An unrecorded species of Allium (Alliaceae) in Korea: A. pseudojaponicum Makino (부추속(부추과) 미기록 식물 1종: 갯부추)

  • Here we report a previously unrecorded species of Korean Allium (Alliaceae) from Geomundo, Yeosu-si, Jeollanam-do. This taxon, A. pseudojaponicum Makino which has been known to distribute only in the southern part of Japan up to date, is easily distinguished from A. thunbergii G. don by lustrous evergreen leaves and lateral scapes as well as chromosome number (2n=32). The common name, 'Gaet-bu-chu', was newly given considering the property of habitat, which is the dry and rocky grasslands facing to the sea in Korea and Japan. In this study, we redescribe morphological characters, and provide illustrations of habit as well as photographs of habitat.

Systematics of Korean Thalictrum L. based on a morphological cladistic analysis (형태학적 분계분석에 의한 한국산 꿩의다리속(Thalictrum L.) 식물의 계통학적 연구)

  • We used Maximum parsimony and Neighbor-joining to investigate phylogenetic relationship of the genus Thalictrum in Korea with 39 morphological characters. The plant materials were used for ingroup 21 taxa and outgroup 1 taxon including a Korean endemic plant. This study confirmed that Sect. Thalictrum and Sect. Camptonotum formed monophyletic group by 100% and 83% bootstrap values respectively, and Sect. Camptonotum appeared to be Sect. Thalictrum's sister group. Sect. Erythrandra and Sect. Physocarpum formed paraphyletic groups. Sect. Physocarpum except T. osmorhizoides appeared to be polytomy. Sect. Tripterium and Sect. Omalophysa formed an independent clade. The shape of stamen is the important character representing the phylogenetic pattern of the genus Thalictrum; the bat and filiform types might be derived from clavate type. The pollination patterns (entomophily, anemophily), and presence of calyx at flowering time may also be related to the evolution of the Thalictrum flowers.

Ethnobotany of Jeju Island, Korea (제주도 지역의 민속식물)

  • The ethnobotanical study was conducted to record and conserve the traditional knowledge of botanical taxa in Jeju island, Korea from March to November, 2011. According to the survey results, derived from 1,774 sheets of 177 residents at 78 places, the ethnobotanical plants in Jeju island consisted of a total 356 taxa; 103 families, 260 genera, 312 species, 2 subspecies, 37 varieties and 5 form. The analysis of usage for 365 taxa showed that the medicinal use was the highest with 284 taxa, followed by edible with 145 taxa, ritual with 36 taxa, craft with 31 taxa and timber with 31 taxa respectively. The leaf of plant was the most useful part, followed by stem and whole plant. The most frequently mentioned taxon was Plantago asiatica (34 citations), followed by Artemisia princeps (32 citations) and Lonicera japonica (30 citations).

Floristic Study of Yeoseo-do (Isl.) in Korea (여서도의 관속식물상)

  • We investigated the flora of Yeoseo-do (N $33^{\circ}58^{\prime}$, E $126^{\circ}55^{\prime}$), located in Wando-gun, Jeollanam-do, South Korea and discussed the vascular plants found there. Four separate field trips were completed from April to October of 2013. The vascular plants were summarized as 463 taxa; 112 families, 306 genera, 409 species, 6 subspecies, 42 varieties, 5 forms and 1 hybrids. The number of endemic species of Korea were 6 taxa, and 12 taxa designated by the Ministry of Environment as red data of endangered vascular plants were investigated in this region. There were a total of 98 taxa of floristic regional indicator plants and one taxon of level I endangered species, Neofinetia falcata (Thunb.) Hu, as designated by the Ministry of Environment. 33 taxa have shown their southern distributional limit ranges, and 10 taxa of halophytes were confirmed in the Yeoseo-do (Isl.). In additon, 27 taxa of the naturalized plants were recorded.

Assessment of Soil Microbial Communities in Carotenoid-Biofortified Rice Ecosystem

  • This study was conducted to investigate the effect of Psy-2A-CrtI (PAC), a genetically modified (GM) rice with enhanced ${\beta}$-carotene, on the soil microbial community. The soil used to cultivate GM rice and its wild-type, Nakdong, was analyzed for population density, denaturing gradient gel electrophoresis (DGGE), and pyrosequencing. It was found that the bacterial, fungal and actinomycetes population densities of the PAC soils were within the range of those of the non-GM rice cultivar, Nakdong. The DGGE banding patterns of the GM and non-GM soils were also similar, suggesting that the bacterial community structures were stable within a given month and were unaffected by the presence of a GM plant. The pyrosequencing result showed a temporal difference in microorganism taxon and distribution ratio, but no significant difference between GM and non-GM was found. The persistence of the transgene DNA in the plant and surrounding soil were investigated for different time periods. There were differences in the persistence within the plant depending on the gene, but they could not be detected after 5 weeks. Also the transgenes were not detected in the surrounding soil. These results indicate that soil microbial communities are unaffected by the cultivation of a PAC rice within the experimental time frame.

Physiological responses to salt stress by native and introduced red algae in New Zealand

  • Intertidal macroalgae are regularly exposed to hypo- or hypersaline conditions which are stressful. However, red algae in New Zealand are generally poorly studied in terms of salinity tolerance. Consequently, two native (Bostrychia arbuscula W. H. Harvey [Ceramiales], Champia novae-zelandiae [J. D. Hooker & Harvey] Harvey [Rhodymeniales]) and one introduced red algal taxon (Schizymenia spp. J. Agardh [Nemastomatales]) were exposed for 5 days in a controlled salt stress experiment to investigate photosynthetic activity and osmotic acclimation. The photosynthetic activity of B. arbuscula was not affected by salinity, as reflected in an almost unchanged maximum quantum yield (Fv/Fm). In contrast, the Fv/Fm of C. novae-zelandiae and Schizymenia spp. strongly decreased under hypo- and hypersaline conditions. Treatment with different salinities led to an increase of the total organic osmolyte concentrations with rising salt stress in B. arbuscula and Schizymenia spp. In C. novae-zelandiae the highest organic osmolyte concentrations were recorded at SA 38, followed by declining amounts with further hypersaline exposure. In B. arbuscula, sorbitol was the main organic osmolyte, while the other taxa contained floridoside. The data presented indicate that all three red algal species conspicuously differ in their salt tolerance. The upper intertidal B. arbuscula exhibited a wide salinity tolerance as reflected by unaffected photosynthetic parameters and strong sorbitol accumulation under increasing salinities, and hence can be characterized as euryhaline. In contrast, the introduced Schizymenia spp. and native C. novae-zelandiae, which preferentially occur in the mid-intertidal, showed a narrower salinity tolerance. The species-specific responses reflect their respective vertical positions in the intertidal zone.

Genetic Diversity of Polydora haswelli (Polychaeta, Spionidae) in Korean Shellfish using cox1 Marker (cox1 분자마커를 이용한 한국산 패류 천공성 다모류 Polydora haswelli (Polychaeta, Spionidae) 유전자 다양성 발굴)

  • Harmful shell-boring species of the genus Polydora (Polychaeta: Spionidae) were frequently reported from commercially important mollusk species in Korea, Japan and China. The traditional approach based on the morphological characteristics showed limitations for species discrimination among shell-boring species. Therefore, DNA barcoding was adopted to identify Polydora species using molecular markers. Two Polydora species (P. haswelli and P. hoplura) in abalone shells were reported from our previous molecular phylogenetic study. In this study, we additionally reported the presence of shell-boring Polydora haswelli in commercially sold shellfish. The taxon-specific cox1 marker used in this study successfully allowed the isolation of P. haswelli from cockle Scapharca subcrenata, mussel Mytilus galloprovincialis, oyster Crassostrea gigas and scallop Argopecten irradians. Polydora hoplura was not found in these shellfish species. The genetic variations were found on the intraspecific level of P. haswelli and the same genotype was also detected in different shellfish species. This result can provide information on a new host and accurate parasitic Polydora species. Moreover, this report can be used as the biodiversity data of Polydora species on the invasion and transition of harmful Polydora species in mollusk aquaculture farms.

Tuber borchii Shapes the Ectomycorrhizosphere Microbial Communities of Corylus avellana

  • In this study, eight-month-old ectomycorrhizae of Tuber borchii with Corylus avellana were synthesized to explore the influence of T. borchii colonization on the soil properties and the microbial communities associated with C. avellana during the early symbiotic stage. The results showed that the bacterial richness and diversity in the ectomycorrhizae were significantly higher than those in the control roots, whereas the fungal diversity was not changed in response to T. borchii colonization. Tuber was the dominant taxon (82.97%) in ectomycorrhizae. Some pathogenic fungi, including Ilyonectria and Podospora, and other competitive mycorrhizal fungi, such as Hymenochaete, had significantly lower abundance in the T. borchii inoculation treatment. It was found that the ectomycorrhizae of C. avellana contained some more abundant bacterial genera (e.g., Rhizobium, Pedomicrobium, Ilumatobacter, Streptomyces, and Geobacillus) and fungal genera (e.g., Trechispora and Humicola) than the control roots. The properties of rhizosphere soils were also changed by T. borchii colonization, like available nitrogen, available phosphorus and exchangeable magnesium, which indicated a feedback effect of mycorrhizal synthesis on soil properties. Overall, this work highlighted the interactions between the symbionts and the microbes present in the host, which shed light on our understanding of the ecological functions of T. borchii and facilitate its commercial cultivation.

Exploring natural hybridizations among Asplenium ruprechtii and related taxa in Korea

  • The purported four hybrid origins of Asplenium in Korea were tested based on morphological, cytological and DNA sequence data. Asplenium castaneo-viride, A. ${\times}$ uiryeongse, A. ${\times}$ montanus, and A. ${\times}$ kitazawae share several morphological characteristics with the Asian walking fern A. ruprechtii and related taxa as parents and show a sympatric distribution with the putative parents, raising the possibility of hybrid origins: A. castaneo-viride (A. ruprechtii and A. incisum), A. ${\times}$ uiryeongse (A. ruprechtii and A. pekinense), A. ${\times}$ montanus (A. ruprechtii, A. trichomanes, and A. incisum), and A. ${\times}$ kitazawae (A. ruprechtii and A. sarelii). We investigated flow cytometry and chloroplast DNA sequence data (rbcL, rps4-trnS, and rps4-trnS intergenic spacer) to clarify the hybridization and origin of each hybrid. In the flow cytometry analyses, A. ruprechtii shows diploid (2x) only, whereas A. castaneo-viride (3x, 4x), A. ${\times}$ uiryeongse (3x), A. ${\times}$ montanus (3x, 4x), and A. ${\times}$ kitazawae (2x, 4x) exhibit polyploidy, suggesting hybrid events along speciation. The rbcL and rps4-trnS and rps4-trnS intergenic spacer data suggest that A. ruprechtii is one the maternal ancestors of all four hybrids. In addition, the rps4-trnS and rps4-trnS intergenic spacer data indicate that A. incisum is also the maternal ancestor of A. ${\times}$ kitazawae and A. ${\times}$ montanus, proposing multiple hybridization events for these two hybrids. In A. ${\times}$ montanus, morphological features such as the leaf forms and sympatric distributions of the species also support the multimaternal hypothesis, but the morphological features of A. ${\times}$ kitazawae must be examined with consideration of hybrid events. To clarify the complex hybrid evolutionary lineages of the four Asplenium hybrids, further research with taxon sampling and molecular markers should be conducted.

Classification of Korean Benthic Macroinvertebrate Types Using the TWINSPAN Clustering and Discriminant Analysis of Environmental Factors Affecting the Distribution of the Types (TWINSPAN 기법을 이용한 한국의 저서성 대형무척추동물의 유형분류 및 유형분포에 영향을 미치는 환경요인에 대한 판별분석)

  • Korean benthic macroinvertebrate community from 959 sites and 14 environmental factors that could affect the biological distribution were surveyed and analyzed. The 438 taxa were divided into six groups according to TWINSPAN. Based on the environmental factor values corresponding to each taxon, the average and range of environmental factors for each biological group tended to increase or decrease in eight environmental factors (altitude, slope, current velocity, fraction of fine particles in substrates, turbidity, $BOD_5$, T-N and T-P). As a result of discriminant analysis (hit ratio: 69.6 %), environmental factors that significantly influence the distribution of each biological group were $BOD_5$, altitude, catchment area, T-N, T-P and fraction of fine particles in substrates. The discriminant functions 1 and 2 classified the existing six biological groups (clustered by TWINSPAN) into five (group 1: highland highly sensitive group; group 2: lowland sensitive group, group; 3 and 4: lowland slightly tolerant group; group 5: lowland tolerant group; group 6: lowland highly tolerant group). Most of the benthic macroinvertebrates were identified in the species level. However, a few others were identified in genus or family level, too. Even the organisms belonging to the same genus or family may prefer different environmental factors in the species level, the identification of characteristic of the habitat environment preferred by organism and species level identification of these organisms are utmost. The results of this study may be used as basic information on environmental factors which can be considered in preservation and restoration research for biodiversity.
