In vitro Antioxidant Activity of Mentha viridis L. and Mentha piperita L. (박하의 in vitro 항산화활성)

  • For selecting a new candidate as functional material, this study was conducted on in vitro antioxidant activity and total phenol content of methanol and water extracts prepared from two Mentha species (M viridis L. (M spicata L.) and Mentha piperita L, Extracts of M. viridis showed more efficient scavenging activity on superoxide and DPPH ${({\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl)}$ radical and inhibitory activity on oxidation of human low density lipoprotein (LDL) induced by $CuSO_4$ and auto-oxidation of linoleic acid than those of M piprita. Methanol extract $(65.88%{\sim}77.59%)$ and water extract $(37.69%{\sim}87.21%)$ of M. viridis also exhibited more potent inhibitory activity on LDL oxidation than that of ${{\alpha}-tocopherol\;(28.37%{\sim}66.54%)}$ at ${1{\sim}100\;{\mu}g/ml}$ of final concentration. The total phenol contents of methanol extract and water extract of Mviridis (17.95% & 10.18%, respectively) as tannic acid equivalent were higher than those of M piperita (15.44% & 9.19%). But the yields of methanol and water extracts of M. viridis (13.3 % & 13.5%) were lower than those of M. piperita (14.1 % & 14.6%). The results implies that the extracts from M. viridis (spicata) is more useful material for industrialization as functional food than those from M. piperita.

  • The mycelial mass of K. mutabilis greatly increased at pH 5.5~6.0 but decreased pH 6.0. The linear mycelial growth wsa mostly supported on sawdust of Quercus accutisima and the mycelial density wsa high on sawdust of Q. accutisima and corn cob. Much mycelial distribution could be showen in ray parenchyma cell and ray tracheid. Severe degradation of ray parenchyma cell was found but little degradation of ray tracheid cell was found. The dry weight loss wsa 5.9% after agar-block test. And the pH wsa acidified from 6.07 to 4.31 and hot water extractives was decreased after degradation of Q. serrata sawdust by K. mutabilis.

Antioxidant and Anti-inflammatory Effects of Extracts from the Flowers of Weigela subsessilis on RAW 264.7 Macrophages (RAW 264.7 대식세포에 미치는 병꽃나무 꽃 추출물의 항산화 및 항염증 효과)

  • This study investigated the antioxidant and anti-inflammatory activity of ethanol extract from the flowers of Weigela subsessilis (WS-E) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The total polyphenol and flavonoid content was 719.19±0.04 μg tannic acid equivalents/ml and 644.87±0.02 μg quercetin equivalents/ml, respectively. The antioxidant activities of WS-E were measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide anion radical scavenging activity. The antioxidant activities of WS-E increased markedly, in a dose-dependent manner. To screen for anti-inflammatory agents, the inhibitory effects of WS-E on the production of proinflammatory cytokines in the LPS-stimulated RAW 264.7 macrophages was examined. WS-E had no effect on cell viability at a concentration of 100 μg/ml. Nitric oxide (NO) and interleukin (IL)-6 production were inhibited in a dose-dependent manner (p<0.05). WS-E had no effect on the production of tumor necrosis factor (TNF)-α at a concentration of 0.16–20 μg/ml but induced TNF-α at a concentration of 100 μg/ml. Inducible nitric oxide synthase (iNOS) expression was also inhibited at lower concentrations (p<0.05). In addition, WS-E reduced the activation of nuclear factor (NF)-κB by inhibition of inhibitoy (I) κB phosphorylation in RAW 264.7 macrophages upon stimulation with LPS (100 ng/ml) for 24 h but not that of mitogen-activated protein kinase (MAPK). These results suggest that WS-E may be a useful antioxidant and anti-inflammatory agent in functional cosmetics.

Optimization of Extraction Conditions of Sarcodon aspratus by Response Surface Methodology (반응표면분석에 의한 능이버섯 추출물의 추출조건 최적화)

  • Response surface methodology (RSM) was used to monitor extraction characteristics of extracts from Sarcodon aspratus. Based on a central composite design, independent variables were microwave power (30~150 W), ethanol concentration (0~100%), and extraction time (1~9 min). Dependent variables were yield, electron-donating ability, total phenol contents, and SOD-like activity. Coefficients of determination ($R^2$) for dependent variables ranged from 0.80 at 0.97. The maximum extraction yield was 50.28% under conditions of 125.1 W microwave power, 18.67% ethanolic concentration, and 7.06 min extraction time. The maximum extraction electron-donating ability was 22.14% under conditions of 31.09 W, 45.76%, and 4.32 min. The maximum extraction total polyphenol content was 30.54 mg tannic acid equivalent/g at 122.54 W, 48.05%, and 8.36 min. The maximum extraction SOD-like activity was 33.44% at 121.17 W, 47.42%, and 8.41 min. Based on superimposition of four dimensional RSM with respect to extraction yield, electron-donating ability, total polyphenol content, and SOD-like activity obtained under various extraction conditions, optimum ranges of extraction conditions were found to be microwave power of 78~88 W, ethanol concentration of 39~57%, and extraction time of 3.5~9 min.

Comparison of the Proliferation pattern of Cultured Rat Calvaria Cell on the Resorbable Barrier Membrane (흡수성 차폐막에 배양된 구개관세포의 증식양상의 비교)

  • The purpose of this study is to evaluate the phenomenon of attachment and spreading of the cultured rat calvarial cell inoculated on their surface of different kinds of biodegradable membrane which had been used on tissue regeneration on periodontal defects by using scanning electron microscope. In this experiment 30 Sprague-Dawley male rats (mean BW 150gm) were used to harvest abundant number of cell in the short period. The rats were sacrificed by decapitatioan to obtain the calvaria for bone cell culture. Calvarial cells were cultured with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. Biodegradable barrier membrane were collected with collagen type, and were divided into 3 different kind of surface such as scattered, polarized and fine-net type as their surface texture. Microcover plate which usually used for cell culture was used as control for smooth surface. All the membrane were seeded with cultured calvarial cell on their surface. The number of cell inoculated on the membrane were $1{\times}10^6$ Cells/ml. After the culture as designed time, all the membrane were washed with 0.1 M Phosphate Buffered saline and fuxed with 2.5% Glutaraldehyde. And all specimen were treated with $OsO_4$, and Tannic acid before drying the cell for coating the cell with gold. Scanning Electron Microscope was used to observation. The following results were obtained. I. During the whole period of experiment, the phenomenon of cell attachment and spreading were revealed similar pattern to compare with smooth surface culture plate and ordinary culture dish. 2. The shape of cell attachment and spreading on the surface of barrier membrane were observed no remarked difference pattern between smooth surface culture plate and ordinary culture dish. 3. The cytoplasmic process of cultured calvaria cell extent to the deep portion of barrier membrane like as their own proper shape. 4. There were no remarkable relationships between the degree of cultured cell spreading and surface structure of barrier membrane. 5. Slight starified layer of cultured calvaria cell were observed on the scattered type of resorbable membrane, Conclusively, this study thus suggest that cultured bone cell inoculated onto the biodegradable barrier membrane may have an important role of carrier for many cell which could be used as new tissue regeneration, and those tissue engeering technique may become an new method in the approach to the repair of bone defects.

Antioxidant Activities of Medicinal Plant Extracts (약용식물 추출물의 항산화 효과)

  • The antioxidant activities of ethanolic extracts obtained from medicinal plants (Scutellaria baicalensis Georgi, Acanthopanax sessiliflorum Seeman, Pueraria lobata Ohwi, Portulaca oleracea Linne, Crataegus pinnatifida Bunge var. typica Schneider, Euonymus alatus Apterus, Hovenia dulcis Thunberg, Prunus yedoensis Matsumura, Albizzia julibrissin Durazz., Chrysanthemum indicum Linne) were evaluated for total phenolic content, total flavonoid content, DPPH radicals, nitrites, $Superoxide^-$ radicals, $ABTS^+$ radical scavenging activity, and reducing power. Antioxidant capacities were the highest in Prunus yedoensis Matsumura for DPPH radical scavenging activity ($IC_{50}$ $5.39{\mu}g/mL$), reducing power (2.72, $A_{700}$), and nitrite scavenging activity ($IC_{50}$ $167.94{\mu}g/mL$). Hovenia dulcis Thunberg and Acanthopanax sessiliflorum Seeman were effective for their nitrite scavenging activities (over 90% at 1 mg/mL). The $Superoxide^-$ radical scavenging activity of Prunus yedoensis Matsumura ($IC_{50}$ $43.39{\mu}g/mL$) was stronger than tannic acid ($IC_{50}$ $46.51{\mu}g/mL$). Five samples (Prunus yedoensis Matsumura, Acanthopanax sessiliflorum Seeman, Hovenia dulcis Thunberg, Crataegus pinnatifida Bunge var. typica Schneider, Scutellaria baicalensis Georgi) were effective for their $ABTS^+$ radical scavenging activity (more than 90% at 0.5 mg/mL). These results suggest that the ethanolic extracts of Prunus yedoensis Matsumura could be used as a functional ingredient in food products.

Anti-Diabetic, Alcohol Metabolizing Enzyme, and Hepatoprotective Activity of Acer tegmentosum Maxim. Stem Extracts (산겨릅나무 줄기 추출물의 항당뇨, 알코올 대사 효소 및 간 보호 활성)

  • This study was carried out to investigate the antidiabetic, alcohol metabolism, anti-inflammatory, and hepatoprotective effects of Acer tegmentosum extracts (ATE). A. tegmentosum has been traditionally used as a folk medicine to treat hepatic disorders. The antioxidative activities of ATE were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide (SOD) assay. DPPH radical scavenging and SOD activities of ATE were about 89% and 82.9% at $0.5{\mu}g/mL$, respectively. Alcohol dehydrogenase and acetaldehyde dehydrogenase activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}-Glucosidase$ inhibitory activity of ATE was 75% higher at $50{\mu}g/mL$ and remarkably increased in a dose-dependent manner. Nitric oxide productions in macrophage RAW 264.7 cells stimulated by lipopolysaccharide was reduced to 16.7% by addition of ATE at 1 mg/mL. ATE showed significant protective effects against tacrine-induced cytotoxicity in Hep G2 cells at $100{\mu}g/mL$. Based on our results, we conclude that ATE may be used as a major pharmacological agent and anti-diabetic, anti-hepatitis, and anti-inflammatory remedy.

Anti-inflammatory Effects of Black Cherry Tomato (Lycopersicon esculentum M.) Juice on LPS-induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7 세포에 대한 흑색 방울토마토 주스의 항염증 효과)

  • This study was carried out to investigate the antioxidative, nitrite-scavenging, alcohol-metabolizing, and anti-inflammatory effects of black-cherry tomato juice (BCTJ) on LPS-induced RAW 264.7 cells. The total phenol content of the BCTJ was $156.83{\mu}g\;tannic-acid-equivalent/ml$. The antioxidative effects of BCTJ were measured using DPPH radical-scavenging activity and SOD-like assay. DPPH radical-scavenging activity of BCTJ was increased in a dose-dependent manner (p<0.05) and was 83.39% at 40%. SOD-like activity of BCTJ was 22.01% at 100%. The effects of BCTJ on alcohol-metabolism were determined by measuring generations of reduced nicotinamide adenine dinucleotides (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities were 198.87% and 181.89% at 40%, respectively. Nitric scavenging activities of BCTJ were 85.06%, 58.25%, and 43.68% at pH values 1.2, 3.0, and 6.0, respectively, at 50%. Anti-inflammatory effects were examined in LPS-stimulated RAW 264.7 cells. Nitric-oxide production was reduced to 83.55% by the addition of BCTJ at 10%. These results suggest that black-cherry tomato juice has great potential as a resource for natural health products.

Serological Study on Rice Stripe Virus (벼 줄무늬잎마름병 바이러스에 대한 혈청학적인 검토)

  • This experiment was performed to clarify the concentration of rice stripe virus in the rice Plant leaves by serological test, and was attempted to inspect the virus carrier among small brown planthopper by antibody-sensitized hemagglutination test. The antiserum was prepared by injecting intervenously into the external marginal vein of the ear of a rabbit. The precipitin titer of it was 1 : 16. The rough virus fluid prepared from diseased leaves was centrifuged at 10.000 rpm, and then the supernatant solution was treated at $55^{\circ}C$ for 5 minutes and the solution clarified by removing the agglutinate was used as the antigen solution. Antibody-sensitized erythrocyte solution was prepared from sheep erythrocytes sensitized by rice stripe virus with tannic acid, and its agglutination titer was 1 : 512. The virus concentrations in flag leaves or first leaves just below them showing different symptoms was high with progressing the severity of symptoms. And the concentrations of the virus in leaves of varieties of the rice plant showing same degree symptom were lower in suscetible varieties, Sadominori, Palgoeng, Mangyong and Nihonbare, than in the resistant one, Tongil, but in Yooshin which was known as the resistant, lower rather than in Tongil. The reacton of antibody-sensitized hemagglutination test to inspect the virus carrier, was so highly sensitive that this reaction was recognized as a method which is able to Identify the carrier accurately in short time.

Antioxidative and Physiological Activities of Traditional Korean Teas (한국 전통차의 생리활성 및 항산화작용)

  • This study was investigated the antioxidative and antimicrobial activities of a water extract (70$^{\circ}C$) of traditional Korean teas (rose, chrysanthemum, pine needles, mulberry, persimmon leaves, and green tea). Total phenol contents in rose, chrysanthemum, pine needles, mulberry, persimmon leaves, and green tea were 272.8, 74.6, 153.5, 73.5, 69.5, and 260.8 mg tannic acid/g extract, respectively, whereas total flavonoid contents were 75.1, 47.8, 26.8, 40.0, 27.9, and 99.5 mg quercetin/g extract, respectively. The order of electron donating abilities of Korean traditional tea water extracts (1 mg/mL) were : rose (96.8%) > green tea (95.3%) > pine needles (71.3%) > chrysanthemum (36.8%) > mulberry (28.9%) ${\qeq}$ persimmon (28.8%). The order of nitrite-scavenging abilities at pH 1.2 (2 mg/mL) was green tea > rose > pine needles > chrysanthemum ${\qeq}$ mulberry ${\qeq}$ persimmon. The order of hydroxyl radical scavenging activity (5 mg/g) was chrysanthemum > mulberry > rose > persimmon leaves > pine needles > green tea. Rose extracts showed antimicrobial activity against Micrococcus luteus, Bacillus cereus, Salmonella enteritidis, and E. coli, whereas green tea extracts showed strong antimicrobial activity aginst S. enteritidis.
