Effect of dietary isoflavones of soybean by-product on estrogen and testosterone levels in mouse

  • Boar taint, an unpleasant odor of pork, is associated with two substances including androstenone and skatole. Testosterone is a steroid hormone as well as a strong predictor for androstenone secretion. Isoflavones of soy origin play a role in modulating the metabolism of sex hormones. Although several methods responsible for reducing boar taint are under investigation, the precise mechanism by which isoflavones reduce testosterone has not yet been identified. The objective of the current study was to investigate the effects of isoflavones extracted from a soy by-product on the concentration of serum testosterone in mouse. A total of 24 mice were supplemented with basal diet (control), daidzin plus genistin mix (T1), or isoflavone extracts (T2). After 11 days of treatment, size and weight of testis, as well as the concentration of sex hormones, including testosterone and estrogen, were analyzed. There was no difference in size or weight of testis from mice among control, T1, and T2. Serum testosterone levels were significantly decreased (p < 0.05) both in T1 and T2 when compared with the control group. Furthermore, estrogen concentration in blood was increased (p < 0.05) in T2 (numerically increased in T1) compared with the control group. Taken together, the use of isoflavones extracted from soy by-products would be a plausible strategy for reducing testosterone level, ultimately reducing boar taint without castration of piglets.

DDT Reduced Testosterone and Aromatase Activity Via ER Receptor in Leydig Cell (DDT의 Aromatase 증가에 의한 Testosterone 감소효과)

  • Dichlorodiphenyltrichloroethane (DDT), is a widespread environmental pollutant. In this study, we investigated the effect of DDT on testosterone production through aromatase and investigated its molecular mechanism in testicular leydig cell, R2C. We investigated that the effects of DDT on testosterone production and its effects on aromatase activity in R2C cell by radio immunoassay (RIA). As the results, the potent leyding cell activator LH increased testosterone production compared to the control. DDT exposure significantly decreased testosterone production in R2C cell and DDT alone affected T reduction in a dose-dependent manner in R2C cell slightly. In addition, DDT was found to increase aromatase activity in R2C cell in a dose dependent manner. In order to assess whether the suppressive effects of DDT on LH-inducible testosterone production might be influenced by the ER, ICI 182.780, a pure antiestrogen, was used, and it was found that these inhibitory effects of DDT were antagonized by ICI 182.780, implying that the ER mediates the suppressive effects of DDT. Furthermore, the inducible effects of DDT on aromatase might be influenced by the ER, ICI 182.780 was used, and it was found that these enhancing effects of DDT were antagonized by ICI 182.780, implying that the ER mediates the inducible effects of DDT. Our results indicated that DDT inhibition of LH-inducible testosterone production in R2C is mediated through aromatase. However, the precise mechanisms by which DDT enhance in leyding cell remains unknown. The current study suggests the possibility that DDT might act as a modulator aromatase gene transcription.

THYROID HORMONES AND TESTOSTERONE IN SHEEP AGE RELATED PROFILES OF SERUM THYROXINE, TRI-IODOTHYRONINE AND TESTOSTERONE IN KAGHANI, RAMBOUILLET AND KAGHANI × RAMBOUILLET SHEEP

  • Secretory patterns of serum tri-iodothyronine, thyroxin and testosterone were analysed by specific radioimmunoassay in Kaghani (n = 42), Rambouilet (n = 42) and Kaghani ${\times}$ Rambouillet (n = 42) male sheep to determine age and breed related changes. Animals were examined at 1-2 wk, 8, 10, 12, 20, 22 and 24 months of age (n = 6/age group). All animals were maintained under standard conditions of feeding and management. $T_3$ and $T_4$ concentrations were maximal (p < 0.01) in 1-2 wk old lambs of all breeds studied ($207.4{\pm}14.9-276.8{\pm}20.4ng/dl$ and $5.76{\pm}0.3-7.23{\pm}0.9{\mu}g/dl$, respectively). For all the breeds, concentrations of $T_3$ declined markedly (p < 0.01) at 8 month of age and then varied in a narrow range ($44.5{\pm}14.0-87.0{\pm}4.8ng/dl$) up to the age of 24 month. While, serum $T_4$ concentrations decreased (p < 0.01) to nadir ($0.97{\pm}0.1-1.43{\pm}0.3{\mu}g/dl$) at 10 months of age in all the 3 breeds. Afterwards, $T_4$ levels increased gradually and attained a small peak at 20 months before declining again at 22 and 24 months of age. Serum $T_3$ and $T_4$ concentrations were correlated significantly (p < 0.05) and their ratio was constant at all ages in the different breeds. Age significantly (p < 0.01) influenced the secretion of testosterone in the breeds examined but the breed differences were comparable. Mean serum concentrations of testosterone were low from 1-2 wk to 10 months of age in the Kaghani, Rambouillet and Kaghani ${\times}$ Rambouillet animals. Peak testosterone levels were noted in the 12 months old Kaghani ($1.75{\pm}0.4ng/ml$), Rambouillet ($1.30{\pm}0.2ng/ml$) and their $T_3$ cross animals ($1.16{\pm}0.08ng/ml$). Mean testosterone levels, thereafter, decreased at 20 months and then at 22 and 24 months remained variable but higher than observed at 1-2 wk -10 months of age. The findings of the present study indicate a significant influence of age but not of breed on the peripheral concentrations of thyroid hormones and testosterone. Serum concentrations of $T_3$ and $T_4$ were not related to testosterone levels. High thyroid secretory activity observed at prenatal phase of development may play an important role in preparing the lambs to extrauterine environments.

Korean Red Ginseng (Panax ginseng) Potentiates the Inhibitory Actions of Testosterone on Obesity and Adipogenesis in High Fat Diet-Fed Castrated Mice

  • It has been suggested that ginseng is beneficial for ameliorating the aging males' symptoms, such as weight gain, fatigue, erectile dysfunction, and depression, in elderly men with testosterone deficiency. We thus investigated the effects of Korean red ginseng (Panax ginseng C.A. Meyer; Araliaceae) on obesity in a mouse model of testosterone deficiency (castrated C57BL/6J mice). The effects of ginseng extract (GE) and/or testosterone on obesity and adipogenesis in high-fat diet (HFD)-fed castrated C57BL/6J mice and 3T3-L1 adipocytes were examined using in vivo and in vitro approaches. After feeding mice a HFD for 8 weeks, we found that mice also receiving GE and/or testosterone showed decreased body weight, adipose tissue mass, adipocyte size, and hepatic lipid accumulation compared with untreated HFD-fed mice. Expression of adipogenic genes ($PPAR{\gamma}$, $C/EBP{\alpha}$, and aP2) was decreased by GE and/or testosterone in adipose tissues. Consistent with the in vivo data, lipid accumulation and the mRNA expression of adipogenesis genes in 3T3-L1 adipocytes were decreased by GE, ginsenosides, and testosterone. The inhibitory effects of GE (or ginsenosides) were comparable to those of testosterone, and the effects of co-treatment with GE (or ginsenosides) and testosterone were greater than those of testosterone alone in vivo and in vitro. Our results indicate that ginseng may be able to potentiate the inhibitory effects of testosterone on obesity and adipogenesis in HFD-fed castrated mice, providing possible therapeutic implications in men with testosterone deficiency.

  • This study is to investigate the effects of testosterone on adipogenesis and its molecular mechanism using RT-PCR analysis and transient transfection assays. Castrated(CAST) mice treated with testosterone had lower white adipose tissue weights and expression of adipocyte-specific genes($PPAR{\gamma}$ and aP2) than CAST control mice. Consistent with the in vivo data, testosterone treatment inhibited triglyceride accumulation and expression of adipocyte-specific genes($PPAR{\gamma}$ and aP2) in differentiated 3T3-L1 cells compared with control group. Testosterone-activated androgen receptor(AR) repressed the luciferase reporter gene activity induced by $PPAR{\gamma}$ transfection. Thus, these results suggest that testosterone downregulates the actions of $PPAR{\gamma}$ on adipogenesis through AR.

  • 5${\alpha}$-dihydrotestosterone(DHT)과 testosterone(T)은 남성 생식기관의 주 생성 호르몬들로 그 구조가 매우 비슷하여 이들 각 개를 특이하게 측정(specific determination)하는 방법이 개발되지 않고 있다. 본 연구는 고속액체 크로마토그라피(HPLC)를 이용하여 이들을 분리한 후 섬광면역측 정법(Luminescence immunoassay, LIA)으로 정량하는 방법을 개발하여 이들의 응용 가능성을 검토하고져 하였다. DHT와 T의 retention time은 각각 10.3min, 17.6min이었다. DHT-LIA와 T-LIA에 서 다른 스테로이드들과의 교차반응도는 방사면역측정법(RIA)과 대동소이하였다. 정도관리(quality control) 시료의 intra-assay variation은 DHT-LIA가 8.7%, T-LIA가 6.0%의 변이계수를 나타내었고, inter-assay variation의 변이계수는 각각 12.0% 및 15.3%이었다. 실측치(y)와 기대치(x)간의 관계를 보면, DHT-LIA경우는 Y=0.94X+0.9(r=0.989), T-LIA는 Y=1.01X+0.06(r =0.988)로 두 측정치 사이에는 통계적으로 유의한 차이가 없었다. 위의-측정방법을 이용하여 DHT-enanthate와 T-enanthate 처리후 혈청내 DHT 및 T의 농도변화를 조사한 실험결과 LIA와 RIA의 값사이에 유의한 차이가 없었다. 위의 결과로 보아 본 실험에서 개발된 DHT와 T의 섬광면역측정법은 정립되었다고 사려된다.

Decreased Levels of Plasma Testosterone/LH Ratio in Male Mice Exposed to Sodium Arsenite

  • While it's been shown that arsenic impairs male reproductive function, it remains unclear whether the mechanism involves an effect on testosterone (T) production. We examined plasma T and luteinizing hormone (LH) levels in mice given water containing either 20 or 40 mg/L sodium arsenite (SA). The plasma T levels were lower in SA-treated mice than in controls and correlated well with testicular T levels within individuals. However, SA treatment did not significantly affect plasma LH levels. The ratio of plasma T to LH was reduced by the treatment with 40 mg/L SA. These results suggest arsenic-induced defect in testicular testosterone production in mice.

  • The metabolism of testosterone ($17{\beta}$-hydroxy-androst-4-en-3-one) was confirmed in horse after a single intramuscular administration of testosterone cypionate (750 mg). Solvent extracts of urine obtained with enzymatic hydrolysis and methanolysis were analyzed by GC/MS after oxime t-butyldimethylsilyl (oxime-TBDMS) derivatization. The structures of four urinary metabolite after testosterone administration in horse were determined based on EI mass spectra and $5{\alpha}$-androstane-$3{\beta}$, $17{\alpha}$-diol and $5{\alpha}$-androstane-$3{\beta}$-ol-17-one as major was confirmed with authentic standard. Also the concentrations of $5{\alpha}$-androstane-$3{\beta}$, $17{\alpha}$-diol, $5{\alpha}$-androstane-$3{\beta}$, $17{\beta}$-diol, dehydroepiandrosterone (DHEA), $5{\alpha}$-androstane-$3{\beta}$-ol-17-one and testosterone were determined in the urine of normal subjects and the urine after administration. The recovery and detection limit in the most drugs were 86.3~94.7% and 1~3 ppb, respectively. Correlation coefficients for calibration were in the range of 0.984~0.999. Excretion profile of testosterone presents the rapid and large increasement up to maximum values at days 5 after administration and the slow regression. The relative ratios of testosterone, its metabolites over DHEA were determined for indication of testosterone administration in horse doping.

  • Ralationship between noradrenergic nervous system activity and luteinizing hormone releasing hormone(LHRH) content mediated by testosterone in hypothalamus was tested. Three groups of adult male animals were prepared; (1) Intact; (2) Castration+Vehicle (Cast+V); (3) Castration+Testosterone (Cast+T). Silastic capsule containing vehicle or testosterone was implanted into neck region of animals two weeks following castration. Norepinephrine content, alpha-adrenergic receptor binding characteristics using H$^{3}$-WB4101, and content of LHRH by LHRH RIA procedure were determined. Testosterone replacement to castrated male rats augmented the content of norepinephrine and LHRH. Testosterone replacement increased the alpha-adrenergic receptor density but did not change alpha-receptor affinity. The data from the present study suggest that increase in LHRH content by testosterone may be positively coupled to the activity of central noradrenergic nervous system.

  • The present study was disigned to determine the concentration of bioavailable steroid hormones in the atretic follicular fluid (FF). The concentradons of progesterone (P), testosterone (T), estradiol (E), androstenedione (A), and 5-$\alpha$ dihydrotestosterone (DIlT) were determined by the established methods of luminescent immunoassay (LIA) or radioimmunoassay (RIA). Concentrations of T, A and Diff in human FF from smail (< 6 mm). medium (8-15 mm), and large (> 15 mm) atretic follicles were significandy higher than those of normal ones (p < 0.01). However, the levels of T, A and DHT in smail atretic foflicle were significandy lower than those found in normal one. The concentrations of P in atretic FF from porcine small (< 3 mm), medium (4-6 mm), and large (> 7 mm) follicles were not different from that of normal ones. However, the concentration of E in atretic forncles of each group was significantly lower than that of normal group (p < 0.001 in each group). On the other hand, the percentages of bioavailable T (BI) in human FF were significandy (p <0.001) higher than those in normal groups. The BT in normal or atretic FF was more than 90 % of total T. The present result demonstrates that the bioavailable androgen, but not E levels in atretic follicles is higher than that of normal one, and that the atretic mechanism might be dependent on the ovarian forncle size in the developmental stage and on the animal model system. Moreover, the present study suggests that the steroids found in the FF are the bioavailable forms and the concentration of BT in FF could be used as one of the valuable criteria classifying the ovarian atretic follicle.
