Screening of Cell Cycle-Related Genes of Pleurotus eryngii Using Yeast Mutant Strains

  • Temperature-sensitive yeast mutants were used to screen for cell cycle-related genes from Pleurotus eryngii genomic DNA. A mushroom genomic DNA library was established and each gene was screened for the ability to rescue seven Saccharomyces cerevisiae temperature-sensitive strains. Hundreds of yeast transformants were selected at restrictive temperatures over $30^{\circ}C$. Plasmids from the transformants that survived were isolated and transformed back into their host strains. The temperature sensitivity of the resulting transformants was tested from $30^{\circ}C$ to $37^{\circ}C$. Ten DNA fragments from P. eryngii were able to rescue yeast temperature-sensitive strains, and their DNA sequences were determined.

A pheromone mutant of Schizosaccharomyces pombe displays nucleolar fragmentation

  • Stresses and nutritional starvation are two main external signals for the induction of sex pheromones in the fission yeast Schizosaccharomyces pombe. In an attempt to identify the components involved in transduction of starvation signals, we screened 135 temperature-sensitive (ts) mutants and isolated 6 mutants that induced the pheromone even in the presence of a nitrogen source. These mutants exhibited two distict induction phenotypes: pheromone induction at restrictive but not at permissive temperatures; and pheromone induction at both permissive and restrictive temperatures. The times required for the maximum pheromone induction at the restrictive temperature differed slightly in each mutant. In addition to the pheromone induction phenotype, the ts243 and ts304 mutants exhibited cell-division-cycle defects. The ts304 mutant cells showed an abnormal cytoplasmic DAPI staining pattern. The nucleolus of this mutant seemed to be fragmented, a phenomenon which is typically observed in aged yeast cells. The result of our genetic analysis indicated that the pheromone induction mutants belonged to 6 separate complementation groups. We designated these mutants pws1 to pws6.

Isolation and Characterization of Pre-$tRNA^{Val}$ Splicing Mutants of Schizosaccharomyces pombe

  • A collection of 132 temperature sensitive (ts) mutants was generated by the chemical mutagenesis of Schizosaccharomyces pombe wild type strain and screened for tRNA splicing defects on Northern blots by hybridization with an oligonucleotide that recognizes the exon of the S. pombe tRNA^Val as a probe. We identidied 6 mutants which accumulate precursor $tRNA^{Val}$. Among them, 2 mutants exhibited remarkable morphological differences compared to wild type cells. One tRNA splicing mutant showed elongated cell shape in permissive as well as non-permissive cultures. The other mutant exhibited shortened cell morphology only in nonpermissive culture. The total RNA pattern in the splicing mutants appeared to be normal. Genetic analysis of four $tRNA^{Val}$ splicing mutants demonstrated that the mutation reside in different genes.

Genetic studies of Baculovirus used as a microbial pesticide

  • Sixteen temperature-sensitive mutants of Autographa californica nuclear polyhedrosis virus were isolated. Several interesting phenotypes were observed. A large proportion of the mutants were un-able to form polyhedral occlusion bodies at the nonpermissive temperature (32.5C). At 32.5C, one mutant formed plaques in which the cells lacked polyhedra. Another mutant type was defective in the production of progeny extracellular nonoccluded virus and produced a plaque consisting of only a single cell containing polyhedra at 32.5C. One mutant was defective in plaque formation, progeny nonocluded virus formation, and polyhedra formation at 32.5C. Several mutants produced nonocluded virus but failed to produce plaques or polyhedra at 32.5C. Other phenotypes were also distinguished. Complementation analyses, performed by either measuring the increase in extracellular non-ocludedvirus formation or by oberving polyhedra formation in mixed infections at 32.5C, indicated the presence of 15 complementation groups. A high frequency of recombination was observed. Four of the mutants were found to be host dependent in their temperature sensitivity for polyhedra formation.

Mad1p, a Component of the Spindle Assembly Checkpoint in Fission Yeast, Suppresses a Novel Septation-defective Mutant, sun1, in a Cell Division Cycle

  • Schizosaccharomyces pombe is suited for the study of cytokinesis as it divides by forming a septum in the middle of the cell at the end of mitosis. To enhance our understanding of the cytokinesis, we have carried out a genetic screen for temperature-sensitive S. pombe mutants that show defects in septum formation and cell division. Here we present the isolation and characterization of a new temperature-sensitive mutant, sun1(septum uncontrolled), which undergoes uncontrolled septation during cell division cycle at restrictive temperature $(37^{\circ}C)$. In sun1 mutant, actin ring and septum are positioned at random locations and angles, and nuclear division cycle continues. These observations suggest that the sun] gene product is required for the proper placement of the actin ring as well as precise septation. The sun] mutant is monogenic recessive mutation unlinked to previously known various cdc genes of S. pombe. In a screen for $sunl^+$ gene to complement the sun] mutant, we have cloned a gene, $susl^+$(suppressor of sun1 mutant), that encodes a protein of 689 amino acids. The predicted amino acid sequence of $susl^+$ gene is similar to the human hMadlp and Saccharomyces cerevisiae Mad1p, a component of the spindle checkpoint in eukaryotic cells. The null mutant of $susl^+$ gene grows normally at various temperatures and has the increased sensitivity to anti-microtubule drug, while $susl^+$ mutant shows no sensitivity to microtubule destabilizing drugs. The putative S. pombe Sus1p directly interacts with S. pombe Mad2p in yeast two-hybrid assays. These data suggest that the newly isolated susr gene encodes S. pombe Mad1p and suppresses sun] mutant defective in controlled septation in a cell division cycle.

Bacillus thuringiensis 의 Temperature-sensitive Mutants 분리와 특성 연구 (Growth Patterns of Temperature-sensitive Mutants of Bacillus Thuringiensis)

  • Bacillus thuringiensis 3ab K- 3 균주를 28$^{\circ}C$에서 UV광으로 돌연변이를 유도한 다음에 42$^{\circ}C$에서 24개의 Temperature-sensitive(ts)돌연변이 균주를 분리하여 42$^{\circ}C$에서의 성장과 항생제 저항성에 대한 성격을 연구하였다. 24개의 ts 돌연변이 균주중에 13개의 ts돌연변이체는 42$^{\circ}C$의 제한온도에서 전혀 성장을 하지 않았으며, 나머지 11개의 돌연변이체는 성장이 28$^{\circ}C$에서 보다 1/2 에서 1/4까지 감축되었다. 이러한 성격에 따라서 ts돌연변이체는 치사군과 비 치사군으로 나눌수 있었다. 또한 B. thuringiensis SI K-2을 nitrosoguanidine으로 28$^{\circ}C$에서 처리하여 7개의 Auxotrophic mutant을 분리하였다. A-N28은 Arg$^{-}$ . Gly$^{-}$ , Leu$^{-}$ , Met$^{-}$ , Ser$^{-}$ , Val$^{-}$ 을 요구했고, A-N 65는 Ala $^{-}$ , Gly$^{-}$ , Leu$^{-}$ , Met$^{-}$ , Ser$^{-}$ , Val$^{-}$ 을 요구했으며 A-N 92는 Gly$^{-}$ , Leu$^{-}$ , Met$^{-}$ 을, A-N115는 7개의 아미노산을 A-N156은 8개를 요구했다. B. thuringiensis 3ab K-3와 SI K-2 는 Penicillin, Ampicillin과 Cephalothin에 저항성을 나타냈고 ts-U171, A-N92와 A-N115는 이 세항생제에 민감성을 나타냈다. Temperature - sensitive돌연변이균주를 42$^{\circ}C$에서 24시간 배양후에 28$^{\circ}C$로 옮기어 배양한 결과 ts-U601, -U603, -U604와 -U171은 성장을 더 이상하지 않았다.
