Studies on the Disease Development and Chemical Control of Tobacco Wild Fire (담배 야화병의 발생소장과 그 방제에 관한 연구)

  • Tobacco wild fire caused by Pseudomonas tabaci(Wolf et Foster) Stevens is one of the most important diseases on tobacco plant in Korea. The objectives of the study were to investigate: 1) the relationships between environmental conditions and the disease development, 2) the effect of chemical spray and soil sterilization on the disease development, and 3) the proper date for chemical sprays during the growing season. The appearance of wild fire on tobacco plant was somewhat proportional to temperature and moisture in atmosphere. The first occurrence of the disease was in late part of June when the temperature was $18\;to\;20^{\circ}C$. with $65\;to\;70^{\circ}C$ of relative humidity. The disease was, then, gradually increased during early part of July with the temperature of $22\;to\;26^{\circ}C$. and $85\;to\;86^{\circ}C$ of. relative humidity. The rapid development of the disease occurred after heavy rain or storm during July. Two to three times of Phytomycin sprays(1:300-350) during the epidemic season reduced $28.3\%$ of disease damage when compared with control plot. Soil sterilization with chloropicrin. 10 liter/10a, Prior to transplanting of seedlings reduced not only wild fro damage but also the other tobacco diseases on field. The study showed that the damage caused by tobacco wild fro could be reduced with the use of early varieties and/or 2 or 3 times of Phytomycin sprays during early part of June, and soil sterilization with chloropicrin prior to transplanting.

Overexpression of a Rice Diacylglycerol Kinase Gene OsBIDK1 Enhances Disease Resistance in Transgenic Tobacco

  • A rice diacylglycerol kinase (DGK) gene, OsBIDK1, which encodes a 499-amino acid protein, was cloned and characterized. OsBIDK1 contains a conserved DGK domain, consisting of a diacylglycerol kinase catalytic subdomain and a diacylglycerol kinase accessory subdomain. Expression of OsBIDK1 in rice seedlings was induced by treatment with benzothiadiazole (BTH), a chemical activator of the plant defense response, and by infection with Magnaporthe grisea, causal agent of blast disease. In BTH-treated rice seedlings, expression of OsBIDK1 was induced earlier and at a higher level than in water-treated control seedlings after inoculation with M. grisea. Transgenic tobacco plants that constitutively express the OsBIDK1 gene were generated and disease resistance assays showed that overexpression of OsBIDK1 in transgenic tobacco plants resulted in enhanced resistance against infection by tobacco mosaic virus and Phytophthora parasitica var. nicotianae. These results suggest that OsBIDK1 may play a role in disease resistance responses.

A Cross Sectional Study of Kretek Smoking in Indonesia as a Major Risk to Public Health

  • Background: Tobacco smoking is a major cause of preventable disease and death worldwide. Kreteks are clove-flavored cigarettes made from a combination of tobacco and ground-clove mixed with a sauce, smoked widely in Indonesia. Because health and social consequences of kretek smoking are potentially as great as those of traditional cigarettes, this study examines the prevalence of kretek smoking in Indonesia and associated risk factors. Materials and Methods: The study used nationally representative Indonesia Global Adult Tobacco Survey data. Multiple logistic regression analysis was employed to identify correlates of kretek smoking. Results: One-third of Indonesian adults smoked tobacco of which about 90.0% smoked kreteks. Prevalence of kretek smoking among men (60.9%) was more than 25 times the rate among women (2.3%). Overall, the highest prevalence of kretek use was in the age group 45-54 years (36.5%), followed by 34-44 (35.1%), 25-34 (34.2 %), and 55-64 years (32.8%). By wealth index, prevalence of kreteks smoking among those in the middle index was almost 50% above the rate for the wealthiest group (36.4% vs 24.8% respectively). Logistic regression results showed that being male, being older, having less education, and being less wealthy were significant predictors of kretek smoking, while urban vs rural residence was not. Conclusions: Kretek smoking is common in Indonesia and is entrenched in the sociocultural fabric of the country. However, potential consequences of kretek smoking, particularly as risks for noncommunicable diseases, underscore the importance of a comprehensive approach to tobacco control as outlined in the World Health Organization's MPOWER strategies.

Global School Personnel Survey Among 5200 School Personnel in India: Comparison of the Results for the Years 2009 and 2006

  • Background: The results of the Global School Personnel Survey (GSPS) conducted in India in 2009 are compared with 2006 GSPS to assess any change in 2009 on tobacco use and knowledge and attitudes to tobacco use, training and availability of tobacco control teaching material in schools and the existence of school tobacco control policies. Methods: GSPS is a cross sectional survey conducted twice (2006 and 2009) in entire India. A total of 180 schools were surveyed each time. Results: Of the participating school personnel, 2660 in 2006 and 2575 in 2009, about 95% were teachers and the balance administrators. In 2009, compared to 2006 the prevalence of current smoking of cigarettes (19.6% in 2006 and 10.3% in 2009) and bidis (21.5% in 2006 and 13.9% in 2009) was found to be significantly lower; the percentage of teachers receiving training on preventing youth tobacco use has significantly reduced (16.7% in 2006 and 10.1% in 2009); access of teachers to educational materials on tobacco use and how to prevent its use among youth had not increased (34.6% in 2006 and 37.8% in 2009); there was no change in policy prohibiting tobacco use among students and school personnel; however, ever use of any tobacco on school premises was significantly lower (15.6% in 2006 and 9.6% in 2009). Conclusions: The prevalence of current smoking (cigarettes/bidis) among school personnel and use of any tobacco on school premises were significantly decreased in 2009 as compared to 2006. Necessary action should be planned to increase the number of teachers trained and the availability of teaching materials on preventing youth tobacco use in order to have effective prevention of tobacco use among students.

연초 버어리종 웅성불임 일대잡종 KB 111의 육성경과 및 농경적 특성

  • The vein necrotic strain of Potato Virus Y (PVY - VN) and black shank (Phyto-phthora parasitica var. nicotianae) are the two major diseases causing severe damages especially in burley tobacco (N. tabacum L.) area in Korea. A new tobacco variety, KB 111, resistant to PVY and black shank disease, was developed by Korea Ginseng & Tobacco Research Institute in 1997. It is a male sterile(MS) F$_1$ hybrid of the cross between MS TC 613 and KB 108. KB 111 was compared to Burley 21 on the agronomic characteristics and disease resistances in performance tests: It possessed upright growth habit and flowered two days later than Burley 21. It was resistant to both PVY and black shank and yielded about 3% more cured leaf than Burley 21, but other characteristics are very simiar to those of Burley 21. The chemical composition and physical properties of the cured leaf of KB 111 were as much acceptable as those of Burley 21 while it produced average yield of good quality leaf and appeared to resistant to PVY and black shank disease on regional farm test in 1998.

  • The present study was conducted to compare the efficiency of individual plant selection for resistance to bacterial wilt with 30 dihaploid lines derived by anther culture and Nicotiana africana method in Fl and F2 generation from a cross between Bright Yellow 4 (BY4) and NC95. F2 dihaploid lines were selected from bacterial wilt disease resistant plants screened under the naturally infested filed conditions. The populations of FB - ADH and FB MDH derived from F2 individual plants with bacterial wilt resistance showed higher resistance to the disease than the populations of Fl - ADH and Fl - MDH, respectively, and no difference for the disease resistance appeared by the haploid deriving method within a generation.

  • The hollow stalk, caused by Erwinia carotovors var. carotovora, is probaly the most devastating disease to burley tobacco in Korea. There are two modes of infection to tobacco plants: the primary infection along the plant stem from soil and the secondary one from the topped end of plants. The secondary infection rate could be reduced to almost zero by applying an antibiotic, streptomycin sulfate(400 ppm solution), right after topping.

  • Three kinds of inoculation methods, capillary, root cutting and dipping were compared for an efficient way to screening the resistant tobacco variety against bacterial wilt, Ralstonia solanacearum. The pricking a capillary tube contained the pathogenic bacterial suspension(10$^{7}$ cfu/$m\ell$) to an axillary bud of each tobacco plant showed different resistance well between varieties. The less period was required in inoculating work and in disease development for the inoculation method used with capillary tube than for two other inoculation methods tested also.

Airborne Nicotine Concentrations in the Workplaces of Tobacco Farmers

  • Objectives: Nicotine is a natural alkaloid and insecticide in tobacco leaves. Green tobacco sickness (GTS) is known as a disease of acute nicotine intoxication among tobacco farmers. Until now, GTS has been recognized globally as a disease that results from nicotine absorption through the skin. However, we assumed that GTS might also result from nicotine inhalation as well as absorption. We aimed to measure the airborne nicotine concentrations in various work environments of Korean tobacco farmers. Methods: We measured the nicotine concentrations in the tobacco fields, private curing barns, and joint curing barns of farmers from July to October 2010. All sampling and analyses of airborne nicotine were conducted according to the National Institute for Occupational Safety and Health manual of analytic methods. Results: The airborne nicotine concentrations (geometric mean [geometric standard deviation]) in the tobacco field were $83.4mg/m^3$ (1.2) in the upper region and $93.3mg/m^3$(1.2) in the lower region. In addition, the nicotine concentration by personal sampling was $150.1mg/m^3$. Similarly, the nicotine concentrations in the private curing barn, workers in curing barns, the front yard of the curing barn, and in the joint curing barn were $323.7mg/m^3$(2.0), $121.0mg/m^3$(1.5), $73.7mg/m^3$(1.7), and $610.3mg/m^3$(1.0), respectively. Conclusions: The nicotine concentration in the workplaces of tobacco farmers was very high. Future studies should measure the environmental concentration of nicotine that is inhaled by tobacco farmers.

Inhibition Effect of Avirulent Pseudomonas solanacearum on the Multiplication of Virulent Isolate in Tobacco Plant (비병원성균주 전처리에 의한 담배세균성마름병균(Pseudomonas solanacearum)의 식물체내 침입 및 증식억제)

  • Significant reduction in disease severity of bacterial wilt (Pseudomonas solanacearum) on the susceptible tobacco cultivar BY 4 was observed until mid-July in a naturally infested field when bacterial suspensions of avirulent isolate were applied to tobacco root zones at one day before and fourty days after transplanting into the field. However, rapid increase in disease severity after mid-July resulted in the same severity $(70\%)$ as on cultivar BY 4 without the application of the avirulent bacterial suspension at the end of the season. Yield increase in cultivar BY 4 was $35\%$ due to the treatment, resulting in $10\%$ price increase. The suppression me chanism did not appear to be dependent upon the inhibition of the virulent bacterial multiplication by the avirulent bacteria in tobacco rhizosphere soil because of no significant difference in the density of the patho genic bacteria between treated and untreated plant root zones. However. penetration of the virulent bacteria into the root systems and their multiplication in tobacco stem were inhibited remarkably by preinoculation with avirulent one, suggesting that those are related to the suppression of disease incidence.
