떫은감의 재배지역과 품종에 따른 영양성분 특성 (Characteristics of Nutritional Components in Astringent Persimmons according to Growing Region and Cultivar)

  • 본 연구에서는 떫은감의 대표적인 품종인 대봉감과 반시의 일반성분 및 영양성분을 분석하고 비교하였으며, 대봉감과 반시의 재배지역별 영양성분의 특성을 살펴보았다. 다양한 영양성분은 품종에 따라 다른 함량을 나타냈는데 대봉감 품종은 불용성 식이섬유, 베타카로틴, 과당, 자당, 만니톨, 칼륨, 사과산, 호박산, 총 페놀화합물이 반시 품종에 비해 유의적으로 높은 함량을 나타냈으며, 반대로 반시 품종은 수분 함량, 조단백질 함량, 비타민 C, 칼슘, 망간, 주석산, 총 플라보노이드 함량이 대봉감 품종에 비해 유의적으로 높은 함량을 보였다. 재배지역에 따라 많은 성분들이 유의적인 차이를 나타냈다. 다른 지역에 비해 비교적 높은 함량을 보이는 영양성분은 A지역은 베타카로틴, 솔비톨, 만니톨, 아연, 총 페놀화합물, B지역은 조지방 함량, 리코펜, 자당, 자일리톨, 사과산, C지역은 호박산, D지역은 수분 함량, 망간, 탄닌, E지역은 조단백질 함량, 나트륨, 망간, 칼슘, 주석산, 총 플라보노이드 함량이었다. 이상의 결과로 떫은감은 품종이나 재배지역에 따라 많은 영양성분의 유의적인 차이를 나타낸다는 것을 확인하였으며 이런 차이는 떫은감을 활용한 가공제품 생산 시 고려되어야 할 사항으로 생각된다.

복숭아 품종별 성숙정도에 따른 항산화 및 항염증 효과간의 상관관계 (In vitro correlation and analysis of anti-oxidant and anti-inflammatory activities by fruit ripening of peach cultivars)

  • 본 연구에서는 복숭아의 품종인 미황, 가납암백도 및 천홍을 성숙정도에 따라 미숙과와 성숙과로 분류하여 항산화 및 항염증 효과간의 상관관계 등을 분석함으로서 향후 염증 및 산화스트레스와 관련된 질병의 예방 또는 치료할 수 있는 기능성 소재를 발굴하고자 수행하였다. 총 페놀 및 플라보노이드 함량은 3품종 모두 성숙과에 비해 미숙과에서 함량이 많았고, 미숙과 중에서는 천홍이, 성숙과 중에서는 미황이 유의성 있게 높은 함량을 나타내었다. 항산화 활성은 미숙과 천홍에서 가장 높았고, 성숙과에서는 DPPH 라디칼 소거능 측정에 의해서는 미황이 유의성 있게 높았으나 FRAP과 CUPRAC 측정에서는 3품종간 유의적인 차이가 없었다. RAW 264.7 세포에서는 미황 미숙과 추출물이 약 30%의 NO 생성 억제 효과가 관찰되었고, 가납암백도 미숙과는 약 29%, 천홍 미숙과는 약 20%의 저해활성을 보이지만, 복숭아 성숙과 3품종에서는 NO 생성 억제 효과가 나타나지 않았다. 3품종 모두 성숙과에 비해 미숙과에서 저해 활성이 높게 나타났다. 총 페놀 함량과 플라보노이드 함량에 따른 항산화 활성들 간의 상관관계에서 고도의 정의 상관성을 나타내었으며, 항염증 활성과 항산화 활성, 항염증 활성과 총 페놀 및 플라보노이드 함량은 낮은 상관관계를 나타내었다.

Cordycepin 고함유 동충하초 수용성 추출물의 이화학적 특성과 혈전 용해활성 (Fibrinolytic Activity and Chemical Properties of Cordycepin-Enriched Cordyceps militaris JLM 0636)

  • Cordycepin 고함유 번데기 동충하초(Cordyceps militaris JLM0636, $CM{\alpha}$)와 일반 번데기 동충하초(Cordyceps militaris, CM)의 생리활성 물질 및 영양성분 분석과 혈전 용해 활성을 비교 검토하였다. Total polyphenol compound, flavonoids 및 glutathione 함량은 CM 보다 $CM{\alpha}$에서 높은 함량을 보였다. CM 및 $CM{\alpha}$의 주요 미네랄 구성 성분으로는 K, Ca, Mg, Na이고, 주요 지방산 조성은 linolenic acid, linoleic acid, oleic acid, palmitic acid으로 나타났다. 혈전 용해 활성을 비교 검토한 결과 $CM{\alpha}$이 CM보다 높은 활성을 보였다. 이상의 실험 결과 $CM{\alpha}$의 생리활성물질과 혈전 용해 활성을 이용한 제품 개발에 필요한 기초 자료를 제공하고 유용하게 사용될 수 있을 것으로 사료되어 진다.

Rhus verniciflua Stokes Extract and Its Flavonoids Protect PC-12 Cells against H2O2-Induced Cytotoxicity

  • Rhus verniciflua Stokes (RVS), an herbal medicine found in East Asia, was extracted and further fractionated to investigate its antioxidant capacity and neuroprotective effects. The RVS ethyl acetate (EtOAc) fraction had the highest level of total phenolics and antioxidant capacity among all solvent fractions tested. Pretreatment of PC-12 cells with the EtOAc fraction effectively attenuated $H_2O_2$-induced oxidative damage. Furthermore, the EtOAc fraction significantly attenuated caspase-3 activity, resulting in inhibition of $H_2O_2$-induced apoptosis. We identified and quantified fustin, sulfuretin, and butein in the EtOAc fraction using accurate mass quadrupole time-of-flight mass spectrometry and reversed-phase high-performance liquid chromatography. The intracellular antioxidant capacity and superoxide dismutase (SOD) activity were significantly increased in PC-12 cells treated with the EtOAc fraction and with individual flavonoids. When cells were pretreated with the EtOAc fraction or individual flavonoids and then co-incubated with diethyldithiocarbamic acid (an inhibitor of SOD activity), cell viability against $H_2O_2$-induced oxidative stress was attenuated. These results suggest that the RVS EtOAc fraction and its flavonoid constituents protect PC-12 cells against $H_2O_2$-induced neurotoxicity through their antioxidant properties.

진달래꽃으로부터 분리된 플라보노이드 화합물의 항산화성에 관한 연구 (Antioxidative Activity of Flavonoids Isolated from Jindalrae Flowers (Rhododendron mucronulatum Turcz.))

  • 우리나라에서 식용으로 이용되는 진달래꽃으로부터 7성분의 항산화성이 있는 플라보노이드 화합물이 분리 동정되었다. 이들 화합물의 구조는 IR, UV, FAB-MS, $^1H\;NMR$$^{13}C\;NMR$에 의해서 얻어진 분광학적인 결과에 근거하여 afzelin, ampelopsin, catechin, myricetin, myricitrin, quercetin 및 quercitrin인 것으로 밝혀졌다. 이들 화합물은 2개의 flavonol, 3개의 flavonol glycoside, 1개의 flavane 및 1개의 dihydroflavonol로 이루어졌다. 에틸에테르 및 초산에틸 구분에 존재하는 flavonol glycoside (14.4 g)는 polyamide C-200 관 크로마토피법, 분취용 박층크로마토그래피법, 재결정화법, sephadex LH-20 관 크로마토그래피법을 통해서 양쪽 구분으로부터 최종적으로 회수된 총 flavonoid량 (17.6g의 82%에 달하였다. 항산화성은 티오시안산철의 존재하에서 리놀레산의 에타놀용액 중에서 측정되었다. 항산화 효능성은 afzelin<$\alpha-tocopherol$

Inhibitory Effects of Naringenin, Kaempherol, and Apigenin on Cholesterol Biosynthesis in HepG2 and MCF-7 Cells

  • The inhibitory effects of naringenin, kaempherol, and apigenin on the production of cholesterol in HepG2 KCLB 88065 and MCF-7 KCLB 30022 cells were evaluated. In this study, quercetin was used as a reference reagent. After incubation for 3 days, fat-soluble contents of both cell types were extracted by using the Folch method and the cholesterol contents in both cultured cells were determined by high performance liquid chromatography. The concentration of cholesterol in untreated each tissue cells was $12.2{\pm}0.11$ and $8.83{\pm}0.12\;mg/g$ of lipid, respectively. The total concentration of each flavonoid was adjusted to 0, 35, or $350{\mu}M$ in the culture broth. As the results, the addition of 2% methanol and dimethyl sulfoxide (DMSO) to the media (control for flavonoid solvents) did not significantly affect cell growth; however, DMSO caused an increase in the production of cholesterol. Each flavonoid inhibited the production of cholesterol in both HepG2 and MCF-7 cells at the concentration of $35{\mu}M$ above. In addition, the inhibitory effect of kaempherol on the production of cholesterol in these cells was greater than the other flavonoids tested and HepG2 cells are more sensitive to flavonoids than MCF-7. From the results, the inhibitory effects of flavonoids on cholesterol production are different depending on the cell type.

생육시기에 따른 양마 잎의 항산화 활성 (Antioxdant Activity of Hibiscus cannabinus L. Leaves in Different Growth Time)

  • The results on the useful functional compounds of kenaf (Hibiscus cannabinus L.) leaves cultivated in reclaimed lands and the biological activity effects of extracts were as follows. On 98 days after sowing (DAS) Tainung-2 showed the highest total chlorophyll content (1.68 mg/g), and on 141 DAS Dowling showed higher chlorophyll content (1.50 mg/g) than the other two did. In all cultivars the total chlorophyll content was higher on 141 DAS than on 98 DAS. Total polyphenol and total flavonoid contents were the highest in Tainung-2 (30.50 mg/g and 57.03 mg/g, respectively), and total polyphenol and total flavonoid contents (30.50 mg/g and 57.03 mg/g, respectively) were the highest in 30% ethanol extraction. Ascorbic acid contents were higher on 141 DAS than on 98 DAS in three cultivars. SOD activities of kenaf leaf extract were generally over 90%. DPPH radical scavenging activity of Tainung-2 was higher than others.

쉽싸리(택란) 괴경 열수 추출물의 항산화특성 (Antioxidant Properties of Hot Water Extract of Lycopus lucidus Trucz Tubers)

  • This study was designed to investigate effective preprocess methods for the antioxidant activity of Lycopus lucidus Trucz tubers extract. Lycopus lucidus Trucz tubers (LlTT) were treated with D (Dried), SD (Steamed and Dried), ASD (Alcohol-Steamed and Dried), DR (Dried and Roasted), SDR (Steamed, Dried and Roasted), and ASDR (Alcohol-Steamed, Dried and Roasted). Solid content was high in Lycopus lucidus Trucs tubers, which were roasted after drying (especially SDR, ASDR). Total sugar and reducing sugar contents were high in roasted ones. Total polyphenol and flavonoid contents were high upon alcohol-steaming in both the dried and roasted groups. Antioxidant activities of LlTT extracts showed effective nitrate scavenging ability, ABTs radical scavenging ability, Xanthine oxidase inhibitory activity, and DPPH radical scavenging ability. As a result, roasting or steaming process (especially using alcohol) can be helpful to obtain the highly antioxidative extract of Lycopus lucidus Trucs tubers.

연잎분말을 첨가한 된장소스의 이화학적 품질 특성 (Physicochemical Quality Characteristics of Fermented Soybean Paste Sauce added Lotus Leaf Powder)

  • The purpose of the study is to examine the physicochemical and quality characteristics of fermented soybean paste sauce containing lotus leaf powder at concentrations of 0% (control), 2%, 4%, 6% and 8%. The color of increasing the amount of concentrated lotus leaf powder in the fermented soybean paste sauce tended to increase the pH, total soluble solids, saltinity and viscosity. It also tended to increase total polyphenol, flavonoids content and DPPH radical scavenging ability. The total bacteria numbers increased with storage period, and those of fermented soybean paste sauce with 4% lotus leaf powder were lower compared to the other samples. The results of this study suggest that lotus leaf is a beneficial ingredient for increasing quality characteristics and the functionality of fermented soybean paste sauce.

Phytochemical constituent, percentage yield and phenolic content estimation of different solvent system of Carica papaya leaves

  • Carica papaya is an important medicinal plant used in the management of different disease conditions. Phytochemical screening was carried out using different chemical test, Percentage yield and total phenolic content was evaluated using Folin Ciocalteu method in different solvent system; methanol, ethanol, ethyl acetate, n-butanol and n-hexane respectively. The phytochemical screening of the studies showed the presence of flavonoids, saponins, tannins, terpenoids, glycosides, steroids, carbonhydrate, anthraquinone and alkaloids. The percentage yield of crude extract and total polyphenol content was high in methanol, ethanol and ethyl acetate when compared with n-butanol and n-hexane. The studies show that Carica papaya leave extracts is a potent source of secondary metabolites. The use of the plant in the management of diseases is justified.
