구증구포 맥문동 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl acetate Fraction of the Guzeunggupo-procossed Platycodon grandiflorum A. De Candolle roots in Caenorhabditis elegans)

  • Using the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of the guzeunggupoprocessed Liriope platyphylla F. T. Wang (Liliaceae) tuber was calculated. Between the methanol extracts of guzeunggupo-processed and non-processed L. platyphylla tuber, the processed L. platyphylla tuber showed higher DPPH radical scavenging effect than the non-processed one. The ethyl acetate soluble fraction of the methanol extract of the guzeunggupo-processed L. platyphylla tuber showed the best DPPH radical scavenging activity. The ethyl acetate fraction of the processed sample was measured for the activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species level. In addition, to verify the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction of the processed sample, SOD-3 expression was measured using a transgenic strain (CF1553). Consequently, the ethyl acetate fraction of the processed sample, increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the ethyl acetate fraction of the processed sample-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

The Forkhead Gene fkhB is Necessary for Proper Development in Aspergillus nidulans

  • The forkhead domain genes are important for development and morphogenesis in fungi. Six forkhead genes fkhA-fkhF have been found in the genome of the model filamentous Ascomycete Aspergillus nidulans. To identify the fkh gene(s) associated with fungal development, we examined mRNA levels of these six genes and found that the level of fkhB and fkhD mRNA was significantly elevated during asexual development and in conidia. To investigate the roles of FkhB and FkhD, we generated fkhB and fkhD deletion mutants and complemented strains and investigated their phenotypes. The deletion of fkhB, but not fkhD, affected fungal growth and both sexual and asexual development. The fkhB deletion mutant exhibited decreased colony size with distinctly pigmented (reddish) asexual spores and a significantly lower number of conidia compared with these features in the wild type (WT), although the level of sterigmatocystin was unaffected by the absence of fkhB. Furthermore, the fkhB deletion mutant produced sexual fruiting bodies (cleistothecia) smaller than those of WT, implying that the fkhB gene is involved in both asexual and sexual development. In addition, fkhB deletion reduced fungal tolerance to heat stress and decreased trehalose accumulation in conidia. Overall, these results suggest that fkhB plays a key role in proper fungal growth, development, and conidial stress tolerance in A. nidulans.

Determination of Heavy Metal Content in Astacus leptodactylus caspicus of Anzali Wetland, Iran

  • With attention to different human activities around Anzali wetland and introduction of different pollutants containing heavy metals into this water ecosystem and absorption and accumulation of heavy metals in shell and muscle of Astacus leptodactylus which is native benthose of Anzali wetland and enjoys food and economic importance; Sampling of waters and sediments from 18 stations and shell and muscle of Astacus leptodactylus from 5 stations had been conducted. After biometric evaluation of Astacus leptodactylus, the chemical analysis of the samples by using international and standard methods and then their heavy metals content was determined by Atomic Absorpotion Spectrophotometer (AAS), model P.U 9400 philips. Result of the analysis stated that; 1) The mean values of Cr, Zn, Pb, Ni, V in Anzali waters were 6.4, 184.5, 28.8, 28.9, 47.1 ($\mu$ gr/lit) respectively, which comparing to the international standards, is not suitable for drinking, but it can be used for agricultural and aquacultural purposes. Even though the concentration of heavy metals in sediments were high, there are in range of acceptable limit for aquatics organisms; 2) Comparatively, the concentration of heavy metals in shell and muscle of Astacus leptodactylus in Anzali wetland (Cr, Zn, Pb, Ni, V in shell were 3.48, 109.3, 14.13, 7.17, 36.2 and in muscle were 2.98, 131.98, 3.12, 4.77, 9.05 ppm respectively) were more than what is determined for Aras dam (in shell were 1.6, 224.3, 7.8, 7.6, 10.3 and in muscle were 1.4, 60.2, 0.45, 2.7, 4.4 mg/kgr respectively); 3) For all samples, concentration of heavy metals in all Astacus leptodactylus in Anzali wetland and Aras dam, was at tolerance level for human consumption; 4) There was no association with significant difference between weight and size of Astacus leptodactylus with absorption and accumulation of heavy metals; but there was a significant association between heavy metals content in water and in sediments, and in sediments, shell and muscle of Astacus leptodactylus in Anzali wetland.

Waterlogging Effects on Nitrogen Accumulation and $N_2$ Fixation of Supernodulating Soybean Mutants

  • Soybean is sensitive to waterlogging stress, leading to reduce their growth and yield significantly. The objective of this study was to characterize the relative sensitivities of biomass accumulation and specific nodule activity under waterlogging stress between supernoduating mutants, 'SS2-2' and 'Sakukei 4' and their wild-type soybeans, 'Sinpaldalkong 2' and 'Enrei', respectively. Flooding treatment was performed to soybean plants grown in a pot by waterlogging for 15 days from the beginning bloom(R1) stage under natural light. The nodule number and weight were considerably decreased by waterlogging stress. The bleeding sap rate of waterlogging soybean plants was decreased by 78-80% in supernodulating mutants and 65-74% in their wild types compared to control plants. The relative ureide-N content was also decreased by waterlogging and the reduction was high in supernodulating mutants. This may cause the massive reduction of shoot and root dry weight and leaf area in waterlogged soybean plants. There was a varietal difference in response to the waterlogging stress. During the waterlogging, supernodulating mutants maintained higher spad value than their wild types. Particularly, the difference between soybean varieties was clear in low rank leaves from the top. Also, supernodulating mutants showed a weak waterlogging tolerance than their wild types. Under waterlogging conditions, massive nodules were considerably destroyed and specific nodule activity after waterlogging may not be recovered when compared to their wild-type soybeans. Supernodulating mutants showed lower seed yield than their wild types in waterlogging conditions.

Effect of salt stress on the anthocyanin content and associated genes in Sorghum bicolor L.

  • Abiotic stress is one of the most serious problems in plant productivity because it dramatically delays plant growth and development. One of the abiotic stresses, soil salinity, has an adverse effect on plant growth, particularly in areas where irrigation is necessary like semiarid Asia and Africa. Among several physiological parameters, anthocyanin accumulation is a valuable indicator of the condition of the plant, and it tends to increase under salt stress conditions because of its protective role in such an environment. Consequently, it may be important to search for well adapted genotypes for upcoming climate changes. Anthocyanins are known to have important roles in defense against biotic and abiotic stresses, providing important functions for protecting plant cells from reactive oxygen species. In this study, we investigated the anthocyanin accumulation between two Korean sorghum genotypes, Sodamchal and Nampungchal. The two genotypes were subjected to a regulated salinity condition, and the anthocyanin contents were evaluated in both. In Nampungchal, the anthocyanin content increased with 150 mM NaCl treatment during the time course of the experiment. However, the anthocyanin content of Sodamchal decreased in the same condition. The measured values of the anthocyanin content should be useful to identify the intensity of the salt tolerance in Sorghum bicolor L. Furthermore, we studied gene expression profiling of salt stress related genes with qRT-PCR. These results suggest that Nampungchal is a more tolerant genotype to salt stress compared to Sodamchal. This information should be useful for breeding salt-resistant cultivars in sorghum.

Improvement of K+ and Na+ Ion homeostasis and salt tolerance by Co-inoculation of arbuscular mycorrhizal fungi (AMF) and spore associated bacteria (SAB)

  • Salinity inhibits plant growth and restricts the efficiency of arbuscular mycorrhizal fungi. The selective uptake of nutrients from the soil and their effective transport to host roots make it essential for plant growth and development under salt stress. AMF spore associated bacteria shown to improve mycorrhizal efficiency under stress. Thus, this study aimed to understand the co-inoculation efficiency of AMF and SAB on maize growth and ion homeostasis under salt stress. Two AMF strains and one SAB were inoculated with maize either alone or in combination with one another. The results of our study showed that AMF and SAB co-inoculation significantly improved dry weight and nutrient uptake of maize under salt stress. Co-inoculation significantly reduced proline accumulation in shoots and Na+ accumulation in roots. Co-inoculation treatment also exhibited the high K+/Na+ ratios in roots at 25 mM NaCl. Mycorrhizal colonization showed positive influence for regulation of ZmAKT2, ZmSOS1 and ZmSKOR gene expressions, contributing to K+ and Na+ ion homeostasis. CLSM view showed that SAB were able move and localize into inter and intra cellular spaces of maize roots. In addition, CLSM view of AMF spores showed that gfp-tagged SAB also associated on the spore outer hyaline layer.

Spatial Pattern of Copper Phosphate Precipitation Involves in Copper Accumulation and Resistance of Unsaturated Pseudomonas putida CZ1 Biofilm

  • Bacterial biofilms are spatially structured communities that contain bacterial cells with a wide range of physiological states. The spatial distribution and speciation of copper in unsaturated Pseudomonas putida CZ1 biofilms that accumulated 147.0 mg copper per g dry weight were determined by transmission electron microscopy coupled with energy dispersive X-ray analysis, and micro-X-ray fluorescence microscopy coupled with micro-X-ray absorption near edge structure (micro-XANES) analysis. It was found that copper was mainly precipitated in a $75{\mu}m$ thick layer as copper phosphate in the middle of the biofilm, while there were two living cell layers in the air-biofilm and biofilm-medium interfaces, respectively, distinguished from the copper precipitation layer by two interfaces. The X-ray absorption fine structure analysis of biofilm revealed that species resembling $Cu_3(PO_4)_2$ predominated in biofilm, followed by Cu-Citrate- and Cu-Glutathione-like species. Further analysis by micro-XANES revealed that 94.4% of copper were $Cu_3(PO_4)_2$-like species in the layer next to the air interface, whereas the copper species of the layer next to the medium interface were composed by 75.4% $Cu_3(PO_4)_2$, 10.9% Cu-Citrate-like species, and 11.2% Cu-Glutathione-like species. Thereby, it was suggested that copper was initially acquired by cells in the biofilm-air interface as a citrate complex, and then transported out and bound by out membranes of cells, released from the copper-bound membranes, and finally precipitated with phosphate in the extracellular matrix of the biofilm. These results revealed a clear spatial pattern of copper precipitation in unsaturated biofilm, which was responsible for the high copper tolerance and accumulation of the biofilm.

Pre-adaptation to Cu during Plant Tissue Culture Enhances Cu Tolerance and Accumulation in Begonia (Begonia evansiana Andr.)

  • A simple and efficient protocol was developed for culturing Cu-tolerant and Cu-accumulating plants via pre-adaptation to Cu during plant tissue culture. We induced multiple shoots from begonia (Begonia evansiana Andr.) leaf explants on MS medium supplemented with naphtaieneacetic acid and benzyladenine. After 3 months, small plantlets were transferred to MS medium supplemented with $100{\mu}M\;CuCl_2$ for pre-adaptation to Cu and cultured for 5 months. Then, these plantlets were individually planted in pots containing artificial soil. An additional 500 mg of Cu dissolved in 1/4 strength MS solution was applied to each pot during irrigation over the course of 2 months. We planted pre-adapted and control begonias in soil from the II-Kwang Mine, an abandoned Cu mine in Pusan, Korea, to examine their ability to tolerate and accumulate Cu for phytore-mediation. Pre-adapted begonias accumulated $1,200{\mu}g$ Cu/g dry root tissue over the course of 45 days. On the other hand, non-Cu-adapted controls accumulated only $85{\mu}g$ Cu/g dry root tissue. To enhance Cu extraction, chelating agents, ethylenediamine tetraacetic acid (EDTA)-dipotassiun and pyridine-2,6-dicarboxylic acid (PDA), were applied. While the chelating agents did not enhance accumulation of Cu in the roots of control begonias, EDTA application increased the level of Cu in the roots of pre-adapted begonias twofold (to $2,500{\mu}g$ Cu/g dry root tissue). Because pre-adapted begonias accumulated a large amount of Cu, mainly in their roots, they could be used for phytostabilization of Cu-contaminated soils. In addition, as a flowering plant, begonias can be used to create aesthetically pleasing remediation sites.

Efficacy of Three Different Plant Species for Arsenic Phytoextraction from Hydroponic System

  • Arsenic (As) is one of the heavy metals which causes acute bio-toxicity even at low concentration and has disastrous effect on environment. In some countries, As contamination has become alarming and increasing day by day as consequences of unsustainable management practices. Many existing physical, chemical and biological processes for As removal from water system are not feasible due to techno-economic limitations. The present study highlights the scope of biological strategy for As removal through phytoextraction. Arsenic uptake and accumulation in the biomass of three plant species and their As tolerance abilities have been investigated to develop an efficient phytoextraction system in combination of these plant species. Three non-crop plant species, Pteris vittata; Mimosa pudica, and Eichhornia crassipus were treated with 0-200 mg/L As in liquid nutrient solution for 14 days. P. vittata accumulated total 9,082.2 mg (8,223 mg in fronds) As/kg biomass and Eichhornia total 6,969 mg (4,517 mg in fronds)/kg biomass at 200 mg/L As concentration, respectively. Bioaccumulation factor (BF) and translocation factor (TF) were estimated to differentiate between excluders, accumulators and accumulation in above ground biomass. Pteris and Eichhornia have highest BF (67 and 17) and TF (64 and 3), respectively. In contrast, Mimosa accumulated up to 174 mg As/kg plant biomass which is low in comparison with other two plants, and both BF and TF were ${\leq}1$. This study reveals that Pteris and Eichhornia are As hyperaccumulator, and potential candidates for As removal from water system.
