A Multiplex PCR Method for the Detection of Genetically Modified Alfalfa (Medicago sativa L.) and Analysis of Feral Alfalfa in South Korea

  • Methods for detecting the presence of genetically modified (GM) crops are evolving to comply with legislation and to enhance monitoring by biotechnology companies and regulators. In order to cover a broad range of detection methods for a new GM crop, conventional multiplex PCR methods are required. Based on the genetic information on three GM alfalfa varieties (J101, J163, and KK179), which were recently approved in South Korea, we developed a fast, reliable, and highly specific multiplex polymerase chain reaction (PCR) method with basic PCR equipment and inexpensive reagents. To validate and verify the newly developed multiplex PCR method, we applied a limit of detection assay and random reference material analysis. We also monitored the unintentional environmental release of GM alfalfa in South Korea by performing the multiplex PCR analysis with 91 feral alfalfa specimens collected from 2000 to 2018. Our methodology is a sensitive, simple, quick, and inexpensive tool for detecting and identifying three GM alfalfa varieties.

  • This paper aims to develop a conceptual framework for Green Supply Chain Management (GSCM) that takes into account the effect of GSCM drivers on implementing GSCM practices in Vietnam FDI companies. This study has considered organizational commitment, social network, and government support as GSCM driver factors and proposed a structural model of the relationships between GSCM drivers and GSCM practices in Vietnam FDI companies. The empirical analysis used data from 192 questionnaires which used a comprehensive, valid, and reliable tool (SPSS 26 and SmartPLS 3.0 software) to evaluate rigorous statistical tests including convergence validity, discriminatory validity, reliability, and Average Variance Extracted (AVE) to analyze and verify the gathered data and develop the hypothesis. The result of path analysis shows that GSCM driver factors constitute a structured system with different degrees of influence on GSCM drivers and GSCM practices. Organizational commitment and government support has a positive relationship with both GSCM drivers and GSCM practices, while social network only has a positive relationship on GSCM drivers. As a result, the testing of the relationship between GSCM drivers and GSCM practices has been verified and supported. The findings of this study can help managers and decision-makers to push the implementation of GSCM practices in FDI companies.

  • Game theory is a powerful tool for analyzing the Supply chain (SC) with different conflicting elements. Among them, the Stackelberg game is the one in which a player as leader has more power than the other ones as followers. Since in many SC systems one element has, in essence, more power than the others; the Stackelberg game has found many applications in SC studies. In this paper, we apply the Stackelberg game-theoretic approach and the corresponding equilibrium point to formulate and analyze a two echelon VMI SC. Comprehensive computational results on an experimental case are conducted to numerically analyze the performance of VMI system against three groups of critical parameters. Moreover, a critical comparison demonstrates the poorer performance of decentralized VMI system than centralized one. This naturally necessitates designing proper contracts between VMI partners in order to more effectively implement the realistic decentralized system.

  • Polymerase chain reaction is widely used as a powerful tool in modern molecular biology. As there is agreement that the HPV is an important factor in the head and neck cancers, the detection of HPV DNA sequence in the head and neck cancer tissue has been tried in several ways. We used the PCR to detect the E1 open reading frames of the HPV in paraffin-embedded tissue of the patients with the head neck cancers. Eleven of the fifty-four tested samples (30%) showed positive result. We have analysed the clinical courses and characteristics related with Human Papillomavirus in those patients.

Multiplex PCR Detection of 4 Events of Genetically Modified Soybeans (RRS, A2704-12, DP356043-5, and MON89788)

  • A multiplex polymerase chain reaction (PCR) method was developed for the detection of 4 events of genetically modified (GM) soybean. The event-specific primers were designed from 4 events of GM soybean (RRS, A2704-12, DP356043-5, and MON89788). The lectin was used as an endogenous reference gene of soybean in the PCR detection. The primer pair YjLec-4-F/R producing 100 bp amplicon was used to amplify the lectin gene and no amplified product was observed in any of the 9 different plants used as templates. This multiplex PCR method allowed for the detection of event-specific targets in a genomic DNA mixture of up to 1% GM soybean mixture containing RRS, A2704-12, DP356043-5, and MON89788. In this study, 20 soybean products obtained from commercial food markets were analyzed by the multiplex PCR. As a result, 6 samples contained RRS. These results indicate that this multiplex PCR method could be a useful tool for monitoring GM soybean.

  • Manufacturing companies are implementing a so-called customer-centered supply chain management to have a competitive advantage. In these efforts, collaboration not only within a company but also with suppliers, partners and customers is emphasized. The fast delivery, reducing the total lead-time from development to delivery, is pursued more than ever, though the quality and cost are still importantly regarded. Die manufacturing companies are not exception from these trends, because a die is a necessary tool for almost manufacturing industries. The planning and scheduling system plays an important role in supply chain management. In this study, we address a scheduling problem of a die manufacturing company. The problem is very complex due to many reasons including the uncertainty in environment and the complexity of constraints. Considering the importance of due-date satisfaction and human planners' roles, we designed the solution algorithm and the user interface respectively. In the implementation phase, modeling constructs and basic solution algorithms of ILOG solver and ILOG scheduler are used. In the paper, the problem and the algorithm are described with ILOG constructs, and the experience of use is also addressed.

  • PCR(Polymerase chain reaction) is a technique to replicate and amplify a desired part of DNA. It is used in various aspects such as DNA fingerprint analysis and rare DNA amplification of an extinct animal. Especially in the medical diagnosis field, it provides various measurement methods at the molecular level such as genetic diagnosis, and is a basic tool for molecular diagnostics. The internal shape of the plastic vial tube for PCR analysis used in the DNA detection process, and the surface roughness and internal cleanliness can affect detection and discrimination results. The plastic vial tube demanded by the developer of the PCR analysis equipment should be changed to a structure that eliminates the residual washing solution in the washing process to ensure the internal cleanliness. Thus the internal structure and the internal surface design for improving the PCR amplification efficiency are key issues to develop the plastic vial tube for the DNA detection process.

4-CMTB Ameliorates Ovalbumin-Induced Allergic Asthma through FFA2 Activation in Mice

  • Free fatty acid receptor 2 (FFA2, also known as GPR43), a G-protein-coupled receptor, has been known to recognize short-chain fatty acids and regulate inflammatory responses. FFA2 gene deficiency exacerbated disease states in several models of inflammatory conditions including asthma. However, in vivo efficacy of FFA2 agonists has not been tested in allergic asthma. Thus, we investigated effect of 4-chloro-α-(1-methylethyl)-N-2-thiazoylylbenzeneacetanilide (4-CMTB), a FFA2 agonist, on antigen-induced degranulation in RBL-2H3 cells and ovalbumin-induced allergic asthma in BALB/c mice. Treatment of 4-CMTB inhibited the antigen-induced degranulation concentration-dependently. Administration of 4-CMTB decreased the immune cell numbers in the bronchoalveolar lavage fluid and suppressed the expression of inflammatory Th2 cytokines (IL-4, IL-5, and IL-13) in the lung tissues. Histological studies revealed that 4-CMTB suppressed mucin production and inflammation in the lungs. Thus, results proved that FFA2 functions to suppress allergic asthma, suggesting 4-CMTB activation of FFA2 as a therapeutic tool for allergic asthma.

The Effects of Brand Communication of Chain Hotel Group on Brand Awareness, Brand Attitude, and Brand Loyalty (체인호텔 기업의 브랜드 커뮤니케이션이 브랜드 인지, 태도, 그리고 충성도에 미치는 영향)

  • Purpose: Brand communication plays an important role in the credibility of consumer behavior as it enhances brand equity. This study investigates the effects of brand communication (firm-created communication, consumer-generated communication) on brand awareness, brand attitude, brand loyalty in the hotel business sector by applying the SOR theory (stimulus-organism-response theory). Research design, data, and methodology: This study was analyzed in a quantitative way using the survey results of 400 customers who had experience of visiting hotels. In this study, SmartPLS 4.0 was used to evaluate the research model. The reliability, convergent validity, and discriminant validity of the measurement tool were verified. Result: Result was found that consumer-generated communications had a positive effect on brand awareness and brand attitude, whereas firm-created communications had a significant effect on brand awareness. In addition, brand awareness had a positive effect on both brand attitude and brand loyalty. Finally, brand attitude was found to have a positive effect on brand loyalty. Conclusions: This study redefines the concept of where chain hotel groups should focus when providing consumers with information about their brands and services. As a result, the conceptual framework of brand communication to increase new customer visits to the hotel brand has been expanded.

Slipchip Device Development in Molecular Diagnostics

  • Slipchip offers advantages such as high-throughout, low cost, and simple operation, and therefore, it is one of the technologies with the greatest potential for high-throughput, single-cell, and single-molecule analyses. Slipchip devices have achieved remarkable advances over the past decades, with its simplified molecular diagnostics gaining particular attention, especially during the COVID-19 pandemic and in various infectious diseases scenarios. Medical testing based on nucleic acid amplification in the Slipchip has become a promising alternative simple and rapid diagnostic tool in field situations. Herein, we present a comprehensive review of Slipchip device advances in molecular diagnostics, highlighting its use in digital recombinase polymerase amplification (RPA), loop-mediated isothermal amplification (LAMP), and polymerase chain reaction (PCR). Slipchip technology allows users to conduct reliable droplet transfers with high-throughput potential for single-cell and molecule analyses. This review explores the device's versatility in miniaturized and rapid molecular diagnostics. A complete Slipchip device can be operated without special equipment or skilled handling, and provides high-throughput results in minimum settings. This review focuses on recent developments and Slipchip device challenges that need to be addressed for further advancements in microfluidics technology.
