어성초추출물투여가 흰쥐의 간장과 혈청의 지질 및 과산화지질함량에미치는 효과 (Effects of Extracts of Houttuynia cordata Thunb on the Level of Lipids and Lipid Peroxidation in the Liver and Serum of Cholesterol-fed Rats)

  • The feeding effects of Herba hottuynia extracts on the lipids profiles and the content of TAB-reactive substances were evaluated in rats fed a high cholesterol diet. Hot water or ethanol extracts of the dried leave were evaporated and lyophilized . The experimental animals wer edevided to four groups and fed the following diets for 4 weeks : BAsal (cholesterol-free diet), CHOL(cholesterol-enriched diet),CW (cholesterol plus water extract) and CE(cholesterol plus ethanol extract). Dietary cholesterol increased significantly the activities of serum GOT and GPT, but the extracts feeding (0.5% of diet) did not influence the activities induced by dietary cholesterol. Although dietary cholesterol increased significantly the concentrationof serum andliver cholesterol, it tended to decrease the concentation of serum triglycerides. CHolesterol feeding had a lowering effect on the lipid peroxidation value of serum, but not inliver.Furthermore, the extracts feeding, especially water extract, decreased markedly the liver peroxidation value. The results suggest that Houttyunia cordata extracts have an in vivo antioxidant effect, judged from the TBA value in the liver rats fed a high cholesterol diet.

Evaluation of Interfering Substances in Routine Chemistry Tests Using Toshiba TBA-C8000 Chemistry Analyzer

  • In clinical chemistry tests, the interfering substances such as hemoglobin, lipid, bilirubin, and drugs, etc. can cause the changes of test results performed by spectrophotometrical methods. We evaluated the effects of interfering substances on the test results by adding interfering substances on the samples in the 19 kinds of clinical chemistry tests such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, total protein, albumin, glucose, total cholesterol, total bilirubin, triglyceride, uric acid, calcium, inorganic phosphours, high density lipoprotein cholesterol, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, and C-reactive protein using newly implemented automatic chemical analyzer Toshiba TBA-C8000 under the direction of CLSI EP07-A guideline. Hemolytic samples show increased concentration of total protein, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase and reduced concentration of total bilirubin, alkaline phosphatase by interfering effect. Hyperlipemic samples show increased concentration of total protein and alkaline phosphatase and reduced concentration of low density lipoprotein cholesterol. The samples with conjugated bilirubinemia show increased concentration of inorganic phosphours, otherwise the samples with unconjugated bilirubinemia show no interference or allowable range in the test result.

인삼의 9종 폴리아세틸렌으로 처리한 간소포체 중의 유리 말론알데히드의 HPLC에 의한 분석 (HPLC Analysis of Free Malonaldehyde in Nine Ginseng Polyacetylene-Treated Liver Microsome)

  • Free malonaldehyde was determined in nine kinds of ginseng polyacetylene-treated micro- some by HPLC analysis. Antioxidant activities of some phenolic compounds and ginseng saponin were also drtermined both by a new HVLC method and by THA method. A new HPLC system separaterl malonaldehyde at a retention time 5,6 min and showed a linear relationship between the peak are a and malonaldehyde concentration. Panaxnol showed the strongest activity among nine polyacetylenes and the addition of either chlorine or aletyl group reduced polyacetylene's own activity. Since C14-polyacetylenes such as panaxyne and panaxyne-epoxide had little or no antioxidant activities, S17-structure should be preserved to exert a radical-scvenging or trapping activity. The antioxidant activities of chlorogenic acid, ferulic acid and catechol were much weaker than those of C17-polyacetylenes. Ginseng saponin showed no antioxidant activity. Since TBA reactive substances and malonaldehyde contents were almost the same in peroxiedized microsome. TBA value seems a good indicator for lipid peroxidation in this particular Fe+3 ADP/NADPH system.

매실(Prunus mume)의 항산화성 (The Antioxidant Activity of Maesil (Prunus mume))

  • 매실의 항산화력을 알아보기 위해 매실과육과 매실즙 및 이들의 수용성 추출물을 시료로 하여 과산화물가, TBA가 및 전자공여능을 측정하였다. 과산화물가는 저장 4주 후 매실과즙 추출물의 경우 226.55 meq/kg으로 항산화제로 알려져 있는 BHT(217.38meq/kg) 및 ascorbic acid(223.32meq/kg)와 유사한 값을 나타내 이들과 유사한 항산화력을 갖는 것으로 분석되었다. TBA가를 통해 항산화력을 측정한 결과 합성산화제인 BHT가 45.35%로 가장 높은 항산화력을 보였고 매실과즙 추출물은 25.0%,로 29.94%를 나타낸 ascorbic acid와 유사한 수준의 값을 나타내었다. DPPH를 이용한 전자공여능을 측정한 결과 0.01% 매실과육 추출물은 34.84%, 0.01% 매실과즙 추출물 43.50%를 보였으며 0.02% 매실과육 추출물은 53.21%, 0.02% 매실과즙 추출물은 59.19%로 증가하는 양상을 나타내었으나 BHT(77.82%)와 ascorbic acid(96.69%)에 비해 낮은 활성을 나타내었다. 이상의 결과로 볼 때 매실과즙 추출물의 경우 BHT와 ascorbic acid와 유사한 항산화성을 나타내 천연 항산화제로의 이용 가능성이 있다고 생각되어진다.

화학발광분석기를 이용한 동결건조 난황분말의 산패도 측정법 (A Method for Measuring Lipid Peroxidation of Freeze-dried Egg Yolk by Using Chemiluminescence Analyzer)

  • 본 연구는 TBARS 및 PV 등의 식품중의 지질산패도 측정법을 대체할 수 있는 간단한 측정법을 개발하고자 시행되었다. 이를 위해 선행 연구에서 식용유지의 산패도 측정법 대체가능성을 나타낸 화학발광도 측정법을 고체시료 중에서 축산식품인 동결건조 난황분말에 적용하여 기존의 산패도 측정법과의 상관관계를 조사하여 대체 가능성을 확인하였다. 암소의 $25^{\circ}C$에서 저장기간(0, 12, 24, 48, 72, 144시간) 별로 시료를 채취해 TBARS와 PV 및 화학발광도를 측정하여 비교하였다. 저장시간에 따른 화학발광도 측정 결과는 특히 과산화물가 측정치와 유의적으로 높은 상관관계를 나타내었으며($r^2=0.987$), 상대적으로 높지는 않지만($r^2=0.874$) TBA가와도 유의적으로 상관관계가 있다고 조사되었다. 또한 실제 표준시료를 이용한 분석 결과에서도 유의적으로 높은 상관관계($r^2=0.9214$, 0.9599)를 나타내었다. 따라서 화학발광도를 이용한 지질 산패도 측정법은 여러 식품 등의 지질 산패도 측정에 있어서 POV를 대체할 수 있을 것으로 기대가 되며, 경우에 따라서는 TBARS에 대해서도 대체가 가능할 것으로 기대된다.

Detection of Oxygen Species Generated from Ag2Se-Graphene Heterojunction Photocatalysts with Excellent Visible Light Driven Photocatalytic Performance

  • Reactive oxygen species (ROS) can be produced by interactions between sunlight and light-absorbing substances in natural water environments and can completely destroy various organic pollutants in waste water. In this study, we used graphene oxide modified $Ag_2Se$ nanoparticles to enhance photochemically generated oxygen (PGO) species activity. Surface area and pore volumes of the $Ag_2Se-graphene$ ($Ag_2Se-G$) samples showed catastrophic decrease due to deposition of $Ag_2Se$. The generation of reactive oxygen species was detected through the oxidation reaction of DPCI to DPCO. The photocurrent density and the PGO effect increase in the case of the use of modified graphene. The PGO effect of the graphene modified with $Ag_2Se$ composites increased significantly due to a synergetic effect between graphene and the $Ag_2Se$ nanoparticles. The photocatalytic activity of sample was evaluated by measuring the degradation of organic pollutants such as methylene blue (MB) and industrial dyes such as Texbrite BA-L (TBA) under visible light.

돼지고기에 대한 EM(Effective Microorganism) 함초 발효액의 항산화 효과 (Antioxidant Effect of Fermented Salicornia herbacea L. Liquid with EM (Effective Microorganism) on Pork)

  • EM 함초 발효액의 항산화 효과에 대한 연구를 수행하였다. 함초의 무기질 함량은Na의 경우 높은 함량을 나타났으며 K과 Mg의 경우 자생지에 관계없이 각 부위별로 거의 비슷한 함량을 보였으나, 동결 건조한 시료가 온풍 건조한 시료보다 훨씬 높은 함량을 나타내었다. Ca 함량은 뿌리보다 잎에서 2배 이상 높은 함량을 보였으며, Fe은 함초의 뿌리에 많이 함유되어 있었다. 해안에서 채취한 함초의 동결 건조시킨 잎이 20.19%로 가장 높은 함량을 보여 구운 소금의 약1/5에 미치는 식염 함량을 나타냈다. 함초의 뿌리 부분을 EM으로 발효시킨 EM 함초 발효액이 저장 14일에는 대조구 보다 5.3배 정도의 항산화 효과를 나타냈다.

돈육 지방에 미치는 함초(Saiicoma herbacea L.)의 항산화 효과 (Antioxidative Effect of Glasswort(Salocornia herbacea L.) on the Lipid Oxidation of Pork)

  • 염생식물인 함초는 우리나라에서 가능성 있는 기능성 식품소재의 알려져 있다. 본 연구는 돈육 지방에 대한 기능성 식품소재로서의 가능성을 알아보기 위해 실험을 수행하였다. 해안에서 채취한 함초를 부분적으로 잎, 줄기, 뿌리로 구분하여 항산화 효과를 비교하였다. 시료에 대한 항산화 효과는 TBA 방법을 사용하여 살펴보았다. 해안 함초의 경우 동격 건조한 잎(FLC)이 0일째에 BHT 다음으로 낮은 TBARS치를 나타내어 가장 항산화 효과가 좋았다. 그러나 저장 7일에는 온풍 건조한 잎(OLC)과 TBARS치가 큰 차이가 없었다. 저장 14일에는 온풍 건조한 잎(OLC)이 BHT 다음으로 높은 항산화 효과를 나타냈다. 결론적으로 해안에서 채취한 함초는 항산화 효과를 보였으며, 온풍 건조시킨 시료에서 잎 >뿌리 >줄기 순으로 돈육의 지방에 대한 항산화 효과를 나타냈다. 특히, 함초는 천일염(sea salt)과 죽염(bamboo salt)보다 2배이상의 항산화 효과를 보였다.

Antioxidative Activity of the Korean Wild Leafy Vegetables: Aster scarer mild Ligularia fischeri

  • The purpose of this study was to evaluate the antioxidative potential of Korean wild leafy vegetables in vivo as well as in vitro. Antioxidative activities of Aster scaber and Ligularia fischeri were evaluated against a reference of Spinacia oleracea Forty rats were fed either control diet or respective vegetable diets for four weeks. The level of thiobarbituric acid reactive substances (TBARS) and the activities of catalase and superoxide dismutase (SOD) in liver were compared. The plasma concentrations of total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and triglyceride (TG) were also compared. Korean wild leafy vegetables were assayed for $\beta$-carotene, vitamin C and vitamin E and total phenolic compound including flavonoid and thiobarbituric acid (TBA) value using the linoleic acid model system. SOD activity in rats fed Aster scaber was significantly higher(as much as 289%) than those fed Spinacia oleracea. Compared to control animals, the Aster scaber fed animals had significantly lower TC and lower atherogenic index. Compared to Spinacia oleracea, Aster scaber and Ligularia fischeri had vitamin C concentration of 150% and 400%, flavonoid concentrations of 470% and 310%, and phenolic compound concentrations of 326% and 203%, respectively, but tended to have lower $\beta$-carotene and significantly lower vitamin E concentrations. TBA values were only 18% of the control value in Aster scarier and Ligularia fischeri and 41% of the control value in Spinacia oleracea. These results suggest that Aster scaber could have potent antioxidative activity in vivo as well as in vitro and potential vague as a functional food to improve the plasma lipid profiles. Furthermore, phenolic and flavonoid compounds may be a major contributing factor to the antioxidative potential of Korean wild leafy vegetables.

Novel Bi2S3/TiO2 Heterogeneous Catalyst: Photocatalytic Mechanism for Decolorization of Texbrite Dye and Evaluation of Oxygen Species

  • A heterogeneous $Bi_2S_3/TiO_2$ composite catalyst was synthesized via a green ultrasonic-assisted method and characterized by XRD, SEM, EDX, TEM analysis. The results clearly show that the $TiO_2$ particles were homogenously coated with $Bi_2S_3$ particles, indicating that $Bi_2S_3$ particle agglomeration was effectively inhibited after the introduction of anatase $TiO_2$. The Texbrite BA-L (TBA) degradation rate constant for $Bi_2S_3/TiO_2$ composites reached $8.27{\times}10^{-3}min^{-1}$ under visible light, much higher than the corresponding value of $1.04{\times}10^{-3}min^{-1}$ for $TiO_2$. The quantities of generated hydroxyl radicals can be analyzed by DPCI degradation, which shows that under visible light irradiation, more electron-hole pairs can be generated. Finally, the possible mechanism for the generation of reactive oxygen species under visible-light irradiation was proposed as well. Our result shows the significant potential of $Bi_2S_3$-semiconductor-based $TiO_2$ hybrid materials as catalysts under visible light for the degradation of industry dye effluent substances.
