AN EXPERIMENTAL STUDY ON EFFECT OF RADIATION IN PALATE DEVELOPMENT OF RAT EMBRYO (방사선조사시 태내백서의 구개형성기에 미치는 영향에 관한 실험적 연구)

  • The author observed morphological change in palate development of rat embryo after irradiation of x-ray on the one side of the duplex uterus. The time-matings occured between 6 p.m. and 8 p.m. and all females with copulation plugs at 8 a.m. were isolated and properly marked for evidence of copulation. The lower left abdomen of mothers were exposed to x-radiation on the 7 1/2th, 9 1/2th, 11 1/2th day of gestation, respectively 150, 250, 350, 500rads. At 18 1/2th day of post-conception, the pregnant females were dissected and the contents of the two uteri examined. The translucent sample by Alizarin red S stain were prepared. The results were as follows; 1. The result that groups irradiated by 250rads and 350rads made marked difference in comparison with the control group suggests the x-ray to be a inducing factor of cleft palate. 2. At 11 1/2th day of gestation, incidence of cleft palate induced by x-irradiation was highest. 3. Mortality showed the highest frequency at 7 1/2th day of gestation and tended to decrease in according to increasing of age. 4. Morphology of cleft palate induced by x-irradiation showed similarity in comparison with those induced by other factors having reported ever.

Effects of Gagambojungikgi-tang on the Immune Mediators Regulation and Wound Healing in the rSj26 or Chemical Antigen induced Atopic Dermatitis Model Mice (rSj26 또는 화학 항원 유도 아토피 피부염 마우스 모델에서 가감보중익기탕의 면역 매개물 조절 및 상처치료 효과)

  • Objectives : The aim of this study was to investigative the effects of Gagambojungikgi-tang (GBT), a Korean herbal medicine, on the immune mediators, T cell proliferation and wound healing in the recombinant Sj26 (rSj26) antigen induced atopic dermatitis(AD) model mice. Methods : GBT is the water extracts prepared from mixture of Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Atractylodes Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisia iwayomogi Herba, Scutellaria Radix, Lonicera japonica Flos. This is a modified prescription of Bojungikgi-tang, which has been used for the treatment of indigestion, and immunological disease in east-asian countries. GBT was orally administered or externally applied at difference doses. The levels of immune mediators [(IgE, IgG1, prostaglandin E2 (PGE2), Th1/Th2 cytokines], T cell proliferation, and wound healing in the rSj26 or chemical antigen induced AD model BALB/c were investigated. Results : GBT dose-dependently suppressed the release of TNF-${\alpha}$, IL-$1{\beta}$ (Th1 cytokines), IL-4, IL-10 (Th2 cytokines), PGE2 (inflammatory mediators) and T cell proliferation. But GBT increased the production of IFN-${\gamma}$ (Th1 cytokine). Furthermore, A wound healing effect of GBT was similar to external application of dexamethasone. Conclusions : These results suggest that GBT suppresses the inflammatory mediators and regulates the Thl/Th2 cytokines, and promotes the wound healing. Therefore, these properties may contribute to the strong anti-AD effect of GBT.

Analysis of Bicycle Crossing Times at Signalized Intersections for Providing Safer Right of Bicycle Users (자전거 이용자 행태 반영을 위한 신호교차로에서의 자전거 횡단시간 연구)

  • When allocating traffic signal at the signalized intersection, minimum green time and clearance time for bicyclists should be significantly considered in order to enhance safety aspects to bicyclists when crossing intersections, especially where intersections with exclusive bicycle paths that are physically separated from pedestrians. In this study, field measurements related to bicycle crossing time, including minimum peen time and clearance time, were collected and analyzed according to bicycles crossing types at the signalized intersections where high rate of bicyclists exists. Three types of bicycle crossing are defined as follows 1) stopping: completely stop before crossing (at least one foot on found) 2) riding: crossing with riding bicycle 3) pulling: crossing without riding bicycles. Minimum green time based on pedestrian speeds should be used as crossing time in this case. For bicyclists, speed of bicycle that is applicable to estimate the minimum green time is in the 1.36m/sec($15^{th}$ percentile) to 1.60m/sec($25^{th}$ percentile) range in case of its stopping. Also it is in the 0.75($15^{th}$ percentile) to 0.87($25^{th}$ percentile) range for pulling at crosswalk. In addition, speed of bicycle to consider for calculating the clearance time is in the 2.51m/sec($15^{th}$ percentile) to 2.79m/sec($25^{th}$ percentile). These values also resulted from $15^{th}$ percentile or $25^{th}$ percentile speeds of riding. The results of this study are expected to be supported in traffic signal allocation process, reflecting bicyclists' characteristics.

Lutein Modulates Th2 Immune Response in Ovalbumin-Induced Airway Inflammation (Ovalbumin으로 유도한 천식 생쥐모델에서 lutein의 Th2 면역반응 연구)

  • The general term flavonoids is often used to categorize a family of natural compounds that are highly abundant in all higher plants, and which in recent years have attracted scientific interest as therapeutics. Lutein is a xanthophyll and one of 600 known naturally occurring carotenoids. It is found in green vegetables such as spinach and kale, and has been demonstrated to exert anti-inflammatory activities. However, its anti-allergic effect in the Th1/Th2 immune response is poorly understood. In this study, we attempt to determine whether lutein regulates inflammatory mediators in an ovalbumin (OVA)-induced murine asthma model. To address this, mice were sensitized and challenged with OVA, and then treated with lutein before the last OVA challenge. Administration of lutein significantly suppressed the OVA-induced airway hyper-responsiveness. It also resulted in a significant alleviation of the infiltration of inflammatory cells into the bronchoalveolar lavage. Additionally, lutein attenuated the increased expression of Th2 responses in OVA-challenged mice. These results demonstrate that lutein is a potent inhibitor that reduces Th2 immune responses. Furthermore, they show that the immunopharmacological function is mediated by a pathway that involves and is regulated by Th2 immune response.

Prenatal Development of Tvrosine Hlrdroxvlase-Containing Neurons in the Rat Brain (흰쥐 태아 뇌에서 TH 신경세포의 초기발생과정)

  • The present study attempts to explore the first appearance 8nd subsequent development of %cosine hvdroxvlase (TH)-containing neurons in the rat brain from embryonic day (E) 10.5 to the neonate. To increase the senti노ivity and resolution power, a double bridge peroxidase-anti-peroxidase technique was employed for cellular localization of TH. In situ hybridization histochemistw with synthetic TH oligomer (30-mer) codinB TH was also used to detect TH mRNA. TH-containing neurons were first detected at E11.5 in the intermediate zone of prosencepha1on and mesencephalon. At this stage, TH-immunoreactive neurons were small, ovoid bee and emitted their fibres into their immediate surroundings. From this stage, TH-immunoreactive neurons increased in their number and underwent migration and cell differentiation. At E15.5, the distribution pattern of the maior groups of TH neurons was similar to that of adult catecholaminergic groups, and at E19.5 the external laver of median eminence showed TH-immunoreactive processes.

  • In this paper, we generalize the Catalan number to the (n, k)-th Catalan numbers and find a combinatorial description that the (n, k)-th Catalan numbers is equal to the number of partitions of n(k-1)+2 polygon by (k+1)-gon where all vertices of all (k+1)-gons lie on the vertices of n(k-1)+2 polygon.

Effects of Taraxaci Herba on Cell Death in Breast Cancer Cells (포공영(蒲公英)이 유방암 세포의 사멸에 미치는 영향)

  • Objectives : This study was designed to investigate the effects of Taraxaci Herba (TH) on cell death in breast cancer cells. Methods : In this experiment, the effects of TH on proliferation rates, cell morphology and growth pattern, intracellular reactive oxygen species (ROS) production. In addition, the effects on nuclear condensation, fragmentation and formation of acidic vesicular organelles (AVO) in MCF-7 cells were also investigated. Finally, autophagy related with protein was observed by using western blot method. Results : TH inhibited proliferation of MCF-7 cells, TH elevated intracellular ROS levels significantly. Treatment with TH did not affect nuclear morphologies such as condensation or fragmentation. On the other hand, TH treatment effectively induced AVO. Finally, one of autophagy related with protein, Microtubule-associated proteins 1A/1B light chain 3A (MAP1LC3A, LC3) level was elevated by treatment with TH. Conclusions : These data indicate that TH is able to be used for patient with breast cancer and mechanisms are involved in autophagy through ROS generation.

THE INFLUENCE OF ABUTMENT SCREW LENGTH AND REPEATED TIGHTENING ON SCREW LOOSENING IN DENIAL IMPLANT (치과용 임플랜트에서 지대주 나사의 길이 및 반복 조임 횟수가 지대주 나사의 풀림에 미치는 영향)

  • Statement of problem: One of common problems associated with dental implant is the loosening of abutment screws that retain the implants. Purpose : This study was performed to investigate the influence of abutment screw length and repeated tightening on screw loosening in dental implant. Material and method: Forty nine Hexplants (13mm length, 4.3mm diameter, Ti grade IV, Warantec. Co. Ltd. Seongnam, Korea) and cementation type abutments(straight abutment) and abutment screws (0.4mm/pitch) were divided into 7 groups, depending on abutment screw length. Each implant and abutment was tightened to 30Ncm by torque controller(MGT50, MARK-10 Inc., USA) and the removal torque values were measured during 10 consecutive closure/opening trials. Results and Conclusion: The results of comparing the removal torque value are as follows : 1. There is no significant difference in the removal torque value between groups in 10 consecutive closure/opening trials (p = 0.97). 2. If the fractured abutment screw is engaged in longer than 2.425 thread length, there is no significant difference in the preload between the fractured abutment screw and the new abutment screw when both are equally tightened to 30 Ncm. 3. The removal torque value in the 1st trial(24.510 Ncm) was lower than that in the 2nd, 3rd, 4th, 5th, 6th, 7th trials and the removal torque value in the 2nd trials(25.551 Ncm) was maximum and was decreased in 1311owing trials. The removal torque value in the 1st trial was significantly lower than that in the 2nd, 3rd, 4th trials and was significantly higher than that in the 8th, 9th, l0th trials(p<0.05). 4. In the 2nd, 3rd, 4th, 5th, 6th, 7th trials, the abutment screw was mainly influenced by settling effect and the higher preload was obtained In the 8th, 9th, l0th trials, the abutment screw was mainly influenced by adhesive wear and the progressively lower preload was obtained.

Inhibitory Effects of Tualang Honey on Experimental Breast Cancer in Rats: A Preliminary Study

  • The study was conducted to determine the effect of Malaysian jungle Tualang Honey (TH) on development of breast cancer induced by the carcinogen 7,12-dimethylbenz(${\alpha}$)anthracene (DMBA) in rats. Forty nulliparous female Sprague-Dawley rats were given 80 mg/kg DMBA then randomly divided into four groups: Group 1 served as a Control while Groups 2, 3 and 4 received 0.2, 1.0 or 2.0 g/kg bodyweight/day of TH, respectively, for 150 days. Results showed that breast cancers in the TH-treated groups had slower size increment and smaller mean tumor size (${\leq}2cm^3$) compared to Controls (${\leq}8cm^3$). The number of cancers developing in TH-treated groups was also significantly fewer (P<0.05). Histological grading showed majority of TH-treated group cancers to be of grade 1 and 2 compared to grade 3 in controls. There was an increasing trend of apoptotic index (AI) seen in TH-treated groups with increasing dosage of Tualang Honey, however, the mean AI values of all TH-treated groups were not significantly different from the Control value (p>0.05). In conclusion, Tualang Honey exerted positive modulation effects on DMBA-induced breast cancers in rats in this preliminary study.

Suppressive effects of Th2 cytokines expression and the signal transduction mechanism in MC/9 mast cells by flavonol derived from Ginkgo biloba leaves (비만세포에서 은행잎 플라보놀에 의한 Th2 Cytokine 발현 및 신호전달 억제 기전 효과)

  • The effects of Flavonol contents from Ginkgo biloba leaf on anti-atopy activity have not rarely been verified. This study is to investigate the effects of flavonol on Th2 cytokine production in MC/9 mast cells. For this, flavonol was analyzed by ELISA and Real-time PCR. Analysis results showed that flavonol significantly suppressed production of Th2 cytokines(IL-13, MIP-1a) in a dose dependent manner. The mRNA expression of IL-4, IL-5, IL-13, TNF-a were effectively restrained by Flavonol at the concentration 25,50,$100{\mu}g/m{\ell}$. And decrease of expression of NFAT-1, c-jun protein was confirmed by western blot analysis. These results indicate that flavonol has effects of decreasing the Th2 cytokine production in the MC/9 mast cell causing inhibition of transcription factors such as NFAT-1, c-jun. Thus, we would like to brief that flavonol may have the applicability as therapeutic agent for atopic dermatitis.
