Production of Bacterial Quorum Sensing Antagonists, Caffeoyl- and Feruloyl-HSL, by an Artificial Biosynthetic Pathway

  • A new series comprising phenylacetyl-homoserine lactones (HSLs), caffeoyl-HSL and feruloyl-HSL, was biologically synthesized using an artificial de novo biosynthetic pathway. We developed an Escherichia coli system containing artificial biosynthetic pathways that yield phenylacetyl-HSLs from simple carbon sources. These artificial biosynthetic pathways contained the LuxI-type synthase gene (rpaI) in addition to caffeoyl-CoA and feruloyl-CoA biosynthetic genes, respectively. Finally, the yields for caffeoyl-HSL and feruloyl-HSL were $97.1{\pm}10.3$ and $65.2{\pm}5.7mg/l$, respectively, by tyrosine-overproducing E. coli with a $\text\tiny{L}$-methionine feeding strategy. In a quorum sensing (QS) competition assay, feruloyl-HSL and p-coumaroyl-HSL antagonized the QS receptor TraR in Agrobacterium tumefaciens NT1, whereas caffeoyl-HSL did not.

The Effects of National Research Fund Recognition on the Research Fund Payment Intend in R&D Project (연구비에 대한 인식이 연구비 사용 의도에 미치는 영향에 관한 연구 : RCMS 사용자를 중심으로)

  • As the importance of R&D has recently become more important, the budget for national R&D projects has continued to grow. Along with, there is a growing awareness to use and manage R&D funds transparently. This study examines the relationship among attitude toward the behavior (ATT), subjective norm (SN), perceived behavioral control (PBC) and behavioral Intention (BI) deriving from TPB (Theory of planned Behavior). To empirically prove the hypothesis, the statistical analysis were conducted based on the response from the 2,021 RCMS users, using SPSS 21.0 and AMOS 21.0. The research model variable consists of ATT, SN, PBC, BI, person type, RCMS characteristics. The results of study can be summarized as follows. First, ATT, SN, PBC have the positive effect on BI. Second, According to the type of person (Director, R&D supporter), there is no difference in ATT on BI but there is a difference in SN (or PBC) on BI. Third, The type of person have moderating effects in the relationship between ATT and BI. Finally, All RCMS characteristics have moderating effects in the relationship between ATT and BI. Some RCMS characteristics have moderating effects in the relationship between PBC and BI.

  • The purpose of the study is to establ ish models of land Cover (use) prediction system for development and management of land resources using remotely sensed data as well as ancillary data in the context of multi-dis¬ciplinary approach in the application to CheJoo Island. The model adopts multi-date processing techniques and is a spatial/temporal land-Cover projection strategy emerged as a synthesis of the probability tra-nsition model and the discrimnant-analys is model. A discriminant modelis applied to all pixels in CheJoo landscape plane to predict the most likely change in land Cover. The probability transition model provides the number of these pixels that will convert to different land Cover in a given future time increment. The syntheric model predicts the future change in land Cover and its volume of pixels in the landscape plane.

The Effects of Regular Walking Exercise on Metabolic Syndrome, Cardiovascular Risk Factors, and Depressive Symptoms in the Elderly with Diabetic Mellitus (노인 당뇨병 환자의 규칙적 걷기운동 프로그램이 대사증후군, 심혈관 위험도 및 우울정도에 미치는 효과)

  • Purpose: This study aimed to estimate the effects of a regular walking exercise program on metabolic syndrome, cardiovascular risk factors, and depressive symptoms among the elderly with diabetic mellitus (DM) based on the Theory of Reasoned Action (TRA). Methods: This study has randomized and stratified experimental design with experimental and control groups. We developed a regular walking exercise program suitable for the elderly with DM based on the guidance of AAHPERD. The experimental group participated in the regular walking exercise program, which contains walking exercise 3 times a week and 50 minutes each time for 3 months and education on controlling diet and preventing complications once a week and 20 minutes each time for 4 weeks. Post-test was conducted after 3 months to estimate metabolic syndrome, cardiovascular risk factors, and depressive symptoms. Results: The regular walking exercise program was effective for decreasing the waist size, the level of fasting blood glucose (FBG) and triglyceride (TG), cardiovascular risk factors and the severity of depressive symptoms among the elderly with DM. Conclusion: The incidence of complications would be decreased by applying a regular walking exercise program.

Irrational Factors Affecting the Purchase of Online Game Items

  • Recently, a number of studies have drawn attention to purchasing online game items. Most of the studies are based on the assumption that consumers behave rationally. Accordingly, TRA- or value-based approaches have been mainly employed to understand the online purchases of game items. However, the purchasing behavior of consumers involves not only making rational decisions, but also making irrational decisions. Hence, their purchase behavior is affected by propensities for conspicuous consumption, impulsive consumption, and habitual consumption. Playing games can be highly addictive, and players often display such addictive behaviors. Our study explored both the rational and irrational factors in purchase behavior to understand how they are associated with purchasing game items. A total of 366 pieces of data were collected from Korean online game users through a survey. Regression analyses of the collected data showed that the behavior of buying game items was influenced not only by the intention to purchase which is a rational factor in consumption, but also by such irrational factors as habit, impulse, and ostentation which should be further emphasized in future studies.

  • A 27-year-old, right-hand-dominant woman with a posttraumatic anterior shoulder dislocation 3 months earlier after traYc accident presented because of pain and limited range of motion in the right shoulder. On physical examination, the patient had negative instability tests and a sulcus sign. On arthroscopic examination, a bifurcate long biceps tendon with two limbs was observed about 1 cm distal to the origin in the supraglenoid tubercle. We found no evidence of a tear in the long biceps tendon on probing, and the margin of each limb was smooth and round. Although this anatomic variant may be benign, its presence might be associated with other shoulder pathology. It is interesting to speculate whether the aberrant biceps anatomy in our patient contributed to transfer of injury at dislocation to the rotator cuff rather than to the classic anterior-inferior capsulolabral complex. In addition, recognition of the described anatomic variant on arthroscopy can aid the shoulder surgeon in focusing treatment on the actual pathology.

On the Beam Focusing Behavior of Time Reversed Ultrasonic Arrays Using a Multi-Gaussian Beam Model

  • One of the fundamental features of time reversal acoustic (TRA) techniques is the ability to focus the propagating ultrasonic beam to a specific point within the test material. Therefore, it is important to understand the focusing properties of a TR device in many applications including nondestructive testing. In this paper, we employ an analytical scheme for the analysis of TR beam focusing in a homogeneous medium. More specifically, a nonparaxial multi-Gaussian beam (NMGB) model is used to simulate the focusing behavior of array transducers composed of multiple rectangular elements. The NMGB model is found to generate accurate beam fields beyond the nonparaxial region. Two different simulation cases are considered here for the focal points specified on and off from the central axis of the array transducer. The simulation results show that the focal spot size increases with increasing focal length and focal angle. Furthermore, the maximum velocity amplitude does not always coincide with the specified focal point. Simulation results for the off-axis focusing cases do demonstrate the accurate steering capability of the TR focusing.

  • Han-Bok presents a unique consumption ex-perience different from Western dress which is common to our clothing life. This study is intended to propose the necessity of broad-ening deepening and systematizing a research related to the social aspects of Han-bok. A questionnaire consisted of 10 statements of attitude to Han-Bok 7 statements of the evaluation of the occasional appropriateness of Han-Bok preference questions about style color harmony and decorative materials of han-Bok was developed which includes 5 opinion statement of the importance of tra-ditiona and some demographic variables. During December 1996 and January 1997 the women aged above 20 responded to it and finally 702 data was analyzed. Korean urban women have positive attitude to Han-Bok And the conceit about Han-Bok was significantly higher than the intention to wear it . The most properly evaluated occasion for wearing han-Bok was 'holdays' like Seolnal and chuseok Han-Bok was evaluated im-proper for casual wear and out wear. Factor analysis of 7 occasions determined 3 factors; traditional occasion special occasion and cas-ual occasion. It is needed to identify the ef-ficiency of Han-Bok according to each differ-ent occasion. Korean urban women preferred the style of traditional line soft color and modern har-mony. In a viewpoint of tradition they put more importance in the line of Han-Bok that they do in the color of it.

lacZ- and aph-Based Reporter Vectors for In Vivo Expression Technology

  • Three vectors, pSG1, 2, and 3, which facilitate in vivo expression technology (IVET) in Gram-negative bacteria, were developed. Vectors pSG1and 2 are derivatives of ColE1, and pSG3 is a derivative of an R6K replicon. These vectors contain oriT sites that allow mobilization when the RK2 Tra functions are provided in trans. These vectors contain promoterless lacZ (pl-lacZ) and promoterless aph (pl-aph) transcriptionally fused together, which allow qualitative and quantitative measurements of the expression of genes in the genome of bacterial cells. pSG1 and 3 contain gentamicin-resistance genes, and pSG2 carries a streptomycin-/spectinomycin-resistance gene, allowing for selection of recombinants generated by a single crossover between a library fragment cloned into a pSG vector and the identical region in the genome of a bacterial species from which the library fragment originated. These vectors were successfully applied to the generation of random fusions at high rates in the genomes of four representative Gram-negative bacteria. In addition, the expression level of ${\beta}-galactosidase$ and the degree of resistance to kanamycin in cells with fusions generated by these vectors were found to be linearly correlated, proving that these vectors can be used for IVET.

  • Mobile genetic segments, or transposons, are also referred to as jumping genes as they can shift from one position in the genome to another, thus inducing a chromosomal mutation. According to the target site-specificity of the transposon during a transposition event, the result is either the insertion of a gene of interest at a specific chromosomal site, or the creation of knockout mutants. The former situation includes the integration of conjugative transposons via site-specific recombination, several transposons preferring a target site of a conserved AT-rich sequence, and Tn7 being site-specifically inserted at attTn7, the downstream of the essential glmS gene. The latter situation is exploited for random mutagenesis in many prokaryotes, including IS (insertion sequence) elements, mariner, Mu, Tn3 derivatives (Tn4430 and Tn917), Tn5, modified Tn7, Tn10, Tn552, and Ty1, enabling a variety of genetic manipulations. Randomly inserted transposons have been previously employed for a variety of applications such as genetic footprinting, gene transcriptional and translational fusion, signature-tagged mutagenesis (STM), DNA or cDNA sequencing, transposon site hybridization (TraSH), and scanning linker mutagenesis (SLM). Therefore, transposon-mediated genetic engineering is a valuable discipline for the study of bacterial physiology and pathogenesis in living hosts.
