Changes in Quality of King Oyster Mushroom (Pleurotus eryngii) during Modified Atmosphere Storage (큰느타리버섯의 MA저장중 품질변화)

  • In order to study the effect of modified atmosphere storage on extending shelf life of King Oyster mushroom was wrapped with PVC film and packed with 20$\mu\textrm{m}$polyolefin(PD941), and effects of temperature(0, 5, 10$^{\circ}C$) in packaging conditions on the respiration and keeping qualities were evaluated. Higher respiratory activity and weight loss were observed at higher temperature. The concentration of O$_2$and CO$_2$ of PVC wrap and polyolefin(PD941) packages for all showed 1∼2% and 10∼14%, respectively. Polyolefin(PD941) package wan superior to the PVC wrap packaging method in Hardness, Hunter L value, Hunter b value and sensory qualities, and reducing weight loss at 0$^{\circ}C$, 5$^{\circ}C$ and 10$^{\circ}C$ compared to PVC wrap. It was found that the optimum shelf-life period of King Oyster mushroom packaged by PVC wrap was estimated to be 50, 28 and 12 days at 0, 5 and 10$^{\circ}C$, respectively, and 50, 32 and 21 days in Polyolefin(PD941).

Study on roof tiles of Iksan featuring Stamped-Roof Tiles (인각와를 통해 본 익산의 기와에 대한 연구)

  • Stamped-Roof Tiles, the characteristics of Baekje Tiles are one of the most frequently excavated tiles in Iksan. Considering the types and amount of excavation of the Stamped-Roof Tiles unearthed in Iksan area, it seems that these are closely related with a Royal family. The tiles might be manufactured by a particular gorup of file maker since they are produced with same way. Referring the Ganji carved in the Stamped-Roof Tiles, I suppose that the tiles were produced and used in two time periods, the time around enthronement of The King Mu and the second quarter of the 7th century around Gichuk, AD. 629. In addition, Convex Tiles used in Wanggung-ri Site and Mireuk Site was also produced in same time as the Stamped-Roof Tiles, considering there pattern and the amount of excavation. The social background of tile production was analogized based on the technology and the time of the tile manufacture as well as the characteristics of the Stamped-Roof Tiles which represent the Obu and the Ohang. The production of the tiles was supported by Obu which aided the King Mu-oriented construction project the economic capacity of the Royal family was not enough to promote. The tile production system was totally controled by the Royal family and the inspector, neither producer nor donator, examined the quantitation of the tiles and generated the imprint of a seal with the Stamped-Roof Tiles.

A Comparative Study on Mu-Gwan(武冠) of Koguryo and China (고구려와 중국의 무관(武冠) 비교 연구)

  • Mu-Guan(武冠) is a headgear of northern nomadic people's costume, Ho-Bok(胡服). It became a part of Chinese(中原) Costume through the adoption of Ho-Bok(胡服) by King Muryong(武寧王) of Kingdom of Cho(趙). Chinese(中原人) did not use it as the formal costume of high class but the Costume military or low-level classes because Chinese(中原人) reguad it as a practical costume only for low-level classes. In this process, Mu-Guan(武冠) and it's clothes had got changed. It became high in shape by an influence Hsien-pi(鮮卑族)'s headgear and became generous in it's going with clothes for Chinese(中原) Costume. It needs to attend that Mu-Guan(武冠) could be found in Koguryo(高句麗)'s mural paintings. Mu-Guan(武冠) of Koguryo(高句麗) had kept the same pattern from middle of 4C to late of 5C. Actually Koguryo(高句麗)'s Mu-Guan(武冠) was similar with Han(漢) dynasty's, but It was quite different from Qin(晉) or Wei(北魏) dynasty's which belong to same period with Koguryo(高句麗)'s. It is possible to guess at Koguryo(高句麗) advanced Mu-Guan(武冠) as their own. Koguryo(高句麗) could be aware of 'Changed Mu-Guan(武冠)'. Because there are frequent diplomatic event between Koguryo(高句麗) and Chiese Dynasties, Barbarian Dynasties. Moreover the guess have persuasion by the existence of 'Changed Mu-Guan(武冠)'in Jee-an(集安) district's mural painting as a Costume of holy person. In brief, It could be noticed that Koguryo(高句麗)'s Mu-Guan(武冠) is distinguished from Chinese and Barbarian dynasties' and Koguryo(高句麗) advanced Mu-Guan(武冠) in their own style.

Detection of Babesia spp. in Free-Ranging Pukus, Kobus vardonii, on a Game Ranch in Zambia

  • Babesia spp. were detected from 4 asymptomatic pukus captured on a game ranch in central Zambia in October 2008. Blood smears were examined in 4 species of aymptomatic free-ranging antelopes, namely the puku (Kobus vordanii), reedbuck (Redunca arundinum), bushbuck (Tragelaphus sylvaticus), and kudu (Tragelaphus strepsiceros), and showed the presence of Babesia parasites only in the puku. In the puku, the prevalence of babesiosis was estimated at 33.3% (n=12), while the overall prevalence in all examined animals was 8.5% (n=47). The parasites showed morphological characteristics of paired ring-like stages with the length varying between $1.61{\mu}m$ and $3.02{\mu}m$ ($mean=2.12{\mu}m$, n=27; $SD=0.76{\mu}m$). Both the infected and non-infected pukus showed good body condition scores (BCS), while the dominant tick species detected from all animals were Rhipicephalus appendiculatus, Rhipicephalus spp., and Boophilus spp. To our knowledge this is the first report of Babesia spp. infection in pukus in Zambia. These findings suggest that wildlife could play an important role in the epidemiology of babesiosis in Zambia.

Dibucaine Inhibition of Serum Cholinesterase

  • The dibucaine number (DN) was determined for serum cholinesterase (EC 3.1.1.8, SChE) in plasma samples. The ones with a DN of 79-82 were used, because they had the "usual" SChE variant. The enzyme was assayed colorimetrically by the reaction of 5,5'-dithiobis-[2-nitrobenzoic acid] (DTNB) with the free sulfhydryl groups of thiocholine that were produced by the enzyme reaction with butrylthiocholine (BuTch) or acetylthiocholine (AcTch) substrates, and measured at 412 nm. Dibucaine, a quaternary ammonium compound, inhibited SChE to a minimum within 2 min in a reversible manner. The inhibition was very potent. It had an $IC_{50}$ of $5.3\;{\mu}M$ with BuTch or $3.8\;{\mu}M$ with AcTch. The inhibition was competitive with respect to BuTch with a $K_i$ of $1.3\;{\mu}M$ and a linear-mixed type (competitive/noncompetitive) with respect to AcTch with inhibition constants, $K_i$ and $K_I$ of 0.66 and $2.5\;{\mu}M$, respectively. Dibucaine possesses a butoxy side chain that is similar to the butryl group of BuTch and longer by an ethylene group from AcTch. This may account for the difference in inhibition behavior. It may also suggest the existence of an additional binding site, other than the anionic binding site, and of a hydrophobic nature.

Sclerotium Rot of Cowpea (Vigna sinensis King) Caused by Sclerotium rolfsii (Sclerotium rolfsii에 의한 동부 흰비단병)

  • Sclerotium rot disease on cowpea (Vigna sinensis King) was observed in the exhibition field of Gyeongsangnam-do Agricultural Research and Extension Services in September 2015. Lesions were covered by white mycelial mats, and numerous sclerotia were formed on the stem near the soil line. The sclerotia were globoid in shape, 1~3 mm in size and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation on potato dextrose agar (PDA) was $30^{\circ}C$, with the hyphal width of $4{\sim}8{\mu}m$. For molecular identification, the complete internal transcribed spacer (ITS) rDNA region of the causal fungus was sequenced and analyzed. Based on the mycological characteristics, ITS rDNA sequence analysis, and pathogenicity test, this fungus was identified as Sclerotium rolfsii. This is the first report of sclerotium rot on cowpea caused by S. rolfsii in Korea.

  • 독사 또는 곤충의 독 30여종을 대상으로 SNU-1 위암세포에 대하여 MTT test를 실시한 결과 세포 독성 활성이 제일 높은 킹코브라(Ophiophagus hannah)의 venom을 가지고 세포 독성 물질을 정제하였다. Gel Filtration Chromatography와 Anion Exchange Chromatography로 정제한 4번째 peak만이 MTT/SRB test결과 IC$_{50}$ value 0.947$\mu\textrm{g}$/ml이었다. 음이온 교환 크로마토그라피로 정제한 단백질을 PRO-RPC로 더 분리하여 순수한 단일성분을 얻었으며 맹장암, 대장암, melanoma, fibrosarcoma 세포에 대해 독성을 확인하였고, 광학 및 전자 현미경에 의해 암세포의 분화와 성장이 억제됨을 재확인하였다. 또한 thymidine uptake asaay에서 암세포의 증식이 억제되었고, 또한 EDTA, $Zn^{++}$, $Ba^{++}$ 첨가로 세포 독성 활성이 증가되었다. Detection of BCR/ABL Fusion Gene by Hematological and Cytogenetical Analysis in Chronic Myeloid Leukemia Patients in Quetta, Pakistan

  • Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of pluripotent stem cells, caused by reciprocal translocation between the long arms of chromosomes 9 and 22, t(9;22)(q34;q11), known as the Philadelphia chromosome. Materials and Methods: A total of 51 CML patients were recruited in this study. Complete blood counts of all CML patients were performed to find out their total leukocytes, hemoglobin and platelets. FISH was performed for the detection of BCR-ABL fusion and cryptogenic tests using bone marrow samples were performed for the conformation of Ph (9;22)(q34;q11) and variant translocation mechanisms. Results: In cytogenetic analysis we observed that out of 51 CML patients 40 (88.9%) were Ph positive and 4 (8.88%) had Ph negative chromosomes. Mean values of WBC 134.5 $10^3/{\mu}l$, hemoglobin 10.44 mg/dl, and platelets 288.6 $10^3/{\mu}l$ were observed in this study. Conclusions: In this study, Ph positive translocation between chromosome (9:22)(q34;q11) were observed in 40 (88.9%) CML patients.

Selection of indigenous starter culture for safety and its effect on reduction of biogenic amine content in Moo som

  • Objective: The aims of this study were to select one strain of Lactobacillus plantarum (L. plantarum) for a potential indigenous safe starter culture with low level antibiotic resistant and low biogenic amine production and evaluate its effect on biogenic amines reduction in Moo som. Methods: Three strains of indigenous L. plantarum starter culture (KL101, KL102, and KL103) were selected based on their safety including antibiotic resistance and decarboxylase activity, and fermentation property as compared with a commercial starter culture (L. plantarum TISIR543). Subsequently, the effect of the selected indigenous safe starter culture on biogenic amines formation during Moo som fermentation was studied. Results: KL102 and TISIR 543 were susceptible to penicillin G, tetracycline, chloramphenicol, erythromycin, gentamycin, streptomycin, vancomycin, ciprofloxacin and trimethoprim (MIC90 ranging from 0.25 to $4{\mu}g/mL$). All strains were negative amino acid-decarboxylase for lysis of biogenic amines in screening medium. For fermentation in Moo som broth, a relatively high maximum growth rate of KL102 and TISIR543 resulted in a generation time than in the other strains (p<0.05). These strain counts were constant during the end of fermentation. Similarly, KL102 or TISIR543 addition supported increases of lactic acid bacterial count and total acidity in Moo som fermentation. For biogenic amine reduction, tyramine, putrescine, histamine and spermine contents in Moo som decreased significantly by the addition KL102 during 1 d of fermentation (p<0.05). In final product, histamine, spermine and tryptamine contents in Moo som inoculated with KL102 were lower amount those with TISIR543 (p<0.05). Conclusion: KL102 was a suitable starter culture to reduce the biogenic amine formation in Moo som.

Isolation and characterization of Bacillus amyloliquefaciens TK3 inhibiting causative bacteria of atrophic rhinitis and fowl typhoid (돼지위축성비염과 가금티푸스 병원균을 저해하는 Bacillus amyloliquefaciens TK3의 분리 및 특성 조사)

  • For prevention of atrophic rhinitis of swine by Bordetella bronchiseptica and fowl typhoid by Salmonella gallinarum, bacterial strains showing antimicrobial activity against those pathogenic bacteria were isolated from various samples collected at animal farms. Among 372 bacterial isolates strain TK3 showed the highest antibacterial activity against both pathogens, and was identified as Bacillus amyloliquefaciens by 16S rRNA gene sequence analysis. B. amyloliquefaciens TK3 could inhibit growth of both pathogens by secretion of antibacterial compounds such as siderophore, rhamnolipid and antimicrobial peptide. Production radius of siderophore on Chrome azurol S agar plate by strain TK3 was 0.53 cm after 14 days of incubation, and concentration of siderophore in King's B medium was 1.06 mmol/ml. It also secreted 82.4 mg/L of rhamnolipid, and antimicrobial peptide that completely inhibited growth of both pathogens at concentration of $30{\mu}l/ml$ in LB medium.
