The fisheries resources of the set net fishery on four fishing grounds (Pyuong Dae, Gu Eum, Du Moah, Gang Jeong) in coastal waters of Jeju Island were studied to the determine catch-variation characteristics from January, 1998 to December, 1999. A total catch of 153,862 kg was made through the survey period. The large total was related mainly to the high catch of Trachurus japonicus, which comprised 58.2% of the total. Other dominant species which account for 23.0% of the total catch were Siganus fuscescens, Todarodes pacificus, Loligo chinesis, Seriola lalandi, Sepioteuthis lessoniana, and Seriola quinqueradiata. Catch density was high in Gu Eum (more than 100,000 kg), whereas those in Pyuong Dae, Du Moah, and Gang Jeong were less than 30,000 kg in total. The fishing period of the set net fishery was from April to December, which was also closely related to the variation of the sea water temperature in the fishing grounds. The ranges of water temperature in the four fishing grounds were $18{\sim}26^{\circ}C$ in Pyuong Dae, $16{\sim}26^{\circ}C$ in Du Moah, $15{\sim}26^{\circ}C$ in Gu Eum, and $15{\sim}26^{\circ}C$ in Gang Jeong.

In this study, an approach for the analysis of the five cephalopod species (octopus, long-arm octopus, squid, wet-foot octopus, beka squid) consumed in the Republic of Korea is developed. The samples were collected from the Southeast Asian countries Thailand, Indonesia, Vietnam, and China. The SYBR-green-based real-time qPCR method, based on the mitochondrial DNA genome of the five cephalopods was developed and validated. The intergroup variations in the mitochondrial DNA are evident in the bioinformatic analysis of the mitochondrial genomic DNA sequences of the five groups. Some of the highly-conserved and slightly-variated regions are identified in the mitochondrial cytochrome-c-oxidase subunit I (COI) gene, 16s ribosomal RNA (16s rRNA) gene, and 12s ribosomal RNA (12s rRNA) gene of these groups. To specify each five cephalopod groups, specific primer sets were designed from the COI, 16s rRNA and 12s rRNA regions. The specific primer sets amplified the DNA using the SYBR-green-based real-time PCR system and 11 commercially secured animal tissues: Octopus vulgaris, Octopus minor, Todarodes pacificus, Dosidicus gigas, Sepia esculenta, Amphioctopus fangsiao, Amphioctopus aegina, Amphioctopus marginatus, Loliolus beka, Loligo edulis, and Loligo chinensis. The results confirmed by a conveient way to calculate relative amplification levels between different samples in that it directly uses the threshold cycles (Ct)-value range generated by the qPCR system from these samples. This genomic DNA-based molecular technique provides a quick, accurate, and reliable method for the taxonomic classification of the animal tissues using the real-time qPCR.

This study made a comparative analysis of behavioral reaction of squid to red (624nm), green (524nm), blue (460nm) & white LED light, its arrival time for the shadow section by making the shadow section in the central section of a water tank just like the bottom part of a squid jigging vessel, and on-site catching efficiency of LED fishing lamp with control fishing vessel. The color LED light showing the highest squidgathering rate as against the shadow section was found to be blue LED light with 39.3% rate under the dark (0.05lx) condition. Under the brighter condition than 0.05lx, white LED light was found to have the highest gathering rate of 41.5%. In addition, it was found that squid gathering rate was high at the shadow section which showed 6.3-fold brightness difference between the shadow section and bright section. As for the arrival time for the shadow section, blue LED light was found to be the fastest in attracting squids in 192.7 seconds under the dark condition while the red LED light was the fastest in luring squids in 164.6 seconds under the bright condition. The ratio of the squid-jigging operation and sailing in fuel consumption of the fishing vessel loaded with LED fishing lamp is about 7 to 1, showing most of the fuel is consumed more in sailing than in squid-jigging operation. As for a catch of squid, the control vessel loaded with MH (Metal Halide) fishing lamp had more catch of 600-7,080 squids than the vessel loaded with LED fishing lamp having a catch of 260-1,700 squids. In addition, even in the comparison of a catch per automatic jigging machine, the catch of the vessel loaded with MH fishing lamp excelled that of the vessel loaded with LED fishing lamp in 6 operations of squid jigging out of 9 operations. The ratio of hand-jigging and automatic jigging machine (one line) in the LED fishing lamp vessel was 1:1.1 excepting the case of having a catch only using an automatic jigging machine, showing almost the same with each other in catches, while in case of a MH fishing lamp vessel, its ratio against hand-jigging was 1 to 5.8, showing hand-jigging excelled in catches.

Squid jigging fishery is very important in that there are about 1,000 jigging vessels more than 10 tonnage and about 5,000 ones less than 10 tonnage in Korea. But the cost of oil which is used to light fishing lamps, goes significantly up to almost one hundred million won for 50 tonnage vessels and forty million won in case of vessels less than 10 tonnage. This cost has almost taken 40% of total fishing costs. That is, the fishing business condition of squid jigging fishery is recently in the very difficult situation. As oil price increases, the business condition of the fishery gets worse and worse. Therefore it is very urgent to develop an economical fishing lamp, to solve this problem of fishery's business difficulty. This research aims at developing a fishing lamp for squid jigging fishery using the light emitting diode which has very excellent efficiency and durability. We made a water tank with 20 meters width which is a shape of raceway to research behavioral characteristics of Japanese flying squid to LED light, and made an experiment to investigate optimum wave of LED light to lure squid. The method is to establish LED lamps on both ends of water tank and to observe squid's behavior. Colors and wave lengths of LED lamps, used in experiment, are red(634nm), yellow(596nm), green(523nm), blue(454nm) and white(454nm+560nm). In experiment for attractive capability of LED lamp to squids, Japanese flying squid are highly attracted to blue lamp and white lamp. However, they are dispersed to red and yellow lamps. In addition, Japanese flying squid have moved and stayed in both dark ends of water tank. When compared intermittent lamp with continuous lamp, Japanese flying squid are highly attracted to intermittent lamp when intermittent interval is 0.25 second.

The aim of this study was to develope a fishing technology for fisheries management for improving the survival rate of young fishes escaped from trawl net. Sea experiments were carried out to investigate the survival rate of the fishes in off Geomun island and Korean southern sea. Cover-net fish cage was designed and manufactured to collect fishes escaped from COD(codend) and TED(trawl escapement device). Fish cage was installed in a shallow site where scuba diver can observe the surviving fishes after separating the cage from trawl net. Most of fishes except horse mackerel(Trachurus japonicus) and mackerel(Scomber japonicus) could escaped through TED(bar space 35mm) easier than COD(mesh size 54mm). Especially, escapement rate of redlip croaker(Pseudosciaena polyactis) escaped from TED was the highest as 73.9%. Survival rate of horse mackerel(Trachurus japonicus) escaped from TED was 85.5% at the point of 96 hours over, while the survival rate for COD showed a rapid decrease according to the time elapse as 85.0% at the point of 24 hours, 76.4% for 48 hours, 62.7% for 72 hours and 56.1% for 96 hours. Over 90% of red seabream(Pagrus major) and rock bream(Oplegnathus fasciatus) escaped from TED were survived 96 hours over and the survival rate of those fishes were excellent compared with the other fish species. All korean pomfret(Pampus echinogaster) escaped from both COD and TED were died within 48 hours. Most of redlip croaker(Pseudosciaena polyactis) were also died within 12 hours because of the weak physiological characteristics by water pressure change. Survival rate of fishes like as horse mackerel(Trachurus japonicus) having hard skin was high while it was low for the fishes like as squid(Todarodes pacificus) and hair-tail(Trichiurus lepturus) having soft skin. We could know that there were big differences in the survival rate offish species escaped from trawl net.

This study surveyed the fishing efficiency for Japanese common squid based on improvements made to LED fishing lamps by utilizing the training ship of Gangwon Provincial College. The training ship, Haesong-ho (24 tons), was equipped with seventy-two 150W electric power LED fishing lamps (10.8kW), and their fishing efficiency and fuel consumption level were surveyed for a total of ten times during the period between June 15, 2009 and July 27, 2009. In addition, the training ship was equipped with seventy-one 300W electric power LED fishing lamps (21.3kW) and their fishing efficiency and fuel consumption level were surveyed for a total of five times during the period between January 17, 2010 and August 4, 2010. In order to compare the fishing efficiency of LED fishing lamps, the catch of another fishing vessel equipped with Metal Halide (MH) fishing lamps of 120kW for the same period and at the same fishing grounds. The fuel consumption levels during the fishing operation of Haesong-ho was about 1,047.7 liters which was approximately 19.9% of the total fuel consumption level of 5,262.6 liters, and the fuel consumption level per operation hour was on average 9.2 liters. The number of Japanese common squid caught by the LED fishing lamp-equipped fishing vessel ranged from 12 to 1,640 squid for each fishing trial and the average quantity was 652. The number of Japanese common squid caught by the MH fishing lamp-equipped 10 fishing vessels ranged from 40 to 4,800 squid and the average quantity was 2,055. The fishing of Japanese common squid was done by the use of hand lines and an automatic jigging machine. The number of Japanese common squid caught per hand line and a single roller of the automatic jigging machine was in the proportion of 50.8% to 49.2% with respect to the LED fishing lamp-equipped fishing vessel. However, the number of Japanese common squid caught per hand line and a single roller of the automatic jigging machine was in the proportion of 86.4% to 13.6% with respect to the MH fishing lamp-equipped fishing vessel where most of the catch was made by hand lines. On the other hand, in comparing the number of Japanese common squid caught per automatic jigging machine, the number of squid caught by the LED fishing lamp-equipped fishing vessel was about the same or greater than the number of squid caught by the MH fishing lamp-equipped fishing vessel.
The effects of garlic concentrations on the properties of squid sik-hae such as the chemical and microbial changes were investigated. pH, the amounts of lactic acid showed significant differences at early stages of fermentation and no significant differences at later stages with increasing garlic concentrations. The amount of TMAO decreased with increasing storage periods and garlic concentrations, whereas that of TMA increased significantly at early stages of fermentation and showed no significant differences with increasing garlic concentrations. At later stages of fermentation, the amount of TMA at $2\%$ garlic concentration was higher than those at 3 and $4\%$. The number of lactic acid bacteria increased significantly garlic concentrations at early stages of storage and had no significant differences at later stages. The numbers of proteolytic bacteria, psychrotroph, and fungi increased in proportion to length of storage and garlic concentrations at early stages of storage whereas, the number of bacteria decreased significantly at later stages of storage with increasing garlic concentrations.
The effects of red pepper and grain contents on the properties of squid sik-hae fermented at $20^{\circ}C$ were investigated. The values of pH at $10\%$ red pepper contents and $30\%$ grain contents were lower at early stages of storage than $5\%\;and\;50\%$, respectively, and showed no significant differences at later stages of storage. The amounts of lactic acid and amino-N were not changed significantly at different red pepper contents where as, higher at $30\%$ grain content than at $50\%$ at early stages of storage. The amounts of total arginine were a little higher at $10\%$ red pepper and $30\%$ grain contents than at $5\%\;and\;50\%$, respectively. There were no significant effects of red pepper and grain contents on the amounts of TMAO and TMA during fermentation. Therefore, the effects of red pepper and grain contents on the chemical properties of squid sik-hae during fermentation were not significant.
Food irradiation is one of the successful modern techniques used to preserve food. However, it needs very careful control. Detection of irradiated food is of prime importance to facilitate global trade and consumer assurance, choice, and protection. This study was performed to evaluate the radiation-induced hydrocarbon content of dried squid and octopus by e-beam irradiation. The samples were collected from supermarkets all over South Korea and irradiated with an e-beam at 0, 1, 3, 5, 7, and 10 kGy doses. Lipids were extracted with soxhelt, and the hydrocarbons induced with irradiation were separated via solid phase extraction (SPE) and identified via gas chromatography mass spectrometry (GC/MS). The major induced hydrocarbons in the irradiated dried squid and octopus were 1-tetradecene and pentadecane derived from palmitic acid and 1-hexadecene and heptadecane from stearic acid. The concentration of hydrocarbons differed from the composition of the fatty acid at the same radiation and increased according to the level of the radiation dose. The hydrocarbons induced by e-beam irradiation, including 1-tetradecene, 1-hexadecene, and heptadecane, were confirmed to have been the irradiation marker compounds. Therefore, they can be used to distinguish the e-beam-irradiated dried squid and octopus from the non-irradiated ones.

Squid (Todarodes pacificus) is processed as dried or seasoned-dried products and its catch gradually increased from 270,298 M/T in 2005 to 367,940 M/T in 2008 in Korea. Squid processing by-product (viscera) was usually discarded as a waste resulting in environmental problem. In order to utilize squid viscera for more value-added products, a natural squid seasoning was developed by fermenting with Aspergillus oryzae koji. Squid viscera at 5, 10 and 15% salt concentrations with fixed levels of 5% koji and 30% water was fermented at room temperature. The quality properties of squid fermented products such as amino-N, TMA, VBN, total viable cell count, pH and total acidity were determined at different fermentation periods. The contents of amino-N, TMA, and VBN of squid seasoning at 5% salt concentration fermented for 14 days were the highest. Based on amino-N content, squid viscera at 5% koji fermented for 14 days was selected for further assays: the content of moisture, crude protein, crude lipid, crude ash, and carbohydrate were 5.98, 35.19, 33.08, 11.30, and 14.45%, respectively. The content of glutamate, alanine, leusine and lysine were 7.06, 12.34, 9.90 and 10.22%, respectively. The $IC_{50}$ values of DPPH scavenging and $\beta$-glucuronidase inhibitory activity were 12.89 and 12.58 mg/mL, respectively. A natural squid seasoning was manufactured by mixing fermented squid viscera and an ingredient. Based on the results of sensory evaluation, the fermented squid viscera seasoning was almost equal to other natural complex seasonings such as anchovy, cow meat, and fisheries seasoning.
