Estimation of Reservoir Discharge to Support TMDL Management in the Geum River Basin (금강수계 오염총량관리를 고려한 저수지 방류량산정)

  • This study estimates adequate discharge to meet the specified target water quality concentration using the pollutant load of the Geum river basin given in TMDL (Total Maximum Daily Load) report. During the 1st phase, BOD is chosen as a target water quality constituent under regulation of the Ministry of Environment in Korea. BOD, TN, and TP loads estimated based on the TMDL and provincial zones were re-distributed for 10 major tributaries, and the remaining areas along the main river are classified as 15 incremental flow areas. Water quality modeling was conducted using Qual2E for the low flow period of a year (i.e. $March{\sim}April$). The results of the model simulation showed that about 30 cms from the Daechung dam would be sufficient to satisfy the target water quality in the Geum river downstream of the Daechung multipurpose Dam.

Effects site concentrations of propofol using target-controlled infusion in dental treatment under deep sedation among different intellectual disability types

  • Background: We aimed to assess the dose needed to achieve the propofol effect-site concentration using target-controlled infusion in intellectually disabled patients and to detail the most effective method for achieving a safe level of consciousness without hemodynamic changes as well as detail any resulting adverse effects. Methods: We performed a retrospective review of sedation service records of 138 intellectually disabled patients (51, mental retardation; 36, autism; 30, brain lesion, 12 genetic diseases, 9 dementia) aged over 15 years and weighing over 30 kg. These patients had received propofol via target-controlled infusion in the special care dental clinic of Seoul National University Dental Hospital from May 2008 to September 2018 for restorative treatment (112), minor surgery (13), prosthodontics (7), periodontics treatment (5), and implant (1). Results: For all groups, the duration of dental treatments was $43{\pm}18$ minutes, total sedation time was $73{\pm}23$ minutes, and total BIS values was $57{\pm}12$. The propofol maintenance dosage values for each group were: mental retardation, $3{\pm}0.5(2-4){\mu}g/ml$; autism, $3.1{\pm}0.7(2-5){\mu}g/ml;$; brain lesion, $2.8{\pm}0.7(1.5-5){\mu}g/ml;$; genetic disease, $2.9{\pm}0.9(1-4){\mu}g/ml;$ and dementia $2.3{\pm}0.7(1-3.4){\mu}g/ml;$. Conclusions: The dementia group needed a lower dosage to reach a safe, effective propofol effect-site concentration than the other groups. Since there were no complications, deep sedation is a great alternative to general anesthesia for dental treatment of intellectually disabled patients.

A Study on the VOCs Contamination of Ambient Air int Kyonggi-do Area (III) (경기도 환경대기중 휘발성유기화합물의 오염실태 조사연구 (III))

  • We investigated the concentration degree of VOCs for ambient air in the air environment control area of Kyonggi-do in 2001. The VOCs showing higher concentration were in the ascending order of Toluene > n-Butane > 1-Butene > Benzene > n-Hexane. The concentration distribution ratio mentioned above showed almost same pattern in each city. The relative concentration ratio of Benzene, Toluene, Ethylbenzene, Xylenes which are aromatic VOCs were 1 : 6.3 : 0.6 : 1.6. It was similar to the pattern in 1999 but different from the pattern in 2000. In the aspect of seasons, the highest B.T.E.X concentration was indicated in fall and in the aspect of areas, the higher concentration showed in the ascending order of Buchon > Ansan > Kwangmyong > Shihung > Sungnam. The average concentration of target VOCs for ambient air in the air environment control area of Kyonggi-do showed lower concentration than the metropolis of Seoul but higher concentration than Kwang-ju city and Dae-gu city. The amount of total VOCs was increasing, comparing with it last year and the more increasing tendency is anticipated so that it will need to establish the proper regulative criteria.

Production of Phagocyte Activating Supernatants by Olive Flounder (Paralichthys olivaceus) Leucocytes Stimulated with Genomic DNA of Escherichia coli

  • Effects of Escherichia coli genomic DNA on the production of phagocyte activating supernatants by the head kidney leucocytes isolated from olive flounder (Paralichthys olivaceus) were investigated. Phagocyte activating activity of the supernatants was estimated by. measuring reactive oxygen species (ROS) production in target head kidney phagocytes. All supernatants from olive flounder head kidney leucocytes-stimulated with E. coli DNA induced significantly (P<0.01) higher ROS production from target pagocytes than the unstimulated control supernatant. Maximum enhancement of chemiluminescent response was observed $5.0-10.0{\mu}g\;mL^{-1}$ of bacterial DNA while the increment ability was decreased significantly (P<0.01) at the concentration of $20.0{\mu}mL^{-1}$. The results demonstrate that olive flounder head-kidney leucocytes stimulated with bacterial DNA release a soluble phagocyte activating cytokines capable of enhancing the respiratory burst activity from target phagocytes.

Buffer-Optimized High Gradient Magnetic Separation: Target Cell Capture Efficiency is Predicted by Linear Bead-Capture Theory

  • High gradient magnetic separation (HGMS) is the most commonly used magnetic cell separation technique in biomedical science. However, parameters determining target cell capture efficiencies in HGMS are still not well understood. This limitation leads to loss of information and resources. The present study develops a bead-capture theory to predict capture efficiencies in HGMS. The theory is tested with CD3- and CD14-positive cells in combination with paramagnetic beads of different sizes and a generic immunomagnetic separation system. Data depict a linear relationship between normalized capture efficiency and the bead concentration. In addition, it is shown that key biological functions of target cells are not affected for all bead sizes and concentrations used. In summary, linear bead-capture theory predicts capture efficiency ($E_t$) in a highly significant manner.

Evaluation of Carryover Contamination on Autopipetting System (자동분주기의 이월오염 평가)

  • Purpose: Autopipetting system is an efficient automated equipment pipetting patient samples and reagents for rapid and accurate test. However, it can cause carryover between high concentration sample and low concentration sample. We evaluated carryover contamination of TECAN freedom Evo 100 autopipetting system. Materials and Method: We studied carryover contamination of $\alpha$-fetoprotein (AFP) and carcinoembryonic antigen (CEA) test on TECAN freedom Evo 100 autopipetting system. Very low concentration control samples were pipetted for comparison to the contaminated very low concentration samples. Then, The contaminated very low concentration samples were pipetted following the high concentration samples were pipetted alternately. The difference of low concentration samples represents carryover. The target value to decide carryover was 1ppm (parts per million). Results: For AFP, the mean values of the uncontaminated control samples and the contaminated samples were less than 0.6 IU/mL (the l imit of detection (LoD)). Carryover did not occur even though the high concentration sample which value was 650000 IU/mL. For CEA, the values of the low concentration control samples and the contaminated samples were less than 0.2 ng/mL (LoD). Carryover did not occur even though the high concentration sample which value was 65,000 ng/mL. Conclusions: Sample carryover was not found on TECAN freedom Evo 100 autopipetting system for AFP, CEA. However, carryover is a potential problem with automated instruments and robotic pipetting systems. Therefore, Clinical laboratories must periodically verify carryover contamination for the accurate and confidential test results.

Physical and Electrical Properties of Nanocrystalline Carbon Films Prepared with Ti Concentration for Contact Strip Application of Electric Railway (전기철도 집전판 응용을 위한 Ti 나노금속 함량에 따른 나노결정 탄소박막의 물리적, 전기적 특성)

  • In this work, we have fabricated the nanocrystalline carbon films by using unbalanced magnetron sputtering method with graphite and Ti targets for contact strip application of electrical railway. The power density of graphite target was fixed and the power density was increased for the increase of Ti concentration in TiC films. We investigated the hardness, surface roughness, contact angle, resistivity, HRTEM and XPS of TiC films with Ti concentration. The hardness and resistivity were improved with increasing Ti concentration. These results indicate that the improvement of hardness and resistivity is related to the increase of sp2 clusters in TiC films.

  • Hypertonicity (hypernatremia) of extracellular fluid causes water movement out of cells, while hypotonicity(hyponatremia) causes water movement into cells, resulting in cellular shrinkage or cellular swelling, respectively. In most part of the body, the osmolality of extracellular fluid is maintained within narrow range($285-295 mOsm/kgH_2O$) and some deviations from this range are not problematic in most tissue of the body except brain. On the other hand, the osmolality in the human renal medulla fluctuates between 50 and $1,200 mOsm/kgH_2O$ in the process of urine dilution and concentration. The adaptation of renal medullary cells to the wide fluctuations in extracellular tonicity is crucial for the cell survival. This review will summarize the mechanisms of urine concentration and the adaptation of renal medullary cells to the hyper tonicity, which is mediated by TonEBP transcription factor and its target gene products(UT-A1 urea transporter etc.).

  • Microfluidic preconcentration technologies, which collect or extract low-abundance analytes in a specific location, have been spotlighted in various applications such as medical and bio-engineering. Here, we conducted extensive studies on the variables to be considered when concentrating target samples based on electrophoresis in an electromembrane system utilizing an ion exchange membrane. Using negatively charged Alexa Fluor 488 and positively charged Rhodamine 6G as fluorescent dyes, we examined the effects of flow velocity of the main channel, channel electrolyte concentration, and applied voltage on sample preconcentration. As a result, it was found that the preconcentration of the target sample occurs much better when the flow velocity is slow and the concentration of the main channel containing the sample is high, given that the channel concentration ratio (main and buffer) is constant. In addition, based on the experimental data acquired in this study, the electrophoretic mobility values of Alexa Fluor 488 and Rhodamine 6G were experimentally calculated and compared.

Cu2ZnSnSe4 Thin Films Preparation by Pulsed Laser Deposition Using Powder Compacted Target

  • $Cu_2ZnSnSe_4$ thin films for solar absorber application were prepared by pulsed laser deposition of a synthesized $Cu_2ZnSnSe_4$ compound target. The film's composition revealed that the deposited films possess an identical composition with the target material. Further film compositional control toward a stoichiometric composition was performed by optimizing substrate temperature, deposition time and target rotational speed. At the optimum condition, X-ray diffraction patterns of films showed that the films demonstrated polycrystalline stannite single phase with a high degree of (112) preferred orientation. The absorption coefficient of $Cu_2ZnSnSe_4$ thin films were above 104 cm.1 with a band gap of 1.45 eV. At an optimum condition, films were identified as a p type semiconductor characteristic with a resistivity as low as $10^{-1}{\Omega}cm$ and a carrier concentration in the order of $10^{17}cm^{-3}$.
