g. The overall total microbial count did not significantly differ, but it tended to decrease in sample GD4. Fungus declined overall or was not detected. Coliform groups were not detected in all samples. Lactobacillus and Bacillus cereus tended to decrease over the aging period. The common fungus and bacterium were Aspergillus oryzae and Tetragenococcus halophilus, respectively. After the aging period, the predominant fungi were Candida versatilis, Candida apicola, and Debaryomyces hansenii. The predominant bacteria were Tetragenococcus halophilus and Cronobacter sakazakii.

Cultivation and Nutritional Value of Prominent Pleurotus spp.: An Overview

  • Pleurotus species are commercially essential mushrooms and widely cultivated throughout the world. The production of Pleurotus mushrooms alone accounts for around 25% of that total cultivated mushrooms globally. In America and Europe, Pleurotus species are considered specialty mushrooms, whereas, in Korea, their cultivation is economically profitable, and it is one of the highly consumed species. Pleurotus species are predominantly found in tropical forests and often grow on fallen branches, dead and decaying tree stumps, and wet logs. Biographical studies have shown that the Pleurotus genus is among the more conspicuous fungi that induce wood decay in terrestrial ecosystems worldwide due to its formidable lignin-modifying enzymes, including laccase and versatile peroxidases. Pleurotus species can be grown easily due to their fast colonization nature on diversified agro-substrates and their biological efficiency 100%. Pleurotus mushrooms are rich in proteins, dietary fiber, essential amino acids, carbohydrates, water-soluble vitamins, and minerals. These mushrooms are abundant in functional bioactive molecules, though to influence health. Pleurotus mushrooms are finding unique applications as flavoring, aroma, and excellent preservation quality. Apart from its unique applications, Pleurotus mushrooms have a unique status delicacy with high nutritional and medicinal values. The present review provides an insight into the cultivation of Pleurotus spp. using different agro-waste as growth substances paying attention to their effects on the growth and chemical composition.

노후 주거용 건축물 비구조체 해체 시 환경유해인자 분석 (A Study on the Analysis of Environmental Hazards when Dismantling Non-Structure of Old Residential Buildings)

  • The number of old buildings older than 30 years in Korea continues to increase from 29.9% in 2005 to 38.8% in 2020. Considering the growing urban regeneration projects, urban maintenance projects, the suppression of urban expansion, and the lack of idle land in the city, the dismantling of old buildings is expected to increase further in the future. As major accidents at building dismantling sites continue to occur, related agencies are also strengthening safety management of building dismantling works. While physical safety management such as collapse and fall is strengthened, there is a relative lack of interest in the health of workers at dismantling sites due to environmental hazards. Since relevant laws stipulate that construction waste should be separated and discharged, old buildings need to be considered for environmental hazards such as fine dust, floating bacteria, and floating molds when dismantling. In this study, we intend to find important safety management elements in the management of building dismantling sites, measure environmental factors harmful to dismantling workers, and present basic data for the management of dismantling sites in the future. "Safety management" was the highest priority, followed by "dust," "vibration," "noise," "bacteria," and "smell." The perception of the importance of "physical damage prevention" with workers working on dismantling and managers managing the site came out similar, but the perception of "work efficiency" and "health disorder prevention" through environmental hazard management showed different priorities. In the process of dismantling, floating bacteria and floating mold were collected, cultured, and measured the concentration in the indoor air. The measurement was measured by dividing it into pre-dismantling and during dismantling.

미생물을 활용한 옥수수병의 생물학적 방제 (Biocontrol of Maize Diseases by Microorganisms)

  • Zea mays, known as maize or corn, is a major staple crop and an important source of energy for humans and animals, thus ensuring global food security. Approximately 9.4% of the loss of total annual corn production is caused by pathogens including fungi, bacteria, and viruses, resulting in economic losses. Although the use of fungicides is one of the most common strategies to control corn diseases, the frequent use of fungicides causes various health problems in humans and animals. In order to overcome this problem, an eco-friendly control strategy has recently emerged as an alternative way. One such eco-friendly control strategy is the use of beneficial microorganisms in the control of plant pathogens. The beneficial microorganisms can control the plant pathogens in various ways, such as spatial competition with plant pathogens, inhibition of fungal or bacterial growth via the production of secondary metabolites or antibiotics, and direct attack to plant pathogens via enzyme activity. Here, we reviewed microorganisms as biocontrol agents against corn diseases.

Biological soil crusts impress vegetation patches and fertile islands over an arid pediment, Iran

  • Background: Plant vegetation appears in heterogeneous and patchy forms in arid and semi-arid regions. In these regions, underneath the plant patches and the empty spaces between them are covered by biological soil crusts (moss, lichen, cyanobacteria, and fungi). Biological soil crusts lead to the formation and development of fertile islands in between vegetation patches via nitrogen and carbon fixation and the permeation of runoff water and nutrients in the soil. Results: The present study has investigated the association of biological soil crusts, the development of fertile islands, and the formation of plant patches in part of the Takht-e Soltan protected area, located in Khorasan Razavi Province, Iran. Three sites were randomly selected as the working units and differentiated based on their geomorphological characteristics to the alluvial fan, hillslope, and fluvial terrace landforms. Two-step systematic random sampling was conducted along a 100-meter transect using a 5 m2 plot at a 0-5 cm depth in three repetitions. Fifteen samplings were carried out at each site with a total of 45 samples taken. The results showed that the difference in altitude has a significant relationship with species diversity and decreases with decreasing altitude. Results have revealed that the moisture content of the site, with biocrust has had a considerable increase compared to the other sites, helping to form vegetation patterns and fertile islands. Conclusions: The findings indicated that biological crusts had impacted the allocation of soil parameters. They affect the formation of plant patches by increasing the soil's organic carbon, nitrogen, moisture and nutrient content provide a suitable space for plant growth by increasing the soil fertility in the inter-patch space.

P-hydroxybenzoic acid positively affect the Fusarium oxysporum to stimulate root rot in Panax notoginseng

  • Background: Plant health is directly related to the change in native microbial diversity and changes in soil health have been implicated as one of the main cause of root rot. However, scarce information is present regarding allelopathic relationship of Panax notoginseng root exudates and pathogenic fungi Fusarium oxysporum in a continuous cropping system. Methods: We analyzed P. notoginseng root exudate in the planting soil for three successive years to determine phenolic acid concentration using GC-MS and HPLC followed by effect on the microbial community assembly. Antioxidant enzymes were checked in the roots to confirm possible resistance in P. notoginseng. Results: Total 29 allelochemicals in the planting soil extract was found with highest concentration (10.54 %) of p-hydroxybenzoic acid. The HPLC showing a year-by-year decrease in p-hydroxybenzoic acid content in soil of different planting years, and an increase in population of F. oxysporum. Moreover, community analysis displayed negative correlation with 2.22 mmol. L-1 of p-hydroxybenzoic acid correspond to an 18.1 % population of F. oxysporum. Furthermore, in vitro plate assay indicates that medium dose of p-hydroxybenzoic acid (2.5-5 mmol. L-1) can stimulate the growth of F. oxysporum colonies and the production of macroconidia, as well as cell wall-degrading enzymes. We found that 2-3 mmol. L-1 of p-hydroxybenzoic acid significantly increased the population of F. oxysporum. Conclusion: In conclusion, our study suggested that p-hydroxybenzoic acid have negative effect on the root system and modified the rhizosphere microbiome so that the host plant became more susceptible to root rot disease.

Rumen fermentation and microbial diversity of sheep fed a high-concentrate diet supplemented with hydroethanolic extract of walnut green husks

  • Objective: This study aimed to assess the impact of a hydroethanolic extract of walnut green husks (WGH) on rumen fermentation and the diversity of bacteria, methanogenic archaea, and fungi in sheep fed a high-concentrate diet. Methods: Five healthy small-tailed Han ewes with permanent rumen fistula were selected and housed in individual pens. This study adopted a self-controlled and crossover design with a control period and an experimental period. During the control period, the animals were fed a basal diet (with a ratio of concentrate to roughage of 65:35), while during the treatment period, the animals were fed the basal diet supplemented with 0.5% hydroethanolic extract of WGH. Fermentation parameters, digestive enzyme activities, and microbial diversity in rumen fluid were analyzed. Results: Supplementation of hydroethanolic extract of WGH had no significant effect on feed intake, concentrations of total volatile fatty acids, isovalerate, ammonia nitrogen, and microbial protein (p>0.05). However, the ruminal pH, concentrations of acetate, butyrate and isobutyrate, the ratio of acetate to propionate, protozoa count, and the activities of filter paper cellulase and cellobiase were significantly increased (p<0.05), while concentrations of propionate and valerate were significantly decreased (p<0.05). Moreover, 16S rRNA gene sequencing revealed that the relative abundance of rumen bacteria Christensenellaceae R7 group, Saccharofermentans, and Ruminococcaceae NK4A214 group were significantly increased, while Ruminococcus gauvreauii group, Prevotella 7 were significantly decreased (p<0.05). The relative abundance of the fungus Pseudomonas significantly increased, while Basidiomycota, Fusarium, and Alternaria significantly decreased (p<0.05). However, there was no significant change in the community structure of methanogenic archaea. Conclusion: Supplementation of hydroethanolic extract of WGH to a high-concentrate diet improved the ruminal fermentation, altered the structure of ruminal bacterial and fungal communities, and exhibited beneficial effects in alleviating subacute rumen acidosis of sheep.
