Characterization of Pigment-Producing Pseudoalteromonas spp. from Marine Habitats and Their Optimal Conditions for Pigment Production (해양환경에서 분리한 Pseudoalteromonas 속 균주들의 특징 및 색소 생성 최적 조건)

  • Three marine bacteria producing pigments were isolated from seawater of Jeju-Do and local solar saltern in Korea. Based on phenotypic characteristics and 16S rRNA sequence analysis, the strains were identified as Pseudoalteromonas spp., which produced red (Ju11-1), yellow (Ju14), and orange (TA20) pigments. The pigments showed UV absorption maxima at 537, 378 and 387 nm, respectively. The strains were growing well on Marine broth 2216 culture medium. The productivity of pigments reached the maximum value after 28 hours (Ju11-1, Ju14) and 24 hours (TA20) at $30^{\circ}C$, 2% NaCl and pH 6-7. The best pigment production of strains were supported by 1% of lactose (Ju11-1) and maltose (Ju14, TA20) as a carbon source and 1% of beef extract as a nitrogen source.

PAALD 방법을 이용한 TaN 박막의 구리확산방지막 특성

  • In this study, as Cu diffusion barrier, tantalum nitrides were successfully deposited on Si(100) substrate and SiO2 by plasma assisted atomic layer deposition(PAALD) and thermal ALD, using pentakis (ethylmethlyamino) tantalum (PEMAT) and $NH_3$ as precursors. The TaN films were deposited on $250^{\circ}$C by both method. The growth rates of TaN films were $0.8{\AA}$/cycle for PAALD and $0.75{\AA}$/cycle for thermal ALD. TaN films by PAALD showed good surface morphology and excellent step coverage for the trench with an aspect ratio of h/w - $1.8 : 0.12 \mu\textrm{m}$ but TaN films by thermal ALD showed bad step coverage for the same trench. The density for PAALD TaN was $11g/\textrm{cm}^3$ and one for thermal ALD TaN was $8.3g/\textrm{cm}^3$. TaN films had 3 atomic % carbon impurity and 4 atomic % oxygen impurity for PAALD and 12 atomic % carbon impurity and 9 atomic % oxygen impurity for thermal ALD. The barrier failure for Cu(200nm)/TaN(l0nm)/$SiO_2(85nm)$/Si structure was shown at temperature above $700^{\circ}$C by XRD, Cu etch pit analysis.

Sliced Channel Allocation Method for Multirate Communtiaction Environments using DAMA Scheduler (Multirate 통신환경에서 DAMA 스케줄러를 이용한 슬라이스 채널 할당방법)

  • In this paper the new channel allocation method which is important in designing mobile radio system, is proposed. If the channel is allocated by time-frequency-code sliced and using DAMA scheduler, the blocking probability is lower than TA channel allocation method. With traffic load $\rho$=0.8 and 2% of blocking probability, TFS method can allocate 36 slices compared to 11 slices in TA method. At traffic load $\rho$=0.9 and 1% of blocking probability, the maximum available slices are 13 in TA method, but in TFS method 40 slices are allocated to user, so TFS method is better frequency availability by 3.75 times than TA method. TFS method could be suitable for system planning to multirate communication users.

  • The purpose of CBSD(Component Based Software Development) if to develop software using a reuse component and components assembly. The reused components have to be adapter for satisfaction of requirement specification. This paper represents a component adaption method which reuse previously developed component or legacy system, using an adaptation component called by TA(Transaction Adaptor). The adaptation using TA transmits XML data that is translated into stream type, from a client to on host system. And we introduce RS(Rule Server) which gets some Information from XML data for application program development. Through the Interaction of TA and RS, we can improve effectiveness of component development and maintenance and we can reuse legacy system.

Synergistic anticancer effects of timosaponin AIII and ginsenosides in MG63 human osteosarcoma cells

  • Background: Timosaponin AIII (TA3) is a steroidal saponin extracted from Anemarrhena asphodeloides. Here, we investigated the anticancer effects of TA3 in MG63 human osteosarcoma cells. TA3 attenuates migration and invasion of MG63 cells via regulations of two matrix metalloproteinases (MMPs), MMP-2 and MMP-9, which are involved with cancer metastasis in various cancer cells. TA3 reduced enzymatic activities and transcriptional expressions of MMP-2 and MMP-9 in MG63 cells. TA3 also inhibited Src, focal adhesion kinase, extracellular signal-regulated kinase (ERK1/2), c-Jun N-terminal kinase (JNK), p38, ${\beta}-catenin$, and cAMP response element binding signaling, which regulate migration and invasion of cells. TA3 induced apoptosis of MG63 cells via regulations of caspase-3, caspase-7, and poly(ADP-ribose) polymerase (PARP). Then, we tested several ginsenosides to be used in combination with TA3 for the synergistic anticancer effects. We found that ginsenosides Rb1 and Rc have synergistic effects on TA3-induced apoptosis in MG63 cells. Methods: We investigated the anticancer effects of TA3 and synergistic effects of various ginseng saponins on TA3-induced apoptosis in MG63 cells. To test antimetastatic effects, we performed wound healing migration assay, Boyden chamber invasion assays, gelatin zymography assay, and Western blot analysis. Annexin V/PI staining apoptosis assay was performed to determine the apoptotic effect of TA3 and ginsenosides. Results: TA3 attenuated migration and invasion of MG63 cells and induced apoptosis of MG63 cells. Ginsenosides Rb1 and Rc showed the synergistic effects on TA3-induced apoptosis in MG63 cells. Conclusions: The results strongly suggest that the combination of TA3 and the two ginsenosides Rb1 and Rc may be a strong candidate for the effective antiosteosarcoma agent.

Simultaneous enhancement of thermostability and catalytic activity of phospholipase $A_1$ by evolutionary molecular engineering

  • The thermal stability and catalytic activity of phospholipase A$_1$ from Serratia sp. MK1 were improved by an evolutionary molecular engineering. Two thermostable mutants were isolated after sequential rounds of error-prone PCR to introduce random mutations and filter-based screening of the resultant mutant library, and identified as having six (mutant TA3) and seven (mutant TA13) amino acid substitutions, respectively. Different types of the substitutions were found in two mutants, resulting in the increase of nonploar residues (mutant TA3) or changes between side chains within polar or charged residues (mutant TA13). The wild-type and mutant enzymes were purified, and the effect of temperature on their stability and catalytic activity was investigated. The T$\sub$m/ values of TA3 and TA13 were increased by 7 and 11$^{\circ}C$, respectively. Thus, evolutionary molecular engineering was found to be an effective and efficient approach to increasing thermostability without compromising enzyme activity.

  • Tantalum nitride(TaN) films were deposited by atomic layer deposition(ALD) and plasma assisted atomic layer deposition(PAALD). The deposition of the TaN thin film has been performed using pentakis (ethylmethlyamino) tantalum (PEMAT) and ammonia($NH_3$) as precursors at temperature of $250^{\circ}C$, where the temperature was proven to be ALD window for TaN deposition from our previous experiments. The PAALD deposited TaN film shows better physical properties than thermal ALD deposited TaN film, due to its higher density$(~11.59 g/\textrm{cm}^3$) and lower carbon(~ 3 atomic %) and oxygen(~ 4 atomic %) concentration of impurities.

Korean Medicine approach to traffic accident victims (교통사고 후유증관리에 관한 한의학적 임상 고찰)

  • Objective : Korean medicine treatment method is noted as alternative in treating traffic accidents(TA) victims recently. The main purpose of this research is to make a survey of the effective way of the Korean medicine treatment about TA victims. Methods : In following research, 389 cases of traffic victims who hospitalized in the Youngsaeng Korean Medicine Hospital & Youngsaeng Clinic from March 1, 2001 to November 30, 2001 were surveyed. The research is focused on finding out the distribution, such as sex and age, damaged part of the body of 389 TA victims, nature of damage of 389 TA victims, chief complaint of 389 TA victims, nature of damage by Korean medicine code classification, treatment given to 389 TA victims, Extract(Ex) medicine given to 389 TA victims, days in hospital of 389 TA victims. Results : The patients reached korean medicine hospital in fewer numbers from most of TA victims. As neck pain and L-spin sprain, most of symptoms was light and the treatment of medicine as well was limited for TA. The treatment method which was used for treating TA was acupuncture, cupping therapy, physical therapy, Ex medicine. Few ways could use the Ex medicine as well. Conclusions : The achieving rate of treatment was marked highly by using Haenggi(行氣) Hwalhyeol(活血) Guyea(祛瘀) and we confirmed a possibility for the treatment of TA in korean medicine treatment method. It needs to increase all kinds of treatment-ways which can treat successfully and needs to come into application of insurance. Korean medicine method will have possibility to take part in TA injury admission, by presenting basis to prove effectiveness of treatment-ways.

Transport properties of polycrystalline TaNx thin films prepared by DC reactive magnetron sputtering method

  • We have investigated the electrical transport properties of polycrystalline tantalum nitride (TaNx) films. Various compositions of tantalum (nitride) thin films have been deposited on SiO2 substrates by reactive DC magnetron sputtering while changing the ratio of nitrogen partial pressure. The substrate temperature was maintained at 283 K during deposition. X-ray diffraction analyses indicated the presence of α-Ta and β-Ta phases in the Ta film deposited in pure argon atmosphere, while fcc-TaNx phases appeared in the sputtering gas mixture of argon and nitrogen. The N/Ta atomic ratio in the film increased ranging from 0.36 to 1.07 for nitrogen partial pressure from 7 to 20.7%. The superconducting transition temperatures of the TaNx thin films were measured to be greater than 3.86 K with a maximum of 5.34 K. The electrical resistivity of TaNx thin film was in the range of 177-577 𝜇Ωcm and increased with an increase in nitrogen content. The upper critical filed at zero temperature for a TaN0.87 thin film was estimated to exceed 11.3 T, while it showed the lowest Tc = 3.86 K among the measured superconducting TaNx thin films. We try to explain the behavior of the increase of the residual resistivity and the upper critical field for TaNx thin films with the nitrogen content by using the combined role of the intergrain Coulomb effect and disorder effect by grain boundaries.

Mutagenic Test of Gardenia Yellow Pigment (치자 황색색소에 대한 변이원성 시험)

  • Gardenia yellow pigment produced by Gardenia jasminoides Ellis was tested for reverse mutagenic test in Salmonella typhimurium stains TA1535, TA1537, TA98 and TA100 at concentrations raging form 6.25 to 200$\mu\textrm{g}$/$m\ell$ per plate. No significant reverse mutagenic activity was observed in any of the S. typhimurium strains, in either presence or absence of S9 mix. There was no toxicity to the bacteria. These result indicate that yellow pigment doesn't have mutagenicity.
