Experimental of Cerebral Vasospasm and Measure the Mean Blood Flow Velocity in the Middle Cerebral Artery

  • To determine the appropriate concentration of papaverine hydrochloride(PPV) for therapeutic intraarterial infusion against cerebral vasospasm and to measure the mean blood flow velocity of the middle cerebral artery in rabbits. Vasospasm was induced in the experimental groups (3 days after infusion; group 1, n=3, 7 days after infusion; group 2, n=3) and a control group (n=l) by placing a blood clot in the subarachnoid space around the top of the internal carotid siphon. PPV (5 mg/kg) was infused into the internal carotid artery. The vascular diameters of the internal carotid artery (ICA) and middle cerebral artery (MCA) were measured on angiograms before and after infusion. The mean blood flow velocity in the MCA was measured on transcranial doppler sonograms before and 24 hours after infusion. After fixation, the MCA was dissected out, stained, and examined microscopically. After PPV infusion in both groups, vascular dilatation of about $20\%$ was seen. The mean increase in blood flow velocity in the group $1(30\%)$ was smaller than in the group $2(70\%)$. The mean blood flow velocity in the MCA decreased by about $30\%$ in both groups, but increased again after 24 hours nearly to the level before PPV infusion. PPV infusion may be more effective in early stages of vasospasm when vascular walls have fewer histologic changes.

Complete Recovery of Perfusion Abnormalities in a Cardiac Arrest Patient Treated with Hypothermia: Results of Cerebral Perfusion MR Imaging

  • Therapeutic hypothermia in cardiac arrest patients is associated with favorable outcomes mediated via neuroprotective mechanisms. We report a rare case of a 32-year-old male who demonstrated complete recovery of signal changes on perfusion-weighted imaging after therapeutic hypothermia due to cardiac arrest. Brain MRI with perfusion-weighted imaging, performed three days after ending the hypothermia therapy, showed a marked decrease in relative cerebral blood flow (rCBF) and delay in mean transit time (MTT) in the bilateral basal ganglia, thalami, brain stem, cerebellum, occipitoparietal cortex, and frontotemporal cortex. However, no cerebral ischemia was not noted on diffusion-weighted imaging (DWI) or fluid-attenuated inversion recovery (FLAIR) sequences. A follow-up brain MRI after one week showed complete resolution of the perfusion deficit and the patient was discharged without any neurologic sequelae. The mechanism and interpretation of the perfusion changes in cardiac arrest patients treated with therapeutic hypothermia are discussed.

영양각탕(羚羊角湯)이 뇌혈류량(腦血流量)에 미치는 영향(影響) (Effect of Yongyanggaktang(YYG) on Regional Cerebral Blood Flow in Rats)

  • Yongyanggaktang has been used in oriental medicine as a therapeutic agent for cerebral disease and eclampsia for many years. The purpose of this study was to determine the effect of Yongyanggaktang and its exception of Cornu Saigae Tataricae(羚羊角) in YYG about regional cerebral blood flow(rCBF) in rats. 1. rCBF was increased by Yongyanggaktang in dose dependently. 2. rCBF was increased by exception of Cornu Saigae Tataricae(羚羊角) in YYG at high dosages. 3. rCBF was decreased by Yongyanggaktang pretreated propranolol. 4. L-NNA did not affect rCBF. 5.ODQ did not affect rCBF. Above the results. Yongyanggaktang relates to the symphathetic action.

  • Women were used as their own controls in the comparison of presterilization and poststerilization menstrual patterns. Five parameters were studied: regularity of cycle length, duration and amount of flow and incidence of dysmenorrhea and intermenstrual bleeding. Three parameters in the electrocoagulation group (regularity of cycle length and duration and amount of flow) and one parameter in the tubal ring group (duration of flow) showed significant changes after sterilization. However, by controlling for the effect of previous contraceptive methods used, no significant menstrual pattern changes following sterilization were discerned in either technique group.

Generation and Characterization of a Monoclonal Antibody with Specificity for Mycoplasma arginini

  • Previously, we generated monoclonal antibodies (MAbs) that bound to the surface of human embryonic stem cells (hESCs) in an attempt to discover new hESC-specific surface markers. In this study, MAb 47-235 (IgG1, ${\kappa}$) was selected for further characterization. The MAb bound to the surface of undifferentiated hESCs but did not bind to mouse ESCs or mouse embryonic fibroblast cells in flow cytometric analysis. The antibody immunoprecipitated a 47 kDa protein from the lysates of cell surface-biotinylated hESCs. Identification of the protein by quadrupole time of flight tandem mass spectrometry revealed that 47-235 binds to Ag 243-5 protein of Mycoplasma arginini. BM-Cyclin treatment of the hESCs that reacted with 47-235 resulted in loss of mycoplasma DNA and the reactivity to 47-235. Nevertheless, the hESCs that were reactive to 47-235 maintained self-renewal and pluripotency and thus could be differentiated into three embryonic germ layers.

  • 본 연구는 시집에 의해서 치료 효과가 일어나는 과정을 도식화하여 현대인에게 시집이 문학치료적 효과를 어떻게 일으키고 있는지에 대해 알아보고 시집으로써 더욱 치료효과를 불러올 수 있는 방법을 찾아내기 위한 서술적 연구이다. 시집으로 묶여있는 시들은 하나의 기승전결의 흐름을 가지고 있다. 이 시들이 서로 앞뒤로 섞여 있더라도 독자의 취향에 따라 기승전결의 양식으로 독자의 인지 시스템에 시냅스 되기 때문이다. 시집은 문학치료적 전이로 구조화 되어있다. 이러한 전이의 구조는 동그라미로 현상된다. 이러한 전이적 구조 속에서 시적 내용이 긍정적이고 창조적인 것이라면 독자에게 더욱 안위감과 흥미를 발화시켜 치료효과를 상승시킬 것이기 때문에 그러한 창작품을 가지고 문학치료적 서사를 진행시키는 것이 매우 중요한 것으로 판단된다.

OIP5 is a highly expressed potential therapeutic target for colorectal and gastric cancers

  • Previously, we reported that overexpression of Opa (Neisseria gonorrhoeae opacity-associated)-interacting protein 5 (OIP5) caused multi-septa formation and growth defects, both of which are considered cancer-related phenotypes. To evaluate OIP5 as a possible cancer therapeutic target, we examined its expression level in 66 colorectal cancer patients. OIP5 was upregulated about 3.7-fold in tumors and over 2-fold in 58 out of 66 colorectal cancer patients. Knockdown of OIP5 expression by small interfering RNA specific to OIP5 (siOIP5) resulted in growth inhibition of colorectal and gastric cancer cell lines. Growth inhibition of SNU638 by siOIP5 caused an increase in sub-G1 DNA content, as measured by flow cytometry, as well as an apoptotic gene expression profile. These results indicate that knockdown of OIP5 may induce apoptosis in cancer cells. Therefore, we suggest that OIP5 might be a potential cancer therapeutic target, although the mechanisms of OIP5-induced carcinogenesis should be elucidated.

  • 본 연구는 대도시 남성노숙인 시설에서 알코올 의존 노숙인들을 위한 치료공동체 프로그램을 수료하고 현재 회복 중인 6인의 회복 체험에 대한 현상학적 연구이다. 드러난 회복의 의미는 다음과 같다. 첫째 공간적 회복이다. 이들은 자신들이 누워있던 거리를 떠나 공동체에 터를 잡으면서 자신을 바라보는 새로운 시선에 둘러싸이게 된다. 둘째, 신체적 회복으로 눕던 자세에서 벗어나 몸을 일으키면서 이들은 자신의 몸과 마음의 움직임의 주체가 된다. 셋째, 연구참여자들의 시간적 회복은 과거와 현재를 수용하고 미래를 품는 현상으로 드러난다. 넷째, 관계의 회복으로 연구참여자들은 관계망을 통해 사회 내에서 자신의 자리를 만들어 간다. 이들의 사회내에서의 자리는 지위라기보다는 관계망 속에 존재한다. 이러한 연구결과를 토대로 알코올 의존 노숙인에 대한 개입을 위한 실천 함의를 도출하였다.

폐암환자에서 치료에 대한 반응 예측지표로서의 DNA Ploidy (DNA Ploidy as a Predictive Index of Therapeutic Response in Lung Cancer)

  • 연구배경 : 수술을 시행했던 비소세포폐암에서 유체세포계산법(flow cytometry)으로 DNA ploidy를 측정했을때 aneuploid 종양이 diploid 종양 보다 생존기간이 짧은 것으로 보고되어 있는데, 소세포폐암을 포함한 폐암에서 기관지 솔질에 의한 세포학적 표본을 이용하여 측정한 DNA ploidy가 치료에 대한 반응을 잘 예측할 수 있을 것인지 알아보고자 연구를 하였다. 방법 : 기관지내시경검사시에 기관지 솔질을 해서 얻은 표본으로 DNA ploidy 검사를 시행하였고 조직학적으로 폐암으로 확진되었던 109예를 대상으로 해부적 생리적 병기와 DNA ploidy와의 관계를 검토하였고, 치료를 시작한지 8주 이상 지나서 반응을 검토할 수 있었던 58예를 대상으로 DNA ploidy에 따른 반응의 차이를 검토하였다. 결과 : 1) 세포 형태에 따라 aneuploid 혹은 고증식력(S+G2M>22%)의 발현 빈도는 차이가 없었다. 2) DNA ploidy와 해부적 생리적 병기는 유의한 관계에 없었으나 비소세포폐암에서 고증식력군때는 저증식력군에 비해 해부적 병기가 진행된 경우가 많았다(p<0.05). 3) 치료에 대한 반응은 해부적 병기에 따라 차이가 있었으며(소세포폐암 p=0.10, 비소세포폐암 p<0.005), 생리적 병기에 따라서는 비소세포폐암에서만 차이가 있었다(p<0.05). 4) DNA ploidy와 증식력에 따라서는 치료에 대한 반응에 유의한 차이는 없었다. 결론 : 기관지 솔질 표본을 이용하여 시행한 DNA ploidy 검사는 고증식력 유무가 해부적 병기와 관계가 있었으나 치료에 대한 단기 반응을 예측하는 데는 해부적 생리적 병기만큼 유용하지는 않았다.

거풍도담탕의 백서의 뇌혈류 변화에 미치는 기전연구 (A Mechanism Study of Geopungdodam-tang on the Change of Cerebral Blood Flow in Rats)

  • Geopungdodam-tang(GDT) have been used in oriental medicine for many centuries as a therapeutic agent of apoplexy. The mechanism of GDT on the cerebral blood flow is not known. The purpose of this Study was to investigate effects of GDT on the pial arterial diameter and action mechanism of GDT-induced increased regional cerebral blood flow(rCBF). The changes of regional cerebral blood f1ow(rCBF) was determinated by Laser-Doppler Flowmetry(LDF), and the changes of pial arterial diametet were determinated by video microscopy methods and video analyzer. The results were as follows ; 1. Pial arterial diameter was significantly increased by GDT in a dose-dependent manner. 2. Pretreatment with L-NNA significantly inhibited GDT induced increased rCBF. 3. Pretreatment with methylene blue significantly inhibited GDT induced increased rCBF. 4. Pretreatment with indomethacin inhibited GDT induced increased rCBF. These results suggest that GDT causes a diverse response of cerebral hemodynamics(rCBF and pial arterial diameter). The cerebral hemodynamics is also mediated by nitric oxide synthase, cyclic GMP(guanylyl cydase) and prostaglandin(cyclooxygenase).
