Neural Transdifferentiation: MAPTau Gene Expression in Breast Cancer Cells

  • Background: In tumor cells, aberrant differentiation programs have been described. Several neuronal proteins have been found associated with morphological neuronal-glial changes in breast cancer (BCa). These neuronal proteins have been related to mechanisms that are involved in carcinogenesis; however, this regulation is not well understood. Microtubule-associated protein-tau (MAP-Tau) has been describing in BCa but not its variants. This finding could partly explain the neuronal-glial morphology of BCa cells. Our aim was to determine mRNA expression of MAP-tau variants 2, 4 and 6 in breast cancer cell lines. Materials and Methods: Cultured cell lines MCF-10A, MDA-MB-231, SKBR3 and T47D were observed under phase-contrast microscopy for neural morphology and analyzed for gene expression of MAP-Tau transcript variants 2, 4 and 6 by real-time PCR. Results: Regarding morphology like neural/glial cells, T47D line shown more cells with these features than MDA-MB-231 and SKBR. In another hand, we found much greater mRNA expression of MAP-Tau transcript variants 2, and to a lesser extent 4 and 6, in T47D cells than the other lines. In conclusion, regulation of MAP-Tau could bring about changes in cytoskeleton, cell morphology and motility; these findings cast further light on neuronal transdifferentiation in BCa.

  • Ras oncoproteins are small molecular weight GTPases known for their involvement in oncogenesis, which operate in a complex signaling network with multiple effectors. Approximately 25% of human tumors possess mutations in a member of this family. The Raf1/MEK/Erk1/2 pathway is one of the most intensively studied signaling mechanisms. Different levels of regulation account for the inactivation of MAP kinases by MAPK phosphatases in a cell type- and stimuli-dependent manner. In the present study, using three inducible Ras-expressing NIH/3T3 cell lines, we demonstrated that MKP3 upregulation requires the activation of the Erk1/2 pathway, which correlates with the shutdown of this pathway. We also demonstrated, by applying pharmacological inhibitors and effector mutants of Ras, that induction of MKP3 at the protein level is positively regulated by the oncogenic Ras/Raf/MEK/Erk1/2 signaling pathway.

  • We determine the largest integer i such that $0 and the coefficient of $t^{i}$ is odd in the polynomial $(1+t+t^{2}+{\cdots}+t^{n})^{n+1}$. We apply this to prove that the co-index of the tangent bundle over $FP^{n}$ is stable if $2^{r}{\leq}n<2^{r}+\frac{1}{3}(2^{r}-2)$ for some integer r.

  • Let ${\mathcal{H}}$ be a complex Hilbert space and ${\mathcal{B}}({\mathcal{H}})$ the algebra of all bounded linear operators on ${\mathcal{H}}$. In this paper, we prove that if ${\varphi}:{\mathcal{B}}({\mathcal{H}}){\rightarrow}{\mathcal{B}}({\mathcal{H}})$ is a unital surjective bounded linear map, which preserves m- isometries m = 1, 2 in both directions, then there are unitary operators $U,V{\in}{\mathcal{B}}({\mathcal{H}})$ such that ${\varphi}(T)=UTV$ or ${\varphi}(T)=UT^{tr}V$ for all $T{\in}{\mathcal{B}}({\mathcal{H}})$, where $T^{tr}$ is the transpose of T with respect to an arbitrary but fixed orthonormal basis of ${\mathcal{H}}$.

  • Recently, many cleaning robots have been made with various algorithms for efficient cleaning. One of them is a DmaxCoverage algorithm which efficiently clean for the situation when the robot has a time limit. This algorithm uses Rectangle Tiling method for finding the biggest rectangle that doesn't have any obstacle. When the robot uses grid map, Rectangle Tiling method can find the optimal value. Rectangle Tiling method is to find all of the rectangles in the grid map. But when the grid map is big, it has a problem that spends a lot of times because of the large numbers of rectangles. In this paper, we propose Four Direction Rectangle Scanning(FDRS) method that has similar accuracy but faster than Rectangle Tiling method. FDRS method is not to find all of the rectangle, but to search the obstacle's all directions. We will show the FDRS method's performance by comparing of FDRS and Rectangle Tiling methods.

The Anti-Microbial Activity of Modified Chitosan. (변형 키토산의 항균효과)

  • New type of chitosan derivatives, chitosan-g-MAP, were synthesized by graft copolymerization of mono (2-methacryloyl oxyethyl) acid phosphate (MAP) into chitosan, in order to solubilize chitosan in water. Ceric ammonium nitrate was used as an initiator for graft copolymerization. The optimal conditions for graft copolymerization were determined on the basis of reaction temperature, time, and the concentration of initiator and monomer. The reaction conditions for the highest percentage of grafting were as follows: an initiator concentration, 3.5${\times}$10$\^$-3/ M; monomer concentration, 0.19 M; and reaction temperature, 40$^{\circ}C$ The reaction rate reached the maximum value after 4 hrs of reaction. Antifungal activity was tested against Candida albicans, Trichophyton rubrum and Trichophyton violaceum by using chitosan-g-MAP and two other chitosan samples which have degree of deacetylation of 70% (DA-7) and 90% (DA-90). Their antifungal activities were investigated in weak acidic range. Maximum antifungal activity of them was observed at pH 5.75. Chitosan-g-MAP inhibited thoroughly the growth of Candida albicans and Trichophyton violaceum. Howerver, DA-70 and DA-90 showed higher antifungal activities on Trichophyton rubrum than that of chitosan-g-MAP.

Separation of Inorganic Sludge and MAP from Municipal Wastewater Sludge Using Hydrocyclone (습식 사이클론을 이용한 하수슬러지내 무기성분 및 MAP 분리)

  • The performance of inorganic sludge separation system is evaluated. Anaerobic digester effluent sludge is used for feed sludge of this system and hydrocyclone is used for inorganic sludge separation. For phosphorus removal and recovery $MgCl_2$ is pumped into MAP growth tank, a component of inorganic sludge separation system. Using this system inorganic sludge which contained less than 40 % of organic matter can be discharged stably and the maximum amount of separated inorganic sludge is 13.4 % of influent sludge based on dry solid. The amount of phosphorus recovered as MAP(as P) is 16.7 % to influent T-P.

  • In this study, we show that Mycobacterium avium subsp. paratuberculosis MAP1305 induces the maturation of bone marrow-derived dendritic cells (BMDCs), a representative antigen presenting cell (APC). MAP1305 protein induces DC maturation and the production of pro-inflammatory cytokines (Interleukin (IL)-6), tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$) through Toll like receptor-4 (TLR-4) signaling by directly binding with TLR4. MAP1305 activates the phosphorylation of MAPKs, such as ERK, p38MAPK, and JNK, which is essential for DC maturation. Furthermore, MAP1305-treated DCs transform naive T cells to polarized $CD4^+$ and $CD8^+$ T cells, thus indicating a key role for this protein in the Th1 polarization of the resulting immune response. Taken together, M. avium subsp. paratuberculosis MAP1305 is important for the regulation of innate immune response through DC-mediated proliferation of $CD4^+$ and $CD8^+$ T cells.

  • We prove the global (in time) vorticity existence for the 2-D Euler equations of a perfect incompressible fluid in $B^0_{{\infty},1}({\mathbb{R}}^2){\cap}L^p({\mathbb{R}}^2)$ with 1 < p < 2. Moreover, we prove that the particle trajectory map X(x, t) satisfies the following estimate: for some positive constant C $${\parallel}X^{\pm1}(\cdot,\;t)-id(\cdot){\parallel}_{B^1_{\infty,1}}{\leq}Ce^{e^{Ct}}$$, where id represents the identity map on ${\mathbb{R}}^2$.
