Lipid Peroxidation and the Thiobarbituric Acid Assay: Standardization of the Assay When Using Saturated and Unsaturated Fatty Acids

  • Saturated fatty acids are less vulnerable to lipid peroxidation than their unsaturated counterparts. In this investigation, individual fatty acids of the $C_{16}$, $C_{18}$ and $C_{20}$ families were subjected to the thiobarbituric (TBA) assay. These fatty acids were chosen based on their degree of saturation and configuration of double bonds. Interestingly, an assay threshold was reached where increasing the fatty acid concentration resulted in no additional decrease in the TBARS concentrations. Therefore, the linear range of TBARS inhibition was determined for fatty acids in the $C_{16}$ and $C_{20}$ families. The rate of TBARS inhibition was greater for the saturated than for unsaturated fatty acids, as measured from the slope of the linear range. These findings demonstrate the need to standardize the TBARS assay using multiple fatty acid concentrations when using this assay for measuring in vitro lipid peroxidation.

Determination of MTBE, TBA and BTEX in Soil by Headspace Gas Chromatography-Mass Spectrometry

  • A headspace gas chromatographic mass spectrometric (GC-MS) assay method was developed for the simultaneous determination of methyl tertiary butyl ether (MTBE), $tert$-butyl alcohol (TBA) and benzene, toluene, ethyl benzene and xylene (BTEX) in soil contaminated with gasoline. 2 g of soil sample were placed in a 10 mL headspace vial filled with 5 mL of phosphoric acid solution (pH 3) saturated with NaCl, and the solution was spiked with fluorobenzene as an internal standard and sealed with a cap. The vial was heated in a heating block for 40 min at $80^{\circ}C$. The detection limits of the assay were 0.08-0.12 ${\mu}g$/kg for the analytes. For five independent determinations at 10 and 50 ${\mu}g$/kg, the relative standard deviations were less than 10%. The method was used to analyze fifty six soil samples collected from various regions contaminated with gasoline in Korea. The developed method may be valuable for the monitoring of the analytes in soil.

  • 혐기적 조건밑에서 $FeCl_{3}$에 의해 촉진된 TBA 반응을 이용하여 혈청중 과산화물 측정을 위한 새로운미량 분석법을 설정하였다. 즉. 혈청단백침전물이 현탁된 TBA반응흔합액에 혈청 $10{\mu}l$$1.0{\mu}mole$$FeCl_{3}$를 첨가한 다음 질소기류밑에서 50분간 끊는 물에 중탕한다. 이 분석법의 반응 예민도는 현행 일반 TBA법보다 40여배나 증대되었으며 따라서 미량의 시료$(10{\sim}20{\mu}l)$로도 TBA반응생성물의 농도 측정이 비색법으로도 가능하였다. 그리고 TBA반응생성물의 butanol 추출액을 $(NH_{4})_{2}SO_{4}$염석으로 부분말수시킴으로써 예민성 및 재현성이 개선될 수 있었다. 이 분석법을 이용하여 측정한 건강인들의 혈청 TBA같은 높은 연령군일수록 증가하는 경향을 보였다.

초고압처리에 의한 연해주 대두분말의 저장기한 중의 산패도 변화 (Effect of High Pressure Processing on the Rancidity of Yeonhaeju Soybean (Bazaz) Powder during Storage)

  • 대두에서 산패취를 발생시키는 lipoxygenase 효소를 비가열 전처리 방법인 초고압 처리 방법을 이용하여 효소의 활성도의 변화를 확인하였다. 연구 결과 초고압 처리에 의해 lipoxygenase 효소의 활성도가 감소하는 것을 확인하였다. 또한 lipoxygenase 효소의 활성도 감소에 의해 대두의 저장성에 미치는 영향을 확인하기 위해 국산 대두와 연해주대두를 이용하여 저장 실험을 실시하였다. 실험 결과 국산대두의 경우 초고압 처리에 의해 lipoxygenase 효소의 활성도가 감소하였으나 TBA가에 영향을 주는 자동산화등의 화학반응에 의해 TBA가가 지속적으로 증가함을 보여주었다. 그러나 연해주 대두의 경우에는 초고압 처리를 한 시료의 TBA가의 증가정도가 상대적으로 낮게 측정된 것을 확인하였다. 이는 연해주 대두가 항산화 성분을 국산콩에 비하여 많이 포함하고 있어 초고압 처리에 의한 lipoxygenase 효소의 불활성과 함께 항산화 성분의 영향으로 TBA가의 저장 중 증가 정도가 낮게 측정된 것으로 판단되어진다.

Evaluation of Enzymatic Method using an Automated Chemistry Analyzers for Homocysteine Measurement

  • 호모시스테인은 관상동맥질환, 뇌혈관 질환의 위험인자로 알려지고 있다. 초기의 호모시스테인 측정은 HPLC를 이용하여 일반적인 검사실에서 이용하기에는 어려움이 많았으며 현재 여러 가지 면역분석기가 도입되어 시행되고 있다. 하지만 최근 자동화학분석기를 이용한 새로운 효소법의 시약이 개발되어 이 시약의 유용성 평가를 하기위해 본 연구에서는 기존의 면역분석기와 비교분석하였다. 그 결과 정밀도 평가에서 변이계수는 0.98 - 3.23%, 2.55~4.58%를 나타내었고, 자동화학분석기(TBA 200FR와 Advia 1650)를 이용한 새로운 효소법(HBI)과 기존의 면역분석기(Advia Centaur와 Immulite 2000)간의 상관관계 평가에서 상관계수는 0.9632와 0.9625로 우수한 상관관계를 보였다. 이 결과를 토대로 호모시스테인 측정에 있어 새로운 효소법의 자동화학분석기를 이용한 방법이 제시되어 보다 간편하고 쉬운 routine analysis에 적합하리라 사료된다.

Measurement of Antioxidant Activity of Anserine, Taurine, and L-Histidine in vitro and Content of Anserine, Taurine, and L-Histidine in Mature and Juvenile Rainbow Trout (Onchorhynchus mykiss) Muscle

  • The content of anserine, taurine, and L-histidine was measured by HPNC in the muscle of mature(670~690g) and juvenile(80~120g) rainbow trout fatmed in Chungsun, Korea. The concentration of anserine and taurine was higher in mature rainbow trout than in juvenile, but that of L-histidine was lower in mature than in juvenile. When measured with the chemiluminescence(CL) assay, anserine and taurine showed very powerful antioxidative activity above physiological concentration rainbow trout. Taurine still showed antioxidative activity below physiological concentration, while anserine showed prooxidative activity below that. L-Histidine was prooxidative dose-dependently. In TBA method, while taurine showed very week antioxidative effect, anserine appeared very powerful antioxidant and L-histidine prooxidant at physiological concentration. There was no synergism between anserine and taurine and anserine inhibited prooxidative effect of L-histidine.

Boiled-Water Extract from Hizikia fusiformis Showing Antioxidant Effects

  • Boiled-water extract (BWE) of Hizikia fusiformis has antioxidant effects. To investigate antioxidant effects of BWE, 1,1-dephenyl-2-picrylhydrazyl (DPPH) and thiobarbituric acid (TBA) methods were used. BWE exhibited strong radical-scavenging activity (87.3% at final conc. $2000\;{\mu}g$ BWE/ml in an assay mixture) on DPPH and good inhibitory activity (64% at final conc. $1000\;{\mu}g$ BWE/ml in an assay mixture) on lipid peroxidation. Raw 264.7 cells treated with lipopolysaccharide (LPS) showed 26.7-fold increase in nitric oxide (NO) ($48.9\;{\mu}M$) compared with control group ($1.83\;{\mu}M$). When treated with BWE and LPS, NO production was inhibited by BWE dose-dependently.

In Vivo Anti-Oxidant Activities of Tectochrysin

  • The anti-oxidant activities of tectochrysin, a major compound of propolis, were investigated. Tectochrysin exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4$-intoxication in rats. Tectochrysin tested exhibited a lipid peroxidation causing a significant decrease in MDA production in TBA-reactant assay. Tectochrysin was strong in the increase in the anti-oxidant enzymes such as hepatic cytosolic superoxide dismutase, catalase and glutathione peroxidase activities in $CCl_4$-intoxicated rats. These results suggest that tectochrysin possess not only the anti-oxidant, but also the activities in $CCl_4$-intoxicated rats. Especially, tectochrysin was found to cause significant increases in the rat liver cytosolic SOD, catalase, GSH-px activities as well as a significant decrease in the MDA production.

Analysis of tert-Butanol, Methyl tert-Butyl Ether, Benzene, Toluene, Ethylbenzene and Xylene in Ground Water by Headspace Gas Chromatography-Mass Spectrometry

  • Methyl tert-butyl ether (MTBE) is added to gasoline to enhance the octane number of gasoline, tert-butyl alcohol (TBA) is major degradation intermediate of MTBE in environment, and benzene, toluene, ethyl benzene and xylene (BTEX) are also major constituents of gasoline. In this study, a simplified headspace analysis method was adapted for simultaneous determination of MTBE, TBA and BTEX in ground water samples. The sample 5.0 mL and 2 g NaCl were placed in a 10 mL vial and the solution was spiked with fluorobenzene as an internal standard and sealed with a cap. The vial was placed in a heating block at 85 $^{\circ}C$ for 30 min. The detection limits of the assay were 0.01 ${\mu}$g/L for MTBE and BTEX, and 0.02 ${\mu}$g/L for TBA. The method was used to analyze 110 ground water samples from various regions in Korea, and to survey the their background concentration in ground water in Korea. The samples revealed MTBE concentrations in the range of 0.01 - 0.45 ${\mu}$g/L (detection frequency of 57.3%), TBA concentrations in the range of 0.02 - 0.08 ${\mu}$g/L (detection frequency of 5.5%), and total BTEX concentrations in the range of 0.01 - 2.09 ${\mu}$g/L (detection frequency of 87.3%). The developed method may be used when simultaneously determining the amount of MTBE, TBA and BTEX in water.

고추 기관별 항산화 활성과 quinone reductase 유도활성 (Antioxidant and Quinone Reductase Inductive Activities of Various Organs of Pepper)

  • 고추 부산물의 활용을 위해 고추의 각 기관별 추출물에 대한 항산화와 QR 유도 활성을 측정하였다. 고추의 과실, 뿌리, 줄기, 잎을 각각 메탄올로 추출하여 분석한 결과, 고춧잎이 다른 기관에 비해 플라보노이드와 페놀성분이 높은 것으로 나타났다. DPPH 항산화 실험에서는 기관별로 큰 차이가 없었던 반면, ABTS 항산화 실험에서는 고춧잎이 다른 기관에 비해 유의적으로 높은 항산화능을 보였다. FTC와 TBA 실험에서 줄기와 고춧잎이 다른 기관에 비해 유의하게 높은 지질과산화 억제능을 나타냈다. QR 유도활성에서는 고춧잎, 뿌리, 줄기, 과실순으로 높았다. 고춧잎의 경우 $50-200\;{\mu}g/mL$ 농도에서는 다른 기관에 비해 높은 세포 성장 억제능을 보였다. 결론적으로 고춧잎은 다른 기관에 비해 항산화 활성과 QR 유도 활성이 높아 기능성소재로서의 활용가치가 높다.
