Ethanol Extracts of Rheum undulatum and Inula japonica Protect Against Oxidative Damages on Human Keratinocyte HaCaT cells through the Induction of ARE/NRF2-dependent Phase II Cytoprotective Enzymes (종대황과 선복화 에탄올 추출물의 인간 피부 세포주인 HaCaT 세포에서 NRF2/ARE에 의존적인 유전자 발현의 유도를 통한 항산화 효과)

  • Mammalian cells control cellular homeostasis using a variety of defensive enzymes in order to combat against environmental oxidants and electrophiles. NF-E2-related factor-2 (NRF2) is a transcription factor that, in response to an exposure to oxidative stress, translocates into the nucleus and modulates the inducible expression of various phase II cytoprotective enzymes by binding to the antioxidant response element (ARE). In the present study, we have acquired 400 ethanol extracts of traditional medicinal plants and attempted to find out possible extract(s) that can increase the NRF2/ARE-dependent gene expression in human keratinocytes. As a result, we have identified that ethanol extracts of Rheum undulatum and Inula japonica strongly activated the ARE-dependent luciferase activity in HaCaT- ARE-luciferase cells. Exposure of ethanol extracts of Rheum undulatum and Inula japonica increased the viability and activated transcription and translation of NRF2-dependent phase II cytoprotective enzymes in HaCaT cells, such as heme oxygenase-1 (HO-1) and NAD[P]H:quinone oxidorecutase-1 (NQO1). In addition, ethanol extracts of Rheum undulatum and Inula japonica suppressed 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced generation of intracellular reactive oxygen species (ROS), thereby inhibiting the formation of 8-hydroxyguanosine (8-OHG) and 4-hydroxynonenal (4-HNE) in HaCaT cells. Together, our results demonstrate that ethanol extracts of Rheum undulatum and Inula japonica exert anti-oxidant effects via the induction of NRF2/ARE-dependent gene expression in human keratinocytes.

  • Skin mucous cells in the three regions of the body in Pseudobagrus fulvidaraco and Leiocassius nitidus were investigated using three histochemical methods (PAS, AB-PAS and HID). In the two species, components of mucous cell were not distinguished in each region, but presented a differences in position and type, size, and count in all regions. Mucous cells of P. fulvidraco were located in two layers, a superficial epithelial cell and the space within alarm substance cells (ASCs). Mucous cells of L. nitidus were situated only in the superficial epithelial cell layer. The size of mucous cells in the ASC layer were larger than those of the superficial epithelial cells for all measured values: diameter, length, width, and area. In the superficial epithelial cell layer, the average number of mucous cells within the three regions did not differ significantly between species by unpaired T-test, but the average number of those within two cell layers of P. fulvidraco had a similarity at each region by paired T-test. The dismilarity in dorsal and ventral regions in average number of ASCs between P. fulvidraco and L. niditus was confirmed by unpaired T-test.

  • The mitotic cycle duration and component plase periods of rice (Oryza sativa L. 'Sumjinbyeo and Seokwangbyeo') root merisem cells at 15, 20, and 30$^{\circ}C$ were determined the use of tritiated thymidine, In this work, the time interval between the maxima of sequential mitotic appearances of marked cells was used to estimate the mitotic cycle duration (MCD) of rice, The MCD of rice of the cultivar 'Sumjinbyeo' and 'Seokwangbyeo' at 20. and 30$^{\circ}C$ was 12. and 20hr, respectively. But the MCD of 'Sumjinbyeo' and 'Seokwangbyeo' was 18 and 20hr, at 15$^{\circ}C$. respectively. The MCD decreased with increasing temperature, The duration of component phase of rice cultivar 'Seokwangbyeo' were essentially the same ratio at 20$^{\circ}C$ and 30$^{\circ}C$. but in 'Sumjinbyeo' cultivar the ratio of $G_1$ period was almost doubled while those of $G_2$ and M were decreased by almost two times at 20$^{\circ}C$ and 30$^{\circ}C$. Deoxyribonucleic acid (DNA). Ribonucleic acid (RNA), and protein synthesis were reduced with increasing temperature from 15$^{\circ}C$ to 30$^{\circ}C$ while the MCD was decreased, This result suggest that DNA, RNA and protein synthesis may not affect the MCD from 15$^{\circ}C$ to 30$^{\circ}C$ in rice.

The Effect of Aralia Cordata Thunb and Cimicifuga Heracleifolia on Cartilage Protection by the Regulation of Metabolism in Human Osteoarthric Chondrocytes (퇴행성 관절염에 대한 독활.승마 복합처방의 대사조절을 통한 연골보호 효과)

  • 목적 : 퇴행성 관절염은 염증성 사이토카인인 IL-$1\beta$에 의해 연골관절이 파괴되고 이로 인해 염증성 사이토카인이 더욱 증가하는 질환이다. 퇴행성 관절염을 치료하기 위해서는 연골 파괴를 가속화시키는 catabolic cytokines의 활성을 줄이고, 성장인자인 anabolic factor의 활성을 증가시는 연골 보호 작용이 있어야 한다. 본 연구에서는 독활 승마 처방(OAH19T)이 catabolic/anabolic 대사 조절에 어떤 영향을 미치는지와 그 신호 전달 기전에 대해 연구하였다. 또한 OAH19T를 구성하는 단미재 및 임상에서 사용되는 COX-2 inhibitor인 Celebrex(CEL)와 효능을 비교 실험하였다. 방법 : 배양된 세포에 IL-$1\beta$로 자극한 후 (1) glycosaminoglycan(GAG)의 분해 억제 정도, (2) OAH19T와 CEL에 대하여 MMP-1과 MMP-3의 유전자 발현 및 활성 억제, (3) Aggrecan 및 Aggrecanases의 유전자 발현 및 활성 억제, (4) OAH19T의 growth factor의 조절 능력, (5) MAPK pathway 등을 RT-PCR(reverse transcriptase-polymerase chain reaction), ELISA(Enzyme-linked immunosorbent assay), western blot, viability 측정을 통해 검증했다. 결과 : 사람 관절 세포에서 (1) 독활 승마 각각의 단미재, 임상에서 사용중인 셀레콕시브(CEL), 조인스보다 실험 약물(OAH19T)이 저농도에서 GAG 분해 억제 효과가 우수하였고, 부탄올로 분획한 OAH19B와는 동등한 효과를 보였다. (2) OAH19T는 IL-$1\beta$에 의하여 활성화된 MMP-1과 MMP-3의 발현을 모두 억제하였으나, CEL은 MMP-1의 발현은 억제하였으나 MMP-3의 발현은 억제하지 못하였다. (3) OAH19T는 IL-$1\beta$에 의하여 손상된 Aggrecan을 회복시켰으며 이는 활성화된 Aggrecanase-1과 Aggrecanase-2를 억제시킴으로써 나타난 결과이다. 그러나 CEL의 경우, 손상된 Aggrecan을 회복시키지 못하였다. (4) 배양된 세포는 IL-$1\beta$에 의하여 TGF-$\beta$II및 TGF-$\beta$ receptor II의 발현이 억제되었으나, OAH19T는 TGF-$\beta$II및 TGF-$\beta$ receptor II의 발현을 회복시켜 OAH19T가 anabolic한 조절능력이 있음을 시사한다. 그러나 CEL의 경우 growth factor에 대한 조절 능력이 없었다. (5) 대사 조절 작용에 대한 기전으로서 MAPK pathway에 대해서 연구한 결과 IL-$1\beta$에 의하여 유도된 pERK, pp38 kinase의 활성은 억제하였고, pJNK의 활성은 변하지 않았다. 또한 OAH19T는 연골 세포에 독성이 없었으며 IL-$1\beta$에 의해 유도된 세포 증식만을 억제시켰다. 이 결과로, OAH19T가 OA chondrocyte의 탈분화 및 세포 고사를 억제하여 연골보호 및 회복 효과가 있음을 알 수 있었다. 결론 : OAH19T는 이를 구성하는 단미재 및 CEL보다 연골보호 효과가 월등하였고, 이러한 연골보호 효과는 catabolic cytokines/growth factors의 균형으로 대사조절을 통해 연골세포의 탈분화 및 세포 고사를 억제하여 연골보호 및 회복 효과가 있음을 알 수 있었다.

Effects of Phytol and Small Water Dropwort Extract on the T Subset in the Sarcoma 180-Transplanted Mice (Phytol과 들미나리추출물이 Sarcoma 180마우스의 T Subset에 미치는 효과)

  • Several studies have shown that phytol isolated from perilla leaf and small water dropwort (Oenanthe javanica (BL) D.C., wild type) extract reveal antirumor activities. In the present study we investigated the effect of phytol and the small water dropwort extract on the T subset in the sarcoma 180-transplanted mice in order to elucidate the immunological mechanism of antiturmor activity of these substances. The results obtained from the experiment were as follows : When phytol was injected into the sarcoma-180 transplanted mice (control), the levels of T cell and T subset by inoculation of the tumor cells were enhanced, but that of T cells in case of small water dropwort extract was similar to the control. Asialo GM1$^{+}$ cells were increased when phytol or small water dropwort extract with injected into tumor-transplanted mice. In normal mice the number of asialo GM1$^{+}$ cells increased with phytol injection and decreased with small water dropwort extract injection. L3T4$^{+}$/Lyt-2$^{+}$cell ratios were decreased when phytol was injected into tumor-transplanted mire, but increased in case of small water dropwort extract injection. In normal mice the ratios showed large decreases with phytol or small water dropwort extract injection. These results indicate that phytol or small water dropwort extract ran activate the proliferation of natural killer cells that are effector cells in tumor-bearing mice.

Thaumatin Isolated from Katemfe Fruit of Thaumatococcus daiellii Inhibits 3T3 L1 Adipocytes Differenciation (Thaumatococcus daiellii 열매 유래 토마틴의 3T3-L1 지방전구세포 분화 억제에 의한 항비만 효과)

  • The effects of thaumatin isolated from katemfe fruit of Thaumatococcus daiellii Benth on 3T3-L1 preadipocyte differentiation was investigated in vitro. 3T3-L1 adipocytes were treated with various concentrations of thaumatin ranging in 0-5 ${\mu}M$. Thaumatin reduced fat accumulation in differentiated 3T3-L1 adipocytes in a dose-dependent manner. 3T3-L1 cell proliferation was 97.0 and 88.3% at 1 and 3 ${\mu}M$ after 8 days of thaumatin treatment, respectively. Thaumatin showed a potent inhibitory effect on stained lipid droplets at a concentration of 3 ${\mu}M$. Thaumatin reduced triglyceride accumulation in differentiated 3T3-L1 cells in a dose-dependent manner, compared with positive control cells. This study provides basic information on the anti-obesity activity of thaumatin.

  • This study examines the effects of alginic acid, a source of dietary fiber, in a glucose-derived media. In particular, we examined how the presence or absence of alginic acid affected the differentiation and triglyceride densities of 3T3-L1 cells. We established that the addition of insulin-like growth factor-I (IGE-I) to 3T3-L1 cells results in acceleration of differentiation. We sought to determine the role of alginic acid in the production of fat by adding alginic acid to 3T3-L1 cells and examining its ability to limit or potentiate this stimulatory effects of IGE-I and IGF binding proteins. We have determined that alginic acid restricts 3T3-L1 cell differentiation and the creation of triglycerides, effectively attenuating 3T3-L1 cell metablolism and growth.

  • Lymph node tissues of BALB/C mouse treated with DNCB were immunohistochemically observed to investigate the activation of cell mediated immunity in lymph node of murine with allergic contact dermatitis. The inguinal region of BALB/C mice were sensitized by one application of $25{\mu}l$ of 5% 2,4-dinitrochlorobenzene(DNCB) onto an abdominal skin and 2 weeks later, the mice were challenged with $4{\mu}l$ of 2.5% DNCB. The inguinal lymph node were obtained at hour 24, 48, and 72 after 2nd DNCB treatment and embedded with paraffin, and then stained by following ABC method that used monoclonal antibody including L3T4(CD4), Ly2(CD8), IL-2R(CD25). The distribution of helper T lymphocytes, cytotoxic T lymphocytes and IL-2 receptors began to increase at hour 24 after after 2nd DNCB treatment and these increase appeared in paracortical area and medullary sinius. These increase were greatest at hour 48. These results indicated that the IL-2 secretion began to increase by activation of helper T lymphocytes in lymph node of DNCB re-exposure area and subsequently to activate suppress T lymphocytes.

  • 반복되는 흉막염 및 폐렴을 주소로 내원한 30세 남자환자에서 경기관지 폐생검과 흉수내 림프구에 대한 유세포 분석 및 T세포 수용체 유전자 재배열 분석을 실시하였다. 경기관지 폐생검 조직의 연역조직화학 염색상 대부분의 림프구가 T세포 표식자인 UCHLl 에 대해 강하게 염색되었고, B세포 표식자인 L26에 대해서는 거의 염색되지 않았다. 흉수에서 추출한 림프구의 유세포 분석상 CD3양성 CD2양성인 T림프구가 대부분이었고, 이들 림프구에 대해 중합효소연쇄반응을 이용한 T세포 수용체 유전자 재배열 분석을 하였더니 $TCR{\gamma}$ 유전자 재배열과 클론성을 관찰할 수 있었다. T세포 원발성 폐림프종으로 진단하였고, 문헌고찰과 함께 보고하는 바이다.

Immune Cell Activation and Co-X-irradiation Effect of Eleutherococcus senticosus Maxim Root (가시오갈피 뿌리의 면역세포 활성 및 방사선 병용효과)

  • Purpose: This study was performed to investigate the effects of immune cell activation and the antitumor effect for the combination of treatment with X-irradiation and E/eutherococcus senticosus Maxim Root (ESMR) on mouse tumor cells. Materials and Methods: ESMR (250g) was extracted with 80% methanol, concentrated under decompression and lyophilized. To determine whether ESMR is able to activate the immune cells or not, the proliferation of splenocytes in vitro and the number of B cells and T cells in splenic lymphocytes in ESMR-pretreated mice were evaluated. X-irradiation was given to the mouse fibrosarcoma tumor cells (FSa II) by 250 kv X-irradiation machine. The cytotoxicity of ESMR was evaluated from its ability to reduce the clonogenecity of FSa II cells. In X-irradiation alone group, each 2, 4, 6 and 8 Gy was given to FSa II cells. In X-irradiation with ESMR group, 0.2 mg/ml of ESMR was exposed to FSa II cells for 1 hour before X-irradiation. Results: The proliferation of cultured mouse splenocytes and thymocytes were enhanced by the addition of ESMR in vitro. The number of B cells and T cells in mouse splenic lymphocytes was significantly increased in ESMR pretreated mice in vivo. In FSa II cells that received a combination of 0.2 mg/ml of ESMR with X-irradiation exposure, the survival fraction with a dose of 2, 4 and 6 Gy was $0.39{\pm}0.005$, $0.22{\pm}0.005$ and $0.06{\pm}0.007$, respectively. For FSa II cells treated with X-irradiation alone, the survival fraction with a dose of 2, 4 and 6 Gy was $0.76{\pm}0.02$, $0.47{\pm}0.008$ and $0.37{\pm}0.01$. The difference in the survival fraction of the mouse FSa II cells treated with and without ESMR was statistically significant (p<0.05). Conclusion: Treatment with ESMR increased cell viability of mouse splenocytes in vitro and especially the subpopulation of B cells and T cells in splenocytes in ESMR-pretreated mice. However, treatment with ESMR did not increase the level of Th and Tc subpopulations in the thymocytes. Treatment with the combination of ESMR and X-irradiation was more cytotoxic to mouse tumor cells than treatment with X-irradiation alone; this finding was statistically significant.
