  • The effect of the weight ratio of carbon source to nitrogen source on the growth and the composition change of fatty acids of Thraustochytrium aureum ATCC 34304 was investigated. The cell concentration of 5 days' culture increased and then decreased according to the increase of C/N, The ratioes for the maximum biomass were unique respectively and distributed between 1 and 4 with the initial sugar concentration in the range of 5 g/L to 25 g/L. The biomass yield, $Y_{x/s}$ decreased along with the increase of C/N, but maintained a constant value 0.35 between 10 and 20. The composition of myristic acid $(C_{14:0})$, one component of the lipid synthesized by T. aureum, was not affected with the change of C/N, but the composition of palmitic acid $(C_{16:0})$ was around $20\%$ below 4 of C/N and decreased to $15\%$ according to C/N about 4. The compositions of oleic acid $(C_{18:1})$ and linoleic acid $(C_{18:2})$ increased from 0 to $20\%\;and\;7\%$ respectively. The composition of $\gamma-linoleic$ acid $(C_{18:3})$, however, reduced from $5\%\;to\;2\%$. EPA $(C_{20:5})$ and DPA $(C_{22:5})$ showed a tendency of reduction in the weight composition according to the increase of C/N, but DHA $(C_{22:6})$ had a trend maintaining an approximately constant value, around $40\%$, irrespective of the change of C/N.

  • Polyunsaturated fatty acids, that is PUFAs, are important constituents of membranes particularly found in the retina and central nervous system. In microorganism-based PUFAs biosynthesis, the genus Thraustochytrids is well evaluated for their potential as a promising candidate in the practical production of PUFAs, such as AA and DHA. In this study, we attempted to optimize a method of total nucleic acid extraction from this microorganism as a preliminary experiment. Using the extracted nucleic acid and degenerated primers for direct PCR, we isolated a $\Delta5$ desaturase gene that contained 1320-nucleotide and encoded 439 amino acids. This gene exhibited an expected function, when expressed in P. pastoris in the presence of appropriate exogenous substrate, as an evidence for $\Delta5$ desaturase activity (conversion of DGLA to AA). These results and information could provide a basis for the construction of engineered strains suitable for the practical production of PUFAs.

  • Studies were made on the optimization of media to c비tivate Thraustochytrium aureum A TCC 34304 for the enhanc엉d p production of linoleic acid. The medium optimization was made with the artificial sea water medium. Yeast extract, sodium g glutamate, peptone and tryptone were considered as nitrogen source. The effect of $\infty$ncentration of nitrogen source as well a as initial glucose on the production of linoleic acid were investigated to optimize the media. The maximum yield of lipid was 0 0.302 mg/g cell mass when initial glucose $\infty$ncentration was 10 g/L and sodium glutamate was used as nitrogen source, a and the yield of linoleic acid to unit cell mass was also maximum to be 8 % in that case. The highest linoleic acid c concentration was obtained in the initial glucose concentration 30 g/L regardless of the kinds of nitrogen source and the | linoleic acid concentration was 0.208 g/L when peptone was supplemented to be 2 g/L.

  • The polyunsaturated fatty acids (PUFAs) are mostly synthesized by the marine original microorganisms. In this study, the effect of concentrations of glucose, NaCl, and $MgSO_4{\cdot}7H_2O$ on both the growth and the production of lipid and Docosahexaenoic acid (DHA) was investigated using Thraustochytrium aureum ATCC 34304. $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ increased according to the increase of initial glucose concentration until 15 g/L of glucose. The maximum values of $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ were 0.18 g/g and 0.41 g/g respectively at 15 g/L of glucose. The biomass yield, $Y_{x/s}$, however, was constant as 0.378 g/g regardless of initial glucose concentrations in the range of 5 g/L to 25 g/L. The specific growth rate of T. aureum increased also with initial glucose concentration from 5 g/L and 15 g/L. The maximum value was $0.79\;day^{-1}$ at 15 g/L of glucose, and the growth rate decreased to $0.57\;day^{-1}$ and remained constant according to initial sugar concentration in the range of higher than 15 g/L of glucose. The concentration of NaCl was changed from 0 g/L to 48 g/L, in order to study the characteristics of the growth and the production of lipid and DHA according to NaCl concentration. The growth and the production of lipid as well as DHA stopped all at 0 g/L of NaCl and the maximum values of all the three variables occurred at 24 g/L. The effect of $MgSO_4{\cdot}7H_2O$ concentration was also investigated not to find the growth and the production of lipid and DHA at 0 g/L. However, the growth and the Production with $MgSO_4{\cdot}7H_2O$ concentration increased to reach the maximum values at 18 g/L of $MgSO_4{\cdot}7H_2O$.

  • The effect of rotation speed and culture temperature on the growth of T. aureum and also the biosynthesis of lipid and DHA was investigated. The optimal temperature for the growth was $32^{\circ}C$, but the best temperature for the maximum production of lipid and DHA was $18^{\circ}C$ in the range of $4^{\circ}C\;to\;39^{\circ}C$. In the case that the temperature was higher than $39^{\circ}C$ and lower than $11^{\circ}C$, the growth was very slow and the production of DHA was also very low. However, the lipid content in the biomass became higher with decrease in the culture temperature. The rotation speed for the maximum growth was 100rpm. But the growth and the production of DHA were not affected by the change of rotation speed in the range of 100 to 250 rpm. When the rotation speed was 50 rpm, the growth as well as the production of DHA and lipid was greatly reduced.
