Chitosans were obtained with varying deacetylation times using the ${\beta}$-chitin isolated from Todarodes pacificus, and their deacetylation degrees and average molecular weights were determined. Films prepared with the squid chitosans were characterized by estimating their tensile strengths, percent elongations, water vapor permeabilities, degree of swelling, and temperatures of glass transition and thermal decomposition. The results suggest that the squid chitosan films were comparable to common crustacean chitosan films in regard of mechanical, moisture barrier, and thermal properties, although further, multilateral investigations are necessary to make a more definitive conclusion.

This study investigated some enzymatic properties and bitterness improvement of an aminopeptidase retentate fraction (ARF) from common squid Todarodes pacificus hepatopancreas extract (HPE), obtained by ultrafiltration with a 10 kDa molecular weight cut off membrane. Endoprotease and aminopeptidase (AP) activity, and the purity of the ARF (>10 kDa) increased by 6.69-18.11 U/mg and 1.5-2.6 fold, respectively, compared to HPE (2.63-9.37 U/mg). The AP activity toward LeuPNA was stable at 20-55℃ and pH 5-9, but decreased slightly with increasing concentration of NaCl in the reaction mixture. The ARF was the most active MetPNA and preferentially hydrolyzed Glu, Leu and AlaPNA. The bitterness tryptic casein hydrolysates (BTCHs) were treated with ARF, and the bitterness of ARF-BTCHs significantly decreased with increasing amounts of released amino acids Ala, Val, Met, Ile and Leu, which show strong correlations with bitterness. Therefore, the ARF of T. pacificus HPE obtained by ultrafiltration may have a considerable potential for application in protein hydrolysis and appears to be ideally suited to the purpose of lowing bitterness in protein hydrolysates.
A visceral helminth of the squid, Todarodes pacificus, is reported from the East Sea, the Republic of Korea. Total 39 squid samples were purchased from a fish market in Jumunjin-eup, Gangneung-si (City) from August 2014 to July 2015 and were examined for helminth parasites with naked eyes and under a stereomicroscope after opening the abdominal cavity with a pair of scissors. Whitish larval worms were mainly found in the stomach and abdominal cavity of the squid. They were detected in 25 (64.1%) out of 39 squids examined, and the infection density was 7 larvae per infected squid. Spatula-shaped larvae were $8.2{\times}2.0mm$ in average size, round to slightly flattened anteriorly, with round hatching posteriorly, and had characteristic 4 tentacles with numerous hooklets in the scolex. The larvae were identified as the plerocercoid stage of Nybelinia surmenicola by their morphological features. This finding represents a new host record and the first report of N. surmenicola infection in T. pacificus squids from the east coast of Korea.

This paper reviews comparison analysis of current and latest application for stock identification methods of Todarodes pacificus, and the pros and cons of each method and consideration of how to compensate for each other. Todarodes pacificus which migrates wide areas in western North Pacific is important fishery resource ecologically and commercially. Todarodes pacificus is also considered as 'biological indicator' of ocean environmental changes. And changes in its short and long term catch and distribution area occur along with environmental changes. For example, while the catch of pollack, a cold water fish, has dramatically decreased until today after the climate regime shift in 1987/1988, the catch of Todarodes pacificus has been dramatically increased. Regarding the decrease in pollack catch, overfishing and climate changes were considered as the main causes, but there has been no definite reason until today. One of the reasons why there is no definite answer is related with no proper analysis about ecological and environmental aspects based on stock identification. Subpopulation is a group sharing the same gene pool through sexual reproduction process within limited boundaries having similar ecological characteristics. Each individual with same stock might be affected by different environment in temporal and spatial during the process of spawning, recruitment and then reproduction. Thereby, accurate stock analysis about the species can play an efficient alternative to comply with effective resource management and rapid changes. Four main stock analysis were applied to Todarodes pacificus: Morphologic Method, Ecological Method, Tagging Method, Genetic Method. Ecological method is studies for analysis of differences in spawning grounds by analysing the individual ecological change, distribution, migration status, parasitic state of parasite, kinds of parasite and parasite infection rate etc. Currently the method has been studying lively can identify the group in the similar environment. However It is difficult to know to identify the same genetic group in each other. Tagging Method is direct method. It can analyse cohort's migration, distribution and location of spawning, but it is very difficult to recapture tagged squids and hard to tag juveniles. Genetic method, which is for useful fishery resource stock analysis has provided the basic information regarding resource management study. Genetic method for stock analysis is determined according to markers' sensitivity and need to select high multiform of genetic markers. For stock identification, isozyme multiform has been used for genetic markers. Recently there is increase in use of makers with high range variability among DNA sequencing like mitochondria, microsatellite. Even the current morphologic method, tagging method and ecological method played important rolls through finding Todarodes pacificus' life cycle, migration route and changes in spawning grounds, it is still difficult to analyze the stock of Todarodes pacificus as those are distributed in difference seas. Lately, by taking advantages of each stock analysis method, more complicated method is being applied. If based on such analysis and genetic method for improvement are played, there will be much advance in management system for the resource fluctuation of Todarodes pacificus.
The catch of Todarodes pacificus in the Yellow Sea is commonly known as the winter cohort. So, to understand the transport process of winter cohort of T. pacificus, and to identify whether the simulated individuals which are transported directly into the Yellow Sea (YS) influence these resources immediately, we conducted a Lagrangian-particle-tracking numerical experiments of T. pacificus from 2005 to 2010 using LTRANS and ROMS. The results show that: (1) Most of the released individuals spread out to the open sea by the Kuroshio and the Tsushima Warm Current around 30 days after release. (2) Unlike the hypothesis proposed by Rosa et al. (2011), Around $30-33N^{\circ}$ near Jeju Island simulated the initial position (3) About 0.01% of individuals released in December were transported solely into the YS around 15 days after release. However there were no surviving individuals due to the low temperature less than $12^{\circ}C$. Also the variation of individuals entered into the YS was not significantly correlated with it in YS catches during the experimental period. Therefore, the most of resources in the YS is assumed to be more influenced by diverse factors of the Pacific Ocean and East Sea than the direct transport in the YS of winter cohort.

To determine the horizontal and temporal distribution of common squid larvae, Todarodes pacificus (hereafter T. pacificus), we conducted surveys using an IKMT net (mesh size: $500{\mu}m$) with a Fisheries Research Vessel (FRV, TAMGU 21) in the southwestern part of the East Sea in summer (August and September) and autumn (November) 2015. A total of 228 larvae, ranging in mantle length (ML) from 1.4 mm to 21.9 mm, were collected at 35 stations over the research period. The monthly average mantle length of T. pacificus larvae did not differ significantly in August, September and November. (p > 0.05). Catch densities at positive stations ranged between 0.1 and $7.9inds./1,000m^3$ over the research period. Incidence rates of T. pacificus larvae were similar over three months, in the study area (62.9 % - 68.6 %). The 4 - 5 mm mantle length range had the highest frequency in size-frequency distributions for T. pacificus larvae. The larval survival temperature ($15-24^{\circ}C$) at positive stations for catch densities was located below a 20 m depth in August whereas it was located at the surface of the water in September and November. The survival temperature for larvae existed from the bottom to the surface of the water where larvae were sampled larvae in shallow sea areas. However, the larval survival temperature occurred in a shallower location than the upper layer of the thermocline in deep sea areas at a depth below 100 m.

Population analysis of the common squid, Todarodes pacificus identified three seasonal cohorts in the Korean waters. They were significantly separable in terms of spawning period and fecundity, and they were slightly different in frequency distribution of egg size, while they are not significantly different in mantle length at $50\%$ maturity.

Paralarvae of the common squid, Todarodes pacificus Steenstrup were sampled with the bongo net (diameter: 60 cm, mesh: $333{\mu}m$) by using oblique tow method with the oceanographic research vessel (Tamgu 12) at 21 stations along the southwestern coastal waters of the East Sea in August and September, 2013. Paralarvae were collected 16 ind./$10m^3$ in August and 7 ind./$10m^3$ in September at approximately, and bottom depth was from 70 m to 500 m. Mantle length of paralarvae was from 1.7 to 6.0 mm. Paralavae were distributed mainly in offshore stations in August and near by Cape Janggi in September. Optimum embryo survival temperature ($18-24^{\circ}C$) was formed shallower than 35 m depth for almost stations where paralarvae were existed, and survival temperature ($15-24^{\circ}C$) for embryo stage was located shallower than 75 m.
This study analyzed the influence of climate change on the spawning ground area of the common squid, Todarodes pacificus. To estimate long term changes in the area of the spawning ground of the common squid, water temperature at 50 m deep that can be inferred from sea surface temperature (SST) based on both NOAA/AVHRR (1981.07-2002.12) and MODIS/AQUA (2003.01-2009.12) ocean color data was analyzed. In addition, five climate indices, Arctic Oscillation Index (AO), Siberian High Index (SH), Aleutian Low Pressure Index (ALP), East Asia Winter Monsoon Index (EAWM) and Pacific Decadal Oscillation (PDO) which are the main indicators of climate changes in the northwestern Pacific were used to study the relationship between the magnitude of the estimated spawning ground and climate indices. The area of the estimated spawning ground was highly correlated with the total catch of common squid throughout four decades. The area of the estimated spawning ground was negatively correlated with SH and EAWM. Especially, PDO was negatively correlated with the area of the spawning ground in the northwestern Pacific (r = -0.39) and in the southern part of the East Sea (r = -0.38). There was a positive relationship between the AO and the area of the spawning ground in the northwestern Pacific (r = 0.46) as well as in the southern part of the East Sea (r = 0.32). Temporally, the area of the winter spawning ground in the southern part of the East Sea in the 1980s was smaller than those areas in the 1990s and 2000s, because the area was disconnected with the western coastal spawning ground of Japan in the 1980s, while the area had been made wider and more continuous from the Korea strait to the western coastal water of Honshu in the 1990s and 2000s.
To develop a whitening agent, cytotoxicity of the soluble collagen isolated from Todarodes pacificus (CIT) was evaluated. CIT tested for cytotoxicity on human dermal fibroblast (CCD-986sk) was exhibited very low cytotoxicity. Because tyrosinase is the key enzyme for melanin biosynthesis, the use of various tyrosinase inhibitors is a common practice for whitening purpose in cosmetics. Tyrosinase inhibitory activity and melanin production assay were measured to confirm the whitening effect. The inhibitory effect of MMP-1 in UV-irradiated human dermal fibroblast was also performed. CIT showed strong inhibition potency on tyrosinase by 51.5% at 0.2 mg/mL which increased the inhibition by increasing the concentration of CIT, and showed 69.1% inhibition at 1.0 mg/mL. CIT showed strong inhibition effect on melanin production with 82% at 1.0 mg/mL. The CIT also reduced about 76% expression of MMP-1 in UV-irradiated CCD-986sk cell at 1.0 mg/mL. From the preliminary observations, we suggest that the collagen isolated from CIT could be a potential source of natural skin-whitening and anti-aging agents for the photo-damaged skin.
