Effects of Taurine Supplementation on the Growth Performance of Juvenile Rock Bream Oplegnathus fasciatus

  • An 8-week feeding trial was conducted to investigate the effects of dietary taurine supplementation on the growth performance of juvenile rock bream, Oplegnathus fasciatus. Triplicate groups of 25 fish averaging $2.72{\pm}0.04$ g ($mean{\pm}SD$) were fed one of six experimental diets prepared by adding taurine at 0%, 0.25%, 0.5%, 1.0%, 1.5% and 3.0% (Control, $Tau_{0.25}$, $Tau_{0.5}$, $Tau_{1.0}$, $Tau_{1.5}$ and $Tau_{3.0}$, respectively). At the end of the feeding trial, the weight gain, specific growth rate and protein efficiency ratio of fish fed the $Tau_{0.5}$, $Tau_{1.0}$ and $Tau_{1.5}$ diets were significantly higher (P < 0.05) than those of fish fed the Control and $Tau_{0.25}$ diets. The feed efficiency of fish fed the $Tau_{0.5}$ diet was significantly higher than that of fish fed the Control, $Tau_{0.25}$ and $Tau_{3.0}$ diets. Fish fed the $Tau_{1.0}$ diet had higher whole-body crude protein content than fish fed the Control diet, while the crude lipid content of fish fed the $Tau_{1.5}$ and $Tau_{3.0}$ diets was significantly lower than that of fish fed the Control and $Tau_{0.25}$ diets. An ANOVA suggested that the optimum level of dietary taurine supplementation to improve growth and reduce the body lipid contents of juvenile rock bream, O. fasciatus, was 0.5%, while a broken line analysis of weight gain indicated a level of 0.62%.

  • This study was to explore lived experience of middle-aged women with aging. The ultimate purpose of this inquiry was to discover the essence of middle-aged women's experience of aging and to promote understanding. Method: This inquiry was performed by using Van Manen's hermeneutical phenomenological approach to make more plausible interpretation of experience. First-handed experiences were explored through multi-stage in-depth interview with 6 women aged between 40 and 64. Second- handed experiences were explored with text such as essay, novel, and photographs. Result: As the process of reflecting and analysing the data of experience were performed, essential themes were emerged: striking onset of event, discomfort and tired body and mind, everything in ones mind, age of harvest gaining much more than loss. Conclusion: This inquiry would be a cornerstone for humanistic nursing care for the mid-aged women.

A Study on the Variation of Aerosol Optical Depth according to Aerosol Types in Northeast Asia using Aeronet Sun/Sky Radiometer Data (AERONET 선포토미터 데이터를 이용한 동북아시아 지역 대기 에어로졸 종류별 광학적 농도 변화 특성 연구)

  • This study has developed a technique to divide the aerosol optical depth of the entire aerosol (${\tau}_{total}$) into the dust optical depth (${\tau}_D$) and the pollution particle optical depth (${\tau}_P$) using the AERONET sun/sky radiometer data provided in Version 3. This method was applied to the analysis of AERONET data observed from 2006 to 2016 in Beijing, China, Seoul and Gosan, Korea and Osaka, Japan and the aerosol optical depth trends of different types of atmospheric aerosols in Northeast Asia were analyzed. The annual variation of ${\tau}_{total}$ showed a tendency to decrease except for Seoul where observation data were limited. However, ${\tau}_D$ tended to decrease when ${\tau}_{total}$ were separated as ${\tau}_D$ and ${\tau}_P$, but ${\tau}_P$ tended to increase except for Osaka. This is because the concentration of airborne aerosols, represented by Asian dust in Northeast Asia, is decreased in both mass concentration and optical concentration. However, even though the mass concentration of pollution particles generated by human activity tends to decrease, Which means that the optical concentration represented as aerosol optical depth is increasing in Northeast Asia.

  • In normal cells tau protein is associated with axonal microtubules, whereas in Alxheimer's disease it is immobilized in the somatodendritic compartment of certain nerve cells as a major component of the paired helical filament. As a part of the study to analyze the nature of the paired helical filament (PHF) deposits and some related factors in brain, we have cloned and expressed a human tau gene cDNA in Escherichia coli to obtain the recombinant human tau protein in abundance.

  • We study multivariate extension of Kendall's tau and its statistical interpretation. There exist various versions of multivariate Kendall's tau, for example Scarsini (1984), Joe (1990) and Genest et al. (2011); however, few of them mention its lower bounds. For the bivariate case, the Fr$\acute{e}$chet-Hoeffding lower bound can achieve the lower bound of Kendall's tau. However in the multivariate case, the Fr$\acute{e}$chet-Hoeffding lower bound itself does not exist as a distribution, which makes the interpretation of Kendall's tau unclear when it has negative value. In this paper, we explain sufficient conditions to achieve the lower bound of Kendall's tau and provide real data examples that provide further insights into the interpretation for the lower bounds of Kendall's tau.

Sequestration of sorcin by aberrant forms of tau results in the defective calcium homeostasis

  • Neurofibrillary tangles (NFTs) of microtubule-associated protein tau are a pathological hallmark of Alzheimer's disease (AD). Endoplasmic reticulum (ER) stress has been known to be involved in the pathogenesis of AD. However, the exact role of ER stress in tau pathology has not yet been clearly elucidated. In present study, the possible relationship between tau pathology and ER stress was examined in terms of sorcin, which is a calcium binding protein and plays an important role in calcium homeostasis. Our previous yeast two hybrid study showed that sorcin is a novel tau interacting protein. Caspase-3-cleaved tau (T4C3) showed significantly increased tau-sorcin interaction compared to wild type tau (T4). Thapsigargin-induced ER stress and co-expression of constitutively active $GSK3{\beta}$ ($GSK3{\beta}-S9A$) also exhibited significantly increased tau-sorcin interactions. T4C3-expressing cells showed potentiated thapsigargin -induced apoptosis and disruption of intracellular calcium homeostasis compared to T4-expressing cells. Overexpression of sorcin significantly attenuated thapsigargin-induced apoptosis and disruption of calcium homeostasis. In contrary, siRNA-mediated knock-down of sorcin showed significantly increased thapsigargin-induced apoptosis and disruption of calcium homeostasis. These data strongly suggest that sequestration of sorcin by aberrant forms of tau compromises the function of sorcin, such as calcium homeostasis and cellular resistance by ER stress, which may consequently result in the contribution to the progression of AD.

The Effect of Treadmill Exercise on Tau Hyperphosphorylayion in an Aged Transgenic Mouse Model of Taupathies

  • Alzheimer's disease (AD) is the most common cause of dementia in adults. Microtubule associated protein tau is abnormally phosphorylated in AD and aggregates as paired helical filaments (PHFs) in neurofibrillary tangles (NFTs). NFTs are the most common intraneuronal inclusion in the brains of patients with AD and have been implicated in mediating neuronal cell death and cognitive deficit. Aberrant phosphorylation of tau is an early pathological event in AD, but the underlying mechanisms are unclear. MAP kinases are a family of Serine/Threonine (Ser/Thr) kinases that involved hyper - phosphorylation of tau in AD. The purpose of this study was to investigate the effect of treadmill exercise on phosphorylation of tau level and activation of MAPKs including JNK, ERK, p38-MAPK. To address this, Tg mouse model of AD, Tg-NSE/hTau 23, which expresses human tau 23 in the brain, was chosen. Animals were subjected to treadmill exercise for 12 weeks from 24 months of age. Treadmill exercise in Tg group improved cognitive function compared with Tg-SED group in watermaze test. In addition, treadmill exercised Tg mice significantly reduced the activation of JNK54/46, p38-MAPK and tau (Ser404, Ser202, Thr231), and increased activation of ERK44/42 in cerebral cortex. These results suggest that treadmill exercise may provide a therapeutic potential to alleviate the tau pathology like AD.

The C-terminal Region of Human Tau Protein with Ability of Filament Formation

  • Tau protein is one of the microtubule-associated proteins in the mammalian brain. In Alzheimer's disease, tau protein is immobilized in the somatodendritic compartment of certain nerve cells, where it forms a part of the paired helical filament (PHF). To understand the role of tau protein in the formation of PHF, a recombinant human tau protein expressed in Escherichia coli and five synthetic peptide fragments (peptide 1 to peptide 5), corresponding to the C-terminal region of tau protein, were prepared and their ability in self-assembly to form filamentous structures was examined. The recombinant human tau protein formed short rod-like structures in 0.1M MES buffer containing 1 mM $MgCI_2$, while a synthetic peptide fragment 1 containing 55 amino acid residues could assemble into a lot of long filamentous structures in water and particularly twisted helical structures in 0.1M MES buffer containing 1 mM $MgCI_2$. This suggests that the C-terminal region possesses a filament-forming ability and may be related to the formation of the helical structure by providing a powerful filament-forming driving force.
