Production of monoclonal antibodies specific to the surface antigens of chicken peripheral blood mononuclear cells (닭의 혈액내 단핵세포 표면항원 특이 단클론성 항체 생산)

  • This study was performed to produce monoclonal antibodies (mAb) specifically reacting with chicken leukocyte surface antigens. Popliteal lymph node cells of BALB/c mice previously immunized through foot-pad with peripheral blood mononuclear cells (PBMC) of chickens separated by Ficoll-Histopaque method. They were fused with P3X63Ag14 mouse myeloma cells. A total of 34 hybridomas secreted antibodies specifically binding to the PBMC. According to the reactivity patterns with PBMC, the mAbs were divided into 4 groups. Group 1 mAbs (IIB3, IIB10, IIE10) specifically reacted with non-adherent lymphocytes but not with adherent cells which were mainly composed of thrombocytes and monocytes in PBMC culture. These mAbs were reactive with 25-59% of thymus cells and 42-64% of spleen cells of chickens. They did not show any significant reactivity with cells in the bursa of Fabricius, T-cell (MDCC-MSB1) and B-cell (LSCC-1104B1) lines. These results indicate that Group I mAbs specifically reacted with T-lymphocyte subpopulation. Monoclonal antibodies in Group II (IC6, IG2-2 and IID9) showed specific reactivity with monocytes but not with thrombocytes or non-adherent cells in PBMC culture. These mAbs, though not reacted with the chicken macrophage cell line, HD11, also bound to macrophages of the spleen and lung in immunohistochemical staining. Five mAbs in Group III showed characteristics of binding to lymphocytes and monocytes, but not to thrombocytes. Twenty-three mAbs in Group IV showed specific reactivity to lymphocytes, monocytes, and thrombocytes. Two mAbs (IC3 and IE9) in Group IV reacted with most of PBMC.

A Study on Blood Coagulation and Bleeding Time under Electroacupuncture Anesthesia and Medicament Anesthesia in the Dog (개의 전침마취(電針麻醉)와 약물마취하(藥物麻醉下)에서 혈액응고(血液凝固) 및 출혈시간(出血時間)에 관한 비교실험(比較實驗))

  • Blood coagulation time, bleeding time, clot retraction ability, thrombocytes counts and hematological values under electroacpuncture anesthesis and medicament anesthesia, using 10 mongrel dogs were compared. The results were summarized as follows: 1. Blood coagulation time under electroacupuncture anesthesia was shorter than that under medicament anesthesia (p<0.001). 2. Bleeding time under electroacupuncture anesthesia was shorter than that under medicament anesthesia (p<0.01). 3. Clot retraction ability under electroacupuncture anesthesia was better than that under medicament anesthesia (p<0.01). 4. Thrombocytes counts under electroacupuncture anesthesia was more increased than that under medicament anesthesia (p<0.05). 5. Erythrocytes counts, hematocrit values, hemnglobin contents and leukocytes counts were decreased (p<0.01) under medicament anesthesia in comparison with control group, but there was no significant difference under electroacupuncture anesthesia.

Evaluation of valid time for analysis of complete blood cell in pig blood using the Hemavet 950FS (자동혈구분석기 Hemavet 950FS를 이용한 돼지 혈액 내 혈구분석의 유효시간 평가)

  • This study was conducted to determine the valid time for accurate detection of complete blood cell count (CBC) in pig blood using an automatic blood corpuscle analyzer (Hemavet 950FS). Blood samples were collected from 34 pigs (Duroc) with a 60 kg (${\pm}3.5$) body weight. Ten specimens with CBC parameters in normal range and with no hemolysis were selected among 34 samples and used in this study. Regarding leukocytes parameters, white blood cell (WBC), neutrophil (NE), and lymphocyte (LY) counts showed a low daily variation (coefficient of variation, CV), whereas monocyte (MO), eosinophil (EO), and basophil (BA) CVs were significantly high (19.7, 56.9, and 53.3%, respectively). On the other hand, all parameters of erythrocytes and thrombocytes showed stable daily variation. All parameters of leukocytes and thrombocytes were significantly reduced as storage time passed (P<0.01 or 0.001), except for lymphocytes (P=0.535). However, no significant differences were observed in parameters of erythrocytes from blood up to 120 hours. From above results, we assert that Hemavet 950FS is useful in analyzing CBC, except for MO, EO, and BA. For accurate detection of leukocyte and thrombocyte parameters, analysis should be performed within 4 hours after blood collection when using Hemavet 950FS. On the other hand, parameters of erythrocytes could be stably detected for at least 120 hours after blood collection.

Pathologic Studies of Experimental Bracken Poisoning in Mice (마우스의 실험적(實驗的) 고사리 중독(中毒)에 관한 병리학적(病理學的) 연구(硏究))

  • In order to investigate the effects of bracken fern (Pteridium aquilinum) on animals, a total of 105 mice given a ration containing different percentage of bracken fern were studied pathologically during the experimental period of 90 days. The results obtained were as follow: 1. Mice in the group, given a ration containing 15% of bracken fern, showed 85% of death rate in 90 days, and mice in the group, given a ration containing 25% of it, showed 100% in 30 days 2. The experimental mice showed decreased myeloid cells and significant reduction in erythrocytes, leukocytes, and thrombocytes. 3. Severe congestion and hemorrhages were observed in lungs, liver, kidney, spleen, stomach, intestine, brain, adrenal and testis from the ones that died during the experimental period, but the necropsies of the latter period indicated mild chanties comparatively. 4. Mice in the group, given a ration containing 15% blacken fern showed a decreasing tendency in body weight.

  • Classification of peripheral leucocytes in Korean catfish (Silurus asotus) were performed. The leucocytes were classified on the basis of their morphological, cytochemieal and phagocytic characteristics. Four types of leucocytes were observed in the peripheral blood. Neutrophils were round and 12 - $15{\mu}m$ in diameter with round, oval or 2 - 3 segmented neuclei. Their cytoplam was stained whitish or grey. Lymphocytes were round and 5.5 - $8{\mu}m$ in diameter with large round neuclei which occupied most of the cells. Monocytes were round or irregular cells and 12 - $15{\mu}m$ in diameter with bean - shaped or irreular neuclei. Thrombocytes were oval or spindle - shaped cells with oval or spindle - neucle. The cells with greyish cytoplasm ranged 7.2 to $14.4{\mu}m$ in length and 5.8 to $7.2{\mu}m$ in width. Only neutrophils showed positive reaction in peroxidase and PAS. Neutrophils and monocytes showed active phagocytosis. As the results it is concluded that peripheral leucocytes of Koran catfish could be classified as lymphocytes, neutrophils, monocytes and thrombocytes.

The Comparative Study of the Effects of Fructificatio Inonoti Obliqui Aqueous Extract according to the Extraction Temperature(II) -Anti-oxidativy Activity, anti inflammatory effect and cancer cell multiplication inhibition effect- (차가버섯 물 추출물의 추출온도에 따른 효능 비교 연구(II) -항산화 효능, 소염 및 항암 효과 연구-)

  • Objectives : The present study purposed to compare the antioxidant effect, anti inflammatory effect and cancer cell multiplication inhibition effect of Fructificatio Inonoti Obliqui aqueous extract according to extraction temperature. Methods : We medicated animal models, which had experimental oxidation, with Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low temperature leachate, and performed hematological analysis and blood chemical analysis with measuring SOD, GSH, catalase, NO and MDA content in the liver. In addition, we made comparative observation of anti inflammatory effect and anti-cancer effect. Results : Compared to the control group, both the group medicated with Fructificatio Inonoti Obliqui total extract and with $50^{\circ}C$ low-temperature leachate were found to decrease the number of thrombocytes in blood plasma and NO content while to increase SOD activity and catalase activity significantly. Both groups also showed anti-inflammatory effect against THP-1 cells and a multiplication inhibition effect against liver cancer cells and stomach cancer cells significantly. Conclusions : Both Fructificatio Inonoti Obliqui total extract and Fructificatio Inonoti Obliqui $50^{\circ}C$ low-temperature leachate have significant antioxidant effect, anti inflammatory effect and anti cancer effect.

Research on Effects of Cordyceps Sinensis in Collagen Induced Arthritis Mouse-Model. (동충하초 추출물이 콜라겐으로 유발된 관절염 생쥐 모델에 미치는 영향)

  • Objectives : We examined to know the effect of Cordyceps sinensis(CS) on arthritis mouse induced by type II collagen. Methods : To analyse immunomodulatory effects of CS, arthritis index, incidence, hind paw edema, DTH, spleen weight, the number of hemocytes, and surface-receptor expression of CD4+, CD8+ and CD19+ cells in DBA/IJ mice which is experimental model of arthritis induced by collagen were measured in vivo. Results: CS reduced arthritis index, incidence, hind paw edema and DTH significantly as compared with the control group in experimental model of arthritis induced by collagen. CS enhanced the spleen weight significantly as compared with the control group but didn't enhanced the number of leukocytes and thrombocytes in experimental model of arthritis. CS enhanced the number of activating cells and surface-receptor expression of CD4+, CD8+ cells as compared with the control group but didn't enhanced those of CD19+ cells in experimental model of arthritis. Conclusion : We found out that CS may have a suppressing effect againist auto-immune disease and will be need continuous research in looking for the more effective mechanism in the future.

  • Holstein cows being raised in the Kangweon district were examined on the values of hematology in infertile cows. The results obtained were as follows; 1. Erythrocyte count was 6.23$\pm$1.49$\times$106/㎣ 2. Hemoglobin was 10.6$\pm$2.00g/100ml 3. Hematocrit was 29.68$\pm$3.27ml/100ml 4. Mean corpuscular volume was 49.4$\pm$12.1$\mu$3 5. Mean corpuscular hemoglobin was 17.3$\pm$3.73pg 6. Mean corpuscular hemoglobin concentration was 35.7$\pm$6.37% 7. Thrombocytes were 324.06$\pm$88.11$\times$103/㎣ 8. Total leukocytes was 8.70$\pm$2.23$\times$103/㎣ 9. Neutrophils, lymphocytes, monicytes, eosiniohils and basophis were 34.44$\pm$6.39, 53.82$\pm$5.84, 2.17$\pm$2.18, 9.10$\pm$3.85 and 0.50$\pm$1.80%, respectively.

Experimental Study on the Anti-Depressant Effects of Gammakdaejo-tang Complex Extracts in Rats Induced with Chronic Mild Stress (감맥대조탕 복합추출물의 만성스트레스 유발 생쥐의 항우울 작용에 관한 실험연구)

  • Objectives: To experimentally assess the anti-depressant effects of Gammakdaejo-tang Complex Extracts (GMDJ-Tang) in rats. Methods: Twenty Wistar Hannover (160~170 g) stressed rats were treated with different concentrations of GMDJ-Tang extracts i.e., 0, 62.5 mg/kg, 125 mg/kg, and 250 mg/kg. Chronic mild stress was induced by food deprivation, empty bottles, forced swimming, flickered light, tilt cages, shaking cages, high density breeding, water deprivation, and by soaking the litter cover according to fixed schedule. Blood and brain tissue samples were collected for biochemical analysis. Tests included serotonin and ELISA assays. Results: GMDJ-tang increased the weight of treated rats as well as levels of serotonin, BDNF and TrkB; however, the differences were not significant. In contrast, the extracts significantly decreased blood glucose in stressed rats. GMDJ-tang extracts did not significantly impact Serum AST, ALT, leukocytes, erythrocytes and thrombocytes when comparing treatment groups to control rats. Likewise, hemoglobin, hematocrit and PLT increased in treatment groups following treatment with GMDJ-tang, but this change was without significance. Conclusions: These results suggest that GMDJ-tang can alleviate chronic mild stress in rats, possibly through anti-depressant activity.

Effects of 1,7-Substituted Methylxanthine Derivatives on LPS-Stimulated Expression of Cytokines and Chemokines in Raw 264.7 and HK-2 Cells

  • Chronic kidney diseases are based on uncontrolled immunological and inflammatory responses to pathophysiological renal circumstances such as glomerulonephritis, which is caused by immunological mechanisms of glomerular inflammation with increased production of renal pro-inflammatory cytokines. Pentoxifylline (PTX) exhibits anti-inflammatory properties by inhibiting cytokine and chemokine production through aggregation of erythrocytes and thrombocytes. We synthesized a series of 1,7-substituted methylxanthine derivatives by the Traube purine reaction, and the formation of purine ring was completed through nitrosation, a reduction of the nitroso to the amine by catalytic hydrogenation as derivatives of PTX. Then we studied biological activities such as renal anti-inflammatory effects of the synthesized compounds in the production of cytokines such as nitric oxide (NO), interleukin (IL)-1β, IL-6, and tumor necrosis factor-α (TNF-α) and of chemokines such as monocyte chemoattractant protein-1 and IL-8 in Raw 264.7 and HK-2 cells. Renal antiinflammatory activities of this novel series of N-1 and N-7-substituted methylxanthine showed that the N-7 methyl-group-substituted analogs (S7b) showed selective 61% and 77% inhibition of the production of NO and IL-8. The other replacement of the N-1-(CH2)4COCH3 roup, as in the case of compound S6c, also showed an effective 50% and 77% inhibition of TNF-α and IL-8 production in LPS-stimulated Raw 264.7 and HK-2 cells.
