The TREK2 Channel Is Involved in the Proliferation of 253J Cell, a Human Bladder Carcinoma Cell

  • Bladder cancer is the seventh most common cancer in men that smoke, and the incidence of disease increases with age. The mechanism of occurrence has not yet been established. Potassium channels have been linked with cell proliferation. Some two-pore domain $K^+$ channels (K2P), such as TASK3 and TREK1, have recently been shown to be overexpressed in cancer cells. Here we focused on the relationship between cell growth and the mechanosensitive K2P channel, TREK2, in the human bladder cancer cell line, 253J. We confirmed that TREK2 was expressed in bladder cancer cell lines by Western blot and quantitative real-time PCR. Using the patch-clamp technique, the mechanosensitive TREK2 channel was recorded in the presence of symmetrical 150 mM KCl solutions. In 253J cells, the TREK2 channel was activated by polyunsaturated fatty acids, intracellular acidosis at -60 mV and mechanical stretch at -40 mV or 40 mV. Furthermore, small interfering RNA (siRNA)-mediated TREK2 knockdown resulted in a slight depolarization from $-19.9mV{\pm}0.8$ (n=116) to $-8.5mV{\pm}1.4$ (n=74) and decreased proliferation of 253J cells, compared to negative control siRNA. 253J cells treated with TREK2 siRNA showed a significant increase in the expression of cell cycle boundary proteins p21 and p53 and also a remarkable decrease in protein expression of cyclins D1 and D3. Taken together, the TREK2 channel is present in bladder cancer cell lines and may, at least in part, contribute to cell cycle-dependent growth.

  • Hydrogen peroxide ($H_2O_2$) causes oxidative stress and is considered as an inducer of cell death in various tissues. Two-pore domain $K^+$ ($K_{2p}$) channels may mediate $K^+$ efflux during apoptotic volume decreases (AVD) in zygotes and in mouse embryos. In the present study, we sought to elucidate linkage between $K_{2p}$ channels and cell death by $H_2O_2$. Thus $K_{2p}$ channels (TASK-1, TASK-3, TREK-1, TREK-2) were stably transfected in HEK-293 cells, and cytotoxicity assay was preformed using cell counting kit-8 (CCK-8). Cell survival rates were calculated using the cytotoxicity assay data and dose-response curve was fitted to the $H_2O_2$ concentration. Ionic currents were recorded in cell-attached mode. The bath solution was the normal Ringer solution and the pipette solution was high $K^+$ solution. In HEK-293 cells expressing TREK-1, TREK-2, TASK-3, $H_2O_2$ induced cell death did not change in comparison to non-transfected HEK-293. In HEK-293 cells expressing TASK-1, however, dose-response curve was significantly shifted to the left. It means that $H_2O_2$ induced cell death was increased. In cell attached-mode recording, application of $H_2O_2$ (300μM) increased activity of all $K_{2p}$ channels. However, a low concentration of $H_2O_2$ ($50{\mu}M$) increased only TASK-1 channel activity. These results indicate that TASK-1 might participate in $K^+$ efflux by $H_2O_2$ at low concentration, thereby inducing AVD.

SF Movie Star Trek Series and the Motif of Time Travel (SF영화 <스타트랙> 시리즈와 시간여행의 모티프)

  • The purpose of this article is to elucidate why the motif of time travel is repeated in the science fiction narrative by examining the functions of this motif in the SF movie series of Star Trek in its narrative and non-narrative aspects. Star Trek IV: The Voyage Home (1986) aims to attract the audience's interest in the story through the use of plausible time travel in the form of the slingshot effect which causes the spacecraft to fly at very fast speeds around an astronomical object. The movie also touches upon the predestination paradox that arises from a change of history in which it describes a formula of transparent aluminum that did not exist at the time. The film also serves as an evocation of the ideology of ecology by including humpback whales in the central narrative and responding to the real issue of the whale protection movement of the times. Star Track VIII: First Contact (1996) intends to interest the audience in the narrative with the warp drive, a virtual device that enables travel at speeds faster than that of light and a signature visual of Star Trek, at the time of its birth through time travel. The film emphasizes the continuation of peaceful efforts by warning the destruction of humanity that nuclear war can bring. It tackles with the view of pacifism and idealism by stressing the importance of cooperation between countries in the real world by making the audience anticipate the creation of the United Federation of Planets through encounters with the extraterrestrial. Star Trek: The Beginning (2009) improves interest through the idea of time travel to the past, this time using a black hole and the parallel universe created thereby. The parallel universe functions as a reboot, allowing a new story to be created on an alternate timeline while maintaining the original storyline. In addition, this film repeats the themes pacifism and idealism shown in the 1996 film through the confrontation between Spock (and the Starfleet) and Nero, the destruction of the Vulcan and the Romulus, and the cooperation of humans and Vulcans. Eventually, time travel in three Star Trek films has the function of maximizing the audience's interest in the story and allowing it to develop freely as a narrative tool. It also functions as an ideal solution for commenting on current problems in the non-narrative aspect. The significance of this paper is to stress the possibility that the motif of time travel in SF narrative will evolve as it continues to repeat in different forms as mentioned above.

  • Korea plans to send a pathfinder lunar orbiter to the Moon for the first time in August 2022. And according to the 3rd Basic Plan for Space Development Promotion, the plan is to send a lunar lander to the Moon before 2030. The selection of the lunar landing area can be varied depending on the lunar lander's mission, therefore preliminary research on the lunar landing sites is essential for a successful lunar exploration mission design. This paper analyzed the characteristics of major regions among 14 proposed regions using NASA's MoonTrek based on the data on the candidate areas for the major moon landing proposed sites by the NASA workshop in 2018. And we looked into what kind of future moon landing missions are suitable for these areas. We also looked at the importance of lunar Antarctica area through the recent lunar landing areas of Moon landing countries and Artemis plan.

A Comparative Study of Satisfaction and Pro-environmental Behavior on the Jeju Olle among Jeju Residents and Visitors (제주도민과 제주방문 도보여행객의 제주올레에 대한 만족도와 친환경 행동 비교)

  • The Jeju Olle Trail Route No. 1 has opened in 2007, and over one million visitors per year trek total of 26 Olle trails throughout the island of Jeju as of 2017. The Olle trekking is one of eco-friendly tourism activities in the era of social issues with environmental collapse and threats. Up until now, studies on the Jeju Olle trail are focused on the visitors while the studies on residents who are the other major pillar of Jeju Olle trail have been limited. The study aims to compare the visit types, satisfaction level and pro-environmental behaviors between the visitors and residents on Jeju Olle trails. Total of 290 survey was collected in three month period from December, 2016 to February, 2017. The survey result indicates the Jeju residents trek the Jeju Olle in a larger group than the Jeju Olle visiting trekkers and show lower level in the visit satisfaction and pro-environmental behaviors. The noticeable lower level in the pro-environmental behaviors of residents can directly connect to the issue of Jeju environment as a whole, and the continuing environmental education program and promotion for residents to improve the pro-environmental behavior level is essential.

  • By using DM(Diagnostic Monitoring) data measured at WiBro network, automatic optimization function of WiBro network is implemented in this paper. The optimization function mentioned is able to be run on PC with 2GHz CPU and 1 GB memory. Automatic optimization function is one module of CellTREK that is a wireless network planning and optimization software developed by Infra Lab., KT.

Analysis of Forefoot Bending Angle in Sprint Spikes According to Bobsleigh Start Lap Time for Development of Korean-Specific Bobsledding Shoes

  • Objective: The aim of this study was to analyze effects of the toe-spring angle of bobsleigh shoes on start speed lap time to develop Korean-specific bobsled shoes suitable for winter environments and for domestic players on the basis of sports science and optimized biomechanical performance. Method: Seven Korean bobsleigh athletes participated in this study, with three pairs of sprint spikes from three companies (Type A, Type B, Type C). To analyze sprint lap time and forefoot bending angle for each shoe, participants were instructed to drag a sled 15 meters from the start line at a maximum sprint. forefoot bending angle was collected by a high speed camera, and lap time speed was measured. Results: Lap time for type B shoes was $3.52{\pm}0.17sec$, type A was $3.55{\pm}0.19sec$, and type C was $3.56{\pm}0.18sec$. Forefoot bending angles were: angle 1, $6.88{\pm}5.55^{\circ}$; angle 2, $9.23{\pm}6.38^{\circ}$; angle 3, $15.56{\pm}5.39^{\circ}$; angle 4, $9.54{\pm}3.85^{\circ}$; angle 5, $9.22{\pm}5.08^{\circ}$; angle 6, $7.66{\pm}6.44^{\circ}$; and angle 7, $4.30{\pm}6.24^{\circ}$ (p<.001). Forefoot bending in angle 3 was as follows: type A, $16.47{\pm}6.01^{\circ}$; type B, $14.30{\pm}4.96^{\circ}$; and type C, $15.90{\pm}5.17^{\circ}$. Conclusion: Hard outsoles and midsoles are better than soft type for reduced start lap time when developing a prototype Korean bobsled shoe.

A simplified framework for estimation of deformation pattern in deep excavations

  • To stabilize the excavations in urban area, soil anchorage is among the very common methods in geotechnical engineering. A more efficient deformation analysis can potentially lead to cost-effective and safer designs. To this end, a total of 116 three-dimensional (3D) finite element (FE) models of a deep excavation supported by tie-back wall system were analyzed in this study. An initial validation was conducted through examination of the results against the Texas A&M excavation cases. After the validation step, an extensive parametric study was carried out to cover significant design parameters of tie-back wall system in deep excavations. The numerical results indicated that the maximum horizontal displacement values of the wall (δhm) and maximum surface settlement (δvm) increase by an increase in the value of ground anchors inclination relative to the horizon. Additionally, a change in the wall embedment depth was found to be contributing more to δvm than to δhm. Based on the 3D FE analysis results, two simple equations are proposed to estimate excavation deformations for different scenarios in which the geometric configuration parameters are taken into account. The model proposed in this study can help the engineers to have a better understanding of the behavior of such systems.
