무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. Effects of Normal Diet with or without Naringin Supplement Following Ethanol Diet on Changes in Lipid Profiles and Antioxidant Enzyme Activities in Rats

  • This study was performed to investigate the effect of normal diet with or without naringin supplement on the lipid and antioxidant metabolism in ethanol-treated rats for a short tenn. Male Sprague-Dawley rats were divided into three groups (n=10), which were assigned to one of three dietary categories : $E_8$ : ethanol diet for 8 wks, $E_4N_4$ : ethanol diet for the first 4 wks and normal diet for the last 4 wks, $E_4Nna_4$ : ethanol diet for the first 4 wks and normal diet with naringin supplement for the last 4 wks. Plasma total cholesterol concentrations were significantly higher in ethanol fed rats for 8 weeks. The HDL-C/total-C ratios of the $E_4N_4$ and the $E_4Nna_4$ groups were significantly higher than that of the $E_8$ group, while the atherogenic index was lower in the $E_4N_4$ and the $E_4Nna_4$ groups than in the $E_8$ group. The $E_4N_4$ and $E_4Nna_4$ diets significantly lowered both the hepatic cholesterol and triglyceride levels compared to the $E_8$ group. Accumulation of hepatic lipid droplets was observed to be the highest in the $E_8$ group. In the current study, the naringin supplement to normal diet significantly lowered both the hepatic HMG-CoA reductase and ACAT activities in ethanol pre-treated rats for 4 weeks. Antioxidant enzyme activities were also upregulated when ethanol feeding was ceased. Naringin supplement given for 4 weeks after ethanol cessation resulted in a significant decrease in the plasma cholesterol and hepatic lipids and plasma TBARS as well as the hepatic HMG-CoA reductase and ACAT activities compared to the rats given ethanol diet for the entire 8 weeks. Replacement of normal diet following a short tenn ethanol feeding was effective for the recovery of ethanol-induced fatty liver and for normalizing plasma and hepatic lipid profiles and antioxidant enzyme activities, regardless of an additional phytochemical supplement, naringin. The effect of naringin could seemingly be more evident if its supplementation period had been extended longer than 4 weeks after ethanol cessation.

Rue(Ruta graveolens L.) 추출물의 Helicobacter pylori에 대한 항균활성과 생리활성효과 (Inhibition against Helicobacter pylori and Biological Activities by Rue (Ruta graveolens L.) Extracts)

  • Rue잎을 물과 에탄올로 추출하여 항균활성과 생리활성 효과를 탐색하였다. 그 결과 각 추출물의 총페놀 함량은 물 추출물에서 16.39 mg/g 알콜 추출물에서 17.07 mg/g로 나타났다. Helicobacter pylori에 대한 disc 방법에서 $200\;{\mu}g/mL$의 폴리페놀 첨가 시 물과 알콜 추출물은 각각 12 mm, 13 mm의 직경을 나타내어 항균활성이 있음을 알 수 있었으며, 2-fold dilution 방법으로 항균활성 검증 결과 물 추출물보다 알콜 추출물에서 더 높은 항균활성을 나타내었다. 물과 알콜 각 추출물의 ABTS radical decolorlization과 antioxidant protection factor(PF)를 살펴 본 결과 ABTS는 물 추출물이 약 $96\%$로 높은 저해율을 나타내었으며 PF는 알콜 추출물에서 1.2로 물 추출물 0.8보다 높게 나타났다. DPPH에 대한 전자공여능은 물 추출물이 89.38로 알콜 추출물 $86.90\%$보다 높게 나타났으며 활성 산소중 지방산화를 일으키는데 주요한 역할을 하는 hydroxyl radical에 대한 각 추출물의 영향은 알콜 추출물이 binding능력이 탁월하여 물 추출물에 비해 낮은 TBARS 값을 나타내었다. Angiotensin converting enzyme과 xanthine oxidase 저 해 활성은 알콜 추출물이 물 추출물보다 더 높게 나타났고, HPLC에 의한 페놀 분석 결과 rosemarinic acid의 함량이 알콜 추출물에서 가장 많았으며 다음으로 qurcetin 함량이 높게 나타났다.

Effects of Green Tea Catechin on Mixed Function Oxidase System and Antioxidative Defense System in Rat Lung Exposed to Microwave

  • The purpose of this study was to investigate the effects of green tea catechin on mixed function oxidase system (MFO), lipofuscin contents, carbonyl value, oxidative damage and the antioxidative defense system in lung of microwave exposed rats. Experimental groups were divided to normal group and microwave exposed group. The microwave exposed groups were subdivided into three groups: catechin free diet (MW-0C) group, 0.25% catechin (MW-0.25C) group and 0.5 % catechin (MW-0.5C) group according to the levels of dietary catechin supplementation. The rats were irradiated with microwave at frequency of 2.45 GHz for 15 min. Experimental animals were sacrificed at 6th day after microwave irradiation. The contents of cytochrome P$_{450}$ contents in MW-0C group was increased to 95% , compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 16% and 31%, respectively, compared with MW-0C group. The activity of NADPH-cytochrome P$_{450}$ reductase in MW-0C group was increased to 44%, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 12% and 17%, respectively, compared with MW-0C group. The activity of superoxide dismutase (SOD) in MW-0C group was decreased to 21 %, compared with normal group. MW-0.25C and MW-0.5C group were significantly (p < 0.05) increased, compared with MW-0C group. The activity of glutathione peroxidase (GSHpx) in MW-0C group was significantly decreased, compared with normal group. MW-0.25C and MW-0.5C groups were recovered to the level of normal group. The thiobarbituric acid reactive substances (TBARS) content in MW-0C group was increased to 34 %, compared with normal group. Catechin supplementation groups were maintained the level of normal group. The levels of caybonyl value in MW-0C group was increased to 21 %, compared with normal group. MW-0.25C and MW-0.5C groups were reduced to 14% and 12%, respectively, compared with MW-0C group. The lipofuscin contents in MW-0C group were increased to 23.4 %, compared with normal group. That of MW-0.5C group was significantly reduced, compared with MW-0C group. In conclusion, MFO system was activated and the formation of oxidized protein, lipofuscin was increased and antioxidative defense system was weakened of lung tissue in microwave exposed rats, thus oxidative damage was increased. But it was rapidly recovered to normal level by green tea catechin supplementation.n.

  • This study was investigated the effect of gamma irradiation (GI) on the shelf-life and quality improvement of squid sundae (a traditional Korean sausage) stored at room temperature after 0 (control), 10 and 20 kGy doses of irradiation. The total viable cells decreased with increasing irradiation dose, and no viable cells were detected with the doses of 20 kGy. There were no significant changes in pH compared to the control. The volatile basic nitrogen (VBN) and thiobarbituric acid reactive substance (TBARS) values of GI (20 kGy) squid sundae were significantly lower than those of the control. In addition, the induction period for the GI (20 kGy) squid sundae measured by the Rancimat method was higher level than that of the control. In a sensory evaluation, there were no significant differences between the control and GI samples during storage.

산사(山査)가 식이성 고지혈증 흰쥐의 혈청지질에 미치는 영향 (Effect of CRATAEGII FRUCTUS(山査) on Sarum Lipid in High Cholesterol Diet Induced Hyperlipide Rats)

  • Objectives: CRATAEGII FRUCTUS(山査) is known as the substance which decreases concentration of sarum lipid. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on decreasing concentration of sarum lipid which are triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol in rat high cholesterol diet inducede hyperlipide rats. Methods: Sprague-Dawley rats were divided into 3 groups, Normal group(supplied enough water and feeds only. Normal Group), high fat diet administered group(AIN-76 high fat diet #100496 used beef tallow for 4 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group(AIN-76 high fat diet #100496 used beef tallow and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 4 weeks. SS Group). Rats were sacrificed and concentration of triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol were measured in rat's sarum lipid Results: There was no significant difference in the level of waight between each experimental group(F=0.243, p=0.787, ANOVA test), Concentration of triglyceride has no significant difference in each group(F=0.210, p=0.813, ANOVA test). There was significant increase of total cholesterol in the Control group compared to those of Normal but there was no significant difference in the SS group compared to the Control group(F=5.237, p=0.0l9, ANOVA test). There was a significant increase of HDL-cholesterol in the SS group compared to the Control group and decrease in Control group compared Normal group(F= 11.437, p=0.001, ANDYA test). Concentration of LDL -cholesterol was significantly different in each group(F= 16.377, p=0.0001, ANDYA test). It was significantly increased in the Control group compared with the Normal group and it has no difference in the SS group compared to the control group. TBARS values(Thiobarbituric Acid Reactive Substances) were significantly different in each group(F=5.242, p=0.0019, ANDYA test). It was significantly increased in the Control group compared with the Normal group and significantly decreased in SS group compared with the Control group. Conclusions: According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in decreaseing concentration of sarum lipid in high cholesterol diet induced hyperlipidemic rats.

소리쟁이 추출물의 항산화 활성과 이를 첨가한 양념돈육의 품질특성 (Antioxidant Activities of Rumex crispus Extracts and Effects on Quality Characteristics of Seasoned Pork)

  • 소리쟁이 주정추출물은 총 폴리페놀 및 총 플라보노이드 함량이 각각 $108.24{\pm}3.29\;{\mu}g/mg$, $33.06{\pm}2.24\;{\mu}g/mg$이었고, 환원제로 작용하며 DPPH 소거활성을 가지는 높은 항산화능을 보였다. 소리쟁이 추출물을 첨가한 양념소스에서도 시료첨가량이 증가할수록 DPPH 소거능이 증가함을 확인하였다. 또한 소리쟁이 추출물을 첨가한 양념 돈육을 제조하여 $4^{\circ}C$에서 21일간 저장하면서 품질특성을 검토한 결과, 소리쟁이 첨가 양념돈육은 0.1% BHA 처리구와 유사하게 대조구에 비해 총세균수 및 지방산패도가 감소되었으며, 대장균군은 검출되지 않았다. 특히 소리쟁이 추출물을 1% 첨가한 양념돈육은 대조구보다 냄새, 다즙성, 연도에서 높은 관능값을 보였다. 따라서 우수한 항산화능을 가진 소리쟁이 추출물은 양념 돈육에서 저장성 및 품질특성의 개선에 도움을 줄 수 있는 소재라고 여겨진다.

수영부하 피로에 대한 마늘추출물 및 비타민 B군 강화 급이의 회복 효과 (Recovery Effect of Garlic Extract and Vitamin B group Enhanced Diet on Swimming Fatigue)

  • 마늘추출물 및 비타민 B군 강화 식이를 4주간 급이한 후 흰쥐에 수영부하 실험으로 스트레스를 가하였을 때, 피로회복 인자와 관련된 흰쥐의 체내 지질성분 및 간 기능에 미치는 영향을 분석하였다. 혈당은 대조군($193.41{\pm}3.85$ mg/dl)보다 실험식이군이 $152.85{\pm}6.57{\sim}169.93{\pm}8.75$ mg/dl로 유의적으로 낮았다. 혈청 중 총 지질, 총 콜레스테롤 및 LDL-콜레스테롤은 대조군에 비해 낮은 경향이었으며 마늘추출물과 비타민 B군 강화 시료를 각각 첨가한 SG군과 SV군 보다는 이들을 혼합 급이한 SGV군에서 유의적으로 더 낮았다. 혈청 중의 TBARS 함량은 정상군에 비해 대조군에서 17.25%나 증가하였으며 마늘추출물 및 비타민 B군 강화 급이로 낮아지는 경향이었다. 간조직의 지질 성분 중 총지질은 정상군에 비해 SGV군이 10% 더 낮은 함량이었고, 중성지질은 대조군에 비해 SGV군에서 46% 더 낮게 정량 되었다. 수영부하 스트레스에 의한 간조직 중의 글리코겐 함량은 대조군이 정상군보다 유의적으로 낮았으며 SG, SV 및 SGV 군은 정상군과 유의차가 없었다. 이상의 결과를 종합하여 볼 때 마늘추출물과 비타민 B군 강화급이는 혈중 혈당, 총콜레스테롤 및 LDL-콜레스테롤을 유의적으로 감소시켰으며 또한 수영부하 스트레스로 혈중 및 간장에 다량 생성될 수 있는 지질과산화물 함량도 유의적으로 감소시키므로 피로회복 및 스트레스 개선에 효과가 있을 것으로 판단된다.

녹차, 메밀, 포도잎 추출물이 고콜레스테롤 급여 흰쥐의 지질대사, TBARS 및 혈액응고에 미치는 영향 (Effects of Green Tea, Buckwheat and Grape Leaves Extracts on Lipid Metabolism, Antioxidative Capacity, and Antithrombotic Activity in Rats Fed High Cholesterol Diets)

  • 본 연구에서는 Sprague Dawley 종 수컷 흰쥐에게 콜레스테롤이 1% 함유된 사료를 급여하여 고지혈증을 유발시키면서 녹차추출물, 메밀추출물, 포도잎추출물 추출물을 500 mg/kg B.W.의 농도로 4주간 매일 같은 시각에 경구투여하여 흰쥐의 혈액과 간장 내의 지질조성과 혈액응고기전 및 체내 지질과산화에 미치는 영향을 관찰하여 다음과 같은 결과를 얻었다. 전군에서 식이섭취량과 체중증가량 및 식이효율은 유의한 차이가 없어서 실험식이가 정상적인 성장에 영향을 미치지 않았다. 녹차추출물투여군은 고콜레스테롤 급여 대조군에 비해서 흰쥐의 꼬리에서의 출혈시간을 증가시켜서 항혈전효과가 있었다. 고콜레스테롤 급여로 인해 흰쥐에게 고지혈증이 유발되었으며 실험 시료를 경구투여하는데 따른 혈액중의 지질대사 개선효과는 없었다. 고콜레스테롤 급여로 흰쥐의 간장은 비대해지고 간장내 중성지방과 총콜레스테롤 함량도 현저히 증가하였으며 녹차추출물, 메밀추출물, 포도잎추출물은 간장 내 총콜레스테롤 함량을 감소시켜 간장 내 지질대사의 개선효과가 있었다. 고콜레스테롤의 급여는 혈장 내의 지질과산화물의 함량을 증가시키며 포도잎추출물은 체내 지질과산화를 억제시키는 효과가 있었으며 간에서는 메밀과 포도잎추출물이 지질과산화를 억제하였다.

모델시스템과 가열우육에서 옻나무 추출물의 항산화 효과 (Antioxidant Activity of Rhus verniciflua Stokes Extract in Model Systems and Cooked Beef)

  • 본 연구는 모델시스템과 가열우육에서 옻나무 추출물의 항산화 효과를 구명함으로서 육제품에 이용 가능성을 검토하고자 실시하였다. DPPH에 대한 소거력$(\%)$은 옻나무 에탄올 추출물을 1, 10, 100, 1,000 ppm으로 증가시킴에 따라 14.79, 75.08, 82.02, $83.97\%$로 증가하였다(p<0.05). 옻나무 에탄올 추출물(10 ppm)을 liposome과 육균질물에 첨가하면 대조구보다 높은 항산화 효과를 나타냈으며, 단독 첨가보다 $\alpha-tocopherol$(2 ppm) 혼용 첨가가 억제율이 증가되었다. 육균질물은 pH 6.0에서, liposome은 pH $5.0\~6.0$에서 최대 산화 억제율을 보였다. 또한 옻나무 열수 추출물의 첨가로 인해 가열 우육의 냉장저장 중 TBARS와 POV가 대조구보다 낮은 수준으로 유지되었으며(p<0.05), 소금을 첨가하여 산화를 촉진시켰어도 강력한 항산화 효과를 나타내었다. 따라서 육제품 제조 시 옻나무 추출물이 천연 항산화제로서 이용 가능성이 있다고 판단된다.
