Seed Set in Close Pollination and Backcross of Interspecific F1 of Lilium spp. (백합 근연수분 및 종간 교잡종 F1 여교잡시 종자형성)

  • We studies seed set in the interspecific F1 /backcross hybrids of Lilium species. In the interspecfic hybrid of L. longiflorum cv. Gelria with L.${\times}$ fomolongi cv. Raizan,93% fruit set was obtained by stigmatic pollination in comparison to 53% from cut-style pollination. Accordingly, the number of seed set resulting from stigmatic and cut-style pollination was 147 and 53, respectively. Pollination o( both stigmatic and cut-style pollination resulted in 47% fruit set in the hybrid of L. longiflorum cv. Lorina with L.${\times}$ fomolongi cv. Raizan. However, stigmatic pollination formed 413 seeds, whereas only 24 seeds were obtained by cut-style pollination in this cross. The hybrid of L.${\times}$ fomolongi cv. Raizan with L. longiflorum cv. Come set 40% fruit with a total of 43 seeds by stigmatic pollination. However, no fruit set was observed in cut-style pollination in this hybrid. Backcrossing the F1 hybrid by cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Chicago with the latter parent led to 53% fruit set, and 109 embryos were obtained. Likewise, backcrossing following cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Corsia with the latter parent formed 67% fruits and 107 embryos. However, in the remaining interspecific hybrids, cut-style pollination set no fruit.

Pathogenicity and PCR detection of Vibrio tapetis in Manila clams, Ruditapes philippinarum (양식 바지락, Ruditapes philippinarum에 대한 Vibrio tapetis의 병원성과 PCR법에 의한 진단)

  • Pathogenicity of Vibrio tapetis, the causative bacterium of 'brown ring disease (BRD)' was evaluated in Manila clams (Ruditapes philippinarumi by artificially 0.1 $m\ell$ infection of $1.0\times10^5$cells and $1.0\times10^8$ cells at 20 $^{\circ}C$. A PCR assay based on 16S rRNA to detect the bacteria in clam tissues was established. Accumulative mortality of clams infected with $1.0\times10^7$cells and $1.0\times10^4$ cells per an individual of the bacteria was 67.5% and 7.5%, respectively. However, the deposit of brown pigment in the inner shells by accumulation of chonchiolin was not found. The bacteria were not be able to re-isolate from the infected clams by the conventional agar plate method but were easily detected by PCR assay established in this experiment. In clams artificially infected with 10 species of Vibrio, a 414bp for V. tapetis was detected in PCR assay. The specific band in the clams infected with $1.0\times10^4$cells per an individual of V. tapetis was detected only in gills one day after the infection but never be found in any tissues including gills three days after the infection. In the case of clams infected with $1.0\times10^8$cells per an individual of V. tapetis the specific band was detected in gills and intestine one day after the infection, in all tissues three days after the infection, and then in gills and adductor muscle nine days after the infection. The PCR assay was applied to detect V. tapetis in manila clam, surf clam (Mactra veneriformis), oyster (Crassostrea gigas) and Thomas' rapa whelk (Rapana venosa) taken from Taean and Gochang from April to July 2004. The infection rates were detected to 23.1% and 9.4% in the oyster and surf clam, while manila clam and Thomas' rapa whelk were not found.

Reproductive Cycle of a Rockfish, Sebastes schlegeli (조피볼락의 생식주기)

  • To clarify the annual reproductive cycle in a rockfish, Sebastes schlegeli, monthly changes in gonadosomatic index (GSI), hepatosomatic index (HSI) and histological feature of gonads and plasma levels of sex steroid hormones ($estradiol-l7{\beta},\;17{\alpha},\;20{\beta}-dihydroxy-4-pregnen-3-one,\;testosterone\;and\;11-ketotestosterone$) were investigated. The annual reproductive cycle in females could be divided into 5 periods as follows: 1) recovery period (June to September): serum level of $estradiol-l7{\beta}$ increased gradually; 2) vitellogenesis period (Septemer to february) : vitellogenic oocytes were obsewed, GSI sustained high value, and serum level of $estradiol-l7{\beta}$ increased; 3) gestation period (February-April): developing larva showed in the ovary, and serum levels of $17{\alpha},\;20{\beta}-dihydroxy-4-pregnen-3-one$ and testosterone increased; 4) partrition period (April to May) : larva were delivered, and value of GSI and serum levels of hormones decreased rapidly; 5) resting period (May to June) : value of GSI and serum levels of $estradiol-l7{\beta}$ and testosterone remained low. The annual reproductive cycle in males could be divided into 6 periods; 1) early maturation period (April to June): value of GSI and serum levels of hormones incresed gradually, cyst of spermatogonia incresed in number, and a small number of cyst of spermatocyte was observed; 2) mid-maturation perid (June to September); value of GSI and serum levels of hormones increased, and germ cells in many cysts were undergoing active sperrnatogenesis; 3) late maturation period (September to November) : value of GSI and serum levels of hormones remained high and spermatozoa were released into the lumina of the seminal lobules; 3) spermatozoa dischaging period (Nobember to December) : the lumina of the seminal lobules were enlarged and filled with mature spermatozoa; 4) degeneration period (December to Februauy)i value of GSI decresed and cyst of spermatocyte were decresed in number; 5) resting period (December to April) : no histological changes of testes were observed, and value of GSI and serum levels of hormones remained low. In November, the lumina of the seminal lobules were filled with mature spermatozoa and sperm masses were present in the ovarian cavity. Thus, copulation in this species occurred in November and December.

Trace Metal Contents in Seaweeds from Korean Coastal Area (한국 연안 해조류의 미량금속 함량)

  • In order to assure the safety of the seaweeds, we measured the contents of the trace metals in the seaweeds harvested from Korean coastal area. We collected 620 marine algal samples from fourteen areas, and then analyzed the trace metals such as Hg, Cd, Cr, Cu, Mn, Ni, Pb and Zn. The contents of Hg, Cd, Ni and Mn were higher in the seaweeds collected from Chungnam Taean area. The contents of Cr and Cu were higher in the seaweeds collected from Yeongdeok and Tongyeong area, respectively. And Pb and Zn were highly detected from the seaweeds of Ulsan area. The mean levels of trace metals were high in the order of Zn $(48.02\pm41.20{\mu}g/g)\;>\;Mn (34.63\pm38.95{\mu}g/g)\;>\;Cu(6.29\pm6.52{\mu}g/g)\;>\;Ni(2.17\pm4.00{\mu}g/g)\; >\;Cr(1.61\pm2.35{\mu}g/g)\;>\;Pb(1.28\pm2.37{\mu}g/g)\;>\; Cd(0.67\pm0.75{\mu}g/g)\;>\;Hg(0.02\pm0.02{\mu}g/g)$. Zn was consistently most abundant metal in all samples, followed by Mn and Cu, which are very important for human health. There were higher contents of Hg and Cd in brown algae, the contents of Cr, Cu, Mn, Ni and Pb in green algae, the content of Zn in red algae among them. Additionally, significant linear correlations were observed between a total 28 different pairs of metals, of them, the correlation factor between Cr and Ni showed the highest (r=0.374, p<0.001). The trace metal contents of seaweeds varied with habitats and kinds of samples, nevertheless they showed very safe level in edible seaweeds tested. Scytosiphon lomentaria, which has been known as a little use of food, have shown a clear selectivity for Cr and Pb. Among non-edible seaweeds, Sargassum thunbergii for both Cd and Mn, Lomentaria hakodatensis for Ni and Grateloupia prolongata for Zn have shown a clear selectivity, respectively. Therefore, these seaweeds could be useful as bioindicator for each trace metal pollution.

Improvements in the Marine Environmental Survey on Impact of Seawater Qualities and Ecosystems due to Marine Sand Mining (바다모래 채취 시 해수 수질 및 생태계 영향에 대한 해양환경조사 개선 방안)

  • We reviewed investigation status on turbidity plume in the statement of marine environmental survey(2008 to 2012) associated with marine sand extraction projects. The survey statement from seven marine sand extraction sites (extraction area of Southern EEZ, extraction area of Western EEZ, relocation zone in the Western EEZ, sea area under jurisdiction of Taean-gun, sea area under jurisdiction of Ansan City, and two discrete sea areas under jurisdiction of Ongjin-gun) in the nearshore and offshore of Korea showed that in situ observations were carried out for the dispersion and transport of suspended sediments on two areas (One is a extraction area in the EEZs, the other is an area of coastal sites). However, sampling station and range have not been selected considering physical, geographical factors (tide, wave, stratification, water depth, etc.) and weather conditions (wind direction and velocity, fetch, duration, etc). Especially turbidity plumes originating from three sources, which include suspended sediments in overflow(or overspill) discharged from spillways and reject chutes of dredging vessel, and resuspended sediments from draghead at the seabed, may be transported to a far greater distance outside the boundary of the extraction site and have undesirable impacts on the marine environment and ecosystem. We address that behaviour of environmental pollutants such as suspended solids, nutrients, and metals should be extensively monitored and diagnosed during the dispersion and transport of the plume. Finally we suggest the necessity to supplement the current system of the sea area utilization consultation and establish the combined guidelines on marine sand extraction to collect basic data, to monitor cumulative effects, and to minimize environmental damages incurred by the aftermath of sand extraction.

Antioxidative and Antimicrobial Activities of Aruncus dioicus var. kamtschaticus Hara Extracts (눈개승마(Aruncus dioicus var. kamtschaticus Hara) 용매 추출물의 항산화 및 항균활성)

  • The solvent extracts of Aruncus dioicus var. kamtschaticus Hara, which were extracted by using several solvents with different polarities, were performed to investigate the antioxidant activities, whitening effect and antimicrobial activity. The content of total polyphenol of fractions from Aruncus dioicus var. kamtschaticus Hara extract showed the highest value ($335.88{\pm}2.26$ mg/g GAE) on ethyl acetate fraction. The ethyl acetate and n-butanol fractions were 0.06 mg/mL and 0.25 mg/mL as $IC_{50}$ values on DPPH radical scavenging, and $99.16{\pm}0.09%$ and $89.29{\pm}0.64%$ on ABTS radical scavenging activity, respectively. Also, reducing power and FRAP value were significantly higher on ethyl acetate fraction. The SOD like activity showed $80.76{\pm}0.61%$ on ethyl acetate and $72.34{\pm}0.79%$ on n-butanol. Tyrosinase inhibition activities (at 5 mg/mL) were $59.08{\pm}0.98%$ on ethyl acetate fraction. The chloroform fraction showed the strongest antimicrobial activities against B. cereus (14 mm), B. subtilis (12.5 mm), S. aureus (10.8 mm), E. coli (20.7 mm) at 0.1 mg/disc and the inhibition zone diameter of ethyl acetate fraction was 17.2 mm against E. coli at 0.5 mg/disc. The minimum inhibitory concentrations (MIC) of chloroform fraction against B. cereus and E. coli were 50 and $25{\mu}g$/mL, respectively. From these results, it is suggested that ethyl acetate and n-butanol fractions of Aruncus dioicus var. kamtschaticus Hara could be used as functional material for food additive ingredient and chloroform fraction could be suitable for the development of a food preservative.

Monthly HPLC Measurements of Pigments from an Intertidal Sediment of Geunso Bay Highlighting Variations of Biomass, Community Composition and Photo-physiology of Microphytobenthos (HPLC를 이용한 근소만 조간대 퇴적물내의 저서미세조류 현존량, 군집 및 광생리의 월 변화 분석)

  • In this study, the surveys were carried out from October (2016) to October (2017) along the tidal flat of Geunso Bay, Taean Peninsula of the western edge of Korea. The sampling trips were carried out for a total of 16 times, once or twice a month. In order to investigate the monthly variation of the microphytobenthos (MPB) biomass, community composition and photo-physiology were analyzed by HPLC (High performance liquid chromatography). The total chlorophyll a (TChl a) concentrations used as an indicator of biomass of MPB in the upper 1 cm sediment layer ranged from 40.4 to $218.9mg\;m^{-2}$ throughout the sampling period. TChl a concentrations showed the maximum level on $24^{th}$ of February and remained high throughout March after which it started to declined. The biomass of MPB showed high values in winter and low values in summer. The monthly variations of Phaeophorbide a concentrations suggested that the low grazing intensity of the predator in the winter may have partly attributed to the MPB winter blooming. As a result of monthly variations of the MPB community composition using the major marker pigments, the concentrations of fucoxanthin, the marker pigment of benthic diatoms, were the highest throughout the year. The concentrations of most of the marker pigments except for chlorophyll b (chlorophytes) and peridinin (dinoflagellates) increased in winter. However, the concentrations of fucoxanthin increased the highest, and the relative ratios of the major marker pigments to TChl a except fucoxanthin decreased during the same period. The vertical distribution of Chl a and oxygen concentrations in the sediments using a fluorometer and an oxygen micro-optode Chl a concentrations decreased with oxygen concentrations with increasing depth of the sediment layers. Moreover, this tendency became more apparent in winter. The Chl a was uniformly vertical down to 12 mm from May to July, but the oxygen concentration distribution in May decreased sharply below 1 mm. The increase in phaeophorbide a concentration observed at this time is likely to be caused by increased oxygen consumption of zoobenthic grazing activities. This could be presumed that MPB cells are transported downward by bioturbation of zoobenthos. The relative ratios (DT/(DD+DT)) obtained with diadinoxanthin (DD) and diatoxanthin (DT), which are often used as indicators of photo-adaptation of MPB, decreased from October to March and increased in May. This indicated that there were monthly differences in activity of Xanthophyll cycle as well.

A Study on the Sahyang and Characteristics of Naturalized Citizens in Early Chosun (조선초기 귀화인(歸化人)의 사향(賜鄕)과 특징)

  • This study presents an investigation into the Sahyang process and activities of the individuals whose Sahyang(receiving Gwanhyang from the king) was confirmed in the Shilrok documents of early Chosun among naturalized citizens(Hyanghwains). In early Chosun, there were four Chinese(Oh Jin, Lee Min-do, Dang Seong, and Mae Woo) in the Sahyang cases with more of those cases found among Hoigol-in(Seol Jang-soo, an Uighur) and Oioa-guk nationality(Lee Hyeon) from the countries bordering on Western China and two Japanese(Pyeong Sun and Pi Sang-ui). They were naturalized from the reign of King Chungryeol of Goryeo to the first year of King Jeongjong's reign of Chosun. They were diverse to include the great grandfather, grandfather, and father of the individual that received Gwanhyang and himself. There were one case of Sahyang during the reign of King Taejo, four during the reign of King Taejong, one during the reign of King Sejong, and three during the reign of King Sejo. The Gwanhyang they received was wide spread across the nation including Gyerim, Imju, Haeju, Sangju, Milyang, Chungju, Changwon, Dongrae, and Taean. It's very likely that the place of Sahyang was Cheohyang. Many of those who received Gwanhyang were translators and achieved great feats in Chosun's diplomacy with Ming Dynasty and Japan. There were also those who worked in medicine, art of divination, and articles of a criminal code. Seol Jang-su, who passed the state exam of Chosun, was in charge of Jigonggeo, and Lee Min-do and Dang Seong made a contribution to the establishment of Chosun and became big or small meritorious retainers at the founding of Chosun. It's all thanks to those performances that they had the honor of Sahyang of receiving Gwanhyang. Although they were the Gwanhyang with the honor of Sahyang, there is no confirmation of the descendents of the Lee family of Imju, the Pyeong family of Changwon, and the Pi family of Dongrae. While the descendents of the naturalized Chinese still remain in the nation, those of naturalized Japanese ceased to exist, which must be closely related to Chosun's perceptions of other nations those days.

Effect of Heavy Metals (As, Cr, Pb) on the Population Growth Rates of Marine Diatom, Skeletonema castatum (중금속(As, Cr, Pb)이 해산 규조류(Skeletonema costatum)의 개체군 성장률에 미치는 영향)

  • In this study, we evaluated the toxic effects of heavy metals (As, Cr and Pb) on the population growth rate (r) of the marine diatom, Skeletonema costatum. The r of the species was determined after 96 h (4 days) exposure to As (0, 0.31, 0.63, 1.25, 2.50 and 5.00 mg L-1), Cr (0, 0.16, 0.31, 0.63, 1.25 and 2.50 mg L-1) and Pb (0, 1.25, 2.50, 5.00, 10.00 and 20.00 mg L-1). It was observed that r in the control (absence of As, Cr and Pb) were greater than 0.04, however suddenly decreased with increased heavy metal concentrations. As, Cr and Pb reduced r in a dose-dependent manner and a significant reduction were occurred at concentration of greater than 0.31, 0.31 and 5.00 mg L-1, respectively. Based on the toxicity, the heavy metal were ranked as Cr>As>Pb, with EC50 (50% effective concentration) values of 0.80, 2.57 and 13.88 mg L-1, respectively. The lowest-observed-effective-concentration (LOEC) of r in exposed to As, Cr and Pb were 0.63, 0.63 and 10.00 mg L-1, respectively. Precisely, a concentration of greater than 0.63 mg L-1 of As, 0.63 mg L-1 of Cr and 10.00 mg L-1 of Pb in marine ecosystems induced toxic effect on the r of S. costatum. Based on our results, we suggested that the r of S. costatum might be a useful bio-indicator for the toxicity assessment of heavy metals on the producer in marine ecosystems.

A Study on the Musical Characteristics of Fishing songs in North Korea (북한 어업노동요의 음악적 특징)

  • The musical characteristics of Fishing songs in North Korean which recorded on Anthology of North Korean Folk Songs were examined in this paper. The results are as follows. Among Fishing songs in North Korea, there are "Rowing Song," the "Song for Hauling Nets," and the "Song for Picking Up Fish" in songs related to walleye pollack fishing and in songs related to croaker fishing, the "Song for Dropping Anchor" and the "Full-Load-of-Fish Song" in songs related to croaker fishing, the "Song for Weighing Anchor," the "Song for Shaking Fish," the "Full-Load-of-Fish Rowing Song," and the "Song for Hanging Walleye Pollack" in songs related to walleye pollack fishing. There are the "Song for Setting a sail" and the "Song for catching shells" in Fishing songs in North Korea, too. In the rhythmic elements of Fishing songs in North Korea, the tempo of the "Songs for Setting a sail" is moderato, and that of the "Songs for Picking Up Fish" of songs related to anchovy or herring fishing is quite slow. "Rowing Songs"(croaker fishing) have various tempos but faster than moderato, and usually get faster and faster as time goes by. Most songs have 4/♩. time and there are some of 2, 3 or 5 time. Some songs use irregular time or 6 time overlapping 2 beats. The mode Menari is the most common tonal system used in fishing songs of North Korea. And the mode Susimga, the mode Changbutaryeong, the mode Nanbongga, and the mode Odolttogi are also used in the tonal system of Fishing songs in North Korea. There are shoutting intonations that have indefinite pitch, too. Modulation or interaction by the mode Changbutaryeong appeared in songs of the mode Menari. There is a tendency to favor a certain tonal system by category. Responsorial form is mostly used, but in the most cases, they were sung in solo. In responsorial songs, several calling songs overlap with the responding songs. Durchkomponiert form is sung by antiphon or in solo, but the case sung in solo originally seems to have been sung by antiphon or unison, if we consider the procedure of the work. The "Song for Picking Up Fish" of Seonbong-gun of North Hamgyeong Province and Yangyang-gun of Gangwon Province, the "Full-Load-of-Fish Song-Bongjuktaryeong" of Jeungsan-gun of South Pyeongan Province and "Full-Load-of-Fish Song-Baechigisory" of Taean-gun of South Chungcheong Province, "Rowing Song" of Nampo City and Ganghwa-gun of Gyeonggi Province are resembled. In neighboring areas, even if the songs of different categorys, sometimes share a same melody. In a certain category, sometimes a same melody is shared to a considerable distance. It was first identified that in anchovy or herring fishing or dybowskii' sand eel fishing, the "Song for Picking Up Fish" is widely shared in the East Sea Coast area.
