The presence of considerable amount of enzymes of TCA cycle isocitrate dehydrogenase (ICDH-NADP+, EC1.1.1.42), $\alpha$-ketogluterate dehydrogenase ($\alpha$-KGD, EC1.2.4.2) and malate dehydrogenase (MDH, EC1.1.1.37) in fresh control and in vitro starved adult Isoparorchis hypselobagri establish the functional TCA cycle in this fluke. The major metabolic end products are pyruvate, lactate, oxaloacetate and malate. The ratio of oxaloacetate/malate assess that oxaloacetate is reduced to malate and in this fluke the reverse TCA cycle is active. The pyruvate/lactate ratio shows pyruvate is reduced to lactate and the fluke is homolactate farmenters.
A flux determination methodology has been built which enables to develop constrained stoichiometric relationships and metabolic balances. The analysis differs from those developed for anaerobic growth conditions in that cell mass formation is a significant sink for carbon. When combined with experimental measurements, a determined system of equations results yielded tricarboxylic acid (TCA) cycle and glycolytic fluxes. The methodology was implemented to determine the fluxes of E. coli B/r and K12, and it was found that as the growth rate in a glucose minimal medium increased, the cells became increasing glycolytic and the TCA fluxes either leveled off or declined. The pattern identified for the TCA fluxes corresponded to ${\alpha}$-ketoglutarate dehydrogenase's induction-repression pattern, thereby suggesting that the induction-repression of the enzyme could result in significant flux changes. When the minimum flux solution was contrasted to the glycolytic and TCA fluxes determined, two observations were made. First, the minimum flux could provide the cell's biosynthetic ATP requirements. Second, at a high growth rate in a glucose medium, the excess glycolytic flux exceeded that of the TCA cycle, which appeared to more closely match the biosynthetic needs.
바실러스 서브틸리스 fsrA 유전자는 대장균의 ryhB 유전자와 유사한 역할을 하는 유전자로 철 조절 유전자인 fur 유전자의 조절을 받는다. 철의 농도가 높을 때에는 ryhB 유전자의 전사가 fur에 의해 억제되지만 철의 농도가 낮아지면 세포내의 철을 보다 효율적으로 절약하기 위하여 ryhB의 전사 억제가 풀려 ryhB small RNA가 생성되고 이는 철을 함유하는 sdhCDAB (succinate dehydrogenase) 유전자의 발현을 억제한다. 본 연구에서는 바실러스에서 이에 상응하는 유전자인 fur와 fsrA의 결실 균주들을 대상으로 여러 TCA 회로의 유기산을 탄소원에서의 이들 균주들의 성장을 측정하였다. 실험 결과 대장균의 fur/ryhB와 마찬가지로 succinate를 탄소원으로 할 경우 바실러스 fur 결실 균주는 성장이 매우 적었지만 fur/fsrA 결실 균주는 정상적으로 성장하였다. 또한 succinate 이외에 citrate, fumarate의 경우도 succinate와 유사한 결과를 보이는 반면에 malate의 경우 fur나 fur/fsrA의 결실 균주들에서 성장이 큰 차이를 보이지 않았다. 이 결과 TCA 회로에서 succinate 상부에 해당하는 유전자들은 fsrA에 의해 억제되나 succinate 하부에 해당하는 유전자들은 fsrA에 의해 억제되지 않는 것으로 생각된다.
목적: 이전 연구에서 치은섬유아세포 혹은 성견 구개 연조직에 trichloroacetic acid (TCA)를 적용하는 것이 세포주기진행 유전자 발현의 변화를 일으키는 것으로 밝혀졌다. 이에 따라 본 연구에서는, hydrophobically modified glycol chitosan (HGC)기반의 나노방출제어시스템을 이용한 TCA 및 상피세포성장인자(EGF)의 순차적 방출시스템에서 이 효과를 검증하기 위하여 다양한 세포주기진행 유전자들의 발현을 규명하였다. 재료 및 방법: TCA와 EGF를 담지하는 HGC기반 나노방출제어시스템을 제작하였다. 실험군은 대조군(CON); TCA-담지형 나노방출제어시스템 투여군(EXP1); TCA- 및 EGF-담지형 나노방출제어시스템 투여군(EXP2)으로 정의되었다. 24시간 및 48시간 배양 시 37개 세포주기 유전자들의 발현을 분석하였다. 영향인자로서의 유전자 및 상관관계에 대해서도 분석하였다. 결과: Cyclins (CCNDs), cell division cycles (CDCs), cyclin-dependent kinases (CDKs), E2F transcription factors (E2Fs), extracellular signal-regulated kinases (ERKs)와 같은 다수의 유전자들과 기타 다른 세포주기 유전자들의 발현이 EXP1과 EXP2에서 상향조절되었다. E2F4, E2F5, GADD45G와 같은 세포주기차단 유전자들의 발현도 상향조절되었으나, 또다른 세포주기차단 유전자인 SMAD4의 발현은 하향조절되었다. 다중회귀분석에서 CCNA2, CDK4 그리고 ANAPC4가 ERK 유전자 발현에 가장 영향력 있는 유전자로 선정되었다. 결론: HGC기반 순차적 나노방출제어시스템을 이용한 TCA 및 EGF의 적용은 다양한 세포주기진행 유전자들의 발현을 상향조절함이 밝혀졌고, 이를 토대로 한 구강연조직증대시스템 개발의 가능성이 확보되었다.
본 연구는 서울시 오피스 건물을 대상으로 친환경 건축물 인증등급이 오피스의 가격과 비용에 미치는 영향을 분석하였다. 이를 위해 건축물들의 매매가격을 다중회귀 분석을 통해 규명하였다. 또한 친환경 인증등급이 높은 경우에 어느 정도의 환경부하비용이 저감되는지를 분석하기 위해 LCC(Life Cycle Cost) + LCA(Life Cycle Assessment) 분석을 실시하였다. 실증분석 결과, 친환경 인증등급이 높을수록 업무용 오피스 가격에 유의한 양의 영향을 준 것으로 나타났다. 또한 LCC 분석에서는, 친환경 인증등급을 높게 받은 건물이 그렇지 않은 건물보다 적은 생애비용이 드는 것으로 나타났다. 또한 친환경 인증등급을 높게 받은 오피스 건물이 그렇지 않은 건물보다 환경부하비용(LCA)이 적게 든다는 것을 확인하였다. LCC와 LCA를 통합한 TCA 분석에서는 친환경 인증등급을 높게 받은 건물이 그렇지 않은 건물보다 적은 사회적 총비용을 나타냈다.

The capability of utilizing a TCA cycle intermediates as the sole carbon source by the multi-vitamin auxotrophic yeast Torulopsis glabrata CCTCC M202019 was demonstrated with plate count method. It is indicated that T. glabrata could grew on a medium with one of the TCA cycle intermediates as the sole carbon source, but more colonies were observed when glucose, acetate and one of the TCA cycle intermediates coexisted in the medium. Among the intermediates of the TCA cycle examined in this study, cell growth was improved by supplementing oxaloacetate. Further investigation showed that the presence of acetate was necessary when oxaloacetate was supplemented. By supplementing with 10 g/L of oxaloacetate in pyruvate batch fermentation, dry cell weight increased from 11.8 g/L to 13.6 g/L, and pyruvate productivity was enhanced from $0.96\;gL^{-1}h^{-1}\;to\;1.19 gL^{-1}h^{-1}$ after cultivation of 56 h. The yield of pyruvate to glucose was also improved from 0.63 g/g to 0.66 g/g. These results indicate that under vitamins limitation, the productivity and yield of pyruvate could be enhanced via an increase of cell growth by the supplementation of oxaloacetate.
Almost all of the aerobic organisms contain citric acid cycle (or, tricarboxylic acid cycle). This cycle is involved both in energy metabolism and biosynthetic reactions; generation of NADH which derives the synthesis of chemical energy, ATP, and provision of intermediates needed for the biosynthesis. Because of its importance in the cellular metabolism, the regulation of the TCA cycle and its component enzymes has been extensively studied by many biologists (7,28). Citrate synthase is resposible for the initial step of the cycle and has been recognized as the rate limiting step (14,121,26). Understanding of the mechanism of the expression of citrate synthase should be a key step for the elucidation of the regulation of the TCA cycle in the cell metabolism.
The utilization of acetate by Dunaliella tertiolecta was examined, and the detections and assays of the enzymes of the tricarboxylic acid cycle and the glyoxylate pathway were described. Acetate could not be utilized as a sole carbon source for the growth. The carboxyl carbon of acetate was incorporated more rapidly into CO2 than the methyl carbon. It was identified that malate, succinate, citrate and etc., were accumulated whne [U-14C] acetate was supplied to the cell free homogenate. The following enzyme activities were measured; acetothiokinase, isocitrate dehydrogenase, fumarase, malate dehydrogenase and aconitase. Though isocitratase, malate synthetase, succinate dehydrogenase and oxoglutarate dehydrogenase could not be detected, 14C from succinate was easily contributed to CO2 and cell component. The evidence suggested that the glyoxylate pathway was not operative and showed that the TCA cycle was the all important pathway in the oxidation of acetate to CO2 in Dunaliella.
A novel psychrotolerant Psychrobacillus strain PB01, isolated from an Antarctic iceberg, was comparatively analyzed with five related strains. The complete genome of strain PB01 consists of a single circular chromosome (4.3 Mb) and a plasmid (19 Kb). As potential low-temperature adaptation strategies, strain PB01 has four genes encoding cold-shock proteins, two genes encoding DEAD-box RNA helicases, and eight genes encoding transporters for glycine betaine, which can serve as a cryoprotectant, on the genome. The pan-genome structure of the six Psychrobacillus strains suggests that strain PB01 might have evolved to adapt to extreme environments by changing its genome content to gain higher capacity for DNA repair, translation, and membrane transport. Notably, strain PB01 possesses a complete TCA cycle consisting of eight enzymes as well as three additional Helicobacter pylori-type enzymes: ferredoxin-dependent 2-oxoglutarate synthase, succinyl-CoA/acetoacetyl-CoA transferase, and malate/quinone oxidoreductase. The co-existence of the genes for TCA cycle enzymes has also been identified in the other five Psychrobacillus strains.
