Spermatogenesis and fine structure of the spermatozoon of the marbled sole, Limanda yokohamae were examined by means of the scanning and transmission electron microscopy. The process of spermatogenesis of the marbled tole is similar to that of other teleost with external fertilization. During the spermiogenesis, chromatin that has been became fine]y granular progressively condenses into many large globules and that homogeneously condensed in the spermatozoan head. A spermatozoon consists of head and tail, and the acrosome is absent. The cytoplasmic collar contained eight mitochondria is observed in the posterior part of the head. The well -developed axonemal lateral fins are observed in the tail. In the TEM observation, the cross section of the axial filament shows '9+2' axonemal structure of microtubules, and the numerous vesicles are observed in the cytoplasm.
Initiation and activation of sperm motility are prerequisite processes for the contact and fusion of male and female gametes at fertilization. The phenomena are under the regulation of cAMP and $Ca^{2+}$ in vertebrates and invertebrates. Mammalian sperm requires $Ca^{2+}$ and cAMP for the activation of sperm motility. Cell signaling for the initiation and activation of sperm motility in the ascidians and salmonid fishes has drawn much attention. In the ascidians, the sperm-activating and attracting factors from unfertilized egg require extracellular $Ca^{2+}$ for activating sperm motility and eliciting chemotactic behavior toward the egg. On the other hand, the cAMP-dependent phosphorylation of protein is essential for the initiation of sperm motility in salmonid fishes. A decrease of the environmental $K^+$ concentration surrounding the spawned sperm causes $K^+$ efflux and $Ca^{2+}$ influx through the specific $K^+$ channel and dihydropyridine-sensitive L-/T-type $Ca^{2+}$ channel, respectively, thereby leading to the membrane hyperpolarization. The membrane hyperpolarization induces synthesis of cAMP, which triggers further cell signaling processes, such as cAMP-dependent protein phosphorylation, to initiate sperm motility in salmonid fishes. This article reviews the studies on the physiological mechanisms of sperm motility and its cell signaling in aquatic species.
Rhodopsin, a dim-light receptor, is a model system for the study of G protein-coupled receptors that transduce extracellular signals into cells. To study the molecular mechanisms of visual systems in fish, the rod opsin gene of olive flounder Paralichthys olivaceus was characterized. The full-length P. olivaceus opsin gene was obtained by PCR amplification of genomic DNA, as well as cDNA synthesis. A comparison of clones obtained from both methods indicated that the olive flounder rod opsin gene lacks introns. Sequence analysis of the opsin gene indicated that it contains a 1,056-bp open reading frame encoding 352 amino acids. The deduced amino acid sequence contains features of typical rod opsins, such as sites for Schiff's base formation (K296) and its counterion (E113), disulfide formation (C110 and C187), and palmitoylation (C322 and C323). An opsin sequence alignment showed the highest similarity between P. olivaceus and Solea solea (95.1%), followed by Hippoglossus hippoglossus (94.5%). An opsin phylogenetic tree revealed a close relationship between olive flounder and teleost rod opsins.
The feeding habits of Spanish mackerel (Scomberomorus niphonius) were studied based on the examination of stomach contents of 445 specimens collected from January to December 2004, in the Southern Sea of Korea. The size of Spanish mackerel ranged from 26.1 to 105.4 cm in fork length (FL). Spanish mackerel was a piscivorous fish which mainly consumed teleost fishes such as chub mackerel (Scomber japonicus) and anchovy (Engraulis japonicus). Its diet also included small quantities of shrimps, stomatopods, cephalopods, crabs, polychaetes, amphipods, cumaceans and copepods. Smaller individuals (<60 cm FL) consumed small fishes such as Engraulis japonicus and crustaceans. The portion of these prey items decreased with increasing fish size, and this decrease was paralleled with increased consumption of lager fishes such as Scomber japonicus and Cololabis saira. The prey size increased with the increase of fish size.
The structures of the fertilizedegg envelope in threespecies of cyprinidae, leoparddanio (Brachydanio frankei), cherry barb (Barbustitteya) and white cloudmountain fish (Tanichthys albonubes) were investigated by light and electron microscopy. The fertilized eggs of all three species have colorless, transparent, spherical and non-floted type. The egg envelopes have a single micropylewhich is thought to the pathway of sperm in the area of the animal pole. Also, an outer layersof both cherry barband white cloud mountain fish areadhesive types, but in that of leoparddanio has non-adhesivetype. In leopard danio, an outer surface ofegg envelope is covered by mushroom-likestructure, that of cherry barb is arranged by button-like structure, and that of white cloud mountain fish isarranged by rod-likestructure. Inboth leopard danioand cherrybarb, the eggenvelopes consist of three distinct layers; an outer layer, a middle layer and an inner layer, and that of white cloud mountain fish hastwo layers. An innerlayer of egg envelopein leopard danio consist of 10 layers, cherry barb has 8 layers, and white cloud mountain fish has $5\sim6$ layers.
Euryhaline teleost have extraordinary ability to deal with a wide range of salinity changes. To study the seawater adaptability of rainbow trout Oncorhynchus mykiss (body weight 638±54 g, length 38.6±2 cm) to salinity increase fish were transferred from freshwater to 7, 14, 21, 28 and 32 psu and checked for mortality over 5 days. No mortality was observed in 0-32 psu. In fish transferred to 0-32 psu, blood osmolality was maintained within physiological range. The changes of serum enzyme activities (aspartate transaminase, AST and alanine transaminase, ALT) showed no significant level during experimental period. To explore the underlying molecular physiology of gill and kidney responsible for body fluid regulation, we measured mRNA expression of five genes, Na+/K+/2Cl- cotransporter1 (NKCC1), aquaporin3 (AQP3), cystic fibrosis transmembrane conductance regulator (CFTR), glucocorticoid receptor (GR) and growth hormone receptor (GHR) in response to salt stress. Based on our result, rainbow trout could tolerate gradual transfer up to 32 psu for 5 days without mortality under physiological stress. This study suggests to alleviate osmotic stress to fish, a gradually acclimation to increasing salinity is recommended.
Paired box protein, PAX7, is a key molecule for the specification, maintenance and skeletal muscle regeneration of muscle satellite cells. In this study, we identified and characterized the cDNA and amino acid sequences of PAX7 from black sea bream (Acanthopagrus schlegelii) via molecular cloning and sequence analysis. A. schlegelii PAX7 cDNA was comprised of 1,524 bp encoding 507 amino acids and multiple sequence alignment analysis of the translated amino acids showed that it contained three domains including paired DNA-binding domain, homeobox domain and OAR domain which were well conserved across various animal species investigated. Pairwise Sequence Alignment indicated that A. schlegelii PAX7 had the same amino acid sequences with that of yellowfin seabream (A. latus) and 99.8% identity and similarity with that of gilt-head bream (Sparus aurata). Molecular phylogenetic analysis confirmed that A. schlegelii PAX7 formed a monophyletic group with those of teleost and most closely related with those of the fish that belong to Sparidae family including A. latus and S. aurata. In the investigation of its tissue specific mRNA expression, the expression was specifically identified in skeletal muscle tissue and a weak expression was also shown in gonad tissue. The cultured cells derived from skeletal muscle tissues expressed PAX7 mRNA at early passage but the expression was not observed after several times of subculture.

These gonad become hermaphrodite and then ditermine the sex of female and male. These results show the red sea bream is juvenile hermaphrodite and undif-ferentiated gonochoristic teleost. Male and female differentiation type of gonad is divided in undifferentiation stage, oogonia-like stage, ovary-like stage, ovary development stage, hermaphroditic testis stage, hermaphroditic ovary stage, and testis development stage. Undifferentiation stage is continued total lenth 18cm at 13 months after hatching. ovary-like stage is continued total length 11~18cm at 13 months after hatching. Ovary-like stage is continued total length 14~26cm at 10~14 months after hatching. Ovary development stage begins from total length 20cm, 14 months after hatching. At 20 months after hatching, 44 percent of total sampled individuals had ovary. Hermaphroditic ovary stage first begins total length 19~20 cm at 15 months after hatching, but it is not observed total length 28~29cm at 20months after hatching. Hermaphroditic testis stage first begins total length 21~22cm at 20months after hatching and is continued for 20months. Testis development stage first begins total length 20~21cm at 20 months after hatching, and is occupied 33 percent total length 28~29cm at 20 months. The beginning of sex differentiation more than 50 percent is from total length 16cm at 11 months after hatching. Sex determination begins total length 20cm, 14months after hatching in female and total length 20cm, 15 months after hatching in male. Sex determination more than 50 percent begins total length 23cm,, 17 months after hatching. Undifferentiated gonadal part of red sea bream consist gonad and fat body. As differentiation is going on and gonad is growing, fat body shrinks. This appearence is showed the same tendency in 3-year old red sea bream. 1.9mm larvae after hatching grow about 19mm larvae for 47 days. The relationship between the total length and body weight of larvae and juveniles in $BW=4.45{\times}10^{-6}TL^{3.4718}$ r=0.9820. Fishes in cage culture grow to maximum total length 28.4cm. The relationship between the total length and body weight of these fishes is $BW=2.36{\times}10^{-2}TL^{2.9180}$, r=0.9971. Undifferentiated gonadal part of red sea bream consist gonad and fat body. As differentiation is going on and gonad is growing, fat body shrinks.
The fine structure of spermatozoa of Pungtungia herzi was examined with scanning and transmission electron microscopies. The spermatozoa of p. herzi are approximately 37.4 ${\mu}{\textrm}{m}$ in length and a relatively simple cell with a spherical nucleus, a short midpiece and a tail. The acrosome is not present as in most teleost fishes. The ultrastructure of spermatozoa represents typical characteristics of cyprinid spermatozoa including the lateral insertion of flagellum, the organization of centriolar complex in shallow nuclear fossa, and the occurrence and asymmetrical arrangement of mitochondria. In the nuclear envelope and mitochondrion, however there were some morphological differences for their ultrastructure. The nuclear envelope is severely undulated and the shallow nuclear fossa contains two centrioles which are at the angle of some 130$^{\circ}$ each other. The most significant feature can be observed with the mitochondrion; five or more mitochondria, which are shown in primary spermatocyte, fuse to form a single one in the mature spermatozoon. The mitochondrial aspect is different from that of other cyprinid spermatozoa, where their mitochondria have a conventional aspect and never fuse to form a mitochondrial derivative. In terms of sperm evolution the fused mitochondria are regarded as the apomorphic character in comparison with the separate mitochondria. The single mitochondrion is not reported in cyprinid spermatozoon except the case of Rhodeus.
The delicate histological structure of gill in teleosts can be easily affected by a variety of biological, chemical or physical detrimental agents because it is directly exposed to the surrounding water. The epithelium of secondary lamella is thin to allow efficient gaseous exchange and this also renders it particularly vulnerable to various pathogens. As well as the main respiratory role, the gill has other various important functions such as acid-base balance, osmoregulation or the excretion of nitrogenous waste products. Thus destruction of epithelial integrity such as epithelial necrosis or thickening can render a fish very vulnerable to respiratory, secretory and excretory difficulties. This article was tried to describe in detail the common processes of pathological responses correlated to the normal histological structures of the gill in teleosts.
