Rhus Verniciflua Stokes Extract Suppresses Expression of Metalloproteinases, iNOS and COX-2 in THP-1 Cells Via Inhibiting NF-𝜅B and MAPK Phosphorylation

  • Objectives: The aim of this study is to investigate the mechanisms involved in the anti-inflammatory and anti-tumor effects of Rhus verniciflua Stokes (RVS) on PMA-differentiated human monocytic leukemia THP-1 cells. Methods: Cells were treated with various concentrations of RVS decoction (0-300㎍/ml) for 24, 48, and 72h. Cell viability was evaluated by MTS/PMS assay. The expressions of MMP-2, MMP-9, TIMP-1, TIMP-2, iNOS and COX-2 mRNA and proteins were measured using RT-PCR and western blotting, respectively. Results: RVS suppressed expression of MMP-2 and MMP-9 mRNA. It also down-regulated iNOS and COX-2 mRNA and protein expression. RVS inhibited NF-𝜅B p65 activity and the phosphorylation of Akt and MAPK (ERK and p38 MAPK). Instead, the phosphorylation of JNK is increased at a very low concentration but decreased at higher concentrations. Conclusion: RVS is regarded to inhibit the expression of MMP and TIMP as well as iNOS and COX-2 gene expression via directly inhibiting the activation of NF-𝜅B and phosphorylation of MAPK pathway in THP-1 cells. This suggests RVS have potential to be used as a therapeutic agent for acute myeloid leukemia (AML).

Resolvin D5, a Lipid Mediator, Inhibits Production of Interleukin-6 and CCL5 Via the ERK-NF-κB Signaling Pathway in Lipopolysaccharide-Stimulated THP-1 Cells

  • One of the omega-3 essential fatty acids, docosahexaenoic acid (DHA), is a significant constituent of the cell membrane and the precursor of several potent lipid mediators. These mediators are considered to be important in preventing or treating several diseases. Resolvin D5, an oxidized lipid mediator derived from DHA, has been known to exert anti-inflammatory effects. However, the detailed mechanism underlying these effects has not yet been elucidated in human monocytic THP-1 cells. In the present study, we investigated the effects of resolvin D5 on inflammation-related signaling pathways, including the extracellular signal-regulated kinase (ERK)-nuclear factor (NF)-κB signaling pathway. Resolvin D5 downregulated the production of interleukin (IL)-6 and chemokine (C-C motif) ligand 5 (CCL5). Additionally, these inhibitory effects were found to be modulated by mitogen-activated protein kinase (MAPK) and NF-κB in lipopolysaccharide (LPS)-treated THP-1 cells. Resolvin D5 inhibited the LPS-stimulated phosphorylation of ERK and translocation of p65 and p50 into the nucleus, resulting in the inhibition of IL-6 and CCL5 production. These results revealed that resolvin D5 exerts anti-inflammatory effects in LPS-treated THP-1 cells by regulating the phosphorylation of ERK and nuclear translocation of NF-κB.

Inhibitory Effects of Zerumbone on MCP-1-Induced THP-1 Migration (MCP-1에 의해 유도된 THP-1 유주에 미치는 Zerumbone의 영향)

  • This study examined the effects of zerumbone on monocyte migration. Monocytes are recognized as important mediators of various inflammatory diseases, and the possibility of controlling inflammatory diseases by regulating the monocyte functions, such as activity and mobility, has been reported. MCP-1, which is a chemokine with levels that increase upon inflammation, causes the migration of the monocyte cell line, THP-1. Migration occurred at a concentration of 10 ng/mL MCP-1, and the highest migration occurred at 100 ng/mL and 200 ng/mL. MCP-1-induced THP-1 migration decreased by more than 40% in the presence of zerumbone. The concentration of cAMP, an important secondary messenger of the CCR2 signaling pathway, the MCP-1 receptor, was increased in the culture medium after a zerumbone treatment. The concentrations of cAMP decreased significantly under the MCP-1 treatment condition only. On the other hand, an increase in cAMP was observed when zerumbone and MCP-1 were treated simultaneously. Erk phosphorylation induced by an MCP-1 treatment was also found to decrease with the zerumbone treatment. This study introduces the possibility of controlling inflammatory diseases through the function of zerumbone, which regulates the migration of monocytes.

The Effects of Rheum palmatum(RHP) Extract on the the Alzheimer's Disease Model (대황(大黃)이 Alzheimer's Disease 병태(病態) 모델에 미치는 영향(影響))

  • This experiment was designed to investigate the effect of Rheum palmatum(RHP) on the Alzheimer's disease. The effects of RHP extract on amyloid precursor proteins(APP), acetylcholinesterase(AChE), glial fibrillary acidic protein(GFAP) mRNA of PC-12 cell treated by $A{\beta}\;and\;IL-1{\beta},\;IL-6,\;TNF-{\alpha}$ mRNA of THP-1 cell treated by LPS and AChE activity of PC-12 cell lysate treated by $A {\beta}$and behavior of memory deficit rats induced by scopolamine and mice glucose, uric acid, AChE activity of memory deficit rats induced by scopolamine were investigated, respectively. The results were summarized as follows ; 1. RHP extract suppressed APP, AChE, GFAP mRNA in PC-12 cell treated by $A{\beta} $. 2. RHP extract suppressed $IL-1{\beta} $, IL-6 $TNF-{\alpha}$ mRNA in THP-1 cell treated by LPS. 3. RHP extract suppressed AChE activity in cell lysate of PC-12 cell treated by $A{\beta}$. 4. HP extract increased glucose, decreased uric acid and AChE significantly in the serum of the memory deficit rats induced by scopolamine. 5. RHP extract group showed significantly inhibitory effect on the memory deficit of mice induced by scopolamine in the experiment of Morris water maze. According to the above results, it is suggested that RHP extract might be usefully applied for prevention and treatment of Alzheimer’s disease and memory deficit symptom.

  • Four quinolone alkaloids were isolated by bioactivity-guided fractionation from the methanol extracts of Evodiae fructus fruits. Structures of compounds were elucidated by spectroscopic analysis ($^1H$-, $^{13}$C-NMR and MS), as follows: 1-methyl-2-undecyl-4(1H)-quinolone (1), evocarpine (2), dihydroevocarpine (3) and mixture of [1-methyl-2-[(Z)-10-pentadecenyl]-4(1H)-quinolone and 1-methyl-2-[(Z)-6-pentadecenyl]-4(1H)-quinolone] (4). They inhibited the interaction of sICAM-1 and LFA-1 in THP-1 cells at $IC_{50}$ values of >150 (1), 109.8 (2), >150 (3) and $40.9 {\mu}M$ (4), respectively,

Anti-Inflammatory Effects of Ethyl Acetate Fraction from Cnidium officinale Makino on LPS-Stimulated RAW 264.7 and THP-1 Cells

  • This work aimed to elucidate the anti-inflammatory effects of ethyl acetate fraction from Cnidium officinale Makino with a cellular system of LPS-stimulated RAW 264.7 and THP-1 cells. Some key pro-inflammatory cytokines and mediators including NO, iNOS, $PGE_2$, COX-2, TNF-${\alpha}$, NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65 were studied by sandwich ELISA and western blot analysis. Ethyl acetate fraction could significantly inhibit the production of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 in LPS-stimulated cell than that of single LPS-stimulated. And ethyl acetate fraction suppresses the activation of NF-${\kappa}B$ p50 and NF-${\kappa}B$ p65. All the results showed that ethyl acetate fraction had a good anti-inflammatory effect on LPS-stimulated RAW264.7 and THP-1 cells. Taken together, the anti-inflammatory actions of ethyl acetate fraction from Cnidium officinale Makino might be due to the down-regulation of NO, $PGE_2$, TNF-${\alpha}$, iNOS and COX-2 via the suppression of NF-${\kappa}B$ activation.

Different invasion efficiencies of Brucella abortus wild-type and mutants in RAW 264.7 and THP-1 phagocytic cells and HeLa non-phagocytic cells

  • Brucellosis is one of the common zoonoses caused by Brucella abortus (B. abortus). However, little has been reported on factors affecting invasion of B. abortus into host cells. To investigate cell-type dependent invasion of B. abortus, phagocytic RAW 264.7 and THP-1 cells and non-phagocytic HeLa cells were infected with wild-type and mutant B. abortus, and their invasion efficiencies were compared. The invasion efficiencies of the strains were cell-type dependent. Wild-type B. abortus invasion efficiency was greater in phagocytic cells than in epithelial cells. The results also indicated that there are different factors involved in the invasion of B. abortus into phagocytic cells.

Characterization of Matrix Metalloproteinase Expression in Triglyceride Treated THP-1 Macrophages

  • Elevated blood triglyceride (TG) levels correlate with development of atherosclerosis suggesting that TG may promote the development of this disease. During atherosclerosis, TG is taken up by tissue macrophages which result in dramatic changes in various secreted factors. One such factor is the family of matrix metalloproteases (MMP) which are involved in tissue remodeling during both physiological and pathological processes. In this study, we examined the MMP expression profile in PMA-differentiated THP-1 macrophages treated with TG. We found that TG-treated THP-1 macrophages showed decreased expression of MMP-3, MMP-7, MMP-8 and MMP-9 in a time- and dose-dependent manner. In contrast, expression of MMP-1, MMP-2, and MMP-10 remained relatively unchanged after TG treatment. In addition, we found that expression of select MMPs was affected by various inhibitors of signaling pathways. In particular, expression of MMP-3 was slightly recovered by cRAF and PLC signaling pathway inhibitors. These data suggests a possible role of MMPs in macrophages during TG-induced atherosclerosis.

The Experimental Study about Antioxidant Activities of Alismatis Rhizoma Herbal Acupuncture (택사약침(澤瀉藥鍼)의 항산화효과(抗酸化效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • Objective : The purpose of this study is to investigate the antioxidant activities of Alismatis Rhizoma herbal acupuncture by experimental methods. Methods : For this purpose, first, we put an emphasis in the control of enzymes of the antioxidant system in various changes inside the cell; these changes caused by the proliferation or the activation of the cell which were brought about by the handling of PMA and $TNF-{\alpha}$ into the THP-1 monocyte cell of the body each other. After that, we caused the acute oxidant symptom by the injection of AAPH into the mouse' abdominal cavity, and then applied the herbal acupuncture on S36 point(足三里), and finally, we measured the change of blood ingredient and the resistance against the activated oxygen of the red blood cell membrane, MDA, SOD, and catalase. Results : In vitro the revelation of $IL-1{\beta}$, IL-8, $TNF-{\alpha}$, NOS II and IL-6 were decreased and the revelation of IL-10, $TGF-{\beta}$, GM-CSFIL-12, GM-CSF and SOD were increased. The DNA-binding of $NF-{\kappa}B$ and AP-1 were activated and the formation of ROS in the THP-1 cell line was decreased. In vivo $IL-1{\beta}$ among producing the cytokine inside the plasma was meaningfully dwindled and the $INF-{\gamma}$ was meaningfully increased. The resistance of red blood cell membrane against the activated oxygen was meaningfully increased and the MDA formation was meaningfully dwindled, In the activation of hepatic antioxidase, the SOD was meaningfully increased. Conclusion : Alismatis Rhizoma herbal acupuncture by experimental methods has effected on the antioxidant activities.
