Identification of Isoleucine-Accepting tRNA in Maize Mitochondria

  • Maize mitochondrial tRNAs for isoleucine have been isolated using a putative $tRNA^{Ile}$ gene probe which has been previously isolated and characterized. It contains the 5'-CAT anticodon which would normally recognize the AUG methionine codon. The nucleotide sequence of one of these tRNAs has been partially determined, and contains a modified nucleotide at the first position of the anticodon. This type of posttranscriptional modification event could change the specificity of amino acid acceptance of a tRNA, unlike that deduced from the corresponding gene. An aminoacylation experiment also demonstrated that these purified tRNAs have isoleucine acceptance activity but no methionine-accepting activity.

A FRET Assay for Celiac Disease

  • To provide a basis for a homogeneous fluorescence resonance energy transfer (FRET) immunoassay for celiac disease, we carried out a FRET experiment using guinea pig tissue transglutaminase (tTG) and antibodies to tTG (anti-tTG) purified from rat serum. Fluorescein was utilized as the probe, and a nonfluorescent dye, QSY 7 served as the quencher. We labeled anti-tTG and tTG with fluorescein isothiocyanate and QSY 7 succinimidyl ester, respectively. Fluorescein-labeled anti-tTG was the donor, and QSY 7-labeled tTG was the acceptor of the FRET experiment. When we titrated fluorescein-labeled anti-tTG with QSY 7-labeled tTG, we observed a large decrease in the steady-state fluorescence intensity, which was due to strong FRET from fluorescein-labeled anti-tTG to QSY 7-labeled tTG. Using time-resolved fluorescence spectroscopy, we could also observe a decrease in the fluorescence lifetime, which confirms the steady-state data. We expect that these results might be useful in the development of a novel fluorescence immunoassay for an easy screening and follow-up of celiac patients.

Signal Analysis of Eddy Current Test Using T/R Coil Probe for Inspection of Steam Generator Tube in NPP (T/R코일프로브를 이용한 원전 SG세관 검사의 와전류탐상 신호해석)

  • In this paper the signal analysis of eddy current test using transmit-receive(T/R) pancake coil of ECT array probe using electromagnetic finite element method(FEM) is performed. For characteristics analysis, the notch defect is used. The depth of defect is 40[%] of steam generator tube thickness, and inside defect and outside defect are used as simulation examples. The signal analysis is performed according to the variation of receive coil position. The receive coil is positioned $0[^{\circ}]$, $30[^{\circ}]$, $60[^{\circ}]$, $90[^{\circ}]$ of circumferential position of transmit coil. To obtain e electromagnetic characteristics of robes, the governing equation is derived from Maxwell's equations, and the problem is solved using the 3-dimensional finite element method. The signal magnitude of inside defect is bigger than that of outside defect, and the signal difference can be seen according to the variation of position of receive coil. The experimental signal and numerical signal of ASME standard tube show similar results. The results in this paper can be helpful when the ECT signals from ECT array probe are evaluated and analyzed.

Clinical Significance of Dual-probe Esophageal pH Monitoring in Pathological Gastroesophageal Reflux Disease with Recurrent Respiratory Symptoms (재발성 호흡기 증상을 동반한 병적 위식도 역류 질환에서 이중 채널 식도내 pH 검사의 의의)

  • Purpose: The aim of this study was to determine clinical significance of dual-probe esophageal pH monitoring and to compare four pH monitoring parameters between proximal and distal esophagus in pathological gastroesophageal reflux disease with recurrent respiratory symptoms. Methods: Among the thirty-four patients who were performed 24 hr pH monitoring, seventeen patients with pathological distal reflux were classified into two groups: Group I (n:12) had recurrent respiratory symptoms and Group II (n:5) hadn't recurrent respiratory symptoms. The ambulatory dual-probe esophageal pH monitoring was performed for 18~24 hr. A pathologic GER was defined when reflux index (percent of the investigation time a pH<4) exceeded the 95th percentile of normal value. Results: Among the sixteen patients with recurrent respiratory symptoms, twelve patients (75%) have pathological distal reflux. Whereas among the eighteen patients without recurrent respiratory symptom, five patients (28%) have pathological distal reflux. In the Group I, the significant differences between proximal and distal esophageal pH recordings persisted for all parameters, but didn't persist in group II except for longest episode. Comparing esophageal pH four parameters between group I and group II at the proximal esophageal site, all parameters didn't show statistically significant differences. Conclusion: Regardless of respiratory symptoms, patients with pathological distal reflux didn't show statistically significant differences in the all parameters at the proximal esophageal site. Therefore we may reconsider usefulness of dual probe pH meter in patients with recurrent respiratory symptoms.

Development and Characterization of Vertical Type Probe Card for High Density Probing Test (고밀도 프로빙 테스트를 위한 수직형 프로브카드의 제작 및 특성분석)

  • As an increase of chip complexity and level of chip integration, chip input/output (I/O) pad pitches are also drastically reduced. With arrival of high complexity SoC (System on Chip) and SiP (System in Package) products, conventional horizontal type probe card showed its limitation on probing density for wafer level test. To enhance probing density, we proposed new vertical type probe card that has the $70{\mu}m$ probe needle with tungsten wire in $80{\mu}m$ micro-drilled hole in ceramic board. To minimize alignment error, micro-drilling conditions are optimized and epoxy-hardening conditions are also optimized to minimize planarity changes. To apply wafer level test for target devices (T5365 256M SDRAM), designed probe card was characterized by probe needle tension for test, contact resistance measurement, leakage current measurement and the planarity test. Compare to conventional probe card with minimum pitch of $50{\sim}125{\mu}m\;and\;2\;{\Omega}$ of average contact resistance, designed probe card showed only $22{\mu}$ of minimum pitch and $1.5{\Omega}$ of average contact resistance. And also, with the nature of vertical probing style, it showed comparably small contact scratch and it can be applied to bumping type chip test.

Effect of Korean Mistletoe Lectin on Gene Expression Profile in Human T Lymphocytes: A Microarray Study

  • Korean mistletoe has a variety of biological effects, such as immunoadjuvant activities. This study investigates the effects of Korean mistletoe lectin (Viscum album L. var. coloratum agglutinin, VCA) on human T lymphocytes to determine whether VCA acts as an immunomodulator. Purified human T-lymphocytes were cultured with VCA and RNA from each point was analyzed using Affymetrix human genome chips containing 22,500 probe sets which represents more than 18,000 transcripts derived from 14,500 human genes. As a result, there was a striking upregulation of genes coding for chemokines. Seventeen genes out of 50 coding for proteins with chemokine activity were upregulated including CXCL9 and IL-8 which are related to the treatment of cancer. In addition, 28 cytokine genes were upregulated including IL-1, IL-6, IL-8, IFN-$\gamma$, and TNF-$\alpha$. Taken together, the data suggest that Korean mistletoe lectin, in parallel with European mistletoe, has an ability to modulate human T cell function.

Implementation of a blood flow simulator and a blood flowmeter probe using self-mixing effect of the laser diode (혈류 시뮬레이터와 LD의 자기혼합효과를 이용한 혈류계 프로브의 구현)

  • In this paper, the authors have implemented a blood flow simulator and a blood flowmeter probe using self-mixing effect of the laser diode. The purpose of the blood simulator is to simulate microvascular blood flow in tissue. It consists of melinex film (thickness = $125{\mu}m$) which has similar optical characteristics to epidermis and porous polyethylene filter (Vyon, porosity 35%, mean pore size $50{\mu}m$, thickness=1 mm) which has similar optical characteristics to dermis. The blood flowmeter probe consists of laser diode(5 mW, 780 nm wavelength), CD lens(focal lenght 12 mm), current-to-voltage converter, highpass filter, and preamplifier. It doesn't need optical fiber, therefore, implementation of the probe is simpler than conventional probe using optical fiber.

  • In this paper, we developed the process for depth-probe type silicon microelectrode arrays. The process consists of four mask steps only. The steps are for defining sites, windows, and for shaping probe using plasma etch from above, and for shaping using wet etch from below, respectively. The probe thickness is controlled by dry etching, not by impurity diffusion. We used gold electrodes with a triple dielectric system consisting of oxide/nitride/oxide. The shank of the probe taper from 200um to tens of urn tip and has 30 um thickness.
