Structural Basis of the Disease-related Proteins: Target Oriented Structural Proteomics

  • To discover new drugs more quickly and more efficiently, pharmaceutical companies and biotechnology firms are increasingly turning to the genomics and the structural proteomics technologies. Structural-proteomics can provide a foundation for this through the determination and analysis for protein structure on a genomics scale. Among many structures determined by CGI, we will present with the representative examples drawn from our work on novel structures or complex structures of the disease-related proteins. The alpha subunit of Hypoxia-inducible factor (HIF) is targeted for degradation under normoxic conditions by an ubiquitin-ligase complex that recognizes a hydroxylated proline residue in HIF, Hydroxylation is catalysed by HIF prolyl 4-hydroxylases (HIFPH) which are Fe(II) and 2-oxoglutarate (2-OG) dependent oxygenases. Here, we discuss the first crystal structure of the catalytic domain of HIFPH in complexes, with the Fe(II)/2-OG at 1.8 ${\AA}$. These structures suggest that the L1 region (residues 236-253), which is also conserved in mammals, form a ‘lid’ that closes over the active site. The structural and mutagenesis analyses allow us to provide a focus for understanding cellular responses to hypoxia and a target for the therapeutic manipulation.

An Important Role of Nrf2-ARE Pathway in the Cellular Defense Mechanism

  • The antioxidant responsive element (ARE) is a cis-acting regulatory element of genes encoding phase II detoxification enzymes and antioxidant proteins, such as NAD(P)H: quinone oxidoreductase 1, glutathione S-transferases, and glutamate-cysteine ligase. Interestingly, it has been reported that Nrf2 (NF-E2-related factor 2) regulates a wide array of ARE-driven genes in various cell types. Nrf2 is a basic leucine zipper transcription factor, which was originally identified as a binding protein of locus control region of ss-globin gene. The DNA binding sequence of Nrf2 and ARE sequence are very similar, and many studies demonstrated that Nrf2 binds to the ARE sites leading to up-regulation of downstream genes. The function of Nrf2 and its downstream target genes suggests that the Nrf2-ARE pathway is important in the cellular antioxidant defense system. In support of this, many studies showed a critical role of Nrf2 in cellular protection and anti-carcinogenicity, implying that the Nrf2-ARE pathway may serve as a therapeutic target for neurodegenerative diseases and cancers, in which oxidative stress is closely implicated.

A study on design of instantaneous field of view of rosette scanning infrared seeker (로젯 주사 적외선 탐색기의 순시 시계 설계에 관한 연구)

  • The rosette-scan seeker is a device mounted on the infrared guided missile. It offers the positions and iamges of target to missiles servo system by scanning a space about target in rosette pattern with a single detector. An instantaneous field of view (IFOV), which is a diameter of a detector moving along the path of the rosette pattern, has the property that its smaller size provide the less interference of background signals and detector noise. If its size is too small to voer the total field of view (TFOV), however, it produces the invisible regions in the TFOV. In this case, the invisible regions cause the performance of the seeker to deteriorate. For full scan-coverage, it is necessary to design the small IFOV without the invisible regions in the TFOV, as possible. In this paper, we propose the new method of designing the smaller IFOV than the conventional method and verify full coverage of the scanned region. By comparing the nose equivalent flux density (NEFD) of the proposed method with the that of the conventional one, we confirm that the former is better than the latter in terms of performance.

  • This study aims to provide the system to help decision making by finding key indicators used in land supply and demand surveys, and by developing storage priority areas and projects. As problems arose such as the rising cost of public projects or disruptions to business progress due to the rapid rise in land prices and the cost of compensation for public projects in the past, a new land policy was adopted and the land reserve system was established in 2009. At that time, The bank's goal was to conserve 2 trillion won of land per year, but the result was sluggish as it accumulated a total of 1.6 trillion won from 2009 to 2015. The reason for this sluggish result is that the type and quantity of indicators are still extensive and the survey of land supply and demand has a problem of poor utilization. In order to make up for the sluggishness, we made key indicators used in land supply and demand surveys using the question investigation, and analyzed the priority of the project areas. This study provide the system to help decision making by finding key indicators used in land supply and demand surveys, and the priorities of project areas.

  • This paper proposes an AVI system which recognizes the license plate and the driver's face simultaneously using passive AVI techniques. For this, firstly, the pro-processing algorithm independent of the environment is proposed and region extraction of the car number plate and the driver's face is described. Secondly, characters are separated and recognition parameters are extracted from target regions. Thirdly, template matching of car number plate is performed and the fuzzy relation matrix of driver face is made for the final recognition processes. The merits of the proposed system are following : Pre-processing is accomplished regardless of the environment. The application areas of conventional AVI system can be expanded in the content that the driver's face is also recognized in the proposed system compared with only the number plast is recognized in the existing systems.

Fast Remote Detection Algorithms for Chemical Gases Using Pre-Detection with a Passive FTIR Spectrometer (수동형 FTIR 분광계에서 초동 탐지 기법을 이용한 고속 원거리 화학 가스 탐지 알고리즘)

  • In this paper, we propose a fast detection and identification algorithm of chemical gases with a passive FTIR spectrometer. We use a pre-detection algorithm that can reduce the spatial region effectively for gas detection and the candidates of the target. It is possible to remove background spectra effectively from measured spectra with the least-squares method. The CC(Correlation Coefficients) and the SNR(Signal-to-Noise Ratio) methods are used for the detection of target gases. The proposed pre-detection algorithm allows the total process of chemical gas detection to be performed with lower complexity compared with the conventional algorithms. This paper can help developing real-time chemical detection instruments and various applications of FTIR spectrometers.

Analysis of the Characteristics of Thermal Environment Change Due to Urban Stream Restoration (도심 하천 복원에 따른 주변지역 열환경 변화 특성 분석)

  • The purpose of this study is to quantitatively analyze the effects of a restoration project on the decrease in the temperature in the surrounding areas. The thermal environment characteristics of the investigation area were analyzed using the meteorological data from the Busanjin Automatic Weather System which is closest to the target area. The terrain data of the modeling domain was constructed using a digital map and the urban spatial information data, and the numerical simulation of the meteorological changes before and after the restoration of the stream was performed using the Envi-met model. The average temperature of the target area in 2016 was $15.2^{\circ}C$ and was higher than that of the suburbs. The monthly mean temperature difference was the highest at $1.1^{\circ}C$ in November and the lowest in June, indicating that the temperatures in the urban areas were high in spring and winter. From the Envi-met modeling results, reductions in temperature due to stream restoration were up to $1.7^{\circ}C$ in winter, and decreased to $3.5^{\circ}C$ in summer. The effect of temperature reduction was seen in the entire region where streams are being restored.

Resumption of School Face-to-Face Classes and Analysis of Secondary Infected Persons in COVID 19 : Applying the Monte-Carlo Method (학교 대면 수업 재개와 2차 감염자 분석 : 몬테카를로 기법 적용을 중심으로)

  • In this study, we estimated the number of secondary COVID-19 infections caused by students with potential transmission potential home. When the existing Monte Carlo method was applied to Korean data, the average number of household members of the second COVID-19 infected was predicted. The summary of this study is as follows. First, in general, the number of secondary infections by students returning home from school is greatly influenced by the virus infection rate of each student group they contact while returning home from school. Korea-based empirical research on this is needed. Second, the number of secondary infections by Korean students was relatively lower than that of previous studies. This can be interpreted as being due to the domestic furniture structure. Third, unlike previous studies that assumed the distribution of secondary infected individuals as normal distribution, assuming a negative binomial distribution, the number of secondary infected individuals was sensitively changed according to the estimated parameters. Interpretation of this result shows that the number of secondary infections may vary depending on the time of decision making, the target region, and the target student group. Finally, according to the results of this analysis, a proposal was made to support education policy decisions.

High-accuracy quantitative principle of a new compact digital PCR equipment: Lab On An Array

  • Digital PCR (dPCR) is the third-generation PCR that enables real-time absolute quantification without reference materials. Recently, global diagnosis companies have developed new dPCR equipment. In line with the development, the Lab On An Array (LOAA) dPCR analyzer (Optolane) was launched last year. The LOAA dPCR is a semiconductor chip-based separation PCR type equipment. The LOAA dPCR includes Micro Electro Mechanical System that can be injected by partitioning the target gene into 56 to 20,000 wells. The amount of target gene per wells is digitized to 0 or 1 as the number of well gradually increases to 20,000 wells because its principle follows Poisson distribution, which allows the LOAA dPCR to perform precise absolute quantification. LOAA determined region of interest first prior to dPCR operation. To exclude invalid wells for the quantification, the LOAA dPCR has applied various filtering methods using brightness, slope, baseline, and noise filters. As the coronavirus disease 2019 has now spread around the world, needs for diagnostic equipment of point of care testing (POCT) are increasing. The LOAA dPCR is expected to be suitable for POCT diagnosis due to its compact size and high accuracy. Here, we describe the quantitative principle of the LOAA dPCR and suggest that it can be applied to various fields.

Development of mRNA Vaccines/Therapeutics and Their Delivery System

  • The rapid development of mRNA vaccines has contributed to the management of the current coronavirus disease 2019 (COVID-19) pandemic, suggesting that this technology may be used to manage future outbreaks of infectious diseases. Because the antigens targeted by mRNA vaccines can be easily altered by simply changing the sequence present in the coding region of mRNA structures, it is more appropriate to develop vaccines, especially during rapidly developing outbreaks of infectious diseases. In addition to allowing rapid development, mRNA vaccines have great potential in inducing successful antigen-specific immunity by expressing target antigens in cells and simultaneously triggering immune responses. Indeed, the two COVID-19 mRNA vaccines approved by the U.S. Food and Drug Administration have shown significant efficacy in preventing infections. The ability of mRNAs to produce target proteins that are defective in specific diseases has enabled the development of options to treat intractable diseases. Clinical applications of mRNA vaccines/therapeutics require strategies to safely deliver the RNA molecules into targeted cells. The present review summarizes current knowledge about mRNA vaccines/ therapeutics, their clinical applications, and their delivery strategies.
