This paper reports that the ultrastructural nature of the interface process between the implants and surrounding bone has been studied after in vivo 1, 4, 8, 12 weeks of implantation of smooth machined implants into rabbit tibias. There was no indication of the fibrous connective tissue formation around the implant that imply intolerance of the bone tissue towards the implant after 1 week of implantation. The regions showing direct bone tissue bonding to the smooth machined implant contained osteoblast activating across the interface in the direction after 4 weeks of implantation. The reaction of a smooth machined implant caused in the first instance formation of an amorphous woven bone, which transformed into a mineralized bone containing collagen fibers. After 8 weeks of implantation, the activities of osteoblast initiated osseointegration forming bone matrix at the interface. During this period, the osteoblast surrounded with a matrix consisting of collagen bundles running in various directions. In the interface area between newly formed bone tissue and implants which has been inserted in rabbit tibias for 12 weeks, the implant and mineralized bone was separated by an amorphous electron dense material layer about $1{\sim}1.5{\mu}m$ in thickness.
Objectives : To evaluate the DNA damage by hair dyeing in human lymphocytes Methods : Comet assays were carried out to evaluate the DNA damage in lymphocytes by hair dyeing. Twenty subjects were selected from women volunteers whose age ranged from 55 to 67 year old. All subjects had no smoking history. Blood samples were collected before and 6 hours after hair dyeing. DNA damage was evaluated by means of the tail moments, which were quantified by a KOMET 4.0 image analysis system. Results : The tail moments before hair dyeing showed no significant differences among subjects except for the high frequency group. The mean values of the tail moments in subjects with low and high frequencies of hair dyeing were 1.39 and 1.77, respectively (p<0.05). The tail moments after hair dyeing increased significantly, The mean values of tail moments in subjects before and after hair dyeing were 1.45 and 1.79, respectively (p<0.01). However, the difference levels of DNA damage in lymphocytes before and after hair dyeing were found to be slightly lower in both the dietary supplement taking group and high frequency group. Conclusions : The high frequency group appears to have a higher level of DNA damage than the low frequency group before hair dyeing. DNA damage in lymphocytes was found to be significantly higher in the volunteers after hair dyeing. In this study, the related factors such as high frequency and taking dietary supplements appeard to reduce DNA damage in lymphocytes after hair dyeing.
The protective effects of onion extract (OE), onion powder extracted in ethanol for 2 days. on carbon tetrachloride ($CCl_4$)-induced hepatotoxicities and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with OE prior to the administration of $CCl_4$ significantly reduced the increase in serum alanine and aspartate aminotransferase activities and hepatic lipid peroxidation in a dose-dependent manner. In addition, pretreatment with OE significantly prevented the depletion of reduced glutathione content in the liver of $CCl_4$-intoxicated mice. $CCl_4$-induced hepatotoxicity was also prevented, as indicated by a liver histopathologic findings. The effects of OE on the cytochrome P450 (P450) 2E1, the major isozyme involved in $CCl_4$ biotransformation were investigated. Treatment of mice with OE resulted in a significant decrease in P450 2E1-dependent p-nitrophenol and aniline hydroxylation in a dose-dependent manner. Consistent with these observations, the P450 2E1 expressions were also decreased, as determined by immunoblot analysis. OE also exhibited antioxidant effects in FeCl$_2$-ascorbate induced lipid peroxidation in rat liver homogenates and in superoxide radical scavenging activity. These results show that the protective effects of OE against the $CCl_4$-induced hepatotoxicity may be due to its ability to block bioactivation of $CCl_4$, mainly tty inhibiting the expression and activities of P450 2E1 and by scavenging free radicals.
This thesis aimed to determine the effecting factors(eating pattern, working condition, stress, health care) that help maintain the health of the cosmetologists' digestive system and the diseases involved (acid reflux, indigestion, gastritis, constipation & diarrhea). The research methods included survey and statistical analysis. The survey was conducted on 242 cosmetologists from August 30 to October 30 2014. The data analysis included frequency, cross table, ${\chi}^2$-test, and regression with SPSS(V. 14). The results were as follows ; (1) The cosmetologists' health of digestive system and eating pattern are related. Regular and enough meal times cause less digestive disease. Acid reflux, indigestion, gastritis, constipation and diarrhea are differently related with the type of usual eating pattern, especially, instant food is not good for digestive health. (2) The cosmetologists' health of digestive system and working condition are related. Longer daily working hours and exposure time to chemical odors are more likely to be associated with digestive diseases; whereas, longer time of standing and talking with colleagues are less likely to be associated with digestive diseases. (3) The cosmetologists' health of digestive system and stress are related. Headache, boredom, conflict of pay and compensation, and insomnia are not good for digestive health. (4) The cosmetologists' health of digestive system and health care are related. Periodical medical examination and usual body stretching are correlated with digestive diseases; whereas, people who do regular exercise and bowel movement are less likely to have digestive diseases.
Manganese (Mn) is used as main materials in various chemical processes of industry, but it suggested that Mn brings about its toxicant by fume or dust through respiratory system and skin barrier. Mn toxicant induces the loss of mental health and life quality by cerebrovascular and skin diseases. Nevertheless, it lefts much unknown on the mechanism and the effectively therapeutic methods about Mn toxicant. Therefore, this study was evaluated the cytotoxicity induced by manganese dioxide ($MnO_2$) in cultured NIH3T3 fibroblasts, and also, the correlation between $MnO_2$-induced cytotoxicity and oxidative stress was examined. While, the effect of Eclipta prostrata L. (EP) extract belong to Compositae was assessed against $MnO_2$-induced cytotoxicity in the view of antioxidative effect for searching the natural resources mitigating or preventing the $MnO_2$-induced cytotoxicity. In this study, $MnO_2$-induced cytotoxicity was revealed as mid-toxic by Borenfreud and Puerner's toxic criteria, and catalase (CAT), an antioxidant prevented $MnO_2$-induced cytotoxicity by the remarkable increase of cell viability in these cultures. While, in the protective effect of EP extract on $MnO_2$-induced cytotoxicity, EP extract effectively prevented the cytotoxicity induced by $MnO_2$ via antioxidative effects such as xanthine oxidase (XO) inhibitory ability and DPPH-radical scavenging ability. From the above results, EP extract showed the effective prevention against $MnO_2$-induced cytotoxicity correlated with oxidative stress by antioxidative effects. Conclusively, this study may be useful to research or development the alternatively therapeutic agent from natural resources like EP extract for the treatment of diseases resulted in oxidative stress.
The morphological characteristics of the cut sections of hair using razor, clipper and scissors were investigated with scanning electron microscopy. With the razor, the cutting section had a long elliptical form and a rhythmical ribbon shape. It was curved outward since the end part of it became thinner. With the scissors, the cutting section had a flat, circular form. It seemed to be because the scissors imposed a constant strength on the hair. With the clipper, the cutting section had also a circular form but a layer of the macrofibril was generated in the middle part of the cortex. It seemed to be generated while the rapidly moving blades of the clipper cut the hair in an instant.
Computerized image processing and analysis system was used for quantitative assessment of skin tissue components in color histological sections. The purpose of this study was to determine that the changes in the collagen fiber density and elastic fiber density in dermis in the rat skin as aging. And also to determine that the correlation between fibroblast density and collagen fiber density and elastic fiber density in the aged rat skin. Ten weeks old ($130{\sim}150g$) eight and fifty-four weeks old ($300{\sim}350g$) eight female Sprague-Dawley rats were used. The full-thickness skin biopsy specimens were prepared serial sections and stained with hematoxylin and eosin, Masson's trichrome and Verhoeff-van Gieson. The collagen fiber and the elastic fiber were identified using the image analysis processing system and then calculated the collagen fiber density rate and the elastic fiber density rate in the dermis. It also identified fibroblast and calculated fibroblast density in the dermis. By using a Student's t-test, a decrease in the collagen fiber density rate (t=-4.650, P<0.001) and the elastic fiber density rate (t=-6.494, P<0.001) of dermis can be observed in aged rats as compared with the young rats. A Student's t-test showed a significantly less fibroblast density in the aged rats than the young rats (t=-4.497, P<0.001). There were significantly positive correlation between the fibroblast density and the collagen fiber density rate (r=.69, P<0.001) and the elastic fiber density rate (r=.91, P<0.001). These results indicate that the aging may decrease the collagen fiber density and elastic fiber density due to reduced the proliferative and synthetic activity of fibroblast in the dermis.
To construct an amylolytic industrial strain of Saccharomyces cerevisiae, the glucoamylase cDNA gene (GAl) from Aspergillus awamori was expressed under the control of the alcohol dehydrogenase gene promoter (ADC1p) and integrated into the chromosomes of industrial S. cerevisiae. An integrative cassette lacking bacterial ampicillin resistance gene but containing the GA1 gene, $\delta$ sequences of Ty1 retrotransposon as target sites for homologous recombination and S. cerevisiae aureobasidin A resistance gene (AUR1-C) as the selection marker was constructed to obtain a strain eligible for commercial use. Industrial S. cerevisiae transformed with this 15-integrative cassette efficiently secreted glucoamylase into the medium and grew on starch as the sole carbon source. The transformants were mitotically stable for 100 generations in nonselective medium.
In this study, we investigated tensile properties and ultrastructural changes of straight permed hair using the rheometer and the scanning electron microscopy. First, we compared the morphological characteristic between the virgin hair and the straight permed hair after testing tensile strength. Cuticle cells were heavily lifted off in straight permed hair than in the virgin hair. Cuticle cells were separated by the destruction of intercellular membrane complex and no destruction or damage were found in cytoplasm. In the comparative test for tensile characteristic between the virgin hair and the straight permed hair, tensile distance of the straight permed hair was decreased by 24.5% or 3.05 mm than the virgin hair. The tensile strength was decreased by 34.63% or $5.62\;g/cm^2$ and the maximum stress by 34.59% or 56.12 g. As a result, the tensile property dropped to the lowest level with the straight permed hair than with the bleached hair or the permanent dyed hair of previous studies.
The purpose of this study was to examine the effect of vanillic acid on the cell viability and melanogenesis in melanocytes damaged by reactive oxygen species (ROS). The human skin melanoma cells (SK-MEL-3) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cell viability for $H_2O_2$-induced cytotoxicity or vanillic acid against $H_2O_2$ was measured by XTT assay in these cultures. For the effect of vanillic acid on the melanogenesis, the tyrosinase inhibitory activity was measured by colorimetric assay at a wavelength of 490 nm, and melanin synthesis activity were assessed after cells were cultured in the media with or without various cencentrations of vanillic acid. In this study, $H_2O_2$ decreased cell viability dose- and time-dependent manners and $XTT_{50}$ was determined at a concentration of 80 ${\mu}M$, $H_2O_2$. Vanillic acid increased the cell viability dose dependently in human skin melanoma cells damaged by $H_2O_2$-induced cytotoxicity. In the tyrosinase inhibitory activity, vanillic acid supresssed tyrosinase activity in dosedependent manner, and also decreased significantly melanin synthesis activity compared with $H_2O_2$-treated group. From these results. It is suggested that $H_2O_2$-mediated cytotoxicity was highly by the toxic criteria of Borenfreund and Puerner and also, vanillic acid has the protective effect on ROS-induced cytotoxicity and melanogenesis in these cultures.
