Plant Regeneration and Effect of Auxin and Cytokinin on Adventitious Shoot Formation from Seedling Explant of Taraxacum platycarpum (민들레 [Taraxacum platycarpum]유식물 절편으로부터 부정아 발생에 미치는 auxin과 cytokinin의 영향 및 식물체 재생)

  • Taraxacum platycarpum has been used as a medicinal plant. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on medium with various kinds of growth regulators. Adventitious shoot formation was achieved when cytokinin was used alone. Shoot formation was higher on MS medium containing 2 mg/L BAP compared to that with 2 mg/L kinetin and 2 mg/L 2-ip. Among root, hypocotyl and cotyledon, roots were the best explant for the adventitious shoot induction. Adventitious shoot formation from roots declined markedly by the combination of both 0.1 mg/L NAA and 2 mg/L BAP, while shoot formation from cotyledons was stimulated by the same combination. Root formation from the regenerated shoots was achieved on 1/3MS medium containing 0.2 mg/L NAA. Regenerated plantlets was acclimatized and transplanted to the soil, showing 100% survival.

Antidepressant effect of water extract of Taraxacum platycarpum through BDNF, ERK and CREB pathway (BDNF, ERK 및 CREB 경로를 통한 포공영 추출물의 항우울 효과)

  • Objectives : Taraxacum platycarpum H. Dahlstedt has been reported to have several biological properties such as skin hydration and antiinflammation. The purpose of this study was to examine the antidepressive effects of water extract of T. platycarpum (WTP) on an animal model of depression. Methods : In the present study, normal ICR mice (4 weeks) were used, and orally administered with WTP (25, 50 and 100 mg/kg). Depression-like behavior was monitored the forced swimming test (FST) and tail suspension test (TST) in mice. The locomotor activity was evaluated to eliminate the false-positive activity in the open field test (OFT). Fluoxetine, the selective serotonin reuptake inhibitor, as a positive control was intraperitoneally administered at a dose of 15 mg/kg at 30 min before starting the behavioral test. Moreover, we evaluated the effects of WTP on the expression of brain-derived neurotrophic factor (BDNF) and the extracellular signal-regulated kinase (ERK)/ cyclic AMP response-element binding protein (CREB) signaling pathway in the hippocampus using Western blot. Results : The administration of WTP (50 and 100 mg/kg) significantly (P < 0.05, respectively) reduced the immobility time during FST and TST without accompanying changes in locomotor activity by OFT. Furthermore, WTP at dose of 100 mg/kg increased the BDNF expression and the phosphorylation of ERK and CREB in the hippocampus region. Conclusions : These results suggest that WTP has a useful anti-depressant effect through the regulation of BDNF/ERK/CREB signaling pathway.

Effect of Presowing Seed Treatments on Germination and Seedling Emergence in Taraxacum platycarpum (민들레 종자의 파종전 처리가 발아율 및 입모율에 미치는 영향)

  • Several presowing seed treatments have been used to facilitate the seed germination and seedling emergence. The study was performed to design a model of presowing seed treatments in Taraxacum platycarpum. Germination and seedling growth responses to temperature, drying, chilling, $GA_3$ and priming were investigated to know the optimum germination conditions and presowing treatment. The optimum temperature condition for germination and seedling growth were $15{\sim}20^{\circ}C$ at dark condition. The seeds exposed to infra red light, however, did not germinate. The moisture content of seeds dried at $35^{\circ}C$ for 3.5 hours was returned to the level of unimbibed seeds. The germination of seeds dried under dark condition was better than those dried under infra red light. Prechilling treatment imposed for 6 weeks showed higher germination rate than $GA_3$ treatment or priming. In field test, the highest field emergence was observed in the seeds dried after prechilled for 6 weeks and followed by drying followed by $GA_3$ > low temperature > $KNO_3$ priming. Imbibition with 0.1 mM $GA_3$ solution for 2 hours then drying at 35 for 3.5 hour under dark condition was effective to the practical presowing treatment for Taraxacum seeds.

Anti-Inflammatory Activities of Extracts from Fermented Taraxacum platycarpum D. Leaves Using Hericium erinaceum Mycelia (노루궁뎅이버섯 균사체로 발효한 민들레잎 추출물의 항염증 활성)

  • This study investigated the fermentation effect of Taraxacum platycarpum Dahlst. leaf extracts using Hericium ernaceum mycelia to test antioxidant and anti-inflammatory activities in vitro. The antioxidant activities of fermented or non-fermented extracts of T. platycarpum leaves were determined by 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, ferric reducing antioxidant power, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity. The leaf extract of T. platycarpum showed higher antioxidant activity than extract of fermented leaves. However, ethanolic extract of fermented T. platycarpum leaves decreased levels of nitric oxide production and pro-inflammatory cytokines such as interleukin-6 and tumor necrosis factor-${\alpha}$ in lipopolysaccharide-stimulated RAW 264.7 cells. Moreover, fermented leaf extract suppressed protein expression of inducible nitric oxide synthase in RAW 264.7 cell culture. Therefore, the enhanced anti-inflammatory activity of ethanolic extracts of fermented T. platycarpum leaves might be attributed to the molecular conversion of leaf ingredients during fermentation and the active ingredients might have specific affinity with ethanol during extraction.

Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.

Screening for Herbicidal Medicinal Plants against Digitaria Sanguinalis and Taraxacum Platycarpum in Turf (잔디밭 잡초 바랭이와 민들레 방제를 위한 살초활성 약용식물의 탐색)

  • Crabgrass (Digitaria sanguinalis L.), Dandelion (Taraxacum platycarpum H. DAHLST.), Annual bluegrass (Poa annua L.), Gree kyllinga (Kyllinga brevifolia var.), Red clover (Trifolium pratense L.), Annual lespedeza (Kummerowia striata (Thunb.)Schindl.), Mugwort (Artemisia capillaris Thunb.), Horseweed (Erigeron canadensis L.), Field horsetail (Equisetum arvense L.) are major weeds in turf. In this study, 23 medicinal plant extracts were tested for herbicidal activities against crabgrass and dandelion in the course of major weed in turf. In pot experiment, we implemented pre-emergence application used medicinal plant extracts showed herbicidal activity in petridish experiment. In effect, inhibition rates of germination were 61.0% at Curcuma longa extract and 62.5% at Cnidium officinale extract in crabgrass pot experiment, and 77.8% at Ailanthus altissima extract in dandelion pot experiment.

The Inhibitory Effects of Chloroform Fraction Extracted from the Dandelion (Taraxacum platycarpum D.) against Lactic Acid Bacteria and Yeast Related to Kimchi Fermentation (포공영 클로로포름 분획의 김치 유산균 및 효모에 대한 저해 효과)

  • This study was conducted to investigate the effects of solvent extracts from medicinal plants on the fermentation of kimchi. Five microorganisms related to kimchi fermentation were selected and the antimicrobial activities of solvent fractions from medicinal plants were investigated. The chloroform fraction from the methanol extract of dandelion (Taraxacum platycarpum D.) exhibited inhibitory activity against five strains such as Lactobacilli plantarum, Lactobacilli brevis, Leuconostoc mesenteroides, Enterococcus faecalis and Saccharomyces cerevisiae. The chloroform fraction from the methanol extract of Dandelion inhibited the growth of E. faecalis and Leu. mesenteroides at the concentration of 80 mg/mL. Scanning electron micrographs of Leu. mesenteroides and E. faecalis treated with chloroform fraction 80 mg/mL exhibited morphological changes, including irregularly contracted cell surface.

The Study on Antioxidant and Anti-inflammatory Effects of Taraxacum platycarpum H. Dahlstedt, Lonicera japonica Thunberg and Leonurus japonicus Houtt. Complex (포공영, 금은화, 익모초 혼합물의 항산화 및 항염증 효과에 관한 연구)

  • Objectives: This study was designed to experiment with the antioxidant and anti-inflammatory effects of Taraxacum platycarpum H. Dahlstedt, Lonicera japonica Thunberg, and Leonurus japonicus Houtt. complex (TLL) in LPS-induced RAW264.7 cell. Methods: The antioxidant activity of TLL was measured by FRAP assay, DPPH radical scavenging activity, ABTS radical scavenging activity. Total polyphenol and flavonoid contents of TLL were measured by using standard methods. The anti-inflammatory effects of TLL were measured by NO production, biomarker production (PGE2, IL-1β, IL-6, TNF-α), mRNA expression level (iNOS, COX-2, IL-1β, IL-6, TNF-α) and protein expression level (ERK, JNK, p38). Results: Total polyphenol and flavonoid contents in TLL were 58.03±1.02 mg of Gallic acid equivalents (GAE)/g and 16.58±0.60 mg of Quercetin equivalents (QE)/g respectively. In FRAP assay, DPPH and ABTS radical scavenging activity, a concentration-dependent increase in TLL was observed. To explore antioxidant and anti-inflammatory effects of TLL, RAW 264.7 cells were treated with TLL and LPS for 24 hours. Cell viability of RAW 264.7 cells were measured by adding EZ-Cytox, It was remarkably increased at 50, 100, 200 ㎍/㎖ concentrations of TLL. NO, ROS, iNOS, IL-1β, IL-6, TNF-α, ERK, JNK and p38 were remarkably decreased at 50, 100, 200 ㎍/㎖ concentrations of TLL compared to the control group. PGE2 and COX-2 were remarkably decreased at 100, 200 ㎍/㎖ concentrations Conclusion: These results suggest that TLL complex has antioxidant and anti-inflammatory effects.

Vegetation Characteristics of Main Herbaceous Flowers for Ecological Restoration (생태복원용 주요 초화류의 녹화특성)

  • Research was initiated to investigate vegetation coverage and growth of main herbaceous flowers under low maintenance condition like a wild state. Eleven entries were comprised of 1 annual, 2 biennials and 8 perennials from widely used herbaceous flowers in Korea. Vegetation coverage and germination rate were high on annual and biennials when compared to perennials. Callistephus chinensis showed high vegetation coverage but lasted only for a year in 2001 and disappeared in 2002 as an annual. Oenothera odorata and Oenothera lamarckiana were also high vegetation coverage species with persistent only for two years as biennials. Therefore, these species should not be used in a large amount for ecological restoration when mixed with perennials because of affecting the vegetation coverage of perennials. There were considerable variations in vegetation coverage with perennial herbaceous flowers : Lotus corniculatus and Taraxacum platycarpum-high vegetation coverage species, Platycodon grandiflorum, Aster tataricus and Aster koraiensis-medium vegetation coverage species, and Aster yomena, Patrinia scabiosaefolia and Chrysanthemum indicum-low vegetation coverage species. Growth types of herbaceous flowers tested in this research were divided into 4 types : Callistephus chinensis, Oenothera lamarckiana and Aster koraiensis-wide leaf and tall type, Aster tataricus, Patrinia scabiosaefolia, Oenothera odorata, Taraxacum platycarpum, and Aster yomena-medium leaf and medium tall type, Platycodon grandiflorum-fine leaf and dwarf type, and Lotus corniculatus-fine leaf and tall type.
