Cold Storage Characteristics of Early Variety of Citrus unshiu Produced in Cheju with Various Treatments (처리조건에 따른 조생온주밀감의 저온저장 특성)

  • The conditions of cold storage of Citrus unshiu Marc. var. miyagawa produced in Cheju were investigated. Compared to the citrus fruits stored at room temperature, the content of soluble solids, total sugar, vitamin C and specific gravity decreased slightly on the fruits stored at $4^{\circ}C$ and 87% relative humidity. Decay ratio and weight loss were below 10% on keeping freshness relatively till late of March during cold storage. Weight loss, decrease of firmness and soluble solids occured gradually during cold storage. Nevertheless seal-packaging with 0.02 mm LDPE film or wax-coating of citrus fruits were effective on weight loss and appearance, decay ratio increased for long-term storage. It was needed to control humidity in cold chamber for preventing from decay and weight loss. Edible part ratio was decreased gradually by respiration, and peel and tissue of fruits were softened slightly by 60 days of cold storage. Decay ratio and weight loss of full-ripened citrus fruits were little during the storage for 100 days, compared to the fruits harvested early. Appearance and taste of citrus fruits stored for 4 month were good relatively, but decay occured rapidly on seal-packaging fruits putting out of cold room for a few days. Because of the difficulty of long-term storage for early variety of Citrus unshiu, the conditions and periods of cold storage would be determined after considering the physicochemical properties of fruits.

Characterization of Streptomyces Species Causing Potato Scab in Korea: Distribution, Taxonomy, and Pathogenicity

  • From 1996 to 1999, potato-growing areas in Korea were surveyed for identification and distribution of potato scab pathogens. Potato scab was widely distributed in the mass cultivation areas, especially in Jriu island, southern areas of Chonnam and Gyounggi provinces, and the alpine area of Gangwon province. Jeju island was the most affected area by this disease. A total of 55 Streptomyces strains were isolated from potato scab lesions, among which 40 strains were pathogenic on progeny tubers. Among the pathogenic strain, 21 strains were identified as previously described S. scabies, 7 Strains as S. turgidiscabies, and 5 Strains as S. acidiscabies, while 7 strains were observed as having distinct phenotypic properties. These strains were classified into six distinct clusters based on phenotypic characteristics and selected representative strains for each cluster. S. scabies (S33) had grey spores in a spiral chain. Mean-while, S. turgidiscabies (S27) had grey spores, S. acidiscabies (S71) had white spores, S. luridiscabiei (S63) had yellow-white spores, S. puniciscabiei (S77) had purple-red spores, and S. niveiscabiei (S78) had thin and compact white spores, all in a rectiflexuous chain. Pathogenicity was determined by the production of thaxtomin A and homologs of necl and ORFtnp genes. In TLC, representative strains S27, S71, S63, S77, and S78 produced a yellow band that co-migrated with the authentic thaxtomin A. However, thaxtomin A was not detected in chloroform extracts from oatmeal broth culture and Slice tuber tissue of S. luridiscabiei (S63) and S. puniciscabiei (S77) by HPLC analysis. In addition, no homologs of necl and ORFtnp genes in S. acidiscabies (S71), S. luridiscabiei (S63), S. puniciscabiei (S77), and S. niveiscabiei (S78) were detected by PCR and Southern hybridization analysis.

Effect of Cow Manure Originated from the Clay Mineral Feeding on the Growth of Zea mays Gk 729 (점토광물 혼합 사료 급여로 발생된 우분 시용에 따른 옥수수 GK729의 생장)

  • This study was conducted to examine the effects of cow manure originated from the clay mineral (porphyrite, zeolite and bentonite) feeding on the growth of Zea mays GK729. CEC of the manure after bentonite, zeolite, and porphyrite feeding were 147.5, 137.0, and 114.0 cmol/Kg, respectively. These values were higher than that of non-mineral treated manure, 107.5 cmol/Kg. After 3 month growing, there was significant difference among treatment in the biomass prodcution and the content of minerals in plant tissue of Z. mays, but the physico-chemical properties of soils were not showed significant difference. For these results suggested that the clay mineral enhanced the grwoth of Z. mays according to the unknown mechanism, the further studies on the physiological changes will be needed.

  • A new technique for the preparation of porous polyurethane vascular prostheses was investigated. Synthetic vascular grafts with porous wall have been widely proposed, claiming that strength, suture retention, kink resistance, and other handling properties are improved over those with nonporous solid wall. Related to these facts, the control of pores and compliance match have been very important and interesting issues. Two kinds of polymer sheets were compared. One was the porous PU-sheet made at room temerature by the solvent/non-solvent exchange. And the other was the porous PU-sheet fabricated by thermal phase transition and solvent/non-solvent exchange in the thermal controlled bath. According to the result of the above experiments, polyurethane solution was injected into a mold designed for U-type graft. After freezing at low temperature, solvent was dissolved out with alcohol at < $0^{\circ}C$ and water at room temperature to form porous vessels. The average pore size and pore occupation were easily changed by changing polyurethane concentration and freezing rate. This technique can give a proper pore size for tissue ingrowth, and suitable compliances for matching with arteries and veins. In addition, the fabrication of more complicated shaped vessels such as the U-type vascular grafts is easily controlled by using a mold. This method might give a desired compliact graft for artificial implantaion with the commercially available medical polymers.

Prevention of Pectinolytic Softening of Kimchi Tissue (펙틴 분해효소를 이용한 김치 조직의 연화 방지)

  • Polygalacturonase(PG) and pectinesterase(PE) were extracted from Chinese cabbage and physicochemical properties of the enzymes were characterized. The preheating conditions for maximum retention of Kimchi texture were also studied. The activity of PE was highest at $50^{\circ}C$ and at 0.02M $CaCl_2$ but decreased in 0.2M $CaCl_2$, PG exhibited maximum activity at $65^{\circ}C$ with 0.3mM $CaCl_2$ but was inhibited by $CaCl_2$ at 0.5mM. Both of the enzymes, however, exhibited the maximum activity with 0.25M NaCl. Optimum preheating treatment was determined for minimum PG activity and maximum PE activity. Thus a maximum crispness and firmness was obtained with preheating in 0.05M $CaCl_2$ solution at $50^{\circ}C$ for 1.5hr results indicated that PE activity and calcium ion were very effective in preserving firmness.

Physicochemical Characterization and Changes in Nutritional Composition of Onions Depending on Type of Freezing Process (냉동 조건에 따른 양파의 이화학적 특성 및 영양성분 변화)

  • Innovative freezing technology is currently applied to preserve foodstuffs for long-term storage. Generally, the quality of frozen food is closely related to the types of freezing and thawing processes. In this study, we characterized the physicochemical properties of onions depending on freezing rate. When onions were frozen at $-40^{\circ}C$, freezing rates were 0.1, 0.5, and $0.7^{\circ}C/min$ depending on air-blast quick freezer mode. Onions were thawed by microwave irradiation at 400 W. Hardness of onion dramatically decreased after freezing and thawing compared with blanched onion. However, the fastest freezing rate did not affect hardness. Thawing loss of onion decreased with a faster freezing rate. For morphological observation, onion frozen at a faster rate showed a smaller ice-crystal size. Vitamin C content decreased upon blanching or freezing, but there was no significant difference according to freezing rate. Although free sugar content also decreased upon blanching and freezing, its highest content was at $0.7^{\circ}C/min$ freezing. Among organic acids, malic acid content was highest at $0.7^{\circ}C/min$ freezing. Based on this study, it could be suggested that a faster freezing rate is effective to improve frozen food quality in accordance with preventing tissue damage or minimizing destruction of nutrients.

Influence of Wood Decaying Fungi for Termite Ecology (목재부후균이 흰개미 서식에 미치는 영향)

  • The white-rot fungus (Trametes versicolor) and brown-rot fungus (Tyromyces palustris), which cause damage to a variety of wooden cultural properties and buildings, such as drying of the wood tissue, decay and cracks, sponge, and discoloration, give rise to serious structural and aesthetic problems. Moreover, pest termites (Reticulitermes speratus kyushuensis Morimoto) can cause damages like hollowing the outdoor beams or pillars of wooden buildings and finally causing such buildings to collapse due to the termites' destruction of the inside of the beams or pillars, leaving only a thin layer on their surfaces. This study was conducted to determine the impact of the wood-decay fungus, a representative damage-causing microorganism, and of insects and termites on the termite ecology. The damage ratio was calculated as the weight of the timber, and the overall total mass was reduced from two kinds of rot fungi. The white-rot fungi reduced the total wood mass, but the brown-rot fungi were observed to have had an increasing tendency to do so. The wood mass was measured after drying to determine the destruction capacity of termites. As a result, the wood mass consumed by the brown-rot fungi was shown to be greater.

  • To understand the properties of the cauliflower mosaic virus (CaMV) 35S promoter and the effect of the deleted maize alcohol dehydrogenase I-S (Adhl-S) intron 1 on the expression of the CaMV $35S{\beta}-glucuronidase$ (GUS) gene in potato (Solanum tuberosum L. cv. Superior), we constructed a chimeric gene and transferred it into potato with Agrobacterium tumefaciens mediated method. The pLS201, a gene transfer vector of 17.7 kilobase pairs, was composed of the CaMV 35S promoter, the 249 base pairs of deleted maize Adhl-S intron 1, the GUS reporter gene, and the kanamycin resistance gene as a selectable marker for transformation. The GUS activity was examined by histochemical and spectrophotometric assay in transformed potato plants. The GUS activity was found primarily around the vascular tissue cells in stem and root. In the spectorophotometric assay, the level of GUS activity of transgenic potato transformed with CaMV 35S/249 bp of intron 1 fragment-GUS (pLS201) was compared with that of potato transformed with CaMV 35S-GUS (pBI121). The quantitative spectrophotometric assay showed that the level of GUS activity in potato transformed with pLS201 was higher in leaf, stem and root by 30-, 34- and 42-fold, respectively than those in potato transformed with pBI121. This results indicate that the inclusion of the deleted maize Adhl-S intron 1 resulted in increament of the GUS gene expression in transgenic potato.potato.

EFFECT OF POLYPHOSPHATE IN ROOT CANAL SEALERS ON THE GROWTH OF ORAL BACTERIA (Polyphosphate가 함유된 근관충전재가 구강세균의 성장에 미치는 영향)

  • Eliminating the infecting bacteria of the root canal system and preventing reinfection must be the main objectives of all endodontic works. None of commercially available root canal sealers have the properties of desirable tissue compatibility and strong antibacterial activity. The purpose of this study is to develope an ideal root canal sealer using commercially available polyphosphate (polyP), Calgon, which is known to be antibacterial and safe. For the study. resin type AH26, zinc oxide eugenol type Tubli Seal. Ca(OH)$_2$ type Apexit as base sealers for polyP (0~3%) and para formaldehyde containing N2 as a control base were selected. Specimens (3$\times$4mm) of the sealers were prepared in a 37$^{\circ}C$ incubator for 3 and 10 days and their antibacterial activity against streptococci and black pigmented anaerobic rods was observed using an agar diffusion method. The result were as follows: 1. Among 3 day old root canal sealers. N2 as a positive control showed the strongest antibacterial effect. followed by AH26. Tubli Seal and. Apexit which barely showed antibacterial activity against the test bacteria. In contrast. 10 day old AH26 showed a greater antibacterial activity than 10 day old N2. 2. All sealer specimens showed a greater antibacterial activity against black pigmented anaerobic rods than streptococci. Three day old ones appeared to be more antibacterial than 10 day old ones except for Apexit. 3. As compared to N2, 3 day old AH26 demonstrated a similar antibacterial activity against black pig mented anaerobic rods but to a lesser extent to streptococci. Ten day old AH26 showed a greater antibacterial activity against black pigmented anaerobic rods than 10 day old N2. 4. As compared to AH26. Tubli Seal generally revealed a lower antibacterial activity but it showed a greater antibacterial activity aginst S. gordonii Challis. 5. Enhancement of antibacterial activity by polyP was more clearly observed when it was added to Ca(OH)$^{\circ}C$ based root canal sealers. Tubli Seal and N2. 6. The addition of polyP enhanced the antibacterial activity of 3 day old AH26 against S. gordonii G9B (16%) and Challis (29%), and P. gingivalis 2561 (24%) only. Moreover, polyP failed to increase antibacterial activity of 10 day old AH26 against the test strains but P. gingivalis A7A1 28(13%). 7. The addition of polyP increased the antibacterial effect of 3 day old Tubli Seal on several test bacteria including s. mutans GS 5 (50%). s. gordonii G9B (47%) and Challis (122%). and all the test strains of P. gingivalis (13~35%) except for 9 14K 1. The addition of polyP to 10 day old Tubli Seal increased antibacterial activity of the root canal sealer against most test strains. 8. 3 day old Apexit failed to show antibacterial activity. if any very little against S. mutans GS 5 and Pr. intermedia ATCC 49046. However. polyP increased its antibacterial activity by 50 and 69%, respectively. Increase of antibacterial activity of 10 day old Apexit by polyP was more clearly observed than that of 3 day old one.

Changes in Physiological and Chemical Properties of the Potatoes during Storage (감자 저장중(貯藏中)에 일어나는 생리화학적(生理化學的) 변화(變化)에 관(關)한 연구(硏究))

  • The water content, respiration rate, solid, and starch contents in the potate tuber of May Queen and Namjak stored at $5^{\circ}C$ and $20^{\circ}C$ were determined during 27 week period. Changes in appearance of the surface and internal tissue also were observed. The dormancy period of May Queen stored at $5^{\circ}C$ and $20^{\circ}C$ was 12 seeks, but Namjak at $5^{\circ}C$ and $20^{\circ}C$ were 9 weeks and 21 weeks, respectively. Greening developed from 12 weeks in the tuber of May Queen at $5^{\circ}C$ and browning of Namjak stored at $5^{\circ}C$ was observed from 6 weeks. In both potatoes stored at $20^{\circ}C$, the rates of respiration were higher than those stored at $5^{\circ}C$, and the rates increased continuously after sprouting at $20^{\circ}C$ but maintained at the low level at $5^{\circ}C$. Solid and starch contents of Namjak were always higher than those of May Queen during 27 weeks, but the levels did not change significantly with time. Solid and starch contents at $20^{\circ}C$ maintained at higher level than at $5^{\circ}C$.
