Interactions between Water-Soluble Polyparacyclophanes and Drugs (II) -Interaction between Water-Soluble Polyparacyclophanes and Fluorescent Hydrophobic Naphthalene Derivatives in Aqueous Solution- (수용성 폴리파라시클로판류와 약물과의 상호작용(제 2보) -수용액중 수용성 폴리파라시클로판류와 형광 소수 나프탈렌 유도체류와의 상호 작용-)

  • A series of water-soluble polyparacyclophanes bearing two diphenylmethane or two diphenyl ether skeletons were investigated to develop useful host compounds by using 1-anilinonaphthalene-6-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) as fluorescent hydrophobic substrates in aqueous solution. It was noteworthy that remarkable fluorescent enhancements and blue shifts of ANS and TNS were observed only in the presence of 1,6,20,25-tetraaza[6.1.6.1] paracyclophane (CPM 44) and 1,6,21,27-tetraaza [7.1.7.1] paracyclophane (CPM 55) for diphenylmethane skeleton, and 1,7,21,27-tetraaza-14,34-dioxa [7.1.7.1] paracyclophane (CPE 55) and 1,8,22,29-tetraaza-15,36-dioxa [8.1.8.1] paracyclophane (CPE 66) for diphenyl ether skeleton, comparing with ${\alpha}-\;and\;{\beta}-cyclodextrins$. However, their acyclic analogues such as 4,4'-dimethylaminodiphenylmethane and 4,4'-dimethylaminodiphenyl ether, and paracyclophanes whose cavities were smaller showed only small effects under the same conditions. These facts suggested that hosts and substrates were in an intimate contact which would not occur without larger structures, and thus that guest molecules were strongly incorporated in the hydrophobic cavities of these larger paracyclophanes. The effects of pH on the fluorescent intensity of ANS-CPM 44, ANS-CPM 55, ANS-CPE 55, ANS-CPE 66, TNS-CPM 44, TNS-CPM 55, TNS-CPE 55 and TNS-CPE 66 systems were not significant below pH 2.0, but their fluorescent intensities were markedly reduced with increasing ionic strength.

  • Complex formation of water-soluble polyparacyclophanes bearing two diphenylmethane or two diphenyl ether skeletons with l-anilinonaphthalene-8-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) was investigated quantitatively to develop useful host compounds comparing with ${\alpha}\;-\;and\;{\beta}-cyc1odextrins$$({\alpha}-\;and\;{\beta}-CyDs$) in aqueous solution. Benesi-Hildebrand type analysis of the fluorescent intensity showed that the dissociation constants (Kd) of paracyclophane-ANS complexes were $1.55\;{\times}\;10^{-4}M$ for 1,6,20,25-tetraaza[6.1.6.1]paracyclophane(CPM 44) and $1.23\;{\times}\;10^{-4}M$ for 1,7,21,27-tetraaza[7.1.7.1]paracyclophane (CPM 55), and those of paracyclophane-TNS complexes were $6.99\;{\times}\;10^{-6}M$ for CPM 44 and $6.23\;{\times}\;10^{-5}M$ for CPM 55, in 1:1 molar ratio. On the other hand, the Kd values of 1,7,21,27-tetraaza-14,34-dioxa[7.1.7.1]paracyclophane (CPE 55)-ANS, 1,8,22,29-tetraaza-15,36-dioxa[8.1.8.1]paracyclophane (CPE 66)-ANS, CPE 55-TNS, CPE 66-TNS complexes were $1.75\;{\times}\;10^{-3}M$, $3.07\;{\times}\;10^{-3}M$, $3.75\;{\times}\;10^{-3}M$ and $2.15\;{\times}\;10^{-3}M$, respectively. On the contrary, the Kd values of ${\alpha}-CyD-ANS$, ${\beta}-CyD-ANS$, ${\alpha}-CyD-TNS$ and ${\beta}-CyD-TNS$ complexes were found to be $3.98\;{\times}\;10^{-2}M$, $1.05\;{\times}\;10^{-2}M$, $1.38\;{\times}\;10^{-2}M$ and $3.52\;{\times}\;10^{-4}M$, respectively. These results mean that the complexation of CPMs with ANS or TNS is by 5.6-1,975 fold stronger than that for ${\alpha}-or\;{\beta}-CyDs$, and the complex formation of CPEs with ANS or TNS is nearly same as or somewhat stronger than that for ${\alpha}-or\;{\beta}-CyDs$. From the Kd values determined at different temperatures, thermodynamic parameters were calculated and the complexation was found to be a spontaneous exothermic reaction. The effects of pH on Kd values of CPM 44-ANS, and CPM 55-ANS complexes were negligible in the range of pH 1.2-1.8. However, the Kd values of these complexes increased significantly with increasing ionic strength.

  • 도로교통소음은 많은 지역에 산재해 있으며 특히 주도로변에 거주하는 사람들에게 환경과 관련하여 매우 중요하다. 도로교통으로부터 소음수준을 계산하는데 몇가지 다른 방법들이 이용되고 있다. 이 방법들은 계산방법과 그래프식 그리고 컴퓨터 모델링 기술 등이다. 교통과 교통소음의 영향으로부터 소음을 계산하는 간단한 기술의 예측방법은 여기에 나타내었다. 이 TNS (traffic Noise Screening) 방법은 서로 다른 도로유형에 대한 일련의 도로교통소음레벨의 예측그래프로 전개된 것이다. 이 그래프는 Federal Highway Administration (FHWA) STAMINA 2.0을 이용하여 다양한 시나리오에 대한 소음 예측모델을 계산한 결과를 기초하였다. TNS에 도로의 기하학적 형태, 교통량 주행속도 그리고 도로중앙선의 거리등의 데이터를 입력시킨다. TNS 그래프는 소음영향과 연관된 교통소음예측에서 사용하는 경우 교통소음레벨의 계산을 쉽게 한다. 이 TNS 방법은 STAMINA 2.0과 같은 상세 모델링을 대신하지는 못하지만 상세 모델을 필요로 할 때 도움을 주는 도구이다 만약 소음계산들이 중요하거나 또는 시나리오가 보다 복잡하고 부가된다면 보다 상세한 모델링이 수행되어져 스크린 결과들이 나타난다.

Characterization of Antidiabetic Compounds from Extract of Torreya nucifera (비자나무 추출물의 항당뇨 활성물질의 특성 연구)

  • Natural products have gained increasing attention due to their advantage of long-term safety and low toxicity for a very long time. Torreya nucifera is widespread in southern Korea and Jeju Island and its seeds are commonly used as edible food. Oriental ingredients have often been reported for their insecticidal, antioxidant and antibacterial properties, but there have not yet been any studies on their antidiabetic effect. In this study, we investigated several biological activities of T. nucifera pericarp (TNP) and seeds (TNS) extracts and proceeded to characterize the antidiabetic compounds of TNS. The initial results suggested that TNS extract at 15 and 10 ㎍/ml concentration has inhibitory effects on α-glucosidase and protein tyrosine phosphatase 1B, that is 14.5 and 4.35 times higher than TNP, respectively. Thus, the stronger antidiabetic TNS was selected for the subsequent experiments to characterize its active compounds. Ultrafiltration was used to determine the apparent molecular weight of the active compounds, showing 300 kDa or more. Finally the mixture was then partially purified using Diaion HP-20 column chromatography by eluting with 50~100% methanol. Therefore we concluded that the active compounds of TNS have potential as therapeutic agents in functional food or supplemental treatment to improve diabetic diseases.

New and noteworthy records of the moss flora of Korea

  • A total of 34 taxa, 33 species and one variety, are newly added to the moss checklist of Korea. They were collected on Jejudo Island, Samcheok limestone areas in Gangwon Province, and Socheongdo Island from 2015 to 2018 and organized into 19 families and 26 genera. Additionally, eight genera are new to Korea. This study verifies the distributions of nine species whose distributions and specimens have remained unknown thus far. Finally, two unrecorded taxa, consisting of one species and one variety, are identified by reexamining Korean specimens deposited at the National Museum and Science in Japan (TNS). The specimens were collected in 1942 and 1940 from Mt. Baekdusan and Anbyun in North Korea.

  • Aconitum kaimaense Uyeki & Sakata was first collected by Uyeki & Sakata at Bujeon Plateau in Hamgyongnam-do and reported as a new species and recorded in Enumeration of Korean Plants (Pak, 1949). A. kaimaense was taxonomically treated under the name of Aconitum jaluense Kom in A Revision of Aconitun Subgenus Aconitum (Ranunculaceae) of East asia. (Kadota, 1987), and it was also treated as synonym in Korea(1996, 2007, 2011). A. kaimaense has erect or apically slightly curved stem, dense corymb inflorescence, pubescent pedicels, yellowish white sepals and carpels 5(or 4) pubescent. In this study, to investigate the taxonomic location of A. kaimaense, we conducted literature observation, sample observations in KYO, TNS, KH, SNU, SKK, and field research in Korea. The distribution in Hongcheon, Pyeongchang, Jeongseon, and Yangyang of Gangwon-do was confimed.

  • This paper provides an improvement on our previous study [1] for multi-fault diagnosis in real time in large-scale systems. In the method, fault propagation probability(FPP) and fault propagation time(FPT) in a fuzzy sense are additively used to describe the fault propagation model(FPM) in more practical manner. A modified fault diagnosis procedure is also given. This method is applied for diagnosis of the primary system in the Kori nuclear power plant unit 2 under a transient condition in case of unit value of FPP on each branch of the FPM.

Cloning of Small Plasmids from Bacillus thuringiensis Subsp. israelensis Using Plasmid Capture System

  • Recently, we have developed an easy, simple and convenient circular DNA cloning system named plasmid capture system (PCS). To investigate usefulness of PCS in cloning of plasmids from Bacillus thuringiensis strains, PCS donors, pPCS-S and pPCS-L were applied to clone plasmids of B. thuringiensis subsp. israelensis by in vitro transposition using 4{TnsABC^*}$ transposase. In result, 3 small plasmids were cloned, and these were consistent with pTX14-1, pTX14-2 and pTX14-3 reported previously from B. thuringiensis subsp. israelensis. Therefore, the PCS can be successfully applied to clone small plasmids from B. thuringiensis strains.

  • $BaMnAl_{11}O_{19}$ was prepared by urea-induced homogeneous precipitation and characterized by X-ray diffraction and scanning electron microscopy. At increased precipitant concentrations, AlOOH replaced $Al(OH)_3$ as an Al precursor. $BaMnAl_{11}O_{19}$ exhibited enhanced catalytic combustion performance and inhibited CO generation. Catalytic performance was also enhanced by the presence of $BaAl_2O_4$ and $BaMnAl_{11}O_{19}$. Compared to $BaAl_2O_4$, $BaMnAl_{11}O_{19}$ exhibited superior catalytic combustion activity.

Quality Investigation of Commercial Northern Sand Lance, Ammodytes Personatus Sauces (시판 까나리액젓의 품질조사)

  • The quality characteristics of 15 kinds of the commercial northern sand lance, Ammodytes Personatus sauce (CNS) of korean traditional salt-fermented fish sauces were evaluated comparing to the traditional northern sand lance, Ammodytes Personatus sauce (TNS). The ranges of chemical compositions of the CNSs were $66.5\~71.0\%$ moisture, $19.3\~24.6\%$ ash, $4.7\~12.0\%$ crude protein, and the pH and salinity were $5.56\~6.47\%,\;24.0\~32.9\%$, respectively. Total nitrogen, amino nitrogen, total free amino acid and total ATP related compounds (sum of ATP$\~$IMP, HxR, Hx and uric acid) were in the ranges of 0.781$\~$ 1,918 g, 445.9$\~$1037.9 mg, 3,258.9$\~$6:562.6mg in 100 ml CNSs, and 4.766$\~$8.989 $\mu$mol in 1 ml CNSs. The CNSs were higher in content of moisture, TMAO, TMA and pH, but lower in content of crude protein, salinity, total nitrogen, amino nitrogen, total ATP related compounds, absorbance at 453 nm and total free amino acid than the TNSs, Both CNS and TNS samples were rich in free amino acids, such as glutamic acid, lysine, alanine, leucine, valine, aspartic acid and isoleucine in the order.
