Alternaria in Food: Ecophysiology, Mycotoxin Production and Toxicology

  • Alternaria species are common saprophytes or pathogens of a wide range of plants pre- and post-harvest. This review considers the relative importance of Alternaria species, their ecology, competitiveness, production of mycotoxins and the prevalence of the predominant mycotoxins in different food products. The available toxicity data on these toxins and the potential future impacts of Alternaria species and their toxicity in food products pre- and post-harvest are discussed. The growth of Alternaria species is influenced by interacting abiotic factors, especially water activity ($a_w$), temperature and pH. The boundary conditions which allow growth and toxin production have been identified in relation to different matrices including cereal grain, sorghum, cottonseed, tomato, and soya beans. The competitiveness of Alternaria species is related to their water stress tolerance, hydrolytic enzyme production and ability to produce mycotoxins. The relationship between A. tenuissima and other phyllosphere fungi has been examined and the relative competitiveness determined using both an Index of Dominance ($I_D$) and the Niche Overlap Index (NOI) based on carbon-utilisation patterns. The toxicology of some of the Alternaria mycotoxins have been studied; however, some data are still lacking. The isolation of Alternaria toxins in different food products including processed products is reviewed. The future implications of Alternaria colonization/infection and the role of their mycotoxins in food production chains pre- and post-harvest are discussed.

Mannitol Amendment as a Carbon Source in a Bean-based Formulation Enhances Biocontrol Efficacy of a 2,4-diacetylphloroglucinol-producing Pseudomonas sp. NJ134 Against Tomato Fusarium Wilt

  • Fusarium wilt caused by Fusarium oxysporum has become a serious problem world-wide and relies heavily on chemical fungicides. We selected Pseudomonas sp. NJ134 to develop an effective biocontrol strategy. This strain shows strong antagonistic activity against F. oxysporum. Biochemical analyses of ethyl-acetate extracts of NJ134 culture filtrates showed that 2,4-diacetylphloroglucinol (DAPG) was the major compound inhibiting in vitro growth of F. oxysporum. DAPG production was greatly enhanced in the NJ134 strain by adding mannitol to the growth media, and in vitro antagonistic activity against F. oxysporum increased. Bioformulations developed from growth of NJ134 in sterile bean media with mannitol as the carbon source under plastic bags resulted in effective biocontrol efficacy against Fusarium wilt. The efficacy of the bioformulated product depended on the carbon source and dose. Mannitol amendment in the bean-based formulation showed strong effective biocontrol against tomato Fusarium wilt through increased DAPG levels and a higher cell density compared to that in a glucose-amended formulation. These results suggest that this bioformulated product could be a new effective biocontrol system to control Fusarium wilt in the field.

Comparison on Irrigation Management Methods by Integrated Solar Radiation and Drainage Level Sensor in Rockwool and Coir Bag Culture for Tomato (토마토의 암면과 코이어 자루재배시 일사량제어법과 배액전극제어법에 의한 급액제어 방법 비교)

  • Irrigation management methods controlled by integrated solar radiation (ISR) or drainage level sensor were evaluated in rockwool or coir bag culture as tomato (Solanum lycopersicum L.) production system. Substrate water content and drainage percentage were more stable in the drainage level sensor method than in the ISR method regardless of substrate type. Total yield and marketable yield were high in the drainage level sensor method, but not between substrates in the same irrigation management method. Sugar content was affected more by the substrate type than irrigation method. The drainage level sensor method was elucidated to be better than the ISR method regardless of substrate type.

  • During survey of postharvest diseases of vegetables in the middle region of Korea in 2003, 2004 and 2005, new disease symptoms showing watery rot and soft rot were observed. In this study, the disease causal agents were identified as Geotrichum candidum, and their host range and pathogenicity were investigated. G. candidum isolated had wide host range and strong pathogenicity against carrot, cucumber, tomato and pumpkin. The disease occurrence on several vegetables that G. candidum can be a serious threat to stable production of fresh vegetable.

Identification and Characterization of Novel Biocontrol Bacterial Strains

  • Because bacterial isolates from only a few genera have been developed commercially as biopesticides, discovery and characterization of novel bacterial strains will be a key to market expansion. Our previous screen using plant bioassays identified 24 novel biocontrol isolates representing 12 different genera. In this study, we characterized the 3 isolates showing the best biocontrol activities. The isolates were Pantoea dispersa WCU35, Proteus myxofaciens WCU244, and Exiguobacterium acetylicum WCU292 based on 16S rRNA sequence analysis. The isolates showed differential production of extracellular enzymes, antimicrobial activity against various fungal or bacterial plant pathogens, and induced systemic resistance activity against tomato gray mold disease caused by Botrytis cinerea. E. acetylicum WCU292 lacked strong in vitro antimicrobial activity against plant pathogens, but induced systemic resistance against tomato gray mold disease. These results confirm that the trait of biological control is found in a wide variety of bacterial genera.

Antifungal Activity in Cell-Free Culture Fluid of Pseudomons solanacearum Strains Collected from Severe Provinces in the North of Vietnam.

  • A research collection of Pseudomons solanacearum bacteria, a pathogen causing ‘bacteria wilt’ disease of more than 265 plant species, represented for northern provinces of Vietnam has recently been established and was saved for examination of antifungal activity in their culture fluids. All strains used in this work have been isolated from infected tomato, potato, and groundnut collected from production fields and they express different levels of virulence on their host plants. Cell-free culture fluids of these strains were tested for antifungal activity (to inhibit growth of mycelium and to destroy germination tube of fungal spores) on a number of fungi that either infect or associate with vegetable crops of Solanaceae family (tomato, potato, pepers...), fruit plants (banana), and even well-known by Vietnamese traditional medicine herbal plants belonging to Trifoliatus, Schefflera, Homalomena and Panax genera (Araliaceae family) of which roots are used as a resource of the herbal material. The antifungal activity was found in nearly all strains tested. Result of study on chitin, CMC, tween 80 and casein degradation abilities of the latter suggested that antifungal activity of positively-found strains may be due to their ability of extracelluar chitinase's excretion that destroy fungal cell wall.

Database Design for IoT-based Greenhouse Systems

  • Since 2000s, proper utilization of IoT (Internet of Things) technology is a key factor for a firm to become more competitive, and this stream is not exceptional for the food and agriculture industry. Along with this stream, Korea government organization, for example MAFRA (Ministry of Agriculture, Food and Rural Affairs), elected to adopt IoT technology, such as USN and RFID technologies, in the food and agriculture industry. Based on the IoT technology, MAFARA launched six "IoT based farm" project in 2007. IoT based farm project includes IoT based greenhouse system project, and it shows drastic efficiency in terms of cost reduction. When it comes to the productivity, however, the effect of IoT based greenhouse system is still ambiguous. In this regard, this study conducted systems analysis and design for IoT based tomato greenhouse in order to help farmers' decision making related to the productivity by establishing standardized database structure and designing output form to analyze productivity indices. Proposed systems analysis and design can be utilized as a data analysis tools by farmers. Productivity data from the proposed systems is can be used by researchers to identify the relationship among environment, plant growth and productivity. Policy makers also can refer to the data and output forms to predict the quantity of fruit during certain period and to revise production guideline more precisely.

Deep Learning based Rapid Diagnosis System for Identifying Tomato Nutrition Disorders

  • Nutritional disorders are one of the most common diseases of crops and they often result in significant loss of agricultural output. Moreover, the imbalance of nutrition element not only affects plant phenotype but also threaten to the health of consumers when the concentrations above the certain threshold. A number of disease identification systems have been proposed in recent years. Either the time consuming or accuracy is difficult to meet current production management requirements. Moreover, most of the systems are hard to be extended, only detect a few kinds of common diseases with great difference. In view of the limitation of current approaches, this paper studies the effects of different trace elements on crops and establishes identification system. Specifically, we analysis and acquire eleven types of tomato nutritional disorders images. After that, we explore training and prediction effects and significances of super resolution of identification model. Then, we use pre-trained enhanced deep super-resolution network (EDSR) model to pre-processing dataset. Finally, we design and implement of diagnosis system based on deep learning. And the final results show that the average accuracy is 81.11% and the predicted time less than 0.01 second. Compared to existing methods, our solution achieves a high accuracy with much less consuming time. At the same time, the diagnosis system has good performance in expansibility and portability.

Ethylene-Induced Auxin Sensitivity Changes in Petiole Epinasty of Tomato Mutant dgt

  • The tomato (Lycopersicon esculentum Mill.) mutant diageotropica (dgt) lacking normal gravitropic response is known to be less sensitive to auxin compared with its isogenic parent VFN8. Straight growth as well as ethylene production in response to added auxin in hypocotyl segments of dgt was negligible. However, there was no significant difference between the two genotypes in auxin transport in petiole segments and its inhibition by the phytotropin N-1-naphthylphthalamic acid(NPA). Kinetic parameters of NPA binding to microsomal membranes were also non-distinguishable between the two. Its petiolar explants treated with ethylene developed epinastic curvature with the magnitude of response increased about 3 folds over non-mutant wild type. Ethylene-induced epinasty in both dgt and VFN8 was nullified by treatment of explants with the ethylene autagonist 2,5-norbonadiene. Lateral transport of 3H-IAA toward the upper side of ethylene-treated petioles in dgt, however, was not significantly more pronounced than in VFN8, the implications being that auxin sensitivity in the mutant was restored, or even rised above the wild type, by ethylene.

Screening of yeast for alcoholic fermentation of no sugar-added tomato concentrate (토마토 농축액을 이용한 무가당 알코올 발효를 위한 효모의 선발)

  • The purpose of this study was to screen desirable yeast strains for alcoholic fermentation of tomato paste without sugar addition. The moisture, crude protein, crude fat, ash, and soluble nitrogen contents of the tomato paste ($25^{\circ}Brix$) were found to be 67.33%, 1.90%, 0.03%, 0.02%, and 30.72%, respectively. Free sugars found in the paste were fructose and glucose. Most abundant free amino acids of the paste were glutamic, aspartic, and ${\gamma}$-aminobutyric acids. Total seven yeast strains (Saccharomyces cerevisiae KDH (TWA), S. cerevisiae Lalvin ICVD-47 (TWB), S. cerevisiae Lalvin RC-212 (TWC), S. cerevisiae Lalvin K1-V1116 (TWD), S. bayanus Lalvin EC-1118 (TWE), S. cerevisiae Enoferm (TWF), and S. cerevisiae DJ97(KCTC8842P) (TWG)) were tasted for alcohol fermentation of the tomato paste. The highest alcohol content (8.2%) and the lowest residual sugar content ($13.25^{\circ}Brix$) were observed in the tomato paste fermented using the S. cerevisiae Lalvin ICVD-47 strain (TWB) after 3 day and 4 day of fermentation, respectively. Sugar and reducing sugar contents, and pH of the tomato paste were not remarkably affected by the difference in yeast strains used, showing $13.25{\sim}13.45^{\circ}Brix$, 28.37~28.48 mg/mL, and 4.43~4.54, respectively, after 4 day of fermentation. Color and total acid content were significantly affected by the types of yeast strains and fermentation time, but the numerical changes were negligible. These results indicate that TWB would be the suitable strain for alcoholic fermentatiom of tomato paste based on its highest alcohol production and the lowest residual sugar content produced during fermentation.
