Structural Change in Transmembrane Region of Syndecan-4 by Mutation

  • Transmembrane(TM) proteins are closely related to transport, channel formation, signaling, cell to cell interaction, so they are the crucial target of modern medicinal drugs. In order to study the structure and function of these TM proteins, it is important to prepare reasonable amounts of proteins. However, their preparation is seriously difficult and time-consuming due to insufficient yields and low solubility of TM proteins. We tried to produce large amounts of Syndecan-4 containing TM domain(SDC4-TM) that is related to the wound healing and tumor. Also, mutated SDC4-TM was studied to investigate structural change by modification of dimerization motif. We performed the structure determination by the Polarity Index at Slanted Angle (PISA) wheel pattern analysis based on $^{15}N-^1H$ 2D SAMPI-4 solid-state NMR of SDC4-TM and computational modeling using Discovery Studio 2016.

Characterization of the recombinant cellulase B from Thermotoga maritima (Thermotoga maritima 유래 내열성 cellulase B 융합단백질의 특성 규명)

  • A gene encoding thermostable cellulase B (TmCelB) was isolated from Thermotoga maritima. The open reading frame (ORF) of TmCelB gene was 825bp long which predicted to encode 274 amino acid residues with a molecular weight of 31,732 Da. The 17 amino acid residues from N-terminal of the TmCelB was known as signal peptides. To analyze the enzymatic activity and biochemical properties, the ORF of TmCelB gene excluding a putative signal sequence encoding 17 amino acids were introduced into the E. coli expression vector, pRSET-B, and overexpressed in E. coli BL21. The optimum temperature of recombinant TmCelB was around 95 ℃, and the optimum pH of recombinant TmCelB was around pH 4.5. The recombinant TmCelB was stable at temperature below 100 ℃.

Monitoring of Rice Growth by RADARSAT and Landsat TM data (RADARSAT과 Landsat TM자료를 이용한 벼 생육모니터링)

  • The objective of this study is to evaluate the use of RADARSAT and Landsat TM data for the monitoring of rice growth. The relationships between backscatter coefficients($\sigma$$^{0}$ ) of RADARSAT data and digital numbers (DN) of Landsat TM and rice growth parameters were investigated. Radar backscatter coefficients were calculated by calibration process and then compared with rice growth parameters; plant height, leaf area index (LAI), and fresh and dry biomass. When radar backscatter coefficient ($\sigma$$^{0}$ ) of rice was expressed as a function of time, it is shown that the increasing trend ranged from -22--20dB to -9--8dB as growth advances. The temporal variation of backscatter coefficient was significant to interpret rice growth. According to the relationship between leaf area index and backscatter coefficient, backscatter coefficient underestimated leaf area index at the beginning of life history and overestimated, at the reproductive stage. The same increasing trend between biomass and backscatter coefficient was shown. From these results, RADARSAT data appear positive to the monitoring of rice growth. Each band of time-series Landsat TM data had a significant trend as a rice crop grows during its life cycle. Spectral indices, NDVI[(TM4-TM3)/(TM4+TM3)] and RVI(TM4/TM2), derived from Landsat TM equivalent bands had the same trend as leaf area index.

A Study on Superficial Dose of 6MV-FFF in HalcyonTM LINAC: Phantom Study (HalcyonTM 선형가속기 6MV-FFF 에너지의 표재 선량에 대한 고찰: Phantom Study)

  • Purpose: The aims of this study were to compare the superficial dose with Optically Stimulated Luminescence Dosimeter(OSLD) measurement and Treatment Planning System(TPS) calculation for 6MV-Flattening Filter Free(FFF) energy using HalcyonTM and TrueBeamTM. Materials and methods: Phantom study was performed using the CT images of human phantom. In the treatment planning system, the Planning Target Volume(PTV) was contoured which is similar to Glottic cancer. Furthermore, Point(M), Point(R), and Point(L) were contoured at the iso-center of head and neck region and 5mm bolus was applied to the body contour. Each treatment plans using 6MV-FFF energy from HalcyonTM and TrueBeamTM with static Intensity Modulated Radiation Therapy(IMRT) and Volumetric Modulated Arc Therapy(VMAT) were established with eclipse. To reproduce the same position as the TPS, OSLDs were placed at the iso-center point and 5mm bolus was applied to compare the error rate after the dose delivery. Result: The results of the study using human phantom are as follows. In case of HalcyonTM, the mean absolute error rates of the point dose using the treatment planning system and the dose measured by OSLD were 1.7%±1.2% for VMAT and 4.0±2.8% for IMRT. Also TrueBeamTM was identified as 2.4±0.4% and 8.6±1.8% respectively for VMAT and IMRT. Conclusion: Through the results of this study, TrueBeamTM confirmed that the average error rate was 2.4 times higher for VMAT and 3.6 times higher for IMRT than HalcyonTM. Therefore, based on the results of this study, If we need a more accurate dose assessment for the superficial dose, It is expected that using HalcyonTM would be better than TrueBeamTM.

Bi-directional energy transfer process in Er3+-Tm3+-codoped fluorozirconate glasses

  • Energy transfer direction in $Er^{3+}-Tm^{3+}$-codoped fluorozirconate glasses has been studied. For $Er^{3+}-Tm^{3+}$-codoped glasses, the dependence of the green emission intensity on the pump power (Pex) of 800 nm has ranged from (Pex)$^2$ to (Pex)$^3$. From this multistep absorption, a 1.48 $\mu m$ emission from the $^3F_4{\rightarrow}^3H_4$ transition on Tm$^{3+}$ ion has been found to transfer into $^4I_{13/2}$, $^4I_{9/2}$ and $^4S_{3/2}$ on $Er^{3+}$ ion. In case of the 1.06 $\mu m$ pumping, the emissin ratio of $^3H_4$ level in $Tm^{3+}$ to $^4I_{13/2}$ in $Er^{3+}$ showed that the amount of the energy transfer from $Tm^{3+}$ into $Er^{3+}$ increased with the increasing concentration of $Tm^{3+}$ ion. Our two kinds of pumping scheme suggest that the direction of dominant energy transfer between $Er^{3+}$ and $Tm^{3+}$ should be dependent on whether the $^3F_4$ level resonates in $Tm^{3+}$ the level or not.

Topographic Relief Mapping on Inter-tidal Mudflat in Kyongki Bay Area Using Infrared Bands of Multi-temporal Landsat TM Data

  • The objective of this study is to develop a method to generate micro-relief digital elevation model (DEM) data of the tidal mudflats using multi-temporal Landsat Thematic Mapper (TM) data. Field spectroscopy measurements showed that reflectance of the exposed mudflat, shallow turbid water, and normal coastal water varied by TM band wavelength. Two sets of DEM data of the inter-tidal mudflat area were generated by interpolating several waterlines extracted from multi-temporal TM data acquired at different sea levels. The waterline appearing in the near-infrared band was different from the one in the middle-infrared band. It was found that the waterline in TM band 4 image was the boundary between the shallow turbid water and normal coastal water and used as a second contour line having 50cm water depth in the study area. DEM data generated by using both TM bands 4 and 5 rendered more detailed topographic relief as compared to the one made by using TM band 5 alone.

An Analysis of the Landuse Classification Accuracy Using IHS Merged Images from IRS-1C PAN Data and Landsat TM Data (IRS-1C PAN 데이터와 Landsat TM 데이터의 IHS중합화상을 이용한 토지이용분류 정확도 분석)

  • In this study, effective multispectral Landsat TM band combinations for a merging with the high resolution IRS-1C PAN data using the IHS method to improve landuse accuracy is discussed. From the pre-classified image using the merged images with TM all six band images(with the exception of band 6 image) and PAN image, a sample data which has ten classes was generated. An evaluation of the overall classification accuracy for the representative seven merged images which were merged using each TM three-band images and IRS-1C PAN image by IHS method for the sample area. The increase in classification accuracy is most significant with the inclusion of two of TM4, TM5 and TM7 infrared band images. Especially, the largest increase(11.8 percent) in landuse classification accuracy were investigated when Landsat TM247 bands were merged with IRS-1C PAN data. The classification accuracy when TM three band image and PAN image were used without merging is higher than result of the case of using the merged images.

Expression, Purification and NMR Studies on MC4R-TM2 Mutant

  • Melanocortin-4 receptor (MC4R) subtype is associated with obese humans. Especially, in a patient with severe early-onset obesity, novel heterozygous mutation in the MC4R gene was detected, resulting in an exchange of aspartic acid to asparagine in $90^{th}$ amino acid residue located in the predicted second trans-membrane domain (TM2). Mutations in the melanocortin-4 receptor (MC4R) gene are the most frequent monogenic causes of severe obesity which have been described as heterozygous with loss of function. In order to compare structure difference between MC4R wild type (MC4R-TM2-wt) and mutant (MC4R-TM2-D90N), we designed both MC4R-TM2-wt and MC4R-TM2-D90N construct in pET 21b vector. In this study, we optimized high-yield purification procedure for recombinant TM2-D90N. Eluted recombinant protein was resolubilized under urea condition for thrombin cleavage reaction and we conducted the high-performance liquid chromatography (HPLC) with reverse phase column under 1% acetonitrile, 0.01% TFA buffer solution. The molecular size of purified target peptide was confirmed by Tricine-SDS page analysis. To characterize MC4R-TM2-D90N, we have performed $^{15}N$-isotope labeling of peptide using M9 media and purified labeled target peptide for hetero-nuclear NMR spectroscopy.

A Study on the Comparison of LANDSAT-5 TM and MSS Data -laying stress on the landuse mapping of Incheon area- (LANDSAT-5의 TM과 MSS 데이타의 비교에 관한 연구 -인천지역의 토지이용분류를 중심으로-)

  • In this study, practical use of TM (Thematic Mapper) data was evaluated by comparison of LANDSAT- 5 TM and MSS (Multispectral Scanner) data. The comparison of TM and MSS data was achieved by analyzing the result of landuse mapping of Incheon area, and in addition, the comparison of accuracy according to image enhancement method was made. From the results of this study, we found that TM data was more accurate by about 20% than MSS data in landuse mapping, and that smoothing was effective in image enhancement processing of TM data.

THz Filter Using the Transverse-electric (TE1) Mode of the Parallel-plate Waveguide

  • The results of the experimental and theoretical studies conducted on terahertz filtering using two parallel-plate waveguides (PPWGs) are presented herein. The first PPWG with 355 ${\mu}m$ plate separation generates 4 THz bandwidth TM modes ($TM_0,\;TM_2,\;and\;TM_4$), whereas the second PPWG with 100 ${\mu}m$ plate separation operated the THz filter for the generated TM modes with 1.5 THz cutoff frequency. The outgoing THz wave of the two PPWGs was truncated until 1.5 THz, and the system was operated using a high-pass THz filter. The absorption and dispersion of the combined TM and TE modes for the filtering system were calculated. The theoretical calculation and measurement results for the cutoff and oscillation frequency in the spectrum domain agreed well.
