N-terminal GNBP homology domain of Gram-negative binding protein 3 functions as a beta-1,3-glucan binding motif in Tenebrio molitor

  • The Toll signalling pathway in invertebrates is responsible for defense against Gram-positive bacteria and fungi, leading to the expression of antimicrobial peptides via NF-$\kappa$B-like transcription factors. Gram-negative binding protein 3 (GNBP3) detects beta-1,3-glucan, a fungal cell wall component, and activates a three step serine protease cascade for activation of the Toll signalling pathway. Here, we showed that the recombinant N-terminal domain of Tenebrio molitor GNBP3 bound to beta-1,3-glucan, but did not activate down-stream serine protease cascade in vitro. Reversely, the N-terminal domain blocked GNBP3-mediated serine protease cascade activation in vitro and also inhibited beta-1,3-glucan-mediated antimicrobial peptide induction in Tenebrio molitor larvae. These results suggest that the N-terminal GNBP homology domain of GNBP3 functions as a beta-1,3-glucan binding domain and the C-terminal domain of GNBP3 may be required for the recruitment of immediate down-stream serine protease zymogen during Toll signalling pathway activation.

A Study on The Demand Management for Determination of Freeway Toll System (고속도로 통행요금체계 결정을 위한 수요관리방안 연구)

  • Statistics shows that 80% of freeway users travel less than 50km, and only 8% of them do over 100km. It means that the freeway is used for commuting. As a result, the freeway is not used efficiently and social cost is high. The current toll system aims for the efficient usage of the freeway by restraining short-distance trips and inducing long-distance ones. Thus, policies such as minimum toll system and discount for long-distance trips have been carried out. However. these two policies take no account of demand management between the freeway and alternative roads. They merely consider the compensation for charge on users, so the assessment of discount rate and distinction of distance are not reasonable. Consequently, not only the effective demand management but also the evaluation of those policies has been difficult to be achieved. The objective of this paper is to analyze those problematic policies and to establish a reasonable and improved toll system. In addition, this study presents the methodology to minimize social cost, which can be achieved by reducing short trips and encouraging long ones on the freeway. A new methodology of freeway toll system is applied and the results are presented.

Optimum Speed Simulation for Electronic Toll Collection (Electronic Toll Collection 운영속도 시뮬레이션)

  • This study is to analyze the impacts of different High-Pass lane speed limits. Paramics, microscopic simulation software, was used to perform microscopic simulations. For the simulations, Incheon toll gate traffic count data was used. The Paramics simulation results show that higher High-Pass speed limit (70Km/h) produced better traffic flow condition and less air pollution cost compared to existing High-Pass speed limit (30Km/h).

The Anti-Inflammatory Effects of Phytochemicals by the Modulation of Innate Immunity

  • Toll-like receptors (TLRs) induce innate immune responses that are essential for host defense against invading microbial pathogens. In general, TLRs have two major downstream signaling pathways; myeloid differential factor 88 (MyD88) and Toll/IL-1R domain-containing adaptor inducing IFN-${\beta}$ (TRIF) leading to the activation of NF-${\kappa}B$ and IRF3. Numerous studies demonstrated that certain phytochemicals possessing anti-inflammatory effects inhibit NF-${\kappa}B$ activation induced by pro-inflammatory stimuli including lipopolysaccharide and tumor necrosis factor-${\alpha}$ ($TNF{\alpha}$). However, the direct molecular targets for such anti-inflammatory phytochemicals are not fully identified. In this paper, we will discuss about the molecular targets of phytochemicals in TLRs signaling pathways. These results present a novel anti-inflammatory mechanism of phytochemicals in TLRs signaling.

  • The extreme weather events have increased around the world this century. One of the main reasons of frequent occurrence is the change of atmospheric circulation by El nino. also Korea Peninsular is not exception. The 97 % of death toll and 89 % of property loss of total are related with extreme-weather events for the last 10 years. for example the heavy rainfall (1998-4999) and Typhoon Rusa and Mamie. In spite of the percent of death toll by extreme-weather disaster is increasing and the total population is growing. but the number of death toll from natural disasters is decreasing. It shows that the loss of property and life can be minimize by preparing the proper disaster prevention measures. There are several preparations to reduce the damage by extreme-weather events: Public facilities have overall check up, to recognize the weather alert, the awareness of the escape route and the art of measures

Benefits of Using Imperfect Information in Controlling an M/M/1 Queueing System

  • In this paper, we analyze an M / M / 1 queueing system where there are incentive conflicts among customers. Self-interested customers' decisions whether to join the system or not may not necessarily induce a socially optimal congestion level. As a way to alleviate the over-congestion, toll imposition was used in Naor's paper [3]. Instead of using a toll mechanism, we study the usefulness of imperfect information on system state (queue size, for example) as a way to reduce the over-congestion by self-interested customers. The main conclusion of this paper is that by purposefully giving fuzzy or imperfect information on the current queue size we can improve the congestion in the system. This result might look contradictory to rough intuition since perfect information should give better performance than imperfect information. We show how this idea is verified. In deriving this result, we use the concept of Nash equilibrium (pure and mixed strategy) as introduced in game theory. In some real situations, using imperfect information is easier to apply than imposing a toll, and thus the result of this paper has practical implications.

  • 현재 사용되고 있는 ETCS(Electronic Toll Collection Services)는 충전식의 스마트 카드와 카드리더를 이용하여 필요한 정보를 읽음으로 인증 및 과금 결제를 처리하는 방식이다. 혀 방식은 스마트 카드 규격이 다양하여 사용자는 각 규격에 맞는 카드리더를 사용해야 한다. 게다가 충전식이기 때문에 매번 충전해야하는 불편함이 있다. 본 논문에서는 현 ETC(Electronic Toll Collection)시스템의 단점을 극복하고자 스마트 카드 대신 휴대폰과 WAP(Wireless Application Protocol) 서버를 사용하여 통행료 자동징수 시스템을 설계한다. 또한 휴대폰을 사용함으로써 ITS(Intelligent Transfer Service) 시스템과 별도로 지역적인 인증이 가능하여 인중절차를 간소화시킬 수 있으며, ITS 시스템의 부하를 줄일 수 있기 때문에 ETC시스템 처리속도 또한 증가시킬 수 있다.

Ligand Recognition by the Toll-like Receptor Family

  • Toll-like receptor (TLR) family proteins, type I transmembrane proteins, play a central role in human innate immune response by recognizing common structural patterns in diverse molecules from bacteria, viruses and fungi. Recently four structures of the TLR and ligand complexes have been determined by high resolution x-ray crystallographic technique. In this review we summarize reported structures of TLRs and their proposed activation mechanisms. The structures demonstrate that binding of agonistic ligands to the extracellular domains of TLRs induces homo- or heterodimerization of the receptors. Dimerization of the TLR extracellular domains brings their two C-termini into close proximity. This suggests a plausible mechanism of TLR activation: ligand induces dimerization of the extracellular domains, which enforces juxtaposition of intracellular signaling domains for recruitment of intracellular adaptor proteins for signal initiation.

Study on Computation of Optimal Tolls When Externalities Exist (외부비용을 포함한 적정통행료 산정 수단에 관한 연구)

  • It is well known that market transactions do not lead to social optima when externalities exist. Given that previous studies such as RICARDO-AEA(2014) have identified various types of external costs, we must take their magnitudes, or externalities in general, into account in order to make toll prices to achieve social optimum. Little has been done on estimation of externalities in road uses in Korea, to the best of our knowledge. We suggested to use the contingent valuation method (CVM) to estimate overall social benefits and applied it to estimation of benefits of road kill prevention as a pilot study. Our empirical model has considered heteroskedasticity explicitly and its estimation result was that individual drivers were willing to pay 147 KRW on average in addition to current toll prices for prevention of road kills. We provided general discussions of externalities in road use and various internalization measures.

  • Industrialization and urbanization occurred-traffic delay, traffic accident and parking problem etc.-variety of transportation problems. Parking problem is one of the transportation problem then transportation engineers would get involved in trying to solve parking problem. A way to overcome a variety of parking problems needs preliminary data for conditions of parking but it lacks the preliminary data to do so. Thus this study analyzed a characteristic of a parking through a field survey for parking in Daegu Metropolitan City. This study analyzed parking characteristic of parking operation method. This study result reveals difference between a toll parking and a no charge parking characteristic. Thus a close examination for a toll parking characteristic and a no charge parking characteristic is a prerequisite for a rational parking policy.
