Hypericin Contents of Hypericum erectum Thunberg (고추나물의 Hypericin 함량)

  • To explore the Korea native plants to substitute for St. John's wort, which produce hypericin that use commercially for the treatment of mild to moderate depression, hypericin contents of Hypericum erectum and H. ascyron collected in two mountain of Korea were examined. From TLC and TLC-densitometer analysis of hypericin contents and biosynthetic pattern, hypericin was detected in flower and leaf of H. erectum, but not in all organs of H. ascyron. The hypericin content of H. erectum grown 200 m high hill in Mt. Byoung-pung was higher than that of Mt. Ji-ri 500 m and 1300 m high hill. When the seasonal variation of hypericin contents in H. erectum leaf collected from two regions was investigated, leaf collected from both regions on July was higher than other seasons.

Isolation and Identification of Cholesterol Oxidation products in heated tallow by TLC (TLC를 이용한 가열우지중 콜레스테롤 산화생성물의 분리 및 확인)

  • The oxidative stability of cholesterol in tallow heated at different frying temperatures (130$\^{C}$, 150$\^{C}$, and 180$\^{C}$) was studied by identifying cholesterol oxides by thin layer chromatography(TLC). And fatty acid compositions in tallow heated were also measured and compared with cholesterol oxides. Unsaturated fatty acid contents slightly decreased as the heating time increased, whereas saturated fatty acid contents increased This phenomenon became excessive especially by heating to higher temperature. It was found that RF value and spot color of the nonsaponifiable lipids from tallow heated on TLC analysis accorded with the synthetic cholesterol oxides in this experiment. Four kinds of cholesterol oxides were detected in tallow heated for 24 hours at three different temperatures. The oxides were identified as 7-$\alpha$-hydroxycholesterol, 7-$\beta$-hydroxycholesterol, 7-ketocholesterol and cholesterol epoxide. It was found that there was a little difference in oxidative pattern of cholesterol between several heating temperatures.

  • To investigate ginseng oligopeptides with biological activities, the water extract was purified by ultra-filtration, gel filtration, ion-exchange and thin layer chromatography. Ultra-filtered water extract exhibited antilipolytic activity, inhibiting epinephrine-induced lipolysis in the isolated fat cells of rat epididymal adipose tissue. The filtrate was separated into 3 fractions by Sephadex G-25 gel filtration. Peptides were found only in the first fraction(S-FI). Saponine and sugars were also detected in tie fraction. S-FI fraction resolved further into 6 fractions by Dowex 50 ion-exchange chromatography. The sugar and saponine depleted fraction(P-F2) from the second chromatography showed antilipolytic activity. The P-F2 fraction revealed 6 spots on TLC. The 6 spots were isolated by TLC and identified as peptides.

Separating of Falcarinol from Acanthopanax senticosus (가시오가피(Acanthopanax senticosus)에서 polyacetylene계 화합물인 falcarinol의 동정)

  • To detect falcarinol in Acanthopanax senticosus, its stem and bark were extracted with methanol fractionated with petroleum ether and diethyl ether, and separated by silica gel column chromatography, Resulting six crude samples were compared with standard falcarinol through TLC on silica gel plates. Fraction 6 showed $R_{f}$ value of 0.46 similar to that of standard falcarinol. Through analytical reverse phase HPLC/PDA, UV spectra of standard falcarinol and traction 6 recorded between 200 and 340 nm showed identical peaks and UV spectra patterns. GC/MS revealed standard falcarinol and fraction 6 have equal retention times of 7.4 and 8.5 min before and after TMS-derivatization, respectively. Standard falcarinol and fraction 6 showed more similar spectrum patterns after TMS-derivatization than before. These results confirm presence of falcarinol in A. senticosus.

Accuracy of Spirometry at Predicting Restrictive Pulmonary Impairment (제한성 환기장애의 진단에서 폐활량검사의 정확성)

  • Background : Low spirometric forced vital capacity(FVC) in conjunction with a normal or high ratio of the forced expiratory volume at 1 second to the forced vital capacity($FEV_1$/FVC%) has traditionally been classified as a restrictive abnormality. However, the gold-standard diagnosis of a restrictive pulmonary impairment requires a measurement of the total lung capacity (TLC). This study was performed to determine the predictive value of spirometric measurements of the FVC for diagnosing a restrictive pulmonary abnormality. Methods : Test results from 1,371 adult patients who undertook both spirometry and lung volume measurements on the same visit from January 1999 to December 2000 were enrolled in this study. The test values for the FVC, the TLC that was below 80% of predicted value, and a $FEV_1$/FVC% that was below 70%, were classified as being abnormal. Results : Of the 1,371 patients, 353 patients had a reduced a FVC. Of these patients, 186 patients had a reduced TLC. Therefore, the positive predictive value was 52.7%. Of the 196 patients with a normal $FEV_1$/FVC% and a reduced FVC, 148(75.5%) patients had a lower TLC. Thirty eight (24.2%) patients out of 157 patients with a low $FEV_1$/FVC% and a low FVC showed a restrictive defect. Conclusion : Spirometry is useful to rule out a restrictive pulmonary abnormality, but a restrictive pattern on the spirometry dose not mean there is a true restrictive disease. For the patients with a low FVC, TLC measurements are essential for diagnosing a restrictive pulmonary impairment.

  • Traditional Chinese herbal drugs, namely, Atractylodes Rhizome and Atractylodes Rhizome White were identified by means of chemical analysis such as TLC and GC/MS. In Korea, the old-grown and the newly-grown rhizomes of Atractylodes japonica Koidzumi (Compositae) are used as the Atractylodes Rhizome and the Atractylodes Rhizome White, respectively. Both rhizomes contain the atractylon asa major spot on TLC. The content of atractylon in a newly-grown rhizome of A. japonica (Atractylodes Rhizome White called in Korea) appears to contain much larger quantity in comparison with that of an old-growon one (Atractylodes Rhizome). The TLC spot pattern analysis showed that rhizomes of A. japonica and A. macrocephala contain the atractylon as a major indicator spot, but no atractylodin (extreamly small amount). However, rhizomes of A. lancea De Candolle and A. koreana Kitamura contain the atractylodin as a major indicator spot.

  • This study was conducted to determine the inheritance of duvatrienediol(DVTs) and cis-abienol concentration from flesh tobacco leaves. NC744, which has a very low levels of DVTs and no cis-abienol was used as a parent in cross to TI 1068 producing a normal amount of DVTs and cis-abienol. Presence of DVTs and cis-abienol on the leaf surface was determined using thin layer chromatography(TLC). Segregation pattern from F$_2$ and BC$_2$[ (TI 1068 $\times$ NC744) $\times$ NC744J generations revealed that TI 1068 have a single dominant gene controlling DVTs and cis-abienol production. And DVTs production was inherited independently of the ability to produce cis-abienol.

  • Anatomical and physico-chemical properties of some nutrient and tonic crude drugs have been estimated and compared. Resin duct and druse crystal were observed more in Acanthopanax Cortex than in Ginseng Radix. Bast fiber bundle and solitary crystals were abandantly present in Glycyrrhizae Radix. Mucilage crude drugs contained mucilage ducts and bundles of raphides as common components and their arrangements of vascular bundles were different. TLC analysis showed different patterns of saponin, flavonoid, ${\gamma}-aminobutyric$ acid and sitosterols.

Quality comparison of musk products using a chromatographic pattern analysis (사향의 크로마토그램 패턴 분석을 통한 품질비교 연구)

  • Objectives : This study is to check the quality of Moschus products in Korean markets using a chromatographic analysis. Methods : We collected musk products, two genuine products from Russia (A, B), two authentic products from Hong Kong (C) and Korea (D), the Ministry of Food and Drug Safety (MFDS) standard (E), and two fire ants products (E, F). Results : For identification, TLC analysis of 60% ethanol extracts of each product showed that A, B, C, and E have the suitable patterns before color development at 365 nm UV light for the MFDS regulation in Korea. A clear red spot was observed from the E at Rf 0.6 with 365 nm UV light after color development, but this spot was not found in A and B. For the purity test, a distinct violet spot with Rf 0.87 was observed from the A and B in TLC analysis of methanol extracts of each product at white light, however, this spot was not matched with the impurities of E and F. In HPLC-UV pattern analysis, a similar peak pattern was shown in A, B and E, and similar peaks were observed C, although the similarity was weaker than that of A, B, and E. The F and G showed different peak patterns compared with the peak patterns of other samples. Conclusions : Hence, it is considered that the test methods need flexibility in application for identification and purity test depending on the type of sample.

A Study of Salmon Oil Type Analysis by FT-IR and Carbon Isotopes Ratio (FT-IR과 탄소동위원소 분석을 통한 연어유의 구분에 관한 연구)

  • This study analyzes the structure types of salmon oil to evaluate the purity of salmon oil products based on the 38 different types of imported salmon oil products distributed in the Republic of Korea. The major types of omega-3 foods in the salmon oil are ethyl ester (EE) and triglyceride (TG). If the salmon oil contained potential contaminants and was processed in order to remove it, EE type omega-3 fatty acids are found in concentration. This provides a good guide in assessing if products were made with EE type ingredients or re-esterified contaminated materials. The results of the FT-IR analysis showed significant difference in the C=O, C-O band positions in TG and EE. There were 19 TG type products and 19 EE type products. The analysis of carbon isotope ratio was performed on the types of TG and EE. There were different properties in the 19 TG type products. In one product, the carbon isotope ratio was -25.15 and the other 18 products showed -22.15~-23.96. The carbon isotope ratio of all 19 EE type products showed -21.91~-23.74. The results of the TLC analysis showed similar results with FR-IR. The re-esterified TG form was not detected in the TG type products, confirming that the TG type products contained natural salmon oil. This study aimed to provide the basic material in classifying the types of natural salmon oil and re-esterified salmon oil, by analyzing the pattern and proportion of FT-IR spectrum, carbon isotope ratio, and TLC.
